    BFL1 Modulates Apoptosis at the Membrane Level through a Bifunctional And Multimodal Mechanism Showing Key Differences With BCLXL

    BFL1 is a relatively understudied member of the BCL2 protein family which has been implicated in the pathogenesis andchemoresistance of a variety of human cancers, including hematological malignancies and solid tumours. BFL1 is generallyconsidered to have an antiapoptotic function, although its precise mode of action remains unclear. By quantitativelyanalyzing BFL1 action in synthetic membrane models and in cells, we found that BFL1 inhibits apoptosis through threedistinct mechanisms which are similar but not identical to those of BCLXL, the paradigmatic antiapoptotic BCL2 familyprotein. Strikingly, alterations in lipid composition during apoptosis activate a prodeath function of BFL1 that is based onnoncanonical oligomerization of the protein and breaching of the permeability barrier of the outer mitochondrial membrane(OMM). This lipid-triggered prodeath function of BFL1 is absent in BCLXL and also differs from that of the apoptoticeffector BAX, which sets it apart from other BCL2 family members. Ourfindings support a new model in which BFL1modulates apoptosis through a bifunctional and multimodal mode of action that is distinctly regulated by OMM lipidscompared to BCLXL.This work was supported by Grants BFU2011-28566 from the Ministerio de Economia y Competitividad and IT838-13 from Gobierno Vasco.     Shared features and reciprocal complementation of the Chlamydomonas and Arabidopsis microbiota

    Microscopic algae release organic compounds to the region immediately surrounding their cells, known as the phycosphere, constituting a niche for colonization by heterotrophic bacteria. These bacteria take up algal photoassimilates and provide beneficial functions to their host, in a process that resembles the establishment of microbial communities associated with the roots and rhizospheres of land plants. Here, we characterize the microbiota of the model alga Chlamydomonas reinhardtii and reveal extensive taxonomic and functional overlap with the root microbiota of land plants. Using synthetic communities derived from C. reinhardtii and Arabidopsis thaliana, we show that phycosphere and root bacteria assemble into taxonomically similar communities on either host. We show that provision of diffusible metabolites is not sufficient for phycosphere community establishment, which additionally requires physical proximity to the host. Our data suggest the existence of shared ecological principles driving the assembly of the A. thaliana root and C. reinhardtii phycosphere microbiota, despite the vast evolutionary distance between these two photosynthetic organisms

    Risk behaviors of 15–21 year olds in Mexico lead to a high prevalence of sexually transmitted infections: results of a survey in disadvantaged urban areas

    BACKGROUND: Due to the fact that adolescents are more likely to participate in high-risk behaviors, this sector of the population is particularly vulnerable to contracting sexually transmitted infections (STIs) and resultant health problems. METHODS: A survey was carried out among adolescents from poor homes in 204 small-urban areas of Mexico. Information was collected in relation to risk behaviors and socio-economic environment. A sub-group of the participants also provided blood and urine samples which were analyzed to detect sexually transmitted infections. RESULTS: The presence of Chlamydia was detected in nearly 8% of participants who had stated that they were sexually active (18%) and approximately 12% were positive for herpes type 2-specific antibodies. For both, a greater proportion of girls resulted positive compared to boys. The presence of these biological outcomes of sexual risk behavior was associated with other risk behaviors (smoking), but not with self-reported indicators of protected sex (reported use of condom during most recent sexual activity). CONCLUSION: The results presented in this study show a startlingly high prevalence of HSV-2 among sexually active Mexican adolescents in poor urban areas, suggesting that this group has participated to a great extent in risky sexual practices. The relationships between socioeconomic environment and adolescent risk behavior need to be better understood if we are to design preventive interventions that modify the determinants of risk behaviors

    Quince (Cydonia oblonga) in vitro plant root formation through an automated temporary inmersion system, and its acclimation

    Artículo científicoQuince (Cydonia oblonga) is a non-traditional fruit tree found in Costa Rica that has therapeutic and nutritional properties; however its slow growth and root formation prevents the production of a homogeneous population when using conventional farming techniques. Hence, the aim of this research project was to generate uniform plant material in a reduced time span using a temporary immersion bioreactor system (RITAS ®). A semisolid rooting MS culture medium supplemented with 0.1 mg L-1 NAA; 0.3 mg L-1 IBA and 3% sucrose (pH 6.5), developed in the Centro de Investigación en Biotecnología (CIB), Instituto Tecnológico de Costa Rica (ITCR), in Cartago, was used as a reference medium. Four different variations in the sucrose concentration (1%, 2%, 3%, and 4%) were performed in liquid medium. Each trial was evaluated with in vitro plants which had been previously exposed to the culture medium of the corresponding treatments, in a stationary mode and for a 15 day long period, and with in vitro plants without any previous treatment (a total of eight treatments). The comparison of the root formation percentages evidenced the clear effect of sucrose concentration used, with the best results obtained when using the 2% sucrose trial with no pre-treatment (73.3%). The in vitro plants were acclimated in cylinders made out of peat, have previously been disinfected with fungicide, and placed in a humidity chamber at a 20.5°C average temperature and a 75,5% relative humidity for the establishment of weekly fertilizing cycles. The acclimation process generated an 80% survival rate, since several seedlings experienced stem strangulation caused by a fungal attack. The conidiophores identified through optical and scanning electron microscopy evidenced the presence of Cladosporium spp., which was controlled with carbendazim and iprodione fungicides

    Gray Matter Changes in Parkinson's and Alzheimer's Disease and Relation to Cognition

    Purpose of Review We summarize structural (s)MRI findings of gray matter (GM) atrophy related to cognitive impairment in Alzheimer's disease (AD) and Parkinson's disease (PD) in light of new analytical approaches and recent longitudinal studies results. Recent Findings The hippocampus-to-cortex ratio seems to be the best sMRI biomarker to discriminate between various AD subtypes, following the spatial distribution of tau pathology, and predict rate of cognitive decline. PD is clinically far more variable than AD, with heterogeneous underlying brain pathology. Novel multivariate approaches have been used to describe patterns of early subcortical and cortical changes that relate to more malignant courses of PD. New emerging analytical approaches that combine structural MRI data with clinical and other biomarker outcomes hold promise for detecting specific GM changes in the early stages of PD and preclinical AD that may predict mild cognitive impairment and dementia conversion

    Exploiting Phenylpropanoid Derivatives to Enhance the Nutraceutical Values of Cereals and Legumes

    Phenylpropanoids are a diverse chemical class with immense health benefits that are biosynthesized from the aromatic amino acid L-phenylalanine. This article reviews the progress for accessing variation in phenylpropanoids in germplasm collections, the genetic and molecular basis of phenylpropanoid biosynthesis, and the development of cultivars dense in seed-phenylpropanoids. Progress is also reviewed on high-throughput assays, factors that influence phenylpropanoids, the site of phenylpropanoids accumulation in seed, Genotype × Environment interactions, and on consumer attitudes for the acceptance of staple foods rich in phenylpropanoids. A paradigm shift was noted in barley, maize, rice, sorghum, soybean, and wheat, wherein cultivars rich in phenylpropanoids are grown in Europe and North and Central America. Studies have highlighted some biological constraints that need to be addressed for development of high-yielding cultivars that are rich in phenylpropanoids. Genomics-assisted breeding is expected to facilitate rapid introgression into improved genetic backgrounds by minimizing linkage drag. More research is needed to systematically characterize germplasm pools for assessing variation to support crop genetic enhancement, and assess consumer attitudes to foods rich in phenylpropanoids
