Prediction of Renal Acid Load in Adult Patients on Parenteral Nutrition
Metabolic acidosis and metabolic bone disease are frequent complications in patients on
parenteral nutrition (PN). A common contributor to these complications could be a daily high renal
acid load. This study aims to find a method for predicting the potential total acid load (PTAL) and
the pH of the compounded parenteral nutrition mixtures. The pH and titratable acidity (TA) of fifty
compounded mixtures were measured. The potential metabolic acid load (PMAL) was calculated
by considering the amount of nutrients that are acid producers and consumers. The PTAL of the
TPN mixtures was calculated by adding TA to PMAL. Multiple linear regression analyses were used
to develop a predictive model for the TA and pH of the compounded mixtures. The predicted TA
and pH values of the analyzed mixtures agreed with those measured (Passing-Bablok analysis).
The PTAL was >50 mmol/day for 82% of the mixtures, >75 mmol/day for 40% of the mixtures, and
>100 mmol/day for 22% of the mixtures. The prediction of the renal acid load in patients on long-term
PN could allow more appropriate acid-base balancing. Moreover, predicting the pH of such mixtures
could be useful to pharmacists to assess the stability and compatibility of the components in the
compounded mixtures
Liposomes loaded with everolimus and coated with hyaluronic acid: A promising approach for lung fibrosis
Chronic lung allograft dysfunction (CLAD) and interstitial lung disease associated with collagen tissue diseases (CTD-ILD) are two end-stage lung disorders in which different chronic triggers induce activation of myo-/fibroblasts (LFs). Everolimus, an mTOR inhibitor, can be adopted as a potential strategy for CLAD and CTD-ILD, however it exerts important side effects. This study aims to exploit nanomedicine to reduce everolimus side effects encapsulating it inside liposomes targeted against LFs, expressing a high rate of CD44. PEGylated liposomes were modified with high molecular weight hyaluronic acid and loaded with everolimus (PEG-LIP(ev)-HA400kDa). Liposomes were tested by in vitro experiments using LFs derived from broncholveolar lavage (BAL) of patients affected by CLAD and CTD-ILD, and on alveolar macrophages (AM) and lymphocytes isolated, respectively, from BAL and peripheral blood. PEG-LIP-HA400kDa demonstrated to be specific for LFs, but not for CD44-negative cells, and after loading everolimus, PEG-LIP(ev)-HA400kDa were able to arrest cell cycle arrest and to decrease phospho-mTOR level. PEG-LIP(ev)-HA400kDa showed anti-inflammatory effect on immune cells. This study opens the possibility to use everolimus in lung fibrotic diseases, demonstrating that our lipids-based vehicles can vehicle everolimus inside cells exerting the same drug molecular effect, not only in LFs, but also in immune cells
d13C values in archaeological 14C-AMS dated charcoals: assessing mid-Holocene climate fluctuations and human response from a high-resolution isotope record (Arslantepe, Turkey)
RATIONALE: Past climate has always influenced human adaptation to the environment. In order to reconstruct palaeoclimate fluctuations and their role in the evolution of Near Eastern societies during the mid-Holocene, high-resolution Δ13C records from fossil wood remains at the archaeological site of Arslantepe (eastern Turkey) have been developed. METHODS: After chemical treatment, 13C values were measured by sample combustion flow using a FLASH EA-CHNS instrument interfaced with a Delta V isotope ratio mass spectrometer via a CONFLO III. Two replicates per sample were analysed. The measurement precision was evaluated by propagating variations of the 13C values of samples and V-PDB standards, whereas the accuracy was checked by a quality control sample. To account for changes in atmospheric CO2, Δ13C values were calculated. In addition, 14C/12C ratios were measured by means of ann AMS system (3 MV tandem accelerator). RESULTS: Mean Δ13C curves of deciduous Quercus and Juniperus from archaeological levels between 4700 and 2000 BC (Arslantepe periods VIII-VI D) were produced, where the isotope values were ordered by the available RC ages. Interspecific variations of evergreen vs deciduous plants were postulated for the juniper Δ13C values being higher than 3‰. The seasonal rainfall amount was recorded by the juniper remains, while the water table levels were obtained from the oak samples. CONCLUSIONS: The local climate experienced times of enhanced/reduced precipitation in concert with regional trends. Anomalies in the air mass circulation from the Mediterranean basin also produced oscillations of rainfall amount. In such a frame the Rapid Climate Change dry events had a consistent signature in the Arslantepe Δ13C record, thus potentially contributing to social or organisational changes at the site
Using Y-chromosome capture enrichment to resolve haplogroup H2 shows new evidence for a two-Path Neolithic expansion to Western Europe
Uniparentally-inherited markers on mitochondrial DNA (mtDNA) and the non-recombining regions of the Y chromosome (NRY), have been used for the past 30 years to investigate the history of humans from a maternal and paternal perspective.Researchers have preferred mtDNA due to its abundance in the cells, and comparatively high substitution rate. Conversely, the NRY is less susceptible to back mutations and saturation, and is potentially more informative than mtDNA owing to its longer sequence length. However, due to comparatively poor NRY coverage via shotgun sequencing, and the relatively low and biased representation of Y-chromosome variants on capture arrays such as the 1240K, ancient DNA studies often fail to utilize the unique perspective that the NRY can yield. Here we introduce a new DNA enrichment assay, coined YMCA (Y-mappable capture assay), that targets the “mappable” regions of the NRY. We show that compared to low-coverage shotgun sequencing and 1240K capture, YMCA significantly improves the coverage and number of sites hit on the NRY, increasing the number of Y-haplogroup informative SNPs, and allowing for the identification of previously undiscovered variants. To illustrate the power of YMCA, we show that the analysis of ancient Y-chromosome lineages can help to resolve Y-chromosomal haplogroups. As a case study, we focus on H2, a haplogroup associated with a critical event in European human history: the Neolithic transition. By disentangling the evolutionary history of this haplogroup, we further elucidate the two separate paths by which early farmers expanded from Anatolia and the Near East to western Europe.Competing Interest StatementThe authors have declared no competing interest.Introduction Results and Discussion - Validating the performance of YMCA - Application of YMCA to YHG H2 as a case study - Identifying diagnostic SNPs for improved YHG H2 resolution Discussion Materials and Methods - Data - Contamination quality filtering - Method of Y Haplogroup Assignment - Comparing the Performance of our Y-capture Array Phylogenetic Tree Reconstructio
Bullet impacts and built heritage damage 1640–1939
© 2018, The Author(s). Conflict damage to heritage has been thrust into the global spotlight during recent conflict in the Middle East. While the use of social media has heightened and enhanced public awareness of this ‘cultural terrorism’, the occurrence of this type of vandalism is not new. In fact, as this study demonstrates, evidence of the active targeting of sites, as well as collateral damage when heritage is caught in crossfire, is widely visible around Europe and further afield. Using a variety of case studies ranging from the 1640s to the 1930s, we illustrate and quantify the changing impact of ballistics on heritage buildings as weaponry and ammunition have increased in both energy and energy density potential. In the first instance, this study highlights the increasing threats to heritage in conflict areas. Second, it argues for the pressing need to quantify and map damage to the stonework in order to respond to these challenges
Phenolic and furanic compounds of Portuguese chestnut and French, American and Portuguese oak wood chips
Botanical species used on aging process must
be wisely and judiciously chosen, and for this selection, a
basic knowledge of the chemical composition of woods is
warranted. Aiming to contribute to extend the knowledge
of the chemical composition of several wood species useful
for enological purposes, we have focused our studies on
Portuguese chestnut and French, American and Portuguese
oak chips. The profile of low molecular weight phenolic
composition of these chips was achieved, using an optimized
extraction method based on pressurized liquid
extraction, followed by the quantification of phenolic acids,
phenolic aldehydes and furanic derivatives by high-performance
liquid chromatography (HPLC-DAD). The
identification of those compounds was also confirmed by
LC-DAD/ESI-MS. This study allowed the determination of
the low molecular phenolic composition of Portuguese
chestnut and French, American and Portuguese oak wood.
According to our results, the influence of the botanical
species seems to be more relevant than the geographic
origin of the wood species
