Oral serum-derived bovine immunoglobulin improves duodenal immune reconstitution and absorption function in patients with HIV enteropathy.
ObjectivesTo examine the impact of serum-derived bovine immunoglobulin, an oral medical food known to neutralize bacterial antigen and reduce intestinal inflammation, on restoration of mucosal immunity and gastrointestinal function in individuals with HIV enteropathy.DesignOpen-label trial with intensive 8-week phase of bovine serum immunoglobulin (SBI) 2.5 g twice daily with a 4-week washout period and an optional 9-month extension study.MethodsHIV enteropathy was defined as chronic gastrointestinal symptoms including frequent loose or watery stools despite no identifiable, reversible cause. Upper endoscopy for tissue immunofluorescent antibody assay and disaccharide gut permeability/absorption studies were performed before and after 8 weeks of SBI to test mucosal immunity and gastrointestinal function. Blood was collected for markers of microbial translocation, inflammation, and collagen kinetics. A validated gastrointestinal questionnaire assessed changes in symptoms.ResultsAll eight participants experienced profound improvement in symptoms with reduced bowel movements/day (P = 0.008) and improvements in stool consistency (P = 0.008). Gut permeability was normal before and after the intervention, but D-xylose absorption increased in seven of eight participants. Mucosal CD4 lymphocyte densities increased by a median of 139.5 cells/mm2 from 213 to 322 cells/mm2 (P = 0.016). Intestinal-fatty acid binding protein (I-FABP), a marker of enterocyte damage, initially rose in seven of eight participants after 8 weeks (P = 0.039), and then fell below baseline in four of five who continued receiving SBI (P = 0.12). Baseline serum I-FABP levels were negatively correlated with subsequent rise in mucosal CD4 lymphocyte densities (r = -0.74, P = 0.046).ConclusionSBI significantly increases intestinal mucosal CD4 lymphocyte counts, improves duodenal function, and showed evidence of promoting intestinal repair in the setting of HIV enteropathy
Precision radial velocities with CSHELL
Radial velocity identification of extrasolar planets has historically been
dominated by optical surveys. Interest in expanding exoplanet searches to M
dwarfs and young stars, however, has motivated a push to improve the precision
of near infrared radial velocity techniques. We present our methodology for
achieving 58 m/s precision in the K band on the M0 dwarf GJ 281 using the
CSHELL spectrograph at the 3-meter NASA IRTF. We also demonstrate our ability
to recover the known 4 Mjup exoplanet Gl 86 b and discuss the implications for

Sterilization of granulomas is common in both active and latent tuberculosis despite extensive within-host variability in bacterial killing
Over 30% of the world’s population is infected with Mycobacterium tuberculosis (Mtb), yet only ~5–10% will develop clinical disease1. Despite considerable effort, we understand little about what distinguishes individuals who progress to active tuberculosis (TB) from those who remain latent for decades. The variable course of disease is recapitulated in cynomolgus macaques infected with Mtb2. Active disease in macaques is defined by clinical, microbiologic and immunologic signs and occurs in ~45% of animals, while the remaining are clinically asymptomatic2,3. Here, we use barcoded Mtb isolates and quantitative measures of culturable and cumulative bacterial burden to show that most lesions are likely founded by a single bacterium and reach similar maximum burdens. Despite common origins, the fate of individual lesions varies substantially within the same host. Strikingly, in active disease, the host sterilizes some lesions even while others progress. Our data suggest that lesional heterogeneity arises, in part, through differential killing of bacteria after the onset of adaptive immunity. Thus, individual lesions follow diverse and overlapping trajectories, suggesting critical responses occur at a lesional level to ultimately determine the clinical outcome of infection. Exploring Halo Substructure with Giant Stars IV: The Extended Structure of the Ursa Minor Dwarf Spheroidal
We present a large area photometric survey of the Ursa Minor dSph. We
identify UMi giant star candidates extending to ~3 deg from the center of the
dSph. Comparison to previous catalogues of stars within the tidal radius of UMi
suggests that our photometric luminosity classification is 100% accurate. Over
a large fraction of the survey area, blue horizontal branch stars associated
with UMi can also be identified. The spatial distribution of both the UMi giant
stars and the BHB stars are remarkably similar, and a large fraction of both
samples of stars are found outside the tidal radius of UMi. An isodensity
contour map of the stars within the tidal radius of UMi reveals two
morphological peculiarities: (1) The highest density of dSph stars is offset
from the center of symmetry of the outer isodensity contours. (2) The overall
shape of the outer contours appear S-shaped. We find that previously determined
King profiles with ~50' tidal radii do not fit well the distribution of our UMi
stars. A King profile with a larger tidal radius produces a reasonable fit,
however a power law with index -3 provides a better fit for radii > 20'. The
existence of UMi stars at large distances from the core of the galaxy, the
peculiar morphology of the dSph within its tidal radius, and the shape of its
surface density profile all suggest that UMi is evolving significantly due to
the tidal influence of the Milky Way. However, the photometric data on UMi
stars alone does not allow us to determine if the candidate extratidal stars
are now unbound or if they remain bound to the dSph within an extended dark

Probing the extragalactic fast transient sky at minute timescales with DECam
Searches for optical transients are usually performed with a cadence of days
to weeks, optimised for supernova discovery. The optical fast transient sky is
still largely unexplored, with only a few surveys to date having placed
meaningful constraints on the detection of extragalactic transients evolving at
sub-hour timescales. Here, we present the results of deep searches for dim,
minute-timescale extragalactic fast transients using the Dark Energy Camera, a
core facility of our all-wavelength and all-messenger Deeper, Wider, Faster
programme. We used continuous 20s exposures to systematically probe timescales
down to 1.17 minutes at magnitude limits (AB), detecting hundreds of
transient and variable sources. Nine candidates passed our strict criteria on
duration and non-stellarity, all of which could be classified as flare stars
based on deep multi-band imaging. Searches for fast radio burst and gamma-ray
counterparts during simultaneous multi-facility observations yielded no
counterparts to the optical transients. Also, no long-term variability was
detected with pre-imaging and follow-up observations using the SkyMapper
optical telescope. We place upper limits for minute-timescale fast optical
transient rates for a range of depths and timescales. Finally, we demonstrate
that optical -band light curve behaviour alone cannot discriminate between
confirmed extragalactic fast transients such as prompt GRB flashes and Galactic

Bodyweight Perceptions among Texas Women: The Effects of Religion, Race/Ethnicity, and Citizenship Status
Despite previous work exploring linkages between religious participation and health, little research has looked at the role of religion in affecting bodyweight perceptions. Using the theoretical model developed by Levin et al. (Sociol Q 36(1):157–173, 1995) on the multidimensionality of religious participation, we develop several hypotheses and test them by using data from the 2004 Survey of Texas Adults. We estimate multinomial logistic regression models to determine the relative risk of women perceiving themselves as overweight. Results indicate that religious attendance lowers risk of women perceiving themselves as very overweight. Citizenship status was an important factor for Latinas, with noncitizens being less likely to see themselves as overweight. We also test interaction effects between religion and race. Religious attendance and prayer have a moderating effect among Latina non-citizens so that among these women, attendance and prayer intensify perceptions of feeling less overweight when compared to their white counterparts. Among African American women, the effect of increased church attendance leads to perceptions of being overweight. Prayer is also a correlate of overweight perceptions but only among African American women. We close with a discussion that highlights key implications from our findings, note study limitations, and several promising avenues for future research
B Cells Regulate Neutrophilia during Mycobacterium tuberculosis Infection and BCG Vaccination by Modulating the Interleukin-17 Response
We have previously demonstrated that B cells can shape the immune response to Mycobacterium tuberculosis, including the level of neutrophil infiltration and granulomatous inflammation at the site of infection. The present study examined the mechanisms by which B cells regulate the host neutrophilic response upon exposure to mycobacteria and how neutrophilia may influence vaccine efficacy. To address these questions, a murine aerosol infection tuberculosis (TB) model and an intradermal (ID) ear BCG immunization mouse model, involving both the μMT strain and B cell-depleted C57BL/6 mice, were used. IL (interleukin)-17 neutralization and neutrophil depletion experiments using these systems provide evidence that B cells can regulate neutrophilia by modulating the IL-17 response during M. tuberculosis infection and BCG immunization. Exuberant neutrophilia at the site of immunization in B cell-deficient mice adversely affects dendritic cell (DC) migration to the draining lymph nodes and attenuates the development of the vaccine-induced Th1 response. The results suggest that B cells are required for the development of optimal protective anti-TB immunity upon BCG vaccination by regulating the IL-17/neutrophilic response. Administration of sera derived from M. tuberculosis-infected C57BL/6 wild-type mice reverses the lung neutrophilia phenotype in tuberculous μMT mice. Together, these observations provide insight into the mechanisms by which B cells and humoral immunity modulate vaccine-induced Th1 response and regulate neutrophila during M. tuberculosis infection and BCG immunization. © 2013 Kozakiewicz et al

LINCS Canvas Browser: interactive web app to query, browse and interrogate LINCS L1000 gene expression signatures
For the Library of Integrated Network-based Cellular Signatures (LINCS) project many gene expression signatures using the L1000 technology have been produced. The L1000 technology is a cost-effective method to profile gene expression in large scale. LINCS Canvas Browser (LCB) is an interactive HTML5 web-based software application that facilitates querying, browsing and interrogating many of the currently available LINCS L1000 data. LCB implements two compacted layered canvases, one to visualize clustered L1000 expression data, and the other to display enrichment analysis results using 30 different gene set libraries. Clicking on an experimental condition highlights gene-sets enriched for the differentially expressed genes from the selected experiment. A search interface allows users to input gene lists and query them against over 100 000 conditions to find the top matching experiments. The tool integrates many resources for an unprecedented potential for new discoveries in systems biology and systems pharmacology.
