    Nutrition and Rheumatoid Arthritis in the ‘Omics’ Era

    Modern high-throughput ‘omics’ science tools (including genomics, transcriptomics, pro teomics, metabolomics and microbiomics) are currently being applied to nutritional sciences to unravel the fundamental processes of health effects ascribed to particular nutrients in humans and to contribute to more precise nutritional advice. Diet and food components are key environmental factors that interact with the genome, transcriptome, proteome, metabolome and the microbiota, and this life-long interplay defines health and diseases state of the individual. Rheumatoid arthritis (RA) is a chronic autoimmune disease featured by a systemic immune-inflammatory response, in genetically susceptible individuals exposed to environmental triggers, including diet. In recent years increasing evidences suggested that nutritional factors and gut microbiome have a central role in RA risk and progression. The aim of this review is to summarize the main and most recent applications of ‘omics’ technologies in human nutrition and in RA research, examining the possible influences of some nutrients and nutritional patterns on RA pathogenesis, following a nutrigenomics approach. The opportunities and challenges of novel ‘omics technologies’ in the exploration of new avenues in RA and nutritional research to prevent and manage RA will be also discussed

    Adding plant oils to dairy goat diets: Effect on fatty acid content of milk fat

    [ES]: El objetivo del presente trabajo fue investigar el efecto de la adición de aceites vegetales de diferente grado de insaturación a la dieta de cabras lecheras sobre el perfil de ácidos grasos de la grasa láctea. Doce cabras de raza Malagueña fueron asignadas al azar a uno de cuatro tratamientos: dieta basal sin aceite añadido y la misma dieta basal adicionada con 48 g/d de aceite de girasol alto oleico, aceite de girasol normal o aceite de lino. La dieta estuvo compuesta por heno de alfalfa (30%) y un concentrado granulado (70%) en el que se incluyó el aceite correspondiente. Todos los aceites redujeron el contenido de ácidos grasos saturados de cadena media de la grasa láctea (P 0,05). El aceite de girasol normal incrementó el contenido de los ácidos C18:1t10, vaccénico y ruménico y aumentó el valor de la ratio linoleico/α-linolénico (P 0,05). Los resultados obtenidos permiten concluir que la adición de aceite de lino a la dieta de cabras en lactación, en comparación con la de los aceites de girasol alto oleico y normal, modifica el perfil de ácidos grasos de la grasa láctea en un sentido más favorable desde el punto de vista de la salud humana.[EN]: The aim of this study was to investigate the effects of adding differently unsaturated vegetable oils to a dairy goat diet on fatty acid content of milk fat. Twelve Malagueña goats were randomly allocated to one of four treatments: no oil basal diet and the same basal diet supplemented with 48 g/d of high oleic sunflower oil, regular sunflower oil or linseed oil. The basal diet was made of alfalfa hay and a pelleted concentrate (30:70 w/w). The concentrate included the respective oils. Supplemented diets decreased medium chain saturated fatty acid content in milk fat (P 0.05). Regular sunflower oil increased C18:1t10, vaccenic and rumenic acid contents as well as linoleic to α-linolenic acid ratio (P 0.05). It was concluded that compared to high oleic o regular sunflower oils, adding linseed oil in dairy goat diets modifies milk fat fatty acid content more favourably from the point of view of human health.Peer Reviewe

    Exploring the efficiency of gallic acid-based dendrimers and their block copolymers with PEG as gene carriers

    The synthesis of a new family of amino-functionalized gallic acid-triethylene glycol (GATG) dendrimers and their block copolymers with polyethylene glycol (PEG) has recently being disclosed. In addition, these dendrimers have shown potential for gene delivery applications, as they efficiently complex nucleic acids and form small and homogeneous dendriplexes. On this basis, the present study aimed to explore the interaction of the engineered dendriplexes with blood components, as well as their stability, cytotoxicity and ability to enter and transfect mammalian cells. Results show that GATG dendrimers can form stable dendriplexes, protect the associated pDNA from degradation, and are biocompatible with HEK-293T cells and erythrocytes. More importantly, dendriplexes are effectively internalized by HEK-293T cells, which are successfully transfected. Besides, PEGylation has a marked influence on the properties of the resulting dendriplexes. While PEGylated GATG dendrimers have improved biocompatibility, the long PEG chains limit their uptake by HEK-293T cells, and thus, their ability to transfect them.     1 página.-- Trabajo presentado al: IDF International Symposium on Sheep, Goat and other non-Cow Milk (Atenas, Grecia, del 16 al 18 de mayo de 2011).The aim of this study was to evaluate the effect of different forage: concentrate (FC) ratios in dairy ewe diets supplemented with sunflower oil (SO) on animal performance and milk fatty acid (FA) profile, particularly focusing on trans C18:1 FAs and conjugated linoleic acid (CLA). For that purpose, sixty lactating Assaf ewes were randomly assigned to 6 treatments in a 3 x 2 factorial arrangement: 3 FC ratios (30:70, 50:50 and 70:30) and 2 levels of SO addition (0 and 20 g/kg DM). Both the diet FC ratio and SO supplementation affected milk yield, although differences between treatments were fairly small. Although the proportion of concentrate induced limited changes in milk FA profile, dietary SO significantly reduced saturated FAs and enhanced total CLA. Furthermore, the incorporation of SO in ewe diets decreased the atherogenicity index value by about 25% and doubled potentially healthy FA contents such as trans-11 C18:1 and cis-9 trans-11 CLA. However, the inclusion of SO in a high concentrate diet (30:70) could switch linoleic acid biohydrogenation pathways, resulting in a significant increase in trans-10 C18:1, trans-9 cis-11 C18:2 and trans-10 cis-12 C18:2 milk fat percentages.Peer reviewe

    Desarrollo de materiales superabsorbentes biodegradables procesados a partir de subproductos agroindustriales

    Actualmente se admite la necesidad cada vez más apremiante de desarrollar nuevos materiales con mayor sostenibilidad. Los materiales superabsorbentes constituyen un claro ejemplo, lo cual implica que estén basados en materias primas renovables, de menor coste e impacto ambiental. A pesar de ello, la comercialización de productos superabsorbentes basados en materias primas de origen natural, que sean biocompatibles y biodegradables se encuentra aún bastante limitada, existiendo una evidente necesidad de impulsar la investigación en el sector. El trabajo pretende contribuir a satisfacer esa necesidad, estableciéndose como objetivo principal desarrrollar nuevos materiales que presenten gran capacidad de absorción de agua (absorbentes y superabsobentes), que puedan ser procesados mediante técnicas de eficacia contrastada en la industria de fabricación de plásticos. Todo ello a partir de fuentes naturales renovables tales como proteínas procedentes de subproductos de la industria agroalimentaria. Además de su capacidad absorbente estos materiales deben presentar propiedades mecánicas y biodegradabilidad apropiadas para que puedan suponer una alternativa a los fabricados con polímeros sintéticos. Como resultado del trabajo se pretende adquirir el conocimiento científico y establecer las bases tecnológicas necesarias para conseguir matrices absorbentes y superabsorbentes biodegradables. Asimismo, se evaluará su potencial en productos SAB para higiene personal y para horticultura.There is an increasing need to shift towards the development of sustainable and environmentally friendly materials in the world market nowadays. Superabsorbents polymers (SAP) are a good example of this new type of materials, being based on renewable natural components with a lower cost. Notwithstanding, the productionand commercialization of biocompatible and biodegradable SAP are still very limited, which makes the need of research on this field pretty clear. This work pretends to cover that need establishing as its main objective the development of new materials that display a great water absorption capacity (absorbents and superabsorbents). These materials will be produced from natural renewable sources (proteins obtained as agro-food industry byproducts) through techniques commonly used in the plastic industry. These materials not only should possess a great water absorption capacity but they should present proper mechanical properties and biodegradability, so they could represent an attractive choice to those obtained from synthetic polymers. Eventually, the acquisition of scientific knowledge and the establishment of the technological base needed for the success in the development of those biodegradable SAP matrixes will be pursued with the different activities.
