    A general setting for symmetric distributions and their relationship to general distributions

    A standard method of obtaining non-symmetrical distributions is that of modulating symmetrical distributions by multiplying the densities by a perturbation factor. This has been considered mainly for central symmetry of a Euclidean space in the origin. This paper enlarges the concept of modulation to the general setting of symmetry under the action of a compact topological group on the sample space. The main structural result relates the density of an arbitrary distribution to the density of the corresponding symmetrised distribution. Some general methods for constructing modulating functions are considered. The effect that transformations of the sample space have on symmetry of distributions is investigated. The results are illustrated by general examples, many of them in the setting of directional statistics.PostprintPeer reviewe

    The mechanical transmission of hepatitis B virus by the common bedbug (Cimex lectularius L.) in South Africa

    Tests for both hepatitis B surface antigen (HBsAg) and hepatitis e antigen (HBeAg) were carried out on wild-caught and laboratory-colonized bedbugs (Cimex lectularius L.), the latter after hepatitis B virus (HBV)-positive blood-meals. Positivity for both antigens was interpreted as an indication of HBV infectivity. Of 22 pools in which were tested 211 bugs collected in the northern Transvaal, 18 were HBsAg-positive and 17 HBeAg-positive, with estimated infection rates of 156,7 and 137.7 per 1000 bugs respectively. Passage of HBV in bugs, allowing an extrinsic incubation period of 57-69 days, resulted in 19 out of 25 bugs being positive for HBsAg after the first passage; only a small number of these were positive for HBeAg. After the second passage all bugs tested were HBsAg-negative, showing that the virus had disappeared. Tests on the salivary glands and carcass of each bug at intervals up to 31 days after an infective meal showed a positivity rate of 98% (HBsAg) and 17% (HBeAg) for carcasses and 20% (HBsAg) and 0% (HBeAg) for salivary glands. Attempts to detect HBV particles in the salivary glands by electron microscopy failed. Bugs were shown to continue to excrete HBsAg in their faeces up to the 42nd day, and both HBsAg and HBeAg together up to the 30th day. HBsAg particles were only detected by electron microscopy in faeces hal1lested on the 10th day. The results as a whole· indicate that no biological mUltiplication of virus occurs in C. lectularius but that mechanical transmission from inseCts to man could occur by: (i) contamination of a person when crushing infective bugs; (ii) contamination from infected faeces; and (iii) infection by bite due to regurgitation or interrupted feeding

    The stimulus control of local enclosures and barriers over head direction and place cell spatial firing

    This research was supported by a grant to P.A.D. and E.R.W. from the Biotechnology and Biological Sciences Research Council (BB/P002455/1).Objective: Head direction cell and place cell spatially tuned firing is often anchored to salient visual landmarks on the periphery of a recording environment. What is less well understood is whether structural features of an environment, such as orientation of a maze sub-compartment or a polarising barrier, can likewise control spatial firing. Method: We recorded from 54 head direction cells in the medial entorhinal cortex and subicular region of male Lister Hooded rats while they explored an apparatus with four parallel or four radially-arranged compartments (Experiment 1). In Experiment 2, we recorded from 130 place cells (in Lister- and Long-Evans Hooded rats) and 30 head direction cells with 90° rotations of a cue card and a barrier in a single environment (Experiment 2). Results: We found that head direction cells maintained a similar preferred firing direction across four separate maze compartments even when these faced different directions (Experiment 1). However, in an environment with a single compartment, we observed that both a barrier and a cue card exerted comparable amounts of stimulus control over head direction cells and place cells (Experiment 2). Conclusion: The maintenance of a stable directional orientation across maze compartments suggests that the head direction cell system has the capacity to provide a global directional reference that allows the animal to distinguish otherwise similar maze compartments based on the compartment’s orientation. A barrier is, however, capable of controlling spatially tuned firing in an environment in which it is the sole polarising feature.Publisher PDFPeer reviewe

    The cellular microscopy phenotype ontology

    BACKGROUND: Phenotypic data derived from high content screening is currently annotated using free-text, thus preventing the integration of independent datasets, including those generated in different biological domains, such as cell lines, mouse and human tissues. DESCRIPTION: We present the Cellular Microscopy Phenotype Ontology (CMPO), a species neutral ontology for describing phenotypic observations relating to the whole cell, cellular components, cellular processes and cell populations. CMPO is compatible with related ontology efforts, allowing for future cross-species integration of phenotypic data. CMPO was developed following a curator-driven approach where phenotype data were annotated by expert biologists following the Entity-Quality (EQ) pattern. These EQs were subsequently transformed into new CMPO terms following an established post composition process. CONCLUSION: CMPO is currently being utilized to annotate phenotypes associated with high content screening datasets stored in several image repositories including the Image Data Repository (IDR), MitoSys project database and the Cellular Phenotype Database to facilitate data browsing and discoverability

    Activity-Dependent Bulk Endocytosis and Clathrin-Dependent Endocytosis Replenish Specific Synaptic Vesicle Pools in Central Nerve Terminals

    Multiple synaptic vesicle (SV) retrieval modes exist in central nerve terminals to maintain a continual supply of SVs for neurotransmission. Two such modes are clathrin-mediated endocytosis (CME), which is dominant during mild neuronal activity and activity-dependent bulk endocytosis (ADBE), which is dominant during intense neuronal activity. However little is known about how activation of these SV retrieval modes impact on the replenishment of the total SV recycling pool and the pools that reside within it; the readily releasable pool (RRP) and reserve pool. To address this question, we examined the replenishment of all three SV pools by triggering these SV retrieval modes during both high and low intensity stimulation of primary rat neuronal cultures. SVs generated by CME and ADBE were differentially labelled using the dyes FM1-43 and FM2-10 and their replenishment of specific SV pools was quantified using stimulation protocols that selectively depleted each pool. Our studies indicate that while the RRP was replenished by CME-generated SVs, ADBE provided additional SVs to increase the capacity of the reserve pool. Morphological analysis of the uptake of the fluid phase marker horse radish peroxidase corroborated these findings. The differential replenishment of specific SV pools by independent SV retrieval modes illustrates how previously experienced neuronal activity impacts on the capability of central nerve terminals to respond to future stimuli

    A user-centred evaluation framework for the Sealife semantic web browsers

    Background: Semantically-enriched browsing has enhanced the browsing experience by providing contextualised dynamically generated Web content, and quicker access to searched-for information. However, adoption of Semantic Web technologies is limited and user perception from the non-IT domain sceptical. Furthermore, little attention has been given to evaluating semantic browsers with real users to demonstrate the enhancements and obtain valuable feedback. The Sealife project investigates semantic browsing and its application to the life science domain. Sealife's main objective is to develop the notion of context-based information integration by extending three existing Semantic Web browsers (SWBs) to link the existing Web to the eScience infrastructure. / Methods: This paper describes a user-centred evaluation framework that was developed to evaluate the Sealife SWBs that elicited feedback on users' perceptions on ease of use and information findability. Three sources of data: i) web server logs; ii) user questionnaires; and iii) semi-structured interviews were analysed and comparisons made between each browser and a control system. / Results: It was found that the evaluation framework used successfully elicited users' perceptions of the three distinct SWBs. The results indicate that the browser with the most mature and polished interface was rated higher for usability, and semantic links were used by the users of all three browsers. / Conclusion: Confirmation or contradiction of our original hypotheses with relation to SWBs is detailed along with observations of implementation issues
