    From the stable to the exotic: clustering in light nuclei

    A great deal of research work has been undertaken in alpha-clustering study since the pioneering discovery of 12C+12C molecular resonances half a century ago. Our knowledge on physics of nuclear molecules has increased considerably and nuclear clustering remains one of the most fruitful domains of nuclear physics, facing some of the greatest challenges and opportunities in the years ahead. The occurrence of "exotic" shapes in light N=Z alpha-like nuclei is investigated. Various approaches of the superdeformed and hyperdeformed bands associated with quasimolecular resonant structures are presented.     4He decay of excited states in 14C

    A study of the 7Li(9Be,4He 10Be)2H reaction at E{beam}=70 MeV has been performed using resonant particle spectroscopy techniques and provides the first measurements of alpha-decaying states in 14C. Excited states are observed at 14.7, 15.5, 16.4, 18.5, 19.8, 20.6, 21.4, 22.4 and 24.0 MeV. The experimental technique was able to resolve decays to the various particle bound states in 10Be, and provides evidence for the preferential decay of the high energy excited states into states in 10Be at ~6 MeV. The decay processes are used to indicate the possible cluster structure of the 14C excited states.Comment: accepted for publication in PR

    Streamlined design of a self-inactivating feline immunodeficiency virus vector for transducing ex vivo dendritic cells and T lymphocytes.

    BACKGROUND: Safe and efficient vector systems for delivering antigens or immunomodulatory molecules to dendritic cells (DCs), T lymphocytes or both are considered effective means of eliciting adaptive immune responses and modulating their type, extent, and duration. As a possible tool toward this end, we have developed a self-inactivating vector derived from feline immunodeficiency virus (FIV) showing performance characteristics similar to human immunodeficiency virus-derived vectors but devoid of the safety concerns these vectors have raised. METHODS: The pseudotyped FIV particles were generated with a three-plasmid system consisting of: the packaging construct, providing Gag, Pol and the accessory proteins; the vector(s), basically containing FIV packaging signal (psi), Rev responsive element, R-U5 region at both ends, and the green fluorescent protein as reporter gene; and the Env plasmid, encoding the G protein of vesicular stomatitis virus (VSV-G) or the chimeric RD114 protein. Both packaging and vector constructs were derived from p34TF10, a replication competent molecular clone of FIV. The pseudotyped particles were produced by transient transfection in the Crandell feline fibroblast kidney (CrFK) or the human epithelial (293T) cell line. RESULTS: To broaden its species tropism, the final vector construct was achieved through a series of intermediate constructs bearing a longer psi, the FIV central polypurin tract sequence (cPPT), or the woodchuck hepatitis post-regulatory element (WPRE). These constructs were compared for efficiency and duration of transduction in CrFK or 293T cells and in the murine fibroblast cell line NIH-3T3. Whereas psi elongation and cPPT addition did not bring any obvious benefit, insertion of WPRE downstream GFP greatly improved vector performances. To maximize the efficiency of transduction for ex-vivo murine DCs and T-lymphocytes, this construct was tested with VSV-G or RD114 and using different transduction protocols. The results indicated that the FIV construct derived herein stably transduced both cell types, provided that appropriate vector makeup and transduction protocol were used. Further, transduced DCs underwent changes suggestive of an induced maturation. CONCLUSION: In contrast to previously described FIV vectors that were poorly efficient in delivering genetic material to DCs and T lymphocytes, the vector developed herein has potential for use in experimental immunization strategies

    High-frequency monitoring of nitrogen and phosphorus response in three rural catchments to the end of the 2011–2012 drought in England

    This paper uses high-frequency bankside measurements from three catchments selected as part of the UK government-funded Demonstration Test Catchments (DTC) project. We compare the hydrological and hydrochemical patterns during the water year 2011–2012 from the Wylye tributary of the River Avon with mixed land use, the Blackwater tributary of the River Wensum with arable land use and the Newby Beck tributary of the River Eden with grassland land use. The beginning of the hydrological year was unusually dry and all three catchments were in states of drought. A sudden change to a wet summer occurred in April 2012 when a heavy rainfall event affected all three catchments. The year-long time series and the individual storm responses captured by in situ nutrient measurements of nitrate and phosphorus (total phosphorus and total reactive phosphorus) concentrations at each site reveal different pollutant sources and pathways operating in each catchment. Large storm-induced nutrient transfers of nitrogen and or phosphorus to each stream were recorded at all three sites during the late April rainfall event. Hysteresis loops suggested transport-limited delivery of nitrate in the Blackwater and of total phosphorus in the Wylye and Newby Beck, which was thought to be exacerbated by the dry antecedent conditions prior to the storm. The high rate of nutrient transport in each system highlights the scale of the challenges faced by environmental managers when designing mitigation measures to reduce the flux of nutrients to rivers from diffuse agricultural sources. It also highlights the scale of the challenge in adapting to future extreme weather events under a changing climate

    No evidence of an 11.16 MeV 2+ state in 12C

    An experiment using the 11B(3He,d)12C reaction was performed at iThemba LABS at an incident energy of 44 MeV and analyzed with a high energy-resolution magnetic spectrometer, to re-investigate states in 12C published in 1971. The original investigation reported the existence of an 11.16 MeV state in 12C that displays a 2+ nature. In the present experiment data were acquired at laboratory angles of 25-, 30- and 35- degrees, to be as close to the c.m. angles of the original measurements where the clearest signature of such a state was observed. These new low background measurements revealed no evidence of the previously reported state at 11.16 MeV in 12C

    Intrinsic dielectric and spectroscopic behavior of perovskite Ba(Ni1/3Nb2/3)O3–Ba(Zn1/3Nb2/3)O3 microwave dielectric ceramics

    [[abstract]]Ceramics of 0.35Ba(Ni1/3Nb2/3)O3–0.65Ba(Zn1/3Nb2/3)O3 were prepared by the mixed oxide route. The effect of the cooling rate (2 °C–240 °C/h) after sintering on the microwave dielectric properties of the ceramics was examined. While the extrinsic factors, such as porosity and secondary phases, markedly influence the dielectric properties in the low-frequency regime, they have minimal effect on these properties in the high-frequency regime. The mechanisms involved in modifying the high-frequency dielectric properties of the materials were investigated by Fourier transform infrared and Raman spectroscopy, in conjunction with the Rietveld analysis of x-ray diffraction (XRD) spectra. A reduction in the cooling rate after sintering results in an increase in the high-frequency Q×f (product of dielectric Q value and measurement frequency) from 42 to 58 THz in the high-frequency regime (∌1.5 THz). Such behavior correlates very well with the increase in the B-site occupancy by Nb (deduced from the Rietveld analyses of XRD spectra) and the increase in the coherency of the lattice vibration (deduced from the reduction in the full-width-at-half-maximum of the A1g(O) Raman mode). In contrast, the cooling rate after sintering has very limited effect on the relative permittivity (varying from 40.8 to 41.9 at 1.5 THz), which is in accord with the phenomenon that the cell volume and the Raman shift of A1g(O) Raman mode are essentially independent of the cooling rate.[[booktype]]箙

    Development of Feline Immunodeficiency Virus ORF-A (tat) Mutants: In Vitro and in Vivo Characterization

    AbstractA functional ORF-A is essential for efficient feline immunodeficiency virus replication in lymphocytes. We have characterized a series of mutants of the Petaluma strain, derived from p34TF10 and having different combinations of stop codons and increasingly long deletions in ORF-A. Six clones proved fully replicative in fibroblastoid Crandell feline kidney cells and monocyte-derived macrophage cultures but failed to replicate in T cell lines and primary lymphoblasts. Cats inoculated with three selected mutants had considerably milder infections than controls given intact ORF-A virus. In vivo, the mutants maintained growth properties similar to those in vitro for at least 7 months, except that replication in lymphoid cells was strongly reduced but not ablated. One mutant underwent extensive ORF-A changes without, however, reverting to wild-type. Antiviral immune responses were feeble in all cats, suggesting that viral loads were too low to represent a sufficiently powerful antigenic stimulus

    Helium Clustering in Neutron-Rich Be Isotopes

    Measurements of the helium-cluster breakup and neutron removal cross sections for neutron-rich Be isotopes A=10-12,14 are presented. These have been studied in the 30 to 42 MeV/u energy range where reaction measurements are proposed to be sensitive to the cluster content of the ground-state wave-function. These measurements provide a comprehensive survey of the decay processes of the Be isotopes by which the valence neutrons are removed revealing the underlying alpha-alpha core-cluster structure. The measurements indicate that clustering in the Be isotopes remains important up to the drip-line nucleus 14^Be and that the dominant helium-cluster structure in the neutron-rich Be isotopes corresponds to alpha-Xn-alpha.Comment: 5 pages, 2 tables and 3 figure
