    Approaching the domesticated plant holobiont from a community evolution perspective

    Plants establish a pivotal relationship with their microbiome and are often conceptualized as holobionts. Nonetheless, holobiont theories have attracted much criticism, especially concerning the fact that the holobiont is rarely a unit of selection. In previous work, we discussed how the plant microbiome can be considered to be an ‘ecosystem on a leash’, which is subject to the influence of natural selection acting on plant traits. We proposed that in domesticated plants the assembly of the plant microbiome can usefully be conceptualized as being subject to a ‘double leash’, which encompasses both the effect of artificial selection imposed by the domesticator on plant traits and the leash from the plant to the microbiome. Here we approach the domesticated plant holobiont, simply defined as a community of organisms, from a community evolution point of view, and show how community heritability (a measure of community selection) complements the ‘double-leash’ framework in providing a community-level view of plant domestication and its impact on plant–microbe interactions. We also propose simple experiments that could be performed to investigate whether plant domestication has altered the potential for community selection at the holobiont level

    Energy analysis to assess the environmental sustainability of the dairy chain

    In the 1990s, attention was focused on saving energy and water with the aim of reducing production costs. Since the turn of this century, problems relating to the management of greenhouse gases have gradually assumed greater importance. Research has highlighted the problems that may arise regarding energy consumption in an Italian dairy chain. Using life-cycle assessment methods, the main steps along the production chain have been identified: breeding, dairy, and food store (FS). Our analysis shows that the different issues involved are often not easily reconcilable. Energy data need to undergo a careful and specific normalization process when dealing with specific data on different parameters (kWh/tmilk, kWh/tmilk processed, kWh/m2store). This study examined a variety of production cases (2 farms, 2 dairies, and 2 FSs) located in Lombardy, northern Italy, and electric and thermal energy consumption and carbon dioxide emissions were evaluated. A total of 2.8 kgCO2/kgcheese carbon dioxide emissions relating to the production process were recorded (39% breeding, 40% dairy, 1% FS). Further studies are needed in order to provide consumers with more precise and correct information (carbon labeling or green label). This may become an important element in consumer choice

    Toward A Stable Alpha-cycloalkyl Amino Acid With A Photoswitchable Cationic Side Chain

    The N-alkylated indanylidenepyrroline (NAIP) Schiff base 3 is an unnatural a-amino acid precursor potentially useful for the preparation of semisynthetic peptides and proteins incorporating charged side chains whose structure can be modulated via Z/E photoisomerization. Here we report that the heteroallylic protons of 3 led to partial loss of ethanol accompanied by formation of the novel heterocyclic system 4 during attempted deprotection. We also show that the same protons catalyze the thermal isomerization of 3, making the light-driven conformational control concept ineffective for times longer than a few hours. These problems are not present in the previously unreported compound 5 where the acidic methyl group is replaced by an H atom. Therefore, 5, rather than 3, constitutes a promising prototype for the design of building blocks capable to modulate the electrostatic potential of a protein in specific locations via light irradiation

    High-Resolution 3D FEM Stability Analysis of the Sabereebi Cave Monastery, Georgia

    This study assesses the static stability of the artificial Sabereebi Cave Monastery southeast of Georgia's capital, Tbilisi. The cliff into which these Georgian-Orthodox caverns, chapels, and churches were carved consists of a five-layered sequence of weak sedimentary rock—all of which bear a considerable failure potential and, consequently, pose the challenge of preservation to geologists, engineers, and archaeologists. In the first part of this study, we present a strategy to process point cloud data from drone photogrammetry as well as from laser scanners acquired in- and outside the caves into high-resolution CAD objects that can be used for numerical modeling ranging from macro- to micro-scale. In the second part, we explore four distinct series of static elasto-plastic finite element stability models featuring different levels of detail, each of which focuses on specific geomechanical scenarios such as classic landsliding due to overburden, deformation of architectural features as a result of stress concentration, material response to weathering, and pillar failure due to vertical load. With this bipartite approach, the study serves as a comprehensive 3D stability assessment of the Sabereebi Cave Monastery on the one hand; on the other hand, the established procedure should serve as a pilot scheme, which could be adapted to different sites in the future combining non-invasive and relatively cost-efficient assessment methods, data processing and hazard estimation

    Macroorganisms are colonized by microbial communities that exert important biological and ecological functions, the composition of which is subject to host control and has therefore been described as “an ecosystem on a leash”. However, domesticated organisms such as crop plants are subject to both artificial selection and natural selection exerted by the agricultural ecosystem. Here, we propose a framework for understanding how host control of the microbiota is influenced by domestication, in which a double leash acts from domesticator to host and host to microbes.     When the Heart Triggers the Esophagus: Esophageal Spasm after Electrical Cardioversion

    Introduction:“Esophageal spasm” is a generic term widely used to attribute unexplained non-cardiac chest pain and/or dysphagia to an esophageal motility disorder. Patient and methods: The authors present the case of an 86-year-old male patient with complete dysphagia after an elective electrical cardioversion for atrial fibrillation. An upper endoscopy performed shortly after the onset of the clinical picture documented disordered esophageal contractions. The patient became asymptomatic within 12 hours of the administration of a spasmolytic therapy. Results: To the best of our knowledge, this is the first report of esophageal spasm after an electrical cardioversion. Discussion: The temporal correlation supports the explanation of a cause-effect relationship between the clinical presentation and the preceding procedure, thus providing elements for clinicians to recognize and treat this particular condition

    Nutritionally enriched tomatoes (Solanum lycopersicum L.) grown with wood distillate: chemical and biological characterization for quality assessment

    : Bio-based products are nowadays useful tools able to affect the productivity and quality of conventionally cultivated crops. Several bio-based products are currently on the market; one of the newest and most promising is the wood distillate (WD) derived from the pyrolysis process of waste biomass after timber. Its foliar application has been widely investigated and shown to promote the antioxidant profile of cultivated crops. WD was used here as additive for the cultivation of tomato (Solanum lycopersicum L.) plants. The application improved quality (chemical) parameters, minerals, polyphenols, and lycopene contents of tomato fruits. The extracts of WD-treated and untreated tomatoes have been chemically and biologically characterized. The 1 H-NMR and ESI-mass spectrometry analyses of the extracts revealed the presence of different fatty acids, amino acids and sugars. In particular, the WD-treated tomatoes showed the presence of pyroglutamic acid and phloridzin derivatives, but also dihydrokaempferol, naringenin glucoside, cinnamic acid, and kaempferol-3-O-glucoside. When tested in cells, the extracts showed a promising anti-inflammatory profile in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Furthermore, the extracts displayed a slight vasorelaxant activity on rat aorta rings (either endothelium-denuded or endothelium-intact) pre-contracted with phenylephrine or potassium chloride. PRACTICAL APPLICATION: Wood distillate has been used for tomato plant growth. Tomatoes showed improved nutritional parameters, and their extracts displayed antioxidant and anti-inflammatory activities

    Bacterial endophytes of mangrove propagules elicit early establishment of the natural host and promote growth of cereal crops under salt stress

    Mangroves, dominating tropical intertidal zones and estuaries, are among the most salt tolerant plants, and propagate through reproductive units called propagules. Similarly to other plants’ seeds, propagules may harbor beneficial bacteria. Our hypothesis was that mangroves, being able to grow into seawater, should harbor bacteria able to interact with the host and to exert positive effects under salt stress, which could be exploited to improve crop production. Therefore, we isolated bacterial endophytes from mangrove propagules with the aim to test whether these bacteria have a beneficial potential on their natural host and on different crops like barley and rice, cultivated under salt stress. The 172 bacterial isolates obtained were screened for plant growth promotion (PGP) activities in vitro, and the 12 most promising isolates were tested on barley under non-axenic conditions and salt stress. Gordonia terrae KMP456-M40 was the best performing isolate, increasing ear weight by 65%. Basing on the in vivo PGP activity and the root colonization ability, investigated by fluorescence in situ hybridization and confocal microscopy, three strains were additionally tested on mangrove propagule germination and on rice growth. The most effective strain was again G. terrae KMP456-M40, which enhanced the root length of mangrove seedlings and the biomass of salt-stressed rice under axenic conditions up to 65% and 62%, respectively. We demonstrated that propagules, the reproductive units of mangroves, host beneficial bacteria that enhance the potential of mangrove seedlings establishment and confer salt tolerance to cereal crops
