    Detection and mapping of illicit drugs and their metabolites in fingermarks by MALDI MS and compatibility with forensic techniques

    Despite the proven capabilities of Matrix Assisted Laser Desorption Ionisation Mass Spectrometry (MALDI MS) in laboratory settings, research is still needed to integrate this technique into current forensic fingerprinting practice. Optimised protocols enabling the compatible application of MALDI to developed fingermarks will allow additional intelligence to be gathered around a suspect’s lifestyle and activities prior to the deposition of their fingermarks while committing a crime. The detection and mapping of illicit drugs and metabolites in latent fingermarks would provide intelligence that is beneficial for both police investigations and court cases. This study investigated MALDI MS detection and mapping capabilities for a large range of drugs of abuse and their metabolites in fingermarks; the detection and mapping of a mixture of these drugs in marks, with and without prior development with cyanoacrylate fuming or Vacuum Metal Deposition, was also examined. Our findings indicate the versatility of MALDI technology and its ability to retrieve chemical intelligence either by detecting the compounds investigated or by using their ion signals to reconstruct 2D maps of fingermark ridge details

    Developing Legacy System Migration Methods and Tools for Technology Transfer

    This paper presents the research results of an ongoing technology transfer project carried out in coopera- tion between the University of Salerno and a small software company. The project is aimed at developing and transferring migration technology to the industrial partner. The partner should be enabled to migrate monolithic multi-user COBOL legacy systems to a multi-tier Web-based architecture. The assessment of the legacy systems of the partner company revealed that these systems had a very low level of decompos- ability with spaghetti-like code and embedded control flow and database accesses within the user interface descriptions. For this reason, it was decided to adopt an incremental migration strategy based on the reengineering of the user interface using Web technology, on the transformation of interactive legacy programs into batch programs, and the wrapping of the legacy programs. A middleware framework links the new Web-based user interface with the Wrapped Legacy System. An Eclipse plug-in, named MELIS (migration environment for legacy information systems), was also developed to support the migration process. Both the migration strategy and the tool have been applied to two essential subsystems of the most business critical legacy system of the partner company

    Identifying Cloned Navigational Patterns in Web Applications

    Web Applications are subject to continuous and rapid evolution. Often programmers indiscriminately duplicate Web pages without considering systematic development and maintenance methods. This practice creates code clones that make Web Applications hard to maintain and reuse. We present an approach to identify duplicated functionalities in Web Applications through cloned navigational pattern analysis. Cloned patterns can be generalized in a reengineering process, thus to simplify the structure and future maintenance of the Web Applications. The proposed method first identifies pairs of cloned pages by analyzing similarity at structure, content, and scripting code. Two pages are considered clones if their similarity is greater than a given threshold. Cloned pages are then grouped into clusters and the links connecting pages of two clusters are grouped too. An interconnection metric has been defined on the links between two clusters to express the effort required to reengineer them as well as to select the patterns of interest. To further reduce the comprehension effort, we filter out links and nodes of the clustered navigational schema that do not contribute to the identification of cloned navigational patterns. A tool supporting the proposed approach has been developed and validated in a case study

    Cooking influence on physico-chemical fruit characteristics of eggplant (Solanum melongena L.)

    Physico-chemical traits of three eggplant genotypes ("Tunisina", "Buia" and "L 305") were evaluated before and after two cooking treatments (grilling and boiling). Different genotypes revealed different changes after cooking, with "Tunisina" showing a better retention of phytochemicals with respect to other two genotypes. The main physical phenomena were water loss during grilling, and dry matter loss after boiling. Chlorogenic acid, the main phenolic in eggplant, resulted higher in grilled samples, while delphinidin glycosides resulted more retained in boiled samples. Glycoalkaloids, thiols and biogenic amines were generally stable, while 5-hydroxy-methyl-furfural was found only in grilled samples. Interestingly, Folin-Ciocalteu index and free radical scavenging capacity, measured with three different assays, were generally increased after cooking, with a greater formation of antioxidant substances in grilled samples. NMR relaxation experiments clarified the hypothesis about the changes of eggplant compounds in terms of decomposition of larger molecules and production of small ones after cooking

    Colostrum from cows immunized with a veterinary vaccine against bovine rotavirus displays enhanced in vitro anti-human rotavirus activity

    Human rotaviruses represent a major cause of severe diarrheal disease in infants and young children. The limited impact of oral vaccines on global estimates of rotavirus mortality and the suboptimal use of oral rehydration justify the need for alternative prophylactic and therapeutic strategies, especially for immunocompromised hosts. The protective effects of colostrum\u2014the first milk produced during the initial 24 to 48 h after parturition\u2014are well documented in the literature. In particular, the ingestion of hyperimmune bovine colostrum has been proposed as an alternative preventive approach against human rotavirus gastroenteritis. Although the immunization of pregnant cows with human rotavirus boosts the release of specific immunoglobulin G in bovine colostrum, it raises regulatory and safety issues. In this study, we demonstrated that the conventional bovine rotavirus vaccine is sufficient to enhance the anti-human rotavirus protective efficacy of bovine colostrum, thus providing a conservative approach to produce hyperimmune bovine colostrum, making it exploitable as a functional food

    Application of MALDI MS Imaging after sequential processing of latent fingermarks

    Latent fingermarks are routinely visualised by subjecting them to one or more CSI/crime lab processes to maximise the recovery of ridge flow and minutiae permitting an identification. In the last decade mass spectrometric imaging (MSI) techniques have been applied to fingermarks to provide information about a suspect and/or on the circumstances of the crime as well as yielding additional images of the ridge pattern. In some cases, these techniques have shown the ability to provide further ridge detail, "filling in the gaps" of the developed mark. Matrix Assisted Laser Desorption Ionisation Mass Spectrometry Imaging (MALDI MSI) is presently the most advanced of the so-called ‘surface analysis’ techniques, in terms of compatibility with a number of fingermark enhancement processes and implementation in operational casework. However, for the use of this technique in major crimes to become widespread, compatibility with sequential processing must be demonstrated. This short study has assessed compatibility with a number fingermark processing sequences applied to natural marks on the adhesive side of brown (parcel) and clear tapes. Within the study undertaken, the results confirm the possibility to use MALDI MSI in sequence with multiple processes offering in some instances, complementary ridge detail with respect to that recovered from marks developed by conventional sequence processing

    A Structural and Geophysical Approach to the Study of Fractured Aquifers in the Scansano-Magliano in Toscana Ridge, Southern Tuscany, Italy

    Fresh water availability has recently become a serious concern in the Italian Apennines, as various activities rely on a predictable supply. Along the Scansano-Magliano ridge in southern Tuscany the situation is further complicated by contamination of the nearby alluvial aquifers. Aquifers locally consist of thin fractured reservoirs, generally within low-permeability formations, and it can be difficult to plan the exploitation of resources based on conventional techniques. An integrated study based on geological investigate the link between tectonics and groundwater circulation and to better define the hydrological model. After the regional identification of fault and fracture patterns, a major structure was investigated in detail to accurately map its spatial position and to understand the geometry and properties of the associated aquifer and assess its exploitation potential. The subsurface around the fault zone was clearly imaged through Ground Probing Radar, 2D and 3D resistivity tomography, and 3D shallow seismic surveys. The vertical and horizontal contacts between the different geological units of the Ligurian and Tuscan series were resolved with a high degree of spatial accuracy. 3D high-resolution geophysical imaging proved to be a very effective means for characterising small-scale fractured reservoirs

    Geophysical signature of a World War i tunnel-like anomaly in the Forni Glacier (Punta Linke, Italian Alps)

    Global warming and the associated glacier retreat recently revealed the entrance to an ice\u2013rock tunnel, at an altitude of 3c3600 m a.s.l., in the uppermost portion of the Forni Glacier in the Central Italian Alps. The tunnel served as an entrance to an Austro-Hungarian cableway station excavated in the rocks during the Great War just behind the frontline. A comprehensive geophysical survey, based on seismic and ground-penetrating radar profiling, was then undertaken to map other possibleWorldWar I (WWI) remains still embedded in the ice. The ice\u2013rock interface was reconstructed over the entire saddle and in the uppermost portion of the glacier. A prominent linear reflector was surprisingly similar to the common response of buried pipes. The reflector orientation, almost longitudinal to the slope, does not seem to be compatible with a glacial conduit or with other natural features. Numerical simulations of a series of possible targets constrained interpretation to a partly water-filled rounded shape cavity. The presence of a preserved WWI tunnel connecting Mount Vioz and Punta Linke could be considered a realistic hypothesis. The Forni glacier could be still considered polythermal and comprised of cold ice without basal sliding in its top portion

    Hydrodynamics in evaporate-bearing fine-grained successions investigated through an interdisciplinary approach : A test study in southern Italy-hydrogeological behaviour of heterogeneous low-permeability media

    Messinian evaporates are widely distributed in the Mediterranean Sea as outcropping sediments in small marginal basins and in marine cores. Progressive filling of subbasins led to the formation of complex aquifer systems in different regions where hypersaline and fresh water coexist and interact in different manner. It also generates a significant diversification of groundwater hydrochemical signature and different microbial communities. In the case study, the hydrogeology and hydrochemistry of the whole system are influenced by good hydraulic connection between the shallower pyroclastic horizon and the underlying evaporate-bearing fine-grained Messinian succession. This is demonstrated by the merge of hydrogeological, chemical, isotopic, and microbiological data. No mixing with deep ascending waters has been observed. As shown by geophysical, hydraulic, and microbiological investigations, the hydraulic heterogeneity of the Messinian bedrock, mainly due to karstified evaporitic interstrata/lenses, causes the hydraulic head to significantly vary with depth. Somewhere, the head increases with the depth's increase and artesian flow conditions are locally observed. Moreover, the metagenomic investigations demonstrated the existence of a poor hydraulic connection within the evaporate-bearing fine-grained succession at metric and decametric scales, therefore leading to a patchwork of geochemical (and microbiological) subenvironments
