Apoptotic gene expression in neuropathic pain
Pain initiated or caused by a primary lesion or dysfunction in the nervous system is defined as neuropathic pain. It results from direct injury to nerves in the peripheral or central nervous system and is associated with several clinical symptoms. Neuropathic pain treatment is extremely difficult, as it is a very complex disease, involving several molecular pathways. Excitatory or inhibitory pathways controlling neuropathic pain development show altered gene expression, caused by peripheral nerve injury.
This study used several experimental pain models to demonstrate the occurrence of programmed cell death in the centers controlling pain induction and maintenance, such as spinal cord and pre-frontal cortex. We combined behavioural, molecular and morphological approaches to assess the involvement of bcl-2 gene family and caspases in neuropathic pain. Chronic constriction injury (CCI) and spared nerve injury (SNI) of rodent sciatic nerve induced the appearance of pain-like behaviours, such as hyperalgesia and allodynia. An early (2-3 days post-CCI) apoptosis appeared in the spinal cord neurons as the pro-apoptotic bax gene increased (320±19%). The incidence of apoptosis appeared to be limited to the first few days following nerve injury. Subsequently, increased expression of anti-apoptotic bcl-2 family genes may inhibit further neuron loss. SNI triggered apoptotic pathway and caspases activation in pre-frontal cortex 7, 14, and 21 days post-injury. Among the time-points analyzed, RT-PCR analysis showed increased expression of the bax/bcl-2 ratio (40±2%), bid (16±2%), caspase-1 (84±3%), caspase-8 (53±6%), caspase-9 (25±6%), caspase-12 (58±2%), TNF (32±2%) genes in the cortex by 7 days post-injury. Western blot analysis showed increased active Caspase-3 protein levels in the cortex at 3, 7, 14, and 21 post-surgery. This study shows that apoptotic genes could be an useful pharmacological target in neuropathic pain controlling.

STOP: A gamified approach to support obese patients in changing their health habits
Obesity is a challenge for the whole world and it is necessary to find new ways to reduce it and to help people in this situation by facilitating the acquisition of healthy habits to replace unhealthy ones. STOP is a project aimed at facing the challenge of obesity through a digital methodology and through a synergy between experts from the industry and academia. By tracing patients' habits, integrating these data with other data already present in databases and providing personalized paths and feedback the «STop Obesity Platform» can be a valuable help for both patients and Healthcare Professionals. All this is inserted in a gamification frame by the creation of an app that establishes an analogy to the wellknown Dorian Gray mirror with the aim of encouraging the performance of the user or the acquisition of healthy behavior through a stimulating and engaging experience. This paper shows the main objectives of the project, defines the general structure of the app and provides some examples of prototype application
Nuove tecnologie per garantire il benessere fisico: strategie didattiche e motivazionali
The introductory part of this paper will discuss the importance of the use of new technologies, analysing their typologies and benefits, in the context of prevention and treatment of chronic and degenerative diseases, with the aim of guaranteeing physical well-being and the consequent improvement of the quality of life in modern society. In the second section, the contribution, using a narrative theoretical method, will deal with the methodologies applied in the pursuit of the aforementioned aim. In the final section, the benefits found in the population will be highlighted, underlining how the linkbetween new technologies and physical well-being will make them inextricably complementary in the future.La parte introduttiva di questo contributo discuterà l’importanza dell’utilizzo delle nuove tecnologie, analizzandone le tipologie e i benefici, nell’ambito della prevenzione e cura delle malattie croniche e degenerative, con l’obiettivo di garantire il benessere fisico e il conseguente miglioramento della la qualità della vita nella società moderna. Nella seconda sezione, il contributo,utilizzando un metodo teorico narrativo, tratterà le metodologie applicate nel perseguimento del suddetto obiettivo. Nella sezione finale verranno evidenziati i benefici riscontrati nella popolazione, sottolineando come il legame tra le nuove tecnologie e il benessere fisico le renderà indissolubilmente complementari in futuro

Management of gastro-esophageal reflux disease: Practice-oriented answers to clinical questions
: Gastro-esophageal reflux disease (GERD) is a condition which is frequently faced by primary care physicians and gastroenterologists. Improving management of GERD is crucial to maximise both patient care and resource utilization. In fact, the management of patients with GERD is complex and poses several questions to the clinician who faces them in clinical practice. For instance, many aspects should be considered, including the appropriateness of indication to endoscopy, the quality of the endoscopic examination, the use and interpretation of ambulatory reflux testing, and the choice and management of anti-reflux treatments, i.e., proton-pump inhibitors and surgery. Aim of the present review was to provide a comprehensive update on the clinical management of patients with GERD, through a literature review on the diagnosis and management of patients with GER symptoms. In details, we provide practice-oriented concise answers to clinical questions, with the aim of optimising patient management and healthcare resource use
Digital Project management in a multicultural environment: the experience at trasnperfect
The following report is carried out as the final work of the master’s degree in Intercultural Studies for Business at the Porto Accounting and Business School (ISCAP), part of the Polytechnic Institute of Porto. It aims to present a detailed reflection and analysis of my 6-month curricular internship which took place in the Netherlands between January 11th and June 30th 2021 at TransPerfect, an international family of companies offering a wide variety of language services and technology solutions to help global businesses operate worldwide marketplace. As pillars of this company, intercultural communication, and cross-cultural management, as well as their relevance and impact in today's business world, are outlined. The report’s focus will then shift to an overview of the language service industry and an elaborate characterization of the host company. From this perspective, I analysed my role as an intern in a digital project management team that allowed me to confirm the theoretical framework of my research: intercultural competence and an efficient cross-cultural management are the key for the success of an international business, especially in the language service industry.O relatĂłrio seguinte Ă© realizado como trabalho final do Mestrado em Estudos Interculturais para NegĂłcios no Instituto Superior de Contabilidade e NegĂłcios do Porto (ISCAP), parte do Instituto PolitĂ©cnico do Porto. Putting manipulation, grasping and sensing in educational APPs for children
La tesi ha come obiettivo di valorizzare pratiche psico-pedagogiche classiche utilizzando le nuove opportunità fornite dalla tecnologia e dagli strumenti digitali sempre più ubiquitari nella nostra società . La modalità individuata per connettere l’ambito digitale e quello fisico è attraverso lo sviluppo della multisensorialità e dunque la riscoperta dei sensi come l’olfatto, il gusto e il tatto nelle pratiche educative per bambini dai 6 agli 8 anni (primi tre anni della scuola primaria).
Per ottenere questo obiettivo si sono sviluppate metodologie e prototipi (sia hardware, che software – APP) fruibili dai bambini della scuola primaria.
In primo luogo è stato ideato, concettualizzato e sviluppato il modello dell’Hyper Activity Book (HAB), che rappresenta la porzione modellistica fondamentale a cui ispirarsi per l’implementazione di prototipi in un settore così delicato quanto cruciale, dove troppo spesso le applicazioni si sono fermate a miglioramenti puntuali e non sistemici. Il modello dell’Hyper Activity Book ha l’ambizione di formare uno schema entro cui poter applicare in maniera organica i principi del Learning by Doing in stretta comunicazione con il concetto degli ambienti didattici ibridi e delle interfacce-utente di stampo tangibile e dei moduli di Intelligenza Artificiale. A corredo del modello dell’HAB e sulla base dell’analisi dello stato dell’arte condotto, sono state prodotte delle linee guida per la progettazione di Ambienti Ibridi.
La ricerca ha permesso lo sviluppo di due hardware, che hanno rappresentato un’evoluzione e un miglioramento incrementale rispetto alle precedenti realizzazioni.
Si è sviluppato un algoritmo per il riconoscimento di oggetti fisici su schermi touchscreen grazie al loro posizionamento sugli stessi. Il prototipo, denominato TriPOD, contiene l’algoritmo che è in grado di riconoscere gli oggetti fisici sulla base della disposizione delle protrusioni capacitive (i pin capacitivi) in maniera univoca, collegando l’identificativo generato con un oggetto tangibile. Questo prototipo è funzionante e sebbene abbia necessità di divenire ancora più robusto in termini tecnici, è uno sviluppo interessante per le applicazioni nel settore dell’educazione e della riabilitazione, in grado di aprire nuovi scenari applicativi.
L’altro prototipo hardware, sviluppato nel percorso dottorale è rappresentato dall’Activity Board 1.0. La tavoletta attiva che sfrutta la tecnologia RFID è il miglioramento di precedenti versioni, attraverso un complesso e lungo lavoro di riaggiornamenti e miglioramenti incrementali. La nuova versione ha la possibilità di connettersi via Wi-Fi e contiene tutti i materiali al suo interno, fornendo una versione “portatile” e immediatamente fruibile in ogni contesto. La tavoletta ha raggiunto un ottimo grado di robustezza, tanto da permettere sperimentazioni senza bug di sistema e rimuovendo la maggior parte di problematiche tecniche. Un risultato è stato anche quello di ridurre drasticamente il costo del sistema, attraverso un’attenta analisi delle componentistiche sul mercato, con l’obiettivo di mantenere gli standard progettuali definiti dal modello HAB e i requisiti funzionali.
Ulteriore risultato del progetto è stato lo sviluppo di APP educative che sfruttano i principi della multisensorialità . Con queste APP educative si intende estendere il campo rispetto alla manualità che è stata centrale nei progetti precedenti (vedi BlockMagic). L’estensione nei confronti dell’udito, del tatto, del gusto e dell’olfatto hanno aperto nuovi scenari di opportunità per tutto il gruppo di ricerca, proponendo delle modalità di interazione ancora più aderenti alle pratiche psico-pedagogiche a cui si ispire. Le APP educative si sono centrate sul metodo ibrido, di coniugazione tra mondo fisico e reale in campo didattico, creando applicazioni per l’educazione musicale, per l’educazione olfattiva e della conoscenza del mondo, per l’immersione nei processi di storytelling, per l’apprendimento dell’anatomia, per l’apprendimento delle lingue, per l’apprendimento dell’ortografia e per l’apprendimento dei rudimenti della matematica. In questa tesi sono stati presentati solo alcune delle APP educative sviluppate in questi tre anni.
A valle del processo è stata svolta una sperimentazione per verificare l’efficacia e l’accettabilità degli strumenti nei confronti di due modelli già utilizzati nelle classi e negli ambiti informati ossia lo strumento cartaceo e il tablet.
Per quanto riguarda l’accettabilità , gli strumenti multisensoriali sono emersi come i preferiti per il gruppo di partecipanti (121 bambini appartenenti a due scuole).
Per quanto riguarda l’efficacia, si è effettuato uno studio sulla memorizzazione di storie. I dati mostrano come le storie eseguite con il libro e con i materiali multisensoriali permettono un buon grado di memorizzazione mentre con il tablet il ricordo degli avvenimenti scende radicalmente
Management of Barrett’s Esophagus: Practice-Oriented Answers to Clinical Questions
Barrett's esophagus is the most important complication of gastro-esophageal reflux disease and the only known precursor of esophageal adenocarcinoma. The diagnosis and treatment of Barrett's esophagus are clinically challenging as it requires a high level of knowledge and competence in upper gastrointestinal endoscopy. For instance, endoscopists should know when and how to perform biopsies when Barrett's esophagus is suspected. Furthermore, the correct identification and treatment of dysplastic Barrett's esophagus is crucial to prevent progression to cancer as well as it is the endoscopic surveillance of treated patients. Herein, we report practice-oriented answers to clinical questions that clinicians should be aware of when approaching patients with Barrett's esophagus
Leakage Current Mechanisms in SiGe HBTs Fabricated Using Selective and Nonselective Epitaxy
SiGe heterojunction bipolar transistors (HTBs) have been fabricated using selective epitaxy for the Si collector, followed in the same growth step by nonselective epitaxy for the p+ SiGe base and n-Si emitter cap. DC electrical characteristics are compared with cross-section TEM images to identify the mechanisms and origins of leakage currents associated with the epitaxy in two different types of transistor . In the first type, the polysilicon emitter is smaller than the collector active area, so that the extrinsic base implant penetrates into the single-crystal Si and SiGe around the perimeter of the emitter and the polycrystalline Si and SiGe exrtrinsic base. In these transistors, the Bummel plots are near-ideal and there is no evidence of emitter/collector leakage. In the second type, the collector active area is smaller than the polysilicon emitter, so the extrinsic base implant only penetrates into the polysilicon extrinsic base. In these transistors, the leakage currents observed depend on the base doping level. In transistors with a low doped base, emitter/collector and emitter/base leakage is observed, whereas in transistors with a high doped base only emitter/base leakage is observed. The emitter/collector leakage is explained by punch through o fhte base caused by thinning of the SiGe base at the emitter perimeter. The emitter/base leakeage is shown to be due to Poole-Frenkel mechanism and is explained by penetration of the emitter/base depletion region into the p+ polysilicon extrinsic base at the emitter periphery. Variable collector/base reverse leakage currents are observed and a variety of mechanisms are observed, including Shockley-Read-Hall recombination, trap assisted tunneling, Poole Frenkel and band to band tunneling. These result s are explained by the presence of polysilicon grains on the sidewalls of the field oxide at the collector perimeter
