The crude skin secretion of the pepper frog Leptodactylus labyrinthicus is rich in metallo and serine peptidases

Peptidases are ubiquitous enzymes involved in diverse biological processes. Fragments from bioactive peptides have been found in skin secretions from frogs, and their presence suggests processing by peptidases. Thus, the aim of this work was to characterize the peptidase activity present in the skin secretion of Leptodactylus labyrinthicus. Zymography revealed the presence of three bands of gelatinase activity of approximately 60 kDa, 66 kDa, and 80 kDa, which the first two were calcium-dependent. These three bands were inhibited either by ethylenediaminetetraacetic acid (EDTA) and phenathroline; thus, they were characterized as metallopeptidases. Furthermore, the proteolytic enzymes identified were active only at pH 6.0–10.0, and their activity increased in the presence of CHAPS or NaCl. Experiments with fluorogenic substrates incubated with skin secretions identified aminopeptidase activity, with cleavage after leucine, proline, and alanine residues. This activity was directly proportional to the protein concentration, and it was inhibited in the presence of metallo and serine peptidase inhibitors. Besides, the optimal pH for substrate cleavage was determined to be 7.0–8.0. The results of the in gel activity assay showed that all substrates were hydrolyzed by a 45 kDa peptidase. Gly-Pro-AMC was also cleaved by a peptidase greater than 97 kDa. The data suggest the presence of dipeptidyl peptidases (DPPs) and metallopeptidases; however, further research is necessary. In conclusion, our work will help to elucidate the implication of these enzymatic activities in the processing of the bioactive peptides present in frog venom, expanding the knowledge of amphibian biology

A high throughput screening (HTS) methodology for evaluation of cellular lipid content based on Nile red fluorescence reads using black background 96-wells test plates and a plate reader equipment allowed the rapid intracellular lipid estimation of strains from a Brazilian phylloplane yeast collection. A new oleaginous yeast,Meyerozyma guilliermondii BI281A, was selected, for which the gravimetric determination of total lipids relative to dry weight was 52.38%for glucose or 34.97%for pure glycerol. The lipid production was optimized obtaining 108 mg/L of neutral lipids using pure glycerol as carbon source, and the strain proved capable of accumulating oil using raw glycerol from a biodiesel refinery. The lipid profile showed monounsaturated fatty acids (MUFA) varying between 56 or 74% in pure or raw glycerol, respectively. M. guilliermondii BI281A bears potential as a new biodiesel feedstock

Building subsidies to identify signs of sexual abuse against children and adolescents

Sexual abuse against children and adolescents is considered a serious public health problem. Any contact or interaction with a minor that includes touching, any type of sex or caressing is characterized as violence and most of these abuses take place at home and are carried out by family members. This violence is underreported, mainly because it occurs in homes and one of the difficulties encountered by health professionals who deal with situations of violence is the lack of language and concepts that combine medical knowledge with that of other areas, such as social and legal. In view of this perspective, the study sought to create subsidies that could help health and administrative professionals of a Family Health Unit to identify suspicious signs of this type of violence against this public, reporting tools, in addition to the reception of victims and the family. To achieve this objective, a booklet was developed with information about the sexual violence that children and adolescents are susceptible, warning signs and how to act in the face of violence, in addition, an easily accessible online page was created, via QR CODE, containing the educational booklet, social networks of public agencies for the protection of children and adolescents and the contacts necessary to make complaints. Training was also carried out with all the health and administrative professionals of the unit. There were 1337 accesses to the QR CODE and the page, indicating propagation of the material. Data on the increase in reports of sexual violence have not yet been obtained

It is a pilot study, involving 15 children that were commited at the Hospital da Criança: Obras Sociais de Irmã Dulce, Salvador, Bahia. The objective of this study is the investigation of family’s perception about the pediatric patient with chronic illness. The responsible for the patient answered a questionnaire that approached subjects regarding the partner demographic profile, family  knowledge degree about the disease, behavior alterations in child and its family, besides the impression of prognostic and  the medical services that were offered to the researched population. The results evidenced that some kind of behavior alteration were presented by all evaluated population, being more frequent symptoms of: sadness; anxiety; isolation and sleepness. The parents referred that after the appearance of the disease, the more common manifestations were: increase of the familiar union, feeling of fear, anxiety and concern with physical sequels. Regarding the model of hospital care, it was revealed that they were satisfied with the quality and efficiency of the medical service. However, they consider insufficient the psychological support to the child and the family throughout the confrontation of the chronic disease. The next phase of this study should make possible a more consistent and inclusive analysis concerning the proposed theme. There is no specific antiviral therapy recommended for coronavirus disease 2019 (COVID-19). In vitro studies indicate that the antiviral effect of chloroquine diphosphate (CQ) requires a high concentration of the drug.To evaluate the safety and efficacy of 2 CQ dosages in patients with severe COVID-19. This parallel, double-masked, randomized, phase IIb clinical trial with 81 adult patients who were hospitalized with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection was conducted from March 23 to April 5, 2020, at a tertiary care facility in Manaus, Brazilian Amazon. - Patients were allocated to receive high-dosage CQ (ie, 600 mg CQ twice daily for 10 days) or low-dosage CQ (ie, 450 mg twice daily on day 1 and once daily for 4 days). - Label: Main Outcomes and Measures Primary outcome was reduction in lethality by at least 50% in the high-dosage group compared with the low-dosage group. Data presented here refer primarily to safety and lethality outcomes during treatment on day 13. Secondary end points included participant clinical status, laboratory examinations, and electrocardiogram results. Outcomes will be presented to day 28. Viral respiratory secretion RNA detection was performed on days 0 and 4. Out of a predefined sample size of 440 patients, 81 were enrolled (41 [50.6%] to high-dosage group and 40 [49.4%] to low-dosage group). Enrolled patients had a mean (SD) age of 51.1 (13.9) years, and most (60 [75.3%]) were men. Older age (mean [SD] age, 54.7 [13.7] years vs 47.4 [13.3] years) and more heart disease (5 of 28 [17.9%] vs 0) were seen in the high-dose group. Viral RNA was detected in 31 of 40 (77.5%) and 31 of 41 (75.6%) patients in the low-dosage and high-dosage groups, respectively. Lethality until day 13 was 39.0% in the high-dosage group (16 of 41) and 15.0% in the low-dosage group (6 of 40). The high-dosage group presented more instance of QTc interval greater than 500 milliseconds (7 of 37 [18.9%]) compared with the low-dosage group (4 of 36 [11.1%]). Respiratory secretion at day 4 was negative in only 6 of 27 patients (22.2%). - Label: Conclusions and Relevance The preliminary findings of this study suggest that the higher CQ dosage should not be recommended for critically ill patients with COVID-19 because of its potential safety hazards, especially when taken concurrently with azithromycin and oseltamivir. These findings cannot be extrapolated to patients with nonsevere COVID-19
