    COMPARAZIONE DEI PROFILI METABOLICI DI VARIETÀ DI CILIEGIO DOLCE (Prunus avium L.) MEDIANTE TECNICHE DI METABOLOMICA UNTARGETED

    La diffusione di cultivar altamente produttive ha influito negativamente sulla tutela dell’agro-biodiversità di Prunus avium L. determinando disinteresse nei confronti di varietà meno produttive ma meritevoli di attenzione per il valore delle loro caratteristiche organolettiche e del relativo patrimonio genetico. Il mondo scientifico ha mostrato interesse nei confronti di tale tematica e ha condotto azioni di conservazione in situ ed ex situ per preservare il germoplasma ancora esistente. La presente ricerca analizza l’agrobiodiversità del ciliegio dolce in relazione alla diversa composizione metabolomica dei frutti, espressione del genotipo che le caratterizza. L’attività di ricerca svolta in campo ha riguardato l’esplorazione del territorio, la raccolta di dati e il campionamento dei frutti di 20 cultivars coltivate in aree geografiche con differenti condizioni pedoclimatiche mentre l’attività effettuata in laboratorio è stata rivolta allo studio metabolomico. La caratterizzazione quali-quantitativa dei metaboliti secondari e la comparazione del peculiare profilo metabolomico dei frutti delle diverse varietà di ciliegia autoctone e commerciali, coltivate in Sicilia e in Veneto, è stata effettuata mediante indagine di metabolomica untargeted. Le analisi sono state condotte utilizzando la tecnica HPLC accoppiata a spettrometro di massa che ha fornito dati di tipo qualitativo, consentendo la comparazione dei campioni, mentre la validazione di tali dati è stata effettuata con un’analisi di tipo quantitativo utilizzando l’HPLC accoppiata a rivelatore diode array . I risultati ottenuti hanno permesso di identificare la composizione metabolomica che caratterizza ciascuna varietà e ciascun gruppo osservato e di individuare le varietà con le migliori caratteristiche organolettiche, oltre che nutrizionali con riferimento al contenuto di metaboliti in essi presenti. I risultati ottenuti suggeriscono che il recupero e la valorizzazione di vecchie varietà, come Sandra Tardiva e Cappuccia, alla luce della presenza di molecole bioattive rilevate, costituisce un potenziale investimento per la rivalutazione economica della cerasicoltura oltre che per la conservazione del germoplasma di antichi genotipi, attualmente coltivati solamente in ristretti comprensori.The spread of highly productive cultivars had a negative effect on the preservation of agrobiodiversity of Prunus avium L. and generated disinterest towards less productive but still deserving attention varieties, with their subsequent loss. The scientific community has shown interest in this issue and studies with different approaches were conducted through the use of conservative practices in situ and ex situ on the germplasm still available. The present research analyzes the agrobiodiversity of sweet cherry in relation to different metabolomics composition of fruits, expression of the genotype that characterizes them. Research activities were conducted on the field, focused on collection of fruits from 20 cultivars of Prunus avium used as crops in different Italian areas with heterogeneous pedo-climatic conditions; the research objectives were to characterize qualitatively and quantitatively the secondary metabolites and to compare the specific metabolic profiles of fruits the different varieties of autochthonous and commercially relevant cherries cultivated in Italy, in Sicily and Veneto regions, by untargeted metabolomics approaches. Data collected using HPLC- coupled mass-spectrometry provided a qualitative comparison of different samples while data validation and quantitative measurements were obtained by HPLC coupled to diode array. The results obtained allowed the identification of metabolites specific to each cultivar and to identify those with the best organoleptic and nutritional properties with respect to their metabolic content. These results suggest that the recovery and valorization of ancient cultivars of Prunus avium L. , like “Sandra tardiva” and “Cappuccia”, for the presence of bioactive molecules might represent a potential investment for revaluation of cherry cultivation as well as for the germplasm preservation of ancient genotypes currently cultivated only in limited areas

    Morpho-chemical observations of human deciduous teeth enamel in response to biomimetic toothpastes treatment

    Today, biomaterial research on biomimetic mineralization strategies represents a new challenge in the prevention and cure of enamel mineral loss on delicate deciduous teeth. Distinctive assumptions about the origin, the growth, and the functionalization on the biomimetic materials have been recently proposed by scientific research studies in evaluating the different clinical aspects of treating the deciduous tooth. Therefore, appropriate morpho-chemical observations on delivering specific biomaterials to enamel teeth is the most important factor for controlling biomineralization processes. Detailed morpho-chemical investigations of the treated enamel layer using three commercial toothpastes (Biorepair, F1400, and F500) were performed through variable pressure scanning electron microscopy (VP-SEM) and energy dispersive X-ray spectroscopy (EDS) on deciduous teeth in their native state. A new microscopy methodology allowed us to determine the behaviors of silicate, phosphate, and calcium contents from the early stage, as commercially available toothpastes, to the final stage of delivered diffusion, occurring within the enamel layer together with their penetration depth properties.     Phenotypical heterogeneity linked to adipose tissue dysfunction in patients with type 2 diabetes

    Adipose tissue (AT) inflammation leads to increased free fatty acid (FFA) efflux and ectopic fat deposition, but whether AT dysfunction drives selective fat accumulation in specific sites remains unknown. The aim of the present study was to investigate the correlation between AT dysfunction, hepatic/pancreatic fat fraction (HFF, PFF) and the associated metabolic phenotype in patients with Type 2 diabetes (T2D). Sixty-five consecutive T2D patients were recruited at the Diabetes Centre of Sapienza University, Rome, Italy. The study population underwent clinical examination and blood sampling for routine biochemistry and calculation of insulin secretion [homoeostasis model assessment of insulin secretion (HOMA-β%)] and insulin-resistance [homoeostasis model assessment of insulin resistance (HOMA-IR) and adipose tissue insulin resistance (ADIPO-IR)] indexes. Subcutaneous (SAT) and visceral (VAT) AT area, HFF and PFF were determined by magnetic resonance. Some 55.4% of T2D patients had non-alcoholic fatty liver disease (NAFLD); they were significantly younger and more insulin-resistant than non-NAFLD subjects. ADIPO-IR was the main determinant of HFF independently of age, sex, HOMA-IR, VAT, SAT and predicted severe NAFLD with the area under the receiver operating characteristic curve (AUROC)=0.796 (95% confidence interval: 0.65-0.94, P=0.001). PFF was independently associated with increased total adiposity but did not correlate with AT dysfunction, insulin resistance and secretion or NAFLD. The ADIPO-IR index was capable of predicting NAFLD independently of all confounders, whereas it did not seem to be related to intrapancreatic fat deposition; unlike HFF, higher PFF was not associated with relevant alterations in the metabolic profile. In conclusion, the presence and severity of AT dysfunction may drive ectopic fat accumulation towards specific targets, such as VAT and liver, therefore evaluation of AT dysfunction may contribute to the identification of different risk profiles among T2D patients

    Aphanizomenon flos-aquae (AFA) Extract Prevents Neurodegeneration in the HFD Mouse Model by Modulating Astrocytes and Microglia Activation

    Obesity and related metabolic dysfunctions are associated with neurodegenerative diseases, such as Alzheimer's disease. Aphanizomenon flos-aquae (AFA) is a cyanobacterium considered a suitable supplement for its nutritional profile and beneficial properties. The potential neuroprotective effect of an AFA extract, commercialized as KlamExtra®, including the two AFA extracts Klamin® and AphaMax®, in High-Fat Diet (HFD)-fed mice was explored. Three groups of mice were provided with a standard diet (Lean), HFD or HFD supplemented with AFA extract (HFD + AFA) for 28 weeks. Metabolic parameters, brain insulin resistance, expression of apoptosis biomarkers, modulation of astrocytes and microglia activation markers, and Aβ deposition were analyzed and compared in the brains of different groups. AFA extract treatment attenuated HFD-induced neurodegeneration by reducing insulin resistance and loss of neurons. AFA supplementation improved the expression of synaptic proteins and reduced the HFD-induced astrocytes and microglia activation, and Aβ plaques accumulation. Together, these outcomes indicate that regular intake of AFA extract could benefit the metabolic and neuronal dysfunction caused by HFD, decreasing neuroinflammation and promoting Aβ plaques clearanc

    No effects of oral vitamin D supplementation on non-alcoholic fatty liver disease in patients with type 2 diabetes: a randomized, double-blind, placebo-controlled trial

    Background: Non-alcoholic fatty liver disease (NAFLD) is the most common hepatic disorder worldwide, reaching prevalence up to 90 % in obese patients with type 2 diabetes (T2D), and representing an independent risk factor for cardiovascular mortality. Furthermore, the coexistence of T2D and NAFLD leads to higher incidence of diabetes’ complications and additive detrimental liver outcomes. The existence of a close association between NAFLD and hypovitaminosis D, along with the anti-inflammatory and insulin-sensitizing properties of vitamin D, have been largely described, but vitamin D effects on hepatic fat content have never been tested in a randomized controlled trial. We assessed the efficacy and safety of 24-week oral high-dose vitamin D supplementation in T2D patients with NAFLD. Methods: This randomized, double-blind, placebo-controlled trial was carried out at the Diabetes Centre of Sapienza University, Rome, Italy, to assess oral treatment with cholecalciferol (2000 IU/day) or placebo in T2D patients with NAFLD. The primary endpoint was reduction of hepatic fat fraction (HFF) measured by magnetic resonance; as hepatic outcomes, we also investigated changes in serum transaminases, CK18-M30, N-terminal Procollagen III Propeptide (P3NP) levels, and Fatty Liver Index (FLI). Secondary endpoints were improvement in metabolic (fasting glycaemia, HbA1c, lipids, HOMA-IR, HOMA-β, ADIPO-IR, body fat distribution) and cardiovascular (ankle-brachial index, intima-media thickness, flow-mediated dilatation) parameters from baseline to end of treatment. Results: Sixty-five patients were randomized, 26 (cholecalciferol) and 29 (placebo) subjects completed the study. 25(OH) vitamin D significantly increased in the active treated group (48.15 ± 23.7 to 89.80 ± 23.6 nmol/L, P < 0.001); however, no group differences were found in HFF, transaminases, CK18-M30, P3NP levels or FLI after 24 weeks. Vitamin D neither changed the metabolic profile nor the cardiovascular parameters. Conclusions: Oral high-dose vitamin D supplementation over 24 weeks did not improve hepatic steatosis or metabolic/cardiovascular parameters in T2D patients with NAFLD. Studies with a longer intervention period are warranted for exploring the effect of long time exposure to vitamin D

    Itch/β-arrestin2-dependent non-proteolytic ubiquitylation of SuFu controls Hedgehog signalling and medulloblastoma tumorigenesis

    Suppressor of Fused (SuFu), a tumour suppressor mutated in medulloblastoma, is a central player of Hh signalling, a pathway crucial for development and deregulated in cancer. Although the control of Gli transcription factors by SuFu is critical in Hh signalling, our understanding of the mechanism regulating this key event remains limited. Here, we show that the Itch/β-arrestin2 complex binds SuFu and induces its Lys63-linked polyubiquitylation without affecting its stability. This process increases the association of SuFu with Gli3, promoting the conversion of Gli3 into a repressor, which keeps Hh signalling off. Activation of Hh signalling antagonises the Itch-dependent polyubiquitylation of SuFu. Notably, different SuFu mutations occurring in medulloblastoma patients are insensitive to Itch activity, thus leading to deregulated Hh signalling and enhancing medulloblastoma cell growth. Our findings uncover mechanisms controlling the tumour suppressive functions of SuFu and reveal that their alterations are implicated in medulloblastoma tumorigenesis

    Graphene-Based Interconnects for Stable Dye-Sensitized Solar Modules

    We present Z-Type Dye Sensitized Solar Modules (DSSMs) with screen printed graphene-based vertical interconnects. This prevents corrosion of interconnects in contact with electrolytic species, unlike conventional Ag interconnects. By enlarging the width of single cells, or by increasing the number of cells, we get an enhancement of the aperture power conversion efficiency ∼+12% with respect to Ag-based modules, with 1000 h stability under 85 °C stress test. This paves the way to original design layouts with decreased dead area and increased generated power per aperture area

    Bendamustine in combination with gemcitabine and vinorelbine is an effective regimen as induction chemotherapy before autologous stem-cell transplantation for relapsed or refractory Hodgkin lymphoma: Final results of a multicenter phase II study

    Purpose: This multicenter, open-label, phase II study evaluated the combination of bendamustine, gemcitabine, and vinorelbine (BeGEV) as induction therapy before autologous stem-cell transplantation (ASCT) in patients with relapsed or refractory Hodgkin lymphoma (HL). Patients and Methods: Patients with HL who were refractory to or had relapsed after one previous chemotherapy line were eligible. The primary end point was complete response (CR) rate after four cycles of therapy. Secondary end points were: overall response rate, stem-cell mobilization activity, and toxicity. Progression-free and overall survival were also evaluated. Results: In total, 59 patients were enrolled. After four cycles of therapy, 43 patients (73%) achieved CR, and six (10%) achieved partial response, for an overall response rate of 83%. The most common grade 3 to 4 nonhematologic toxicities included febrile neutropenia (n = 7) and infection (n = 4). Regarding hematologic toxicities, grade 3 to 4 thrombocytopenia and neutropenia were each experienced by eight patients (13.5%). CD34+ cells were successfully harvested in 55 of 57 evaluable patients, and 43 of 49 responding patients underwent ASCT. With a median follow-up of 29 months, the 2-year progression-free and overall survival rates for the total population were 62.2% and 77.6%, respectively. The same figures for patients undergoing autograft were 80.8% and 89.3%, respectively. Conclusion: This phase II study demonstrates that BeGEV is an effective salvage regimen able to induce CR in a high proportion of patients with relapsed or refractory HL before ASCT. These data provide a strong rationale for further development of the BeGEV regimen
