    Fixation locations when grasping partly occluded objects

    When grasping an object, subjects tend to look at the contact positions of the digits (A. M. Brouwer, V. H. Franz, D. Kerzel, & K. R. Gegenfurtner, 2005; R. S. Johansson, G. Westling, A. Bäckström, & J. R. Flanagan, 2001). However, these contact positions are not always visible due to occlusion. Subjects might look at occluded parts to determine the location of the contact positions based on extrapolated information. On the other hand, subjects might avoid looking at occluded parts since no object information can be gathered there. To find out where subjects fixate when grasping occluded objects, we let them grasp flat shapes with the index finger and thumb at predefined contact positions. Either the contact position of the thumb or the finger or both was occluded. In a control condition, a part of the object that does not involve the contact positions was occluded. The results showed that subjects did look at occluded object parts, suggesting that they used extrapolated object information for grasping. Additionally, they preferred to look in the direction of the index finger. When the contact position of the index finger was occluded, this tendency was inhibited. Thus, an occluder does not prevent fixations on occluded object parts, but it does affect fixation locations especially in conditions where the preferred fixation location is occluded. © ARVO

    Study on the Effect of Levulinic Acid on Whey-Based Biosynthesis of Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) by Hydrogenophaga pseudoflava

    Background and Objective: Production of polyhydroxyalkanoate copolyesters consisting of 3-hydroxybutyrate and 3-hydroxyvalerate units was for the first time studied using the production strain Hydrogenophaga pseudoflava based on sustainable raw materials. This strategy provides for increased cost efficiency in PHA production and in enhanced material quality.Material and Methods: As a particularity, production of these poly(3-hydroxybutyrate-co-3- hydroxyvalerate) copolyesters was based on a novel substrate/co-substrate combination: whey permeate from dairy industry, on the one hand, acted as substrate for biomass and 3HB biosynthesis; on the other hand, levulinic acid, accessible from various renewable resources, was used as 3HV-related precursor compound. The experiments were carried out on shaking flask scale using defined nutrient media.Results and Conclusion: Applied during nutritionally balanced growth of H. pseudoflava, levulinicacid displays drastic growth inhibition at rather low concentrations of 0.2 g l-1 (growth inhibition constant Ki = 0.032), which suggests the careful supply of this compound in the first phase of cultivation. Under nitrogen-free cultivation conditions, inhibition of the strain´s metabolism by levulinic acid was less pronounced. Here, poly(3-hydroxybutyrate-co- 3-hydroxyvalerate) concentrations up to 4.2 g l-1 and volumetric poly(3-hydroxybutyrate-co-3- hydroxyvalerate) productivities up to 0.06 g l-1 h -1 were achieved in dependence on the precursor supply. Investigating poly(3-hydroxybutyrate-co-3-hydroxyvalerate) composition in setups supplied with differently composed whey/levulinic acid mixtures revealed 3- hydroxyvalerate fractions in the polymer between 0 and 0.6 mol mol-1 . This study successfully demonstrates the feasibility of combined utilization of different waste- and by-products from food industry and agriculture for generation of value-added 2nd generation biopolymers.     Neurodegenerative disease biomarkers Aβ1–40, Aβ1–42, tau, and p-tau181 in the vervet monkey cerebrospinal fluid: Relation to normal aging, genetic influences, and cerebral amyloid angiopathy

    Background:The Caribbean vervet monkey (Chlorocebus aethiops sabaeus) is a potentially valuable animal model of neurodegenerative disease. However, the trajectory of aging in vervets and its relationship to human disease is incompletely understood. Methods:To characterize biomarkers associated with neurodegeneration, we measured cerebrospinal fluid (CSF) concentrations of Aβ1-40, Aβ1-42, total tau, and p-tau181 in 329 members of a multigenerational pedigree. Linkage and genome-wide association were used to elucidate a genetic contribution to these traits. Results:Aβ1-40 concentrations were significantly correlated with age, brain total surface area, and gray matter thickness. Levels of p-tau181 were associated with cerebral volume and brain total surface area. Among the measured analytes, only CSF Aβ1-40 was heritable. No significant linkage (LOD > 3.3) was found, though suggestive linkage was highlighted on chromosomes 4 and 12. Genome-wide association identified a suggestive locus near the chromosome 4 linkage peak. Conclusions:Overall, these results support the vervet as a non-human primate model of amyloid-related neurodegeneration, such as Alzheimer's disease and cerebral amyloid angiopathy, and highlight Aβ1-40 and p-tau181 as potentially valuable biomarkers of these processes

    High resilience of carbon transport in long-term drought-stressed mature Norway spruce trees within 2 weeks after drought release

    Under ongoing global climate change, drought periods are predicted to increase in frequency and intensity in the future. Under these circumstances, it is crucial for tree\u27s survival to recover their restricted functionalities quickly after drought release. To elucidate the recovery of carbon (C) transport rates in c. 70-year-old Norway spruce (Picea abies [L.] KARST.) after 5 years of recurrent summer droughts, we conducted a continuous whole-tree 13^{13}C labeling experiment in parallel with watering. We determined the arrival time of current photoassimilates in major C sinks by tracing the 13^{13}C label in stem and soil CO2_{2} efflux, and tips of living fine roots. In the first week after watering, aboveground C transport rates (CTR) from crown to trunk base were still 50% lower in previously drought-stressed trees (0.16 ± 0.01 m h1^{-1}) compared to controls (0.30 ± 0.06 m h1^{-1}). Conversely, CTR below ground, that is, from the trunk base to soil CO2_{2} efflux were already similar between treatments (c. 0.03 m h1^{-1}). Two weeks after watering, aboveground C transport of previously drought-stressed trees recovered to the level of the controls. Furthermore, regrowth of water-absorbing fine roots upon watering was supported by faster incorporation of 13^{13}C label in previously drought-stressed (within 12 ± 10 h upon arrival at trunk base) compared to control trees (73 ± 10 h). Thus, the whole-tree C transport system from the crown to soil CO2_{2} efflux fully recovered within 2 weeks after drought release, and hence showed high resilience to recurrent summer droughts in mature Norway spruce forests. This high resilience of the C transport system is an important prerequisite for the recovery of other tree functionalities and productivity

    Dynamics of initial carbon allocation after drought release in mature Norway spruce—Increased belowground allocation of current photoassimilates covers only half of the carbon used for fine‐root growth

    After drought events, tree recovery depends on sufficient carbon (C) allocation to the sink organs. The present study aimed to elucidate dynamics of tree-level C sink activity and allocation of recent photoassimilates (Cnew_{new}) and stored C in c. 70-year-old Norway spruce (Picea abies) trees during a 4-week period after drought release. We conducted a continuous, whole-tree 13^{13}C labeling in parallel with controlled watering after 5 years of experimental summer drought. The fate of Cnew_{new} to growth and CO2_{2} efflux was tracked along branches, stems, coarse- and fine roots, ectomycorrhizae and root exudates to soil CO2_{2} efflux after drought release. Compared with control trees, drought recovering trees showed an overall 6% lower C sink activity and 19% less allocation of Cnew_{new} to aboveground sinks, indicating a low priority for aboveground sinks during recovery. In contrast, fine-root growth in recovering trees was seven times greater than that of controls. However, only half of the C used for new fine-root growth was comprised of Cnew_{new} while the other half was supplied by stored C. For drought recovery of mature spruce trees, in addition to Cnew_{new}, stored C appears to be critical for the regeneration of the fine-root system and the associated water uptake capacity

    Grasping isoluminant stimuli

    We used a virtual reality setup to let participants grasp discs, which differed in luminance, chromaticity and size. Current theories on perception and action propose a division of labor in the brain into a color proficient perception pathway and a less color-capable action pathway. In this study, we addressed the question whether isoluminant stimuli, which provide only a chromatic but no luminance contrast for action planning, are harder to grasp than stimuli providing luminance contrast or both kinds of contrast. Although we found that grasps of isoluminant stimuli had a slightly steeper slope relating the maximum grip aperture to disc size, all other measures of grip quality were unaffected. Overall, our results do not support the view that isoluminance of stimulus and background impedes the planning of a grasping movement
