    Legionella pneumophila infection induces programmed cell death, caspase activation, and release of high-mobility group box 1 protein in A549 alveolar epithelial cells: inhibition by methyl prednisolone

    <p>Abstract</p> <p>Background</p> <p><it>Legionella pneumophila </it>pneumonia often exacerbates acute lung injury (ALI) and acute respiratory distress syndrome (ARDS). Apoptosis of alveolar epithelial cells is considered to play an important role in the pathogenesis of ALI and ARDS. In this study, we investigated the precise mechanism by which A549 alveolar epithelial cells induced by <it>L. pneumophila </it>undergo apoptosis. We also studied the effect of methyl prednisolone on apoptosis in these cells.</p> <p>Methods</p> <p>Nuclear deoxyribonucleic acid (DNA) fragmentation and caspase activation in <it>L. pneumophila</it>-infected A549 alveolar epithelial cells were assessed using the terminal deoxyribonucleotidyl transferase-mediated triphosphate (dUTP)-biotin nick end labeling method (TUNEL method) and colorimetric caspase activity assays. The virulent <it>L. pneumophila </it>strain AA100jm and the avirulent <it>dotO </it>mutant were used and compared in this study. In addition, we investigated whether methyl prednisolone has any influence on nuclear DNA fragmentation and caspase activation in A549 alveolar epithelial cells infected with <it>L. pneumophila</it>.</p> <p>Results</p> <p>The virulent strain of <it>L. pneumophila </it>grew within A549 alveolar epithelial cells and induced subsequent cell death in a dose-dependent manner. The avirulent strain <it>dotO </it>mutant showed no such effect. The virulent strains of <it>L. pneumophila </it>induced DNA fragmentation (shown by TUNEL staining) and activation of caspases 3, 8, 9, and 1 in A549 cells, while the avirulent strain did not. High-mobility group box 1 (HMGB1) protein was released from A549 cells infected with virulent <it>Legionella</it>. Methyl prednisolone (53.4 μM) did not influence the intracellular growth of <it>L. pneumophila </it>within alveolar epithelial cells, but affected DNA fragmentation and caspase activation of infected A549 cells.</p> <p>Conclusion</p> <p>Infection of A549 alveolar epithelial cells with <it>L. pneumophila </it>caused programmed cell death, activation of various caspases, and release of HMGB1. The dot/icm system, a major virulence factor of <it>L. pneumophila</it>, is involved in the effects we measured in alveolar epithelial cells. Methyl prednisolone may modulate the interaction of <it>Legionella </it>and these cells.</p

    Effect of shadow availability at pasture on reproductive traits of Nelore bulls (Bos indicus) raised in southeastern Brazil

    A radiação solar é responsável pelo aumento da temperatura corpórea em touros. Uma alternativa para minimizar o estresse térmico é usar sombreamento artificial. Assim, este estudo teve como objetivo avaliar o efeito da redução do estresse térmico através da disponibilização de sombra artificial, sobre características reprodutivas de touros da raça Nelore (Bos indicus). Para isso, dez touros foram divididos em: Disponibilidade de sombra artificial (AS, n = 5) e Não-disponibilidade de sombra artificial (US, n=5). Cada grupo foi mantido em piquete de dois hectares cada, no qual a sombra para o grupo AS foi criada artificialmente. Os animais foram submetidos a avaliações clínicas-reprodutivas e análise seminal. Nenhuma interação foi observada entre os tratamentos (AS e US) e o tempo (8 coletas), para todas as variáveis analisadas (P &gt; 0.05). Nenhum efeito significativo (P&gt;0.05) de tratamento foi observado para os parâmetros analisados. Dessa forma, concluiu-se que a ausência de áreas sombreadas, durante o verão, não afetou negativamente as características reprodutivas como: circunferência escrotal, consistência testicular, motilidade progressiva, percentagem de células com movimentos rápidos (Computer Assisted Semen Analisys - CASA), morfologia e viabilidade espermática em touros da raça Nelore criados na região sudeste do Brasil, considerando que esse resultado pode ser diferente em outras regiões do país, onde as temperaturas ambientais são mais elevadas.Solar radiation is responsible for bull body temperature elevation. An alternative to minimize heat stress is to use artificial shade. Thus, this study aimed to evaluate the effect of thermal stress reduction, through shade availability, on reproductive characteristics of Nellore bulls (Bos indicus). For this, ten bulls were divided in: Available artificial shade (AS, n = 5) and Unavailable shade (US, n = 5). Each group was kept in two hectare paddocks, in which shade availability for group AS was artificially created. Animals were submitted to a clinical-reproductive evaluation and seminal analyses. No interaction was observed between treatments (AS and US) and time (8 collections) for all analyzed variables (P&gt;0.05). No significant effect (P &gt; 0.05) of treatment was observed for all parameters analyzed. So, it can be concluded that the absence of shaded areas during summer does not negatively affect reproductive characteristics such as: scrotal circumference, testicular consistency, progressive motility, percentage of rapidly moving cells (Computer Assisted Semen Analysis - CASA), morphology or sperm viability in Nellore bulls raised in southeastern Brazil, considering that results could be different in other regions of the country where average temperature is higher

    Correlações entre a hemodinâmica testicular e as características espermáticas em carneiros

    A artéria testicular é responsável pelo fluxo de sangue que chega aos testículos e tem grande importância na termorregulação. Mudanças vasculares nos testículos podem levar à queda da produção espermática, refletindo na motilidade e morfologia. O objetivo deste trabalho foi avaliar as correlações entre a vascularização testicular e as características espermáticas. Foram utilizados oito carneiros adultos Santa Inês com diferentes status reprodutivos. A vascularização testicular e as características seminais foram avaliadas por um período de 90 dias. A ultrassonografia Doppler colorida foi utilizada para avaliar a hemodinâmica testicular. Foram calculados os índices de resistência (RI) e os índices de pulsatilidade (PI) com o modo espectral do Doppler. O modo colorido do Doppler foi utilizado para analisar o fluxo sanguíneo do plexo pampiniforme e do parênquima testicular. As características seminais avaliadas foram o volume, concentração, motilidade e morfologia. Os dados foram submetidos ao teste de correlação linear de Person (P &lt; 0.05 foi considerado significativo). Não foram encontradas relações entre motilidade e a hemodinâmica testicular. A porcentagem de defeitos totais correlacionou-se positivamente com o escore de vascularização dos parênquimas direito e esquerdo, e com o RI e PI esquerdos. Também o escore de vascularização dos plexos se correlacionou positivamente com a média de pixels e negativamente com o RI e PI, de ambos os lados. Este trabalho mostrou que o aumento de defeitos espermáticos pode estar correlacionado com o aumento do fluxo sanguíneo nos testículos; contudo, mais estudos são necessários.The testicular artery is responsible for the blood supply that reaches the testis and has great importance in heat radiation. Vascular changes in the testis may lead to damage in sperm production, reflected in sperm motility and morphology. The aim of the present study was to evaluate correlations between testicular vascularity and sperm characteristic. Eight adult Santa Ines rams showing different reproductive status were used. The testicular vascularity and sperm characteristics were evaluated fortnightly during 90 days. Color Doppler ultrasonography was used to evaluate the testicular hemodynamic. Resistance index (RI) and pulsatility index (PI) of the testicular artery were evaluated by spectral-Doppler mode. The color-Doppler mode was used to evaluate the blood flow of the pampiniform plexus and testicular parenchyma. The semen analyses assessed were volume, concentration, motility, and morphology. The data were submitted to Pearson´s linear correlations test (p &lt; 0.05 was considered significant). No correlations were found between motility and testicular hemodynamic. The percentage of total sperm defects was positively correlated to left and right parenchymal score and to left RI and PI. On the other hand, the pampiniform plexus score was positively correlated to the number of colored pixels and negatively correlated to the RI and PI, for both sides. This study showed that the increase of sperm defect can be related to increase of testicular blood flow; however, more studies are need

    The industry has made available in the market a series of substances (nutraceuticals) which intent would be to optimize the use of nutrients in some metabolic paths, influencing positively reproductive performance in animals. However, the response to the use of nutraceuticals varies according to the animal. As the organism is highly complex and in order to achieve a perfect activity of the hypothalamic-pituitary-gonadal axis, an ideal interaction in molecular basis is needed, where the nutraceuticals can have their direct action. The aim of this study was to review the function and research results using the main nutraceuticals (&#946; carotene, vitamin A, L-carnitine, omegas 3, 6 and 9 and Gamma-oryzanol) on reproductive characteristics of bulls and stallions.A indústria tem disponibilizado no mercado uma série de substâncias (nutracêuticos) com a intenção de otimizar a utilização de nutrientes em algumas vias metabólicas, influenciando positivamente o desempenho reprodutivo dos animais. No entanto, a resposta ao uso de nutracêuticos varia de acordo com o animal. Como o organismo é altamente complexo e, a fim de alcançar perfeita atividade do eixo hipotálamo-hipófise-gonadal, uma interação ideal na base molecular é necessária, onde os nutracêuticos podem ter sua ação direta.     The association of medroxyprogesterone to the Ovsynch protocol for the fixed time artificial insemination of cyclic buffaloes (Bubalus bubalis) in the Eastern Amazon

    O presente estudo visou avaliar os efeitos da associação da medroxiprogesterona (análogo sintético da progesterona) ao protocolo Ovsynch sobre o crescimento folicular, a ovulação e a taxa de concepção de búfalas criadas na Amazônia Oriental (Tracuateua-PA). Vinte e sete fêmeas adultas (G1 n=14 e G2 n=13), cíclicas, sem bezerro ao pé e com ECC 3,5 foram submetidas a Ovsynch. Os animais do G2 receberam 60 mg de medroxiprogesterona entre D0 e D7 (D0=início do tratamento). A ultra-sonografia ovariana foi realizada nos D 0, 7, 9 e 10. O contingente de folículos pequenos diferiu no D7 (G1: 4,57±0,60 versus G2: 6,54±0,67; P=0,05). Tempo e tratamento influenciaram o diâmetro folicular no D7. O crescimento do folículo dominante entre D7 e D9 foi maior nos animais tratados (G1: 2,05±0,49 mm/dia versus 3,48±0,41 mm/dia; P<0,05). Mais animais do G1 ovularam precocemente (35,71% versus 30,77%), porém isso não afetou as taxas de concepção (G1: 50,00% e G2: 30,77%; P>0,05). Os achados sugerem que a medroxiprogesterona (1) aumenta recrutamento folicular e retarda o crescimento dos folículos com diâmetro maior que 5,0 mm entre D0 e D7; (2) sua retirada incrementa em 1,7 vezes o crescimento folicular do D7 ao D9; (3) pode contribuir para a ovulação de folículos maiores e, em tese, para maior formação de tecido luteínico; (4) não promove ovulação precoce após o Ovsynch; (5) não eleva as taxas de concepção após sincronização de fêmeas cíclicas e com bom escore corporal, devendo ser avaliada para uso em fêmeas acíclicas ou com ECC mais baixo.The aim of this study was to evaluate the effect of the association of medroxyprogesterone with the Ovsynch protocol on the follicular growth, ovulation, and conception rate of buffaloes in the Eastern Amazon (Tracuateua-PA). Twenty-seven cyclic, non-lactating females (G1 n=14 and G2 n=13) with a 3.5 BCS were synchronized with the Ovsynch. Animals from G2 received a supplementation of 60 mg of medroxyprogesterone from D0 to D7 (D0=starting day of treatment). Ultrasonography evaluation on ovaries was realized on Days 0, 7, 9, and 10. A significant effect of treatment on follicle number was observed on D7 (G1: 4.57±0.60 versus G2: 6.54±0.67; P=0.05). Time and treatment influenced follicular diameter on D7. The growth of the dominant follicular was increased from D7 to D9 on treated females (G1: 2.05±0.49 mm/day versus 3.48±0.41 mm/day; P<0.05). G1 animals (35.71% versus 30.77%) ovulated comparatively earlier than their G2 counterparts, but this did not affect the conception rates (G1: 50,00% and G2: 30,77%; P>0,05). The findings suggest that medroxyprogesterone: (1) increases follicular recruitment and inhibits the growth of follicles whose diameter was larger than 5.0mm from D0 to D7; (2) the withdrawal of medroxyprogesterone speeds up follicular growth 1.7 fold from D7 to D9; (3) would probably contribute to the ovulation of larger follicles thereby forming more luteal tissue; (4) does not stimulate early ovulation after Ovsynch; and (5) does not elevate the conception rate after the synchronization of cyclic females with good body condition, and should be evaluated for the utilization in acyclic/reduced BCS females.Embrapa Amazônia OrientalBanco da Amazôni

    Impulse Control in Finance: Numerical Methods and Viscosity Solutions

    The goal of this thesis is to provide efficient and provably convergent numerical methods for solving partial differential equations (PDEs) coming from impulse control problems motivated by finance. Impulses, which are controlled jumps in a stochastic process, are used to model realistic features in financial problems which cannot be captured by ordinary stochastic controls. The dynamic programming equations associated with impulse control problems are Hamilton-Jacobi-Bellman quasi-variational inequalities (HJBQVIs) Other than in certain special cases, the numerical schemes that come from the discretization of HJBQVIs take the form of complicated nonlinear matrix equations also known as Bellman problems. We prove that a policy iteration algorithm can be used to compute their solutions. In order to do so, we employ the theory of weakly chained diagonally dominant (w.c.d.d.) matrices. As a byproduct of our analysis, we obtain some new results regarding a particular family of Markov decision processes which can be thought of as impulse control problems on a discrete state space and the relationship between w.c.d.d. matrices and M-matrices. Since HJBQVIs are nonlocal PDEs, we are unable to directly use the seminal result of Barles and Souganidis (concerning the convergence of monotone, stable, and consistent numerical schemes to the viscosity solution) to prove the convergence of our schemes. We address this issue by extending the work of Barles and Souganidis to nonlocal PDEs in a manner general enough to apply to HJBQVIs. We apply our schemes to compute the solutions of various classical problems from finance concerning optimal control of the exchange rate, optimal consumption with fixed and proportional transaction costs, and guaranteed minimum withdrawal benefits in variable annuities

    Effect of globular adiponectin on interleukin-6 and interleukin-8 expression in periodontal ligament and gingival fibroblasts

    PURPOSE: Globular adiponectin (gAd) is a type of adipocytokine, which is mainly produced by adipose tissue. It has been reported that gAd acts as a pro- as well as an anti-inflammatory factor. Interleukin (IL)-6 and IL-8 are pro-inflammatory cytokines. To investigate the role of gAd on periodontal tissues, the expression of adiponectin receptor 1 (AdipoR1) and the effect of gAd on the expression of IL-6 and IL-8 were investigated in periodontal ligament (PDL) and gingival fibroblasts. METHODS: PDL and gingival fibroblasts were cultured from human periodontal tissues. gAd derived from Escherichia coli and murine myeloma cells were used. The expression of AdipoR1 was estimated by reverse transcription-polymerase chain reaction and western blot. The expression of cytokines was measured by enzyme-linked immunosorbent assay. RESULTS: PDL and gingival fibroblasts expressed both mRNA and protein of AdipoR1. gAd derived from E. coli increased the production of IL-6 and IL-8, but polymyxin B, an inhibitor of lipopolysaccharide (LPS), inhibited IL-6 and IL-8 production induced by gAd in both types of cells. gAd derived from murine myeloma cells did not induce IL-6 and IL-8 production in those cells. gAd derived from E. coli contained higher levels of LPS than gAd derived from murine myeloma cells. LPS increased production of IL-6 and IL-8 in PDL and gingival fibroblasts, but pretreatment of cells with gAd derived from murine myeloma cells did not inhibit LPS-induced IL-6 and IL-8 expression. CONCLUSIONS: Our results suggest that PDL and gingival fibroblasts express AdipoR1 and that gAd does not act as a modulator of IL-6 and IL-8 expression in PDL and gingival fibroblasts.ope

    Eficiência in vitro de três diluidores para sêmen de coelho

    Foi o objetivo deste trabalho comparar in vitro a eficiência de três diluidores para sêmen de coelhos: solução de Ringer com lactato de sódio, um meio à base de citrato de sódio e gema de ovo e um meio à base de leite desnatado sobre a manutenção da motilidade progressiva e do vigor espermático. Para tanto, foram utilizados 5 coelhos realizando-se 10 colheitas de sêmen de cada (n = 50). O sêmen foi colhido com vagina artificial, e avaliado quanto ao volume, motilidade, vigor e concentração. O sêmen foi diluído (20x106 espermatozóides/mL) em microtubos pré-aquecidos a 37ºC nos três diluidores, e então incubados em banho-maria a 37ºC por 120 minutos, realizando-se avaliações a cada 30 minutos. Imediatamente após a diluição (tempo 0) a motilidade espermática não diferiu (P&gt;;0,05) entre os diluidores, todavia, reduziu (P&lt;0,05) no diluído com Ringer quando comparado ao sêmen in natura. Já o vigor no tempo 0 diminuiu (P&lt;0,05) nos três meios. A motilidade espermática foi melhor preservada (P&lt;0,05) durante a incubação de 30 até 120 minutos para o sêmen diluído nos diluidores citrato-gema e à base de leite do que para aquele diluído em Ringer. A preservação do vigor variou entre os diluidores durante o tempo de incubação in vitro, contudo, foi semelhante entre os três diluidores após 120 minutos de incubação. Com estes resultados, pode-se inferir que os diluidores testados proporcionam um meio que permite manter a viabilidade espermática para que possa ser realizada a inseminação artificial em coelhas até duas horas pós-diluiçãoThe objective of this experiment was to compare in vitro efficiency of three rabbit semen extenders: sodium lactate Ringer solution, sodium citrate and yolk-egg medium, and skim milk-based medium, on maintenance of sperm vigour and motility. To that end 5 rabbits were utilized; ten semen collections were taken from each (n = 50). The semen was collected by artificial vagina, and evaluated for volume, motility, vigour, and concentration. The semen was diluted (20x10(6) spermatozoa/mL) in pre-warmed micro tube at 37ºC in the three extenders, and then it was incubated in water bath at 37ºC during 120 minutes, performing evaluation every 30 minutes. Immediately after the dilution (time 0) the sperm motility was not different among extenders (P&gt;;0.05), however, decreased (P&lt;0.05) in Ringer extender when compared to in natura semen. The vigor in time 0 decreased (P&lt;0.05) in the three extenders. The sperm motility was better preserved (P&lt;0.05) during the incubation from 30 to 120 minutes for the semen diluted in yolk egg-citrate and skim milk-based extenders than for the Ringer extender. The vigour preservation varied among the extenders during the in vitro incubation; however, it was similar among the three extenders after 120 minutes of incubation. Based on these results, it can be deduced that the tested extenders promote a medium that allows the maintenance of sperm viability so that artificial insemination can take place within two hours of post-dilutio

    Explorando os efeitos da sincronização do segundo estro e flushing alimentar sobre a incidência de cistos ovarianos em marrãs utilizando gonadotrofinas exógenas

    Estrus stimulation by exogenous gonadotropins (EG) in association with dietary flushing is an important tool for the improvement of gilt reproductive performance. However, there is evidence associating both flushing and EG with a disturbance in the endocrine balance that could lead to increased ovarian cysts. The aim of this study was to evaluate whether flushing or EG might affect the ovulation rate and the incidence of ovarian cysts. Seventy-one gilts were randomly distributed into 2x2 factorial design with four treatments: flushing and hormone (wFwH); no flushing and hormone (nFwH); flushing without hormone (wFnH); and neither flushing nor hormone (nFnH). Gilts were slaughtered for macroscopic and histopathological ovary examination approximately five days after AI. The characterization of these cysts was performed by optical microscopy in the following: follicular cysts (FC), luteinized cysts (LC) or cystic corpora lutea (CCL). The number of ovulations did not differ between treatments. There was no interaction between the factors in any analyzed variable. The frequency of gilts with CCL and LC was not affected by flushing and EG. No difference was found in the incidence of FC, with 12.5% and 5.88% in gilts from wFwH and nFwH treatments, respectively. There were no differences in the proportion of CCL between FC and LC (9.85 vs. 4.22 and 4.22%, respectively). In conclusion, the use of exogenous gonadotropins for second estrus synchronization in gilts, either alone or in association with dietary flushing, does not increase the incidence of ovarian cysts, nor does it decrease the ovulation rate.A estimulação do estro por gonadotrofinas exógenas (GE) associada ao flushing alimentar é uma ferramenta importante na melhoria do desempenho reprodutivo de marrãs. Contudo, há evidência da associação do flushing com GE levando ao desequilíbrio no sistema endócrino que poderia levar ao aumento de cistos ovarianos. O objetivo deste estudo foi avaliar se o flushing ou GE pode afetar a taxa de ovulação e a incidência de cistos ovarianos. Setenta e uma marrãs foram distribuídas aleatoriamente em arranjo fatorial 2x2 com quatro tratamentos: flushing e hormônio (cFcH); sem flushing e com hormônio (sFcH); com flushing e sem hormônio (cFsH) e sem flushing e hormônio (sFsH). Marrãs foram abatidas para exame macroscópico e histopatológico dos ovários, aproximadamente cinco dias após IA. A caracterização desses cistos foi realizada por microscopia óptica: cistos foliculares (CF), cistos luteinizados (CL) ou corpos lúteos císticos (CCL). O número de ovulações não diferiu entre os tratamentos. Não houve interação entre os fatores em qualquer variável analisada. A frequência de leitoas com CCL e CL não foi afetada pelo flushing e GE. Não houve diferença na incidência de CF, com 12,5% e 5,88 % em leitoas dos tratamentos cFcH e sFcH, respectivamente. Não foram obtidas diferenças na proporção de CCL entre CF e CL (9,85 vs. 4,22 e 4,22%, respectivamente). Em conclusão, a utilização de gonadotrofinas exógenas para sincronização do segundo estro de marrãs, isoladamente ou em associação com o flushing, não aumenta a incidência de cistos ovarianos e não diminui a taxa de ovulação

    Efeitos combinados da restrição alimentar e flushing sobre a fertilidade de marrãs inseminadas artificialmente em diferentes ciclos estrais

    The study evaluated the effect of dietary restriction in prepubertal gilts associated with the use of flushing in the preceding cycle of artificial insemination, 2nd or 3rd estrus on productive performance, number of corpora lutea and fetus, besides the fetus survival rate. A total of 96 gilts were distributed to a completely randomized design with 2x2x2 factorial arrangement of treatments, consisting of: nutritional strategy, ad libitum or restricted feeding, use or not of flushing in the preceding cycle AI, and time of AI, held on the 2nd or 3rd estrus. The age at which gilts expressed the 1st estrus was not influenced by nutritional strategy, but the body weight and weight gain were higher (P &lt; 0.05) in gilts submitted to ad libitum feeding. No interactions were observed as treatment effect on number of corpora lutea and fetus survival rate. The number of fetuses in females on ad libitum feeding was not influenced by time of AI, but the females feed restriction showed an increase (P &lt; 0.05) in the number of fetuses when inseminated on 3rd estrus. Thus, it is concluded that the use of ad libitum nutritional strategy allows for insemination forward 2nd estrus without reducing the number of fetuses, reducing the non-productive days gilt. However, this strategy reduces the rate of fetal survival until day 35 of gestation.Foi avaliado o efeito da restrição alimentar na fase pré-púbere associada ao emprego do “flushing” no ciclo precedente ao da inseminação artificial, sendo realizada no 2° ou 3° estro, sobre o desempenho produtivo, numero de corpos lúteos e de fetos, além da taxa de sobrevivência fetal. Um total de 96 fêmeas foi distribuído em delineamento inteiramente casualizado com arranjo fatorial 2x2x2, sendo considerados os fatores: estratégia nutricional, alimentação ad libitum ou restrita; utilização ou não de “flushing” no ciclo precedente a IA, e momento da IA, realizada no 2° ou 3° estro, constituindo oito tratamentos. A idade em que as marras manifestaram o 1° estro não foi influenciada pela estratégia nutricional, mas o peso e o ganho de peso foram maiores (P &lt; 0,05) na alimentação ad libitum. Não foram observados interação e tão pouco efeito de tratamento no numero de corpos lúteos e a taxa de sobrevivência dos fetos. O numero de fetos em fêmeas mantidas em alimentação ad libitum não foi influenciado pelo momento da IA, contudo as fêmeas em restrição alimentar apresentaram aumento (P &lt; 0,05) no numero de fetos quando inseminadas no 3° estro.
