    Flight service evaluation of an advanced composite empennage component on commercial transport aircraft. Phase 1: Engineering development

    The empennage component selected for this program is the vertical fin box of the L-1011 aircraft. The box structure extends from the fuselage production joint to the tip rib and includes the front and rear spars. Various design options were evaluated to arrive at a configuration which would offer the highest potential for satisfying program objectives. The preferred configuration selected consists of a hat-stiffened cover with molded integrally stiffened spars, aluminum trussed composite ribs, and composite miniwich web ribs with integrally molded caps. Material screening tests were performed to select an advanced composite material system for the Advanced Composite Vertical Fin (ACFV) that would meet the program requirements from the standpoint of quality, reproducibility, and cost. Preliminary weight and cost analysis were made, targets established, and tracking plans developed. These include FAA certification, ancillary test program, quality control, and structural integrity control plans

    Protein structures from the causative agent of anthrax (Bacillus anthracis) are being determined as part of a structural genomics programme. Amongst initial candidates for crystallographic analysis are enzymes involved in nucleotide biosynthesis, since these are recognized as potential targets in antibacterial therapy. Purine nucleoside phosphorylase is a key enzyme in the purine-salvage pathway. The crystal structure of purine nucleoside phosphorylase (DeoD) from B. anthracis has been solved by molecular replacement at 2.24 Ã… resolution and refined to an R factor of 18.4%. This is the first report of a DeoD structure from a Gram-positive bacterium

    Thermodynamic stabilities of ternary metal borides: An ab initio guide for synthesizing layered superconductors

    Density functional theory calculations have been used to identify stable layered Li-MM-B crystal structure phases derived from a recently proposed binary metal-sandwich (MS) lithium monoboride superconductor. We show that the MS lithium monoboride gains in stability when alloyed with electron-rich metal diborides; the resulting ordered Li2(1−x)Mx_{2(1-x)}M_xB2_2 ternary phases may form under normal synthesis conditions in a wide concentration range of xx for a number of group-III-V metals MM. In an effort to pre-select compounds with the strongest electron-phonon coupling we examine the softening of the in-plane boron phonon mode at Γ\Gamma in a large class of metal borides. Our results reveal interesting general trends for the frequency of the in-plane boron phonon modes as a function of the boron-boron bond length and the valence of the metal. One of the candidates with a promise to be an MgB2_2-type superconductor, Li2_2AlB4_4, has been examined in more detail: according to our {\it ab initio} calculations of the phonon dispersion and the electron-phonon coupling λ\lambda, the compound should have a critical temperature of ∼4\sim4 K.Comment: 10 pages, 9 figures, submitted to PR

    Medium-term performance and maintenance of SUDS:a case-study of Hopwood Park Motorway Service Area, UK

    One of the main barriers to implementing SUDS is concern about performance and maintenance costs since there are few well-documented case-studies. This paper summarizes studies conducted between 2000 and 2008 of the performance and maintenance of four SUDS management trains constructed in 1999 at the Hopwood Park Motorway Service Area, central England. Assessments were made of the wildlife value and sedimentation in the SUDS ponds, the hydraulic performance of the coach park management train, water quality in all management trains, and soil/sediment composition in the grass filter strip, interceptor and ponds. Maintenance procedures and costs were also reviewed. Results demonstrate the benefits of a management train approach over individual SUDS units for flow attenuation, water treatment, spillage containment and maintenance. Peak flows, pond sediment depth and contaminant concentrations in sediment and water decreased through the coach park management train. Of the 2007 annual landscape budget of £15,000 for the whole site, the maintenance costs for SUDS only accounted for £2,500 compared to £4,000 for conventional drainage structures. Furthermore, since sediment has been attenuated in the management trains, the cost of sediment removal after the recommended period of three years was only £554 and, if the design is not compromised, less frequent removal will be required in future

    Elliptical-core two mode fiber sensors and devices incorporating photoinduced refractive index gratings

    Results of experiments performed using germanium-doped, elliptical core, two-mode optical fibers whose sensitivity to strain was spatially varied through the use of chirped, refractive-index gratings permanently induced into the core using Argon-ion laser light are presented. This type of distributed sensor falls into the class of eighted-fiber sensors which, through a variety of means, weight the strain sensitivity of a fiber according to a specified spatial profile. We describe results of a weighted-fiber vibration mode filter which successfully enhances the particular vibration mode whose spatial profile corresponds to the profile of the grating chirp. We report on the high temperature survivability of such grating-based sensors and discuss the possibility of multiplexing more than one sensor within a single fiber

    Genome integration and excision by a new Streptomyces bacteriophage, Ï•Joe

    Bacteriophages are the source of many valuable tools for molecular biology and genetic manipulation. In Streptomyces, most DNA cloning vectors are based on serine integrase site-specific DNA recombination systems derived from phage. Because of their efficiency and simplicity, serine integrases are also used for diverse synthetic biology applications. Here we present the genome of a new Streptomyces phage, ϕJoe, and investigate the conditions for integration and excision of the φJoe genome. ϕJoe belongs to the largest Streptomyces phage cluster (R4-like) and encodes a serine integrase. The attB site from S. venezuelae was used efficiently by an integrating plasmid, pCMF92, constructed using the ϕJoe int/attP locus. The attB site for ϕJoe integrase was occupied in several Streptomyces genomes, including S. coelicolor, by a mobile element that varies in gene content and size between host species. Serine integrases require a phage-encoded recombination directionality factor (RDF) to activate the excision reaction. The ϕJoe RDF was identified and its function was confirmed in vivo Both the integrase and RDF were active in in vitro recombination assays. The ϕJoe site-specific recombination system is likely to be an important addition to the synthetic biology and genome engineering toolbox. IMPORTANCE: Streptomyces spp. are prolific producers of secondary metabolites including many clinically useful antibiotics. Bacteriophage-derived integrases are important tools for genetic engineering as they enable integration of heterologous DNA into the Streptomyces chromosome with ease and high efficiency. Recently researchers have been applying phage integrases for a variety of applications in synthetic biology, including rapid assembly of novel combinations of genes, biosensors and biocomputing. An important requirement for optimal experimental design and predictability when using integrases, however, is the need for multiple enzymes with different specificities for their integration sites. In order to provide a broad platform of integrases we identified and validated the integrase from a newly isolated Streptomyces phage, ϕJoe. ϕJoe integrase is active in vitro and in vivo The specific recognition site for integration is present in a wide range of different Actinobacteria, including Streptomyces venezuelae, an emerging model bacterium in Streptomyces research

    Theoretical study of metal borides stability

    We have recently identified metal-sandwich (MS) crystal structures and shown with ab initio calculations that the MS lithium monoboride phases are favored over the known stoichiometric ones under hydrostatic pressure [Phys. Rev. B 73, 180501(R) (2006)]. According to previous studies synthesized lithium monoboride tends to be boron-deficient, however the mechanism leading to this phenomenon is not fully understood. We propose a simple model that explains the experimentally observed off-stoichiometry and show that compared to such boron-deficient phases the MS-LiB compounds still have lower formation enthalpy under high pressures. We also investigate stability of MS phases for a large class of metal borides. Our ab initio results suggest that MS noble metal borides are less unstable than the corresponding AlB2_2-type phases but not stable enough to form under equilibrium conditions.Comment: 14 pages, 15 figure

    Spectra of Discrete Schr\"odinger Operators with Primitive Invertible Substitution Potentials

    We study the spectral properties of discrete Schr\"odinger operators with potentials given by primitive invertible substitution sequences (or by Sturmian sequences whose rotation angle has an eventually periodic continued fraction expansion, a strictly larger class than primitive invertible substitution sequences). It is known that operators from this family have spectra which are Cantor sets of zero Lebesgue measure. We show that the Hausdorff dimension of this set tends to 11 as coupling constant λ\lambda tends to 00. Moreover, we also show that at small coupling constant, all gaps allowed by the gap labeling theorem are open and furthermore open linearly with respect to λ\lambda. Additionally, we show that, in the small coupling regime, the density of states measure for an operator in this family is exact dimensional. The dimension of the density of states measure is strictly smaller than the Hausdorff dimension of the spectrum and tends to 11 as λ\lambda tends to 00
