A PTAS for Bounded-Capacity Vehicle Routing in Planar Graphs
The Capacitated Vehicle Routing problem is to find a minimum-cost set of
tours that collectively cover clients in a graph, such that each tour starts
and ends at a specified depot and is subject to a capacity bound on the number
of clients it can serve. In this paper, we present a polynomial-time
approximation scheme (PTAS) for instances in which the input graph is planar
and the capacity is bounded. Previously, only a quasipolynomial-time
approximation scheme was known for these instances. To obtain this result, we
show how to embed planar graphs into bounded-treewidth graphs while preserving,
in expectation, the client-to-client distances up to a small additive error
proportional to client distances to the depot
Use of professionally administered topical fluorides in Asia
Professionally applied topical fluoride varnish, gel, and solution have been shown to be effective in preventing and in arresting dental caries. Their use in different countries in Asia varies greatly and may not correlate with the dental caries situation of the populations in the countries. In the higher-income countries, use of fluoride varnish and gel is common among dental professionals. In contrast, the use of professionally administered topical fluorides is not common in the lower-income countries. Fluoride varnish, being easy to apply and safe, has been the preferred agent for the prevention of early childhood caries, which is prevalent in many developing countries in Asia. The relatively high cost of professionally administered fluoride agents and the shortage of a dental workforce, especially in lower income countries, have hampered the widespread adoption of these effective caries prevention methods in the private and public dental services. Government health policies should be pursued to lower the cost of treatment, either through incentives for local production and/or elimination of taxes and tariffs on imported fluoride products.published_or_final_versio
Physical disaggregation of numerical model rainfall
International audienceA methodology is presented for the disaggregation of numerical model fields of convective rainfall using a physically based procedure. The scheme uses surface sensible heat flux values derived from high-resolution multichannel satellite radiometer observations. The sensible heat flux values initialise a simple convective model to calculate the convective disaggregation parameter (CDP), which is theoretically proportional to the convective rainfall rate. The CDP diagnostic parameter can be derived as a one-time field if the surface characteristics are invariant, as a seasonal value may be, or it may be evaluated on a case by case basis. Once found, the CDP can be used to disaggregate numerical weather prediction (NWP) convective rainfall fields for as far ahead as such fields are produced
APOE ε4 status is associated with white matter hyperintensities volume accumulation rate independent of AD diagnosis.
To assess the relationship between carriage of APOE ε4 allele and evolution of white matter hyperintensities (WMHs) volume, we longitudinally studied 339 subjects from the Alzheimer's Disease Neuroimaging Initiative cohort with diagnoses ranging from normal controls to probable Alzheimer's disease (AD). A purpose-built longitudinal automatic method was used to segment WMH using constraints derived from an atlas-based model selection applied to a time-averaged image. Linear mixed models were used to evaluate the differences in rate of change across diagnosis and genetic groups. After adjustment for covariates (age, sex, and total intracranial volume), homozygous APOE ε4ε4 subjects had a significantly higher rate of WMH accumulation (22.5% per year 95% CI [14.4, 31.2] for a standardized population having typical values of covariates) compared with the heterozygous (ε4ε3) subjects (10.0% per year [6.7, 13.4]) and homozygous ε3ε3 (6.6% per year [4.1, 9.3]) subjects. Rates of accumulation increased with diagnostic severity; controls accumulated 5.8% per year 95% CI: [2.2, 9.6] for the standardized population, early mild cognitive impairment 6.6% per year [3.9, 9.4], late mild cognitive impairment 12.5% per year [8.2, 17.0] and AD subjects 14.7% per year [6.0, 24.0]. Following adjustment for APOE status, these differences became nonstatistically significant suggesting that APOE ε4 genotype is the major driver of accumulation of WMH volume rather than diagnosis of AD
Stable expression of Escherichia coli {beta}-glucuronidase A (GusA) in Giardia lamblia: application to high-throughput drug susceptibility testing
Objectives In order to create a suitable model for high-throughput drug screening, a Giardia lamblia WB C6 strain expressing Escherichia coli glucuronidase A (GusA) was created and tested with respect to susceptibility to the anti-giardial drugs nitazoxanide and metronidazole. Methods GusA, a well-established reporter gene in other systems, was cloned into the vector pPacVInteg allowing stable expression in G. lamblia under control of the promoter from the glutamate dehydrogenase (gdh) gene. The resulting transgenic strain was compared with the wild-type strain in a vitality assay, characterized with respect to susceptibility to nitazoxanide, metronidazole and-as assessed in a 96-well plate format-to a panel of 15 other compounds to be tested for anti-giardial activity. Results GusA was stably expressed in G. lamblia. Using a simple glucuronidase assay protocol, drug efficacy tests yielded results similar to those from cell counting. Conclusions G. lamblia WB C6 GusA is a suitable tool for high-throughput anti-giardial drug screening
Dynamical approach to spectator fragmentation in Au+Au reactions at 35 MeV/A
The characteristics of fragment emission in peripheral Au+Au
collisions 35 MeV/A are studied using the two clusterization approaches within
framework of \emph{quantum molecular dynamics} model. Our model calculations
using \emph{minimum spanning tree} (MST) algorithm and advanced clusterization
method namely \emph{simulated annealing clusterization algorithm} (SACA) showed
that fragment structure can be realized at an earlier time when spectators
contribute significantly toward the fragment production even at such a low
incident energy. Comparison of model predictions with experimental data reveals
that SACA method can nicely reproduce the fragment charge yields and mean
charge of the heaviest fragment. This reflects suitability of SACA method over
conventional clusterization techniques to investigate spectator matter

Sex-related differences in whole brain volumes at age 70 in association with hyperglycemia during adult life
Longitudinal studies of the relationship between hyperglycemia and brain health are rare and there is limited information on sex differences in associations. We investigated whether glycosylated haemoglobin (HbA1c) measured at ages of 53, 60-64 and 69 years, and cumulative glycemic index (CGI), a measure of cumulative glycemic burden, were associated with metrics of brain health in later life.
Participants were from Insight 46, a sub-study of the Medical Research Council National Survey of Health and Development (NSHD) who undertook volumetric MRI, florbetapir amyloid-PET imaging and cognitive assessments at ages of 69-71. Analyses were performed using linear and logistic regression as appropriate, with adjustment for potential confounders. We observed a sex interaction between HbA1c and whole brain volume (WBV) at all three time points. Following stratification of our sample, we observed that HbA1c at all ages, and CGI were positively associated with lower WBV exclusively in females. HbA1c (or CGI) was not associated with amyloid status, white matter hyperintensities (WMHs), hippocampal volumes (HV) or cognitive outcomes in either sex.
Higher HbA1c in adulthood is associated with smaller WBV at 69–71 years in females but not in males. This suggests that there may be preferential target organ damage in the brain for females with hyperglycemia
New methods of evaluation of the flavour composition in annihilation by double hemisphere tagging at LEP/SLC energies
Two new methods are proposed to extract the flavour contents of the events produced at LEP/SLC, together with the classification matrix of a tagging by hemispheres. By utilising the tagging obtained in both hemispheres, the efficiencies, backgrounds and flavour compositions are directly obtained by fitting the data. A minimal dependence on modelling and a consistent treatment of systematic errors are achieved by applying these methods. The choice of the tagging algorithm is irrelevant in the methods, provided that similar efficiencies are reached. As an example, a multivariate analysis technique combining the tracking information given by a microvertex detector has been applied to extract the Z → b overlineb branching ratio using a standard simulation of a LEP/SLC experiment
Caveolin-1 protects B6129 mice against Helicobacter pylori gastritis.
Caveolin-1 (Cav1) is a scaffold protein and pathogen receptor in the mucosa of the gastrointestinal tract. Chronic infection of gastric epithelial cells by Helicobacter pylori (H. pylori) is a major risk factor for human gastric cancer (GC) where Cav1 is frequently down-regulated. However, the function of Cav1 in H. pylori infection and pathogenesis of GC remained unknown. We show here that Cav1-deficient mice, infected for 11 months with the CagA-delivery deficient H. pylori strain SS1, developed more severe gastritis and tissue damage, including loss of parietal cells and foveolar hyperplasia, and displayed lower colonisation of the gastric mucosa than wild-type B6129 littermates. Cav1-null mice showed enhanced infiltration of macrophages and B-cells and secretion of chemokines (RANTES) but had reduced levels of CD25+ regulatory T-cells. Cav1-deficient human GC cells (AGS), infected with the CagA-delivery proficient H. pylori strain G27, were more sensitive to CagA-related cytoskeletal stress morphologies ("humming bird") compared to AGS cells stably transfected with Cav1 (AGS/Cav1). Infection of AGS/Cav1 cells triggered the recruitment of p120 RhoGTPase-activating protein/deleted in liver cancer-1 (p120RhoGAP/DLC1) to Cav1 and counteracted CagA-induced cytoskeletal rearrangements. In human GC cell lines (MKN45, N87) and mouse stomach tissue, H. pylori down-regulated endogenous expression of Cav1 independently of CagA. Mechanistically, H. pylori activated sterol-responsive element-binding protein-1 (SREBP1) to repress transcription of the human Cav1 gene from sterol-responsive elements (SREs) in the proximal Cav1 promoter. These data suggested a protective role of Cav1 against H. pylori-induced inflammation and tissue damage. We propose that H. pylori exploits down-regulation of Cav1 to subvert the host's immune response and to promote signalling of its virulence factors in host cells
