    Water is one of the basic necessities of life. God has gifted Pakistan with abundant water resources, with rivers flowing down the Himalayas and Karakoram heights from the world’s largest glaciers and free and unique bounty for this land. Pakistan is basically an agrarian economy. Out of its total geographical area of 79.61 million hectares, cultivated area is 22.05 million hectares. The total area under irrigation is 19.02 million hectares [Agricultural Statistics of Pakistan (2005-06)]. Irrigated land supplies more than 90 percent of agricultural production and most of the country’s food. Agriculture sector is regarded as the backbone of Pakistan’s economy. It contributes 25 percent of the GDP. About more than 50 percent labour force is employed in this sector. Agriculture sector is also the major user of water and its consumption will continue to dominate water requirement. Similarly, for industrial development main source of energy is hydropower which is generated by dint of water stored in big dams and reservoirs. Therefore the importance of the water for the survival of our economy cannot be denied

The Hampton Roads’ Building Resilient Communities (BRC) Flood Game is a simulation role-playing game designed to allow players to increase their understanding of the impact of various community response interventions to sea level rise and recurrent flooding. Players will examine and assess the tradeoffs of resiliency investments, the impact policies may have on the population, and the amount of time return on investment takes. The BRC game was designed using research-based game design principles to positively impact the user experience and facilitate the development of the overall game environment

    There is a decrease in general intellectual development level and mastery of a certain range of logical operations when learning. The decrease is compensated with the help of increased motivation and efficiency. When managing students’ cognitive processes and learning motives, it is important to consider the formation stages of logical and theoretical thinking. The authors found that insufficient professional orientation and low level of academic motivation hold a second position among the student dropout reasons, after the lack of desire to study and work in the chosen direction. If we consider the state of a graduate student at an agricultural university from the point of preparedness for the production process, then the professional education and competencies that the future specialist has mastered largely determine their competitiveness on the labor market. Since the current education system has never experienced the problems that the pandemic caused, it is necessary to reconsider the further development of the education system on an innovative base using not only distance (remote) education but also digital technologies

    ABSTRACT: Denial of Service and the Distributed Denial of Service Attacks have recently merged as one of the most newsworthy, if not the greatest, weaknesses of the Internet. This research paper attempts to explain how they work, why they are hard to combat today, and what will need to happen if they are to be brought under control. It is divided into parts for the purpose of easy analysis and understanding. The first is an overview of the current situation and also brief explanatory of the rest of the chapters being covered. The second is a detailed description of exactly how this attack works, and why it is hard to cope with today; of necessity it includes a description of how the Internet works today. The third section is totally about the different attacks in recent years and how they affected the people or the big organizations. The fourth section describes the short-term prospects, the tools which are used to rectify these attacks. The fifth is problems being faced with an explanatory of the percentage of attack in recent years and comparing the problems. The sixth is what can be done today to help alleviate this problem. The seventh section describes the legal actions and also legal actions that can be followed against the attack by the victim; and the eighth section describes the long-term picture, what will change to bring this class of problem under control, if not eliminate it entirely

    Proteomic and Biological Analysis of the Effects of Metformin Senomorphics on the Mesenchymal Stromal Cells

    Senotherapeutics are new drugs that can modulate senescence phenomena within tissues and reduce the onset of age-related pathologies. Senotherapeutics are divided into senolytics and senomorphics. The senolytics selectively kill senescent cells, while the senomorphics delay or block the onset of senescence. Metformin has been used to treat diabetes for several decades. Recently, it has been proposed that metformin may have anti-aging properties as it prevents DNA damage and inflammation. We evaluated the senomorphic effect of 6 weeks of therapeutic metformin treatment on the biology of human adipose mesenchymal stromal cells (MSCs). The study was combined with a proteome analysis of changes occurring in MSCs’ intracellular and secretome protein composition in order to identify molecular pathways associated with the observed biological phenomena. The metformin reduced the replicative senescence and cell death phenomena associated with prolonged in vitro cultivation. The continuous metformin supplementation delayed and/or reduced the impairment of MSC functions as evidenced by the presence of three specific pathways in metformin-treated samples: 1) the alpha-adrenergic signaling, which contributes to regulation of MSCs physiological secretory activity, 2) the signaling pathway associated with MSCs detoxification activity, and 3) the aspartate degradation pathway for optimal energy production. The senomorphic function of metformin seemed related to its reactive oxygen species (ROS) scavenging activity. In metformin-treated samples, the CEBPA, TP53 and USF1 transcription factors appeared to be involved in the regulation of several factors (SOD1, SOD2, CAT, GLRX, GSTP1) blocking ROS

    Evidence suggests that the CXXC type zinc finger (ZF-CXXC) protein 5 (CXXC5) is a critical regulator/integrator of various signaling pathways that include the estrogen (E2)-estrogen receptor α (ERα). Due to its ZF-CXXC domain, CXXC5 is considered to be a member of the ZF-CXXC family, which binds to unmethylated CpG dinucleotides of DNA and through enzymatic activities for DNA methylation and/or chromatin modifications generates a chromatin state critical for gene expressions. Structural/functional features of CXXC5 remain largely unknown. CXXC5, suggested as transcription and/or epigenetic factor, participates in cellular proliferation, differentiation, and death. To explore the role of CXXC5 in E2-ERα mediated cellular events, we verified by generating a recombinant protein that CXXC5 is indeed an unmethylated CpG binder. We uncovered that CXXC5, although lacks a transcription activation/repression function, participates in E2-driven cellular proliferation by modulating the expression of distinct and mutual genes also regulated by E2. Furthermore, we found that the overexpression of CXXC5, which correlates with mRNA and protein levels of ERα, associates with poor prognosis in ER-positive breast cancer patients. Thus, CXXC5 as an unmethylated CpG binder contributes to E2-mediated gene expressions that result in the regulation of cellular proliferation and may contribute to ER-positive breast cancer progression
