    Life cycle performance assessment tool development and application with a focus on maintenance aspects

    Ships are among the most complex systems in the world. The always increasing interest in environmental aspects, the evolution of technologies and the introduction of new rule constraints in the maritime field have compelled the innovation of the ship design approach. At an early design stage, there is the need to compare different design solutions, also in terms of environmental performance, building and operative costs over the whole ship life cycle. In this context, the Life Cycle Performance Assessment (LCPA) tool allows an integrated design approach merging the evaluation of both costs and environmental performances on a comparative basis, among different design solutions. Starting from the first tool release, this work aims to focus on the maintenance of the propulsion system, developing a flexible calculation method for maintenance costs prediction, based on the ship operational profiles and the selected technical solution. After the improvement, the whole LCPA tool has been applied on a research vessel to evaluate, among different propulsion layout solutions, the one with the more advantageous performance in terms of costs during the whole vessel operating life. The identification of the best design solution is strictly dependent on the selection criterion and the point of view of the interested parties using the LCPA tool, e.g., the shipbuilder or the ship-owner

    Influence of shoot number and crop load on potted Chambourcin grapevines. 2. Whole-vine vs. single-leaf photosynthesis

    Two-year-old potted Chambourcin grapevines were trained to one shoot with 0 or 1 cluster (1/0 and 1/1 respectively) or four shoots with 0 or 4 clusters (4/0 and 4/4 respectively) to determine the effects of canopy development rate, canopy morphology, and crop load on whole-vine photosynthesis. Significant differences in canopy development rate, canopy morphology and dry matter partitioning occurred among treatments but whole-vine net photosynthesis (Pn) and dry matter production were not affected. Photosynthetic compensation by leaves of severely pruned vines enabled them to produce quantities of dry matter similar to vines with greater leaf area. Vines bearing crop supported the development of berries by partitioning carbohydrate to fruit at the expense of vegetative tissues so overall vine dry weight was not different among cropped and non-cropped treatments. Whole-vine Pn determinations were linearly related to vine dry mass. By contrast, single leaf measurements used to estimate whole-vine Pn were not related to dry mass. If a similar relationship can be demonstrated in field vines, it may be possible to quantify the influence of biotic and abiotic stresses on vine biomass production and subsequent yields

    HIF-1 alpha-independent hypoxia-induced rapid PTK6 stabilization is associated with increased motility and invasion

    © 2014 Landes Bioscience. PTK6/Brk is a non-receptor tyrosine kinase overexpressed in cancer. Here we demonstrate that cytosolic PTK6 is rapidly and robustly induced in response to hypoxic conditions in a HIF-1-independent manner. Furthermore, a proportion of hypoxic PTK6 subsequently re-localized to the cell membrane. We observed that the rapid stabilization of PTK6 is associated with a decrease in PTK6 ubiquitylation and we have identified c-Cbl as a putative PTK6 E3 ligase in normoxia. The consequences of hypoxia-induced PTK6 stabilization and subcellular re-localization to the plasma membrane include increased cell motility and invasion, suggesting PTK6 targeting as a therapeutic approach to reduce hypoxia-regulated metastatic potential. This could have particular significance for breast cancer patients with triple negative disease

    IN VITRO AMOEBICIDAL ACTIVITY OF SOME MEDICINAL PLANTS OF THE BAMUN REGION (CAMEROON)

    Fifty two medicinal plants belonging to different families selected on the basis of their traditional use against jaundice and various liver disorders were tested for their amoebicidal activities. They were extracted and tested for their antiamoebic activity in vitro using polyxenic culture of Entamoebic histolytica. As the result, 14 exhibited an antiamoebic activity at a dose of 100µg/ml from the second to the fourth day of incubation. The 14 selected extracts were additionaly tested for 6 days at 10, 100 and 500g/ml of concentration, and only the leaves extract of Codiaeum variegatum exhibited a clear antiamoebic activity (EC50=10,74 the second day), and had a more pronounced activity than metronidazole the reference product

    The Automated Inspection of Opaque Liquid Vaccines

    In the pharmaceutical industry the screening of opaque vaccines containing suspensions is currently a manual task carried out by trained human visual inspectors. We show that deep learning can be used to effectively automate this process. A moving contrast is required to distinguish anomalies from other particles, reflections and dust resting on a vial's surface. We train 3D-ConvNets to predict the likelihood of 20-frame video samples containing anomalies. Our unaugmented dataset consists of hand-labelled samples, recorded using vials provided by the HAL Allergy Group, a pharmaceutical company. We trained ten randomly initialized 3D-ConvNets to provide a benchmark, observing mean AUROC scores of 0.94 and 0.93 for positive samples (containing anomalies) and negative (anomaly-free) samples, respectively. Using Frame-Completion Generative Adversarial Networks we: (i) introduce an algorithm for computing saliency maps, which we use to verify that the 3D-ConvNets are indeed identifying anomalies; (ii) propose a novel self-training approach using the saliency maps to determine if multiple networks agree on the location of anomalies. Our self-training approach allows us to augment our data set by labelling 217,888 additional samples. 3D-ConvNets trained with our augmented dataset improve on the results we get when we train only on the unaugmented dataset.Comment: 8 pages, 5 Figures, 3 Tables, ECAI 2020 Conference Proceeding

    Validation of Calcein Violet as a New Marker of Semen Membrane Integrity in Domestic Animals

    peer reviewedMany fluorochromes routinely used in semen quality analysis emit in the green and red channels, limiting their possible combination for multiple parameter analysis. The use of fluorophores emitting in different light channels broadens the possibilities of combination to expand the range of simultaneously evaluated criteria. This is of great interest in cases of small ejaculated volumes, such as those naturally occurring in roosters, small dog breeds and drones (Apis mellifera). The purpose of this experiment is to establish Calcein Violet (CaV), a blue fluorochrome, as a marker of viability and acrosomal integrity in domestic animals in order to free the red and green channels. SYBR®14/Propidium Iodide (PI) was used as reference dye, heat-treated samples as negative controls, serial staining combination for validation and epifluorescence microscopy for observation. Dead spermatozoa marked in red with PI showed no blue fluorescence either from the head or the tail. Live spermatozoa showed a decreasing blue emission from head to tail when single stained with CaV. Unreacted acrosomes showed intense blue fluorescence irrespective of plasma membrane integrity. This needs to be further confirmed for species with small and difficult to observe heads. Establishment of CaV as a marker of membrane integrity by fluorescence microscopy is a decisive first step towards further technical development and use with flow cytometry

    Long-term follow-up including extensive complement analysis of a pediatric C3 glomerulopathy cohort

    BACKGROUND: C3 glomerulopathy (C3G) is a rare kidney disorder characterized by predominant glomerular depositions of complement C3. C3G can be subdivided into dense deposit disease (DDD) and C3 glomerulonephritis (C3GN). This study describes the long-term follow-up with extensive complement analysis of 29 Dutch children with C3G. METHODS: Twenty-nine C3G patients (19 DDD, 10 C3GN) diagnosed between 1992 and 2014 were included. Clinical and laboratory findings were collected at presentation and during follow-up. Specialized assays were used to detect rare variants in complement genes and measure complement-directed autoantibodies and biomarkers in blood. RESULTS: DDD patients presented with lower estimated glomerular filtration rate (eGFR). C3 nephritic factors (C3NeFs) were detected in 20 patients and remained detectable over time despite immunosuppressive treatment. At presentation, low serum C3 levels were detected in 84% of all patients. During follow-up, in about 50% of patients, all of them C3NeF-positive, C3 levels remained low. Linear mixed model analysis showed that C3GN patients had higher soluble C5b-9 (sC5b-9) and lower properdin levels compared to DDD patients. With a median follow-up of 52 months, an overall benign outcome was observed with only six patients with eGFR below 90 ml/min/1.73 m(2) at last follow-up. CONCLUSIONS: We extensively described clinical and laboratory findings including complement features of an exclusively pediatric C3G cohort. Outcome was relatively benign, persistent low C3 correlated with C3NeF presence, and C3GN was associated with higher sC5b-9 and lower properdin levels. Prospective studies are needed to further elucidate the pathogenic mechanisms underlying C3G and guide personalized medicine with complement therapeutics.
