Analysis of reaktivated proviruses of the human endogenous retrovirus familie HERV-K/HML2 and creation of a model for transcriptional control in melanoma and teratocarcinoma cell lines

Humane endogene Retroviren (HERVs) sind Relikte von Infektionen der Keimzellen von Primatenvorläufern mit exogenen Retroviren vor 40 Millionen Jahren. Die meisten HERV Elemente sind defekt durch die Akkumulation von Mutationen, Deletionen und Rearrangements. Lediglich einige Proviren der HERV-K/HML-2 Familie enthalte noch offenen Leserahmen für alle retroviralen Proteine. Die Expression repetitiver endogener Elemente wird in somatischen Zellen unterdrückt. Transkription und Translation von HERV-K Genprodukten tritt in Keimzelltumoren, Melanomen und Eierstockkrebs auf. In der vorliegenden Arbeit wurde die HERV-K exprimierende Melanomlinie UKRV Mel 2-C9 etabliert und charakterisiert. Es konnte gezeigt werden, dass die HERV-K Expressionen durch die dominante Aktivität des Provirus 108 bedingt ist. Die Expression und korrekte Prozessierung der viralen Proteine konnte nachgewiesen werden. Durch Elektronenmikroskopie wurde bestätigt, dass HERV-K 108 virale Partikel mit charakteristischen Strukturen der Hüllproteine produziert. Diese Partikel enthalten virale Volllängen RNA-Genome. Da HERV-K 108 in der Linie UKRV Mel 2-C9 eine Mutation im aktiven Zentrum der Reversen Transkriptase zeigt, sind die Partikel nicht infektiös. Um Einblicke in die der HERV-K Expression in Keimzelltumoren und Melanomen zu Grunde liegenden transkriptionellen Regulationsmechanismen zu erhalten, wurden mittels 5’ und 3’ RACE (rapid amplification of cDNA ends) der Transkriptionsstart und die Terminationsstelle bestimmt. Es konnte gezeigt werden, dass der HERV-K Promotor in den untersuchten Zelllinien TATA-unabhängig agiert obwohl die HERV-K LTR (long terminal repeat) ein TATA-Motiv aufweist. Die transkriptionelle Aktivität des HERV-K Promotors ist abhängig von einer GC-Box in unmittelbarer Nähe zu einem Initiatorelement am Startpunkt der Transkription. Das GC-Sequenzmotiv ist die Bindestelle für die Transkriptionsfaktoren SP1 (specific protein) und SP3. Mutationen des Bindemotivs führten zu einem drastischen Abfall der Promotoraktivität im Luziferaseassay. Durch Proteinknockdown mittels spezifischer siRNA gegen SP1 und SP3 konnte die Promotoraktivität ebenso deutlich reduziert werden. Elektrophoretic mobility shift assays und Chromatinimmunopräzipitationen bestätigten eine Bindung von SP1 und SP3 an die HERV-K LTR. Die Transkriptionsfaktoren SP1 und SP3 sind an der Initiation der basalen Transkription von HERV-K maßgeblich beteiligt.Human endogenous retroviruses (HERVs) resulted from germ line infections of primate ancestors. At present most HERV families are defective but some proviruses of the HERV-K/HML-2 family have maintained open reading frames for all viral proteins. Although usually strictly repressed, HERV-K transcription and translation is re-activated in germ cell tumours and melanoma. For these types of cancer HERV-K is a diagnostic and prognostic marker. We aim to understand the transcriptional regulation of HERV-K. The transcriptional start and termination sites were identified by 5' and 3’-rapid amplification of cDNA ends. The impact of specific transcription factors on the HERV-K LTR was analyzed using luciferase reporter assays, mutational and siRNA approaches, chromatin immuno-precipitation assays (ChIP) and electrophoretic mobility shift assays (EMSA). We demonstrate that the HERV-K LTR is a TATA independent promoter despite the presence of a TATA motif. Mutation of this motif did not abrogate promoter activity. Transcription initiation is mediated by a GC-box, the binding motif of the transcription factors SP1 (specificity protein) and SP3, located near an initiator element. Mutation of the GC-box significantly reduced HERV-K promoter activity as did a knock down of the Sp1 and Sp3 proteins. EMSA and ChIP confirmed that the GC-box represents a binding site for the transcription factors Sp1 and Sp3

Gene editing of SAMHD1 in macrophage-like cells reveals complex relationships between SAMHD1 phospho-regulation, HIV-1 restriction, and cellular dNTP levels

ABSTRACT Sterile α motif and HD domain-containing protein 1 (SAMHD1) is a dNTP triphosphate triphosphohydrolase (dNTPase) and a potent restriction factor for immunodeficiency virus 1 (HIV-1), active in myeloid and resting CD4+ T cells. The anti-viral activity of SAMHD1 is regulated by dephosphorylation of the residue T592. However, the impact of T592 phosphorylation on dNTPase activity is still under debate. Whether additional cellular functions of SAMHD1 impact anti-viral restriction is not completely understood. We report BLaER1 cells as a novel human macrophage HIV-1 infection model combined with CRISPR/Cas9 knock-in (KI) introducing specific mutations into the SAMHD1 locus to study mutations in a physiological context. Transdifferentiated BLaER1 cells harbor active dephosphorylated SAMHD1 that blocks HIV-1 reporter virus infection. As expected, homozygous T592E mutation, but not T592A, relieved a block to HIV-1 reverse transcription. Co-delivery of VLP-Vpx to SAMHD1 T592E KI mutant cells did not further enhance HIV-1 infection indicating the absence of additional SAMHD1-mediated anti-viral activity independent of T592 dephosphorylation. T592E KI cells retained dNTP levels similar to WT cells indicating uncoupling of anti-viral and dNTPase activity of SAMHD1. The integrity of the catalytic site in SAMHD1 was critical for anti-viral activity, yet a poor correlation between HIV-1 restriction and global cellular dNTP levels was observed in cells harboring catalytic core mutations. Together, we emphasize the complexity of the relationship between HIV-1 restriction, SAMHD1 enzymatic function, and T592 phospho-regulation and provide novel tools for investigation in an endogenous and physiological context. IMPORTANCE We introduce BLaER1 cells as an alternative myeloid cell model in combination with CRISPR/Cas9-mediated gene editing to study the influence of sterile α motif and HD domain-containing protein 1 (SAMHD1) T592 phosphorylation on anti-viral restriction and the control of cellular dNTP levels in an endogenous, physiologically relevant context. A proper understanding of the mechanism of the anti-viral function of SAMHD1 will provide attractive strategies aiming at selectively manipulating SAMHD1 without affecting other cellular functions. Even more, our toolkit may inspire further genetic analysis and investigation of restriction factors inhibiting retroviruses and their cellular function and regulation, leading to a deeper understanding of intrinsic anti-viral immunity

Patient-Reported Quality of Care for Osteoarthritis in General Practice in South Tyrol, Italy: Protocol for Translation, Validation and Assessment of the OsteoArthritis Quality Indicator Questionnaire (OA-QI)

Background: Evidence-based recommendations for the treatment of knee and hip osteoarthritis are similar internationally. Nevertheless, clinical practice varies across countries. Instruments for measuring quality have been developed to improve health care through targeted interventions. Studies on health service quality must consider the structural and cultural characteristics of countries, because each of their strengths and weaknesses differ. However, such instruments for health-related patient-reported outcomes for osteoarthritis have not yet been validated in German and Italian languages. Objectives: In order to be able to set targeted measures for the improvement of prevention and non-surgical treatment of osteoarthritis in South Tyrol, Italy, the quality of care must be recorded. Therefore, the aim of the project is to update, translate, and validate the OsteoArthritis Quality Indicator (OA-QI) questionnaire version 2, an established and validated questionnaire in Norwegian and English, for Germany and Italy. The second aim is to determine the quality of care for osteoarthritis of the hip and knee in a sample of patients who consult general practice in South Tyrol, and for comparison with patients who are admitted to rehabilitative spa-treatments for osteoarthritis in the state of Salzburg, Austria. Discussion: The results of this study will enable the identification and closure of gaps in osteoarthritis care. Although it is expected that body weight and exercise will play special roles, other areas of nonsurgical care might also be involved

GPR91 exacerbates rheumatoid arthritis via extracellular succinate released by intact inflammatory macrophages

When SUCNR1/GPR91 expressing macrophages are activated by inflammatory signals they change their metabolism and accumulate succinate. Here we show that during this activation macrophages release succinate into the extracellular milieu. They simultaneously upregulate GPR91 which functions as an autocrine and paracrine sensor for extracellular succinate to enhance IL-1 production. GPR91 deficient mice lack this metabolic sensor and show reduced macrophage activation and production of IL-1 during antigen-induced arthritis (AIA). Succinate is abundant in synovial fluids from rheumatoid arthritis (RA) patients, and these fluids elicit IL-1 release from macrophages in a GPR91-dependent manner. Together, we reveal a GPR91/succinate-dependent feed-forward loop of macrophage activation and propose GPR91 antagonists as novel therapeutic principles to treat RA
