    Mechanistic behaviour and molecular interactions of heat shock protein 47 (HSP47)

    This project involves the study of heat shock protein 47 (HSP47), which is a molecular chaperone crucial for collagen biosynthesis. It exhibits a high degree of sequence homology with members of the serine protease inhibitor (serpin) superfamily, though HSP47 does not possess the inhibitory activity. It is a single-substrate chaperone, and binds only to collagen. ā€˜Knock-outā€™ of the hsp47 gene impairs the secretion of correctly folded collagen triple helix molecules leading to embryonic lethality in mice. Thus the aim of this project was to elucidate the specific mechanism that governs the binding to and release from collagen at the molecular level, known as the ā€˜pH-switch mechanismā€™. Emphasis is given on histidine (His) residues as the HSP47-collagen dissociation pH is similar to the pKa of the imidazole side chain of His residues. Site directed mutagenesis was used to mutate surface His residues, based on a mouse HSP47 homology model. The effects of the mutations on the behaviour of HSP47 were then assessed by collagen binding assays and structural analyses with circular dichroism (CD). All mutants were found to have good solubility and retain their binding ability to collagen like wild-type HSP47 in batch assay, but perturbed behaviour was seen in column experiment. Mutation of His residue at position 191 (H191) causes the shift in the collagen dissociation pH, while mutation of H197 and/or 198 disrupt the specific HSP47-collagen interaction. H191, 197 and 198 are predicted to be located in the region near the C-terminus of strand 3 of Ī²-sheet A (s3A) in the homology model, a region specifically known as the ā€˜breach clusterā€™ in serpin nomenclature. The extent of conformational rearrangement of this region was further investigated by means of intrinsic tryptophan fluorescence spectroscopy using a series of single tryptophan (Trp) mutants. Results from analyses performed on the mutants did not contradict the observation seen in His mutational work, as Trp residues in the ā€˜breachā€™ cluster are likely to be located in the dynamic region of HSP47 pH-triggered conformational change. In conclusion, this study establishes the importance of His residues in the ā€˜breach clusterā€™ to HSP47 pH-switch behaviour. Finally, a model for HSP47 pH-switch mechanism was proposed from data obtained via mutagenesis experiments. The model is hoped to assist future research into HSP47 cellular behaviour and will also be of great use in therapeutic applications involving the molecular chaperone

    Shadira Firdausi is one of the influencers on TikTok social media who uses TikTok social media as a place to build personal branding. Shadira's TikTok account presents DIY and a day in my life content that is uniquely packaged in her style. The @shaturday account has been followed by 4.1 million followers. This research aims to describe the personal branding carried out by Shadira Firdausi through her TikTok social media account. Researchers use the theory of The Eight Laws of Personal Branding by Peter Montoya, where there are 8 concepts in building personal branding. Researchers also use the basic characteristics that must be built when forming personal branding according to Montoya & Vandehey, namely you, promise, and relationship as well as video elements in describing Shadira's personal branding on TikTok social media. Peter Montoya defines personal branding as a process that requires unique skills, personality and characteristics packaged in a strong identity, so that it can be remembered by the public. This research uses a qualitative approach with a descriptive research type and a qualitative content analysis research basis. This research uses documentation techniques in the form of screenshots from the @shaturday account with DIY themed videos. This data was then analyzed using interactive data analysis developed by Miles and Huberman. The research results show that the personal branding built by Shadira Firdausi through her TikTok social media account @shaturday prioritizes Shadira, who is seen as an aesthetic and candid influencer. SDM yang harus ditingkatkan adalah seperti sikap profesional guru dan karyawan, baik secara kognitif, afektif, dan psikomotorik agar dapat menjalankan tugas-tugasnya secara efektif dan efisien. SMP Muhammadiyah 8 Surakarta termasuk salah satu sekolah swasta dengan terakreditasi A. Sekolah ini memiliki kelebihan dalam pembelajaran yang menerapkan sistem fullday school, serta kurikulum sekolah yang memadukan antara kurikulum Depdiknas dan kurikulum Dikdasmen Muhammadiyah. Sekolah yang menerapkan sistem pembelajaran fullday school diharapkan memiliki tenaga pengajar yang lebih profesional dan berkualitas. Kepemimpinan kepala sekolah diharapkan dapat mengedepankan visi dan misi, mengutamakan musyawarah, serta menghargai pendapat dari anggotanya. Tujuan dari penelitian ini adalah untuk mendeskripsikan kepemimpinan kepala sekolah, usaha-usaha dalam meningkatkan SDM, serta problem yang muncul dalam peningkatan SDM dan solusi mengatasinya. Jenis penelitian ini adalah penelitian kualitatif dengan pendekatan deskriptif. Subjek penelitian ini adalah kepala sekolah, guru dan karyawan. Sedangkan teknik pengumpulan data dengan menggunakan metode wawancara, observasi, dan dokumentasi, yang kemudian dianalisis dengan menggunakan metode deskriptif kualitatif. Hasil dari penelitian ini adalah kepemimpinan kepala sekolah terbukti efektif dengan adanya tiga corak gaya kepemimpinan. Usaha-usaha dalam peningkatan SDM melalui MGMP, pengajian, diskusi, pelatihan-pelatihan serta pemberian gaji dan reward.     EDUCATION, INTENSITY OF MOTHERā€™S PERCEPTION SENSORY STIMULATION WITH LANGUAGE DEVELOPMENT OF CHILDREN

    Introduction: Stimulation is an important factor in the childrenā€™s growth and development. According to NHCS-National Center for Health Statistics (2000) in United States, 3.29% of children which is less than 5 years old still have the language development delay. Therefore, the purpose of this study is to analyze the corcorrelation between the education and the intensity of perception sensory stimulation given by the carers and the language development speed of children whose age is 9 ā€“ 12 months in integrated health post of Durung Bedug Baru, Sidoarjo (Integrated Health Post). Method: The design of the study used was a correlative analytic with cross sectional. The population involved 40 mothers and their childrenā€™s age was 9 ā€“ 12 months. 36 respondents were chosen as the samples by using simple random sampling technique. Questionnaires and DDST (Denver Development Screening Test) were used to collect the data which were then analyzed by using Spearmanā€™s Rank statistic test. Moreover, based on the statistic test, the result showed that Ļ = 0.00 < Ī± = 0.05 so that H0 was rejected, it described that there was a corcorrelation between the motherā€™s education and the language development, and Ļ = 0.011 meant that p < Ī± = 0.05. Result: H0 was rejected, it described that there was a corcorrelation between the intensity of carerā€™s perception sensory stimulation and the language development speed of children whose aged were 9 ā€“ 12 months. Discussion: Low education and lack of giving intensity of perception sensory stimulation causes the children have an abnormal language development.     This study aims to analyze the effect of variable number of members and current ratio on the remaining business results (SHU). The sample used is all data for the time series period 2012-2018. The data used are secondary data and the method used is multiple linear regression analysis with the help of SPSS Version 23 program to obtain the influence of the independent variables with the dependent variable.
