    Mastitis y muerte en una oveja Corriedale asociada a una infección con Mycoplasma spp. en la provincia de Buenos Aires

    Mycoplasma spp. infections are associated with different clinical syndromes in ruminants: mastitis, pneumonia, arthritis, otitis, among others. Nevertheless, case reports of clinical diseases associated with Mycoplasma-infections are scarce in small ruminants of Argentina. Mycoplasma agalactiae and Mycoplasma capricolum are the causal agents of contagious agalactia, an exotic disease in the region. Therefore, strict surveillance of these pathogens is needed. Severe mastitis of a 7-years-old Corriedale ewe in a flock from Buenos Aires province is reported. The affected ewe had delivered twin lambs 15 days before the clinical disease was detected and died. During post mortem examination, supramammary lymphadenomegaly was evident. Mammary gland was enlarged and firm, with cyanotic skin and superficial edema; multiple caseous whitish foci were observed in the mammary parenchyma. Similar caseous foci were observed in the caudal-ventral lobes of the right lung. In the histopathological analysis, chronic multifocal necrotizing severe mastitis and fibrinous bronchopneumonia with multifocal necrosis and fibrinous pleurisy were observed. Mycoplasma spp. was isolated from the mammary gland. Mycoplasma spp. nested-PCR resulted positive in mammary gland and lung. Sequencing analysis confirmed the presence of Mycoplasma arginini and Mycoplasma bovis in mammary gland and lung, respectively. The pathological findings were similar to the described in cases of contagious agalactia, therefore, Mycoplasma speciation was decided in order to discard the presence of this exotic disease.Las infecciones con Mycoplasma spp. están asociadas con diferentes cuadros clínicos en rumiantes: mastitis, neumonía, artritis, otitis, entre otros. Sin embargo, existen escasos reportes de cuadros clínicos asociados con estas infecciones en pequeños rumiantes de Argentina. Mycoplasma agalactiae y Mycoplasma capricolum son los agentes causales de la agalactia contagiosa, enfermedad exótica en la región. Entonces, es necesario realizar vigilancia epidemiológica estricta de estos patógenos. Se diagnosticó un cuadro de mastitis severa y muerte en una oveja Corriedale de 7 años de edad, en una majada de la provincia de Buenos Aires. La oveja había parido mellizos 15 días antes que comenzara a manifestar enfermedad clínica y muriera. Durante su necropsia, se observó linfoadenomegalia supramaria, glándula mamaria aumentada de tamaño y firme, con piel cianótica y edema superficial. El parénquima mamario presentaba múltiples focos caseosos blanquecinos. Focos caseosos similares se observaron en el lóbulo caudo-ventral del pulmón derecho. En el análisis histopatológico se observó mastitis necrotizante multifocal severa crónica y bronconeumonía fibrinosa con múltiples focos de necrosis pulmonar y pleuritis fibrinosa. Se aisló Mycoplasma spp. de la glándula mamaria. Mediante nested-PCR, se detectó ADN de Mycoplasma spp. en glándula mamaria y pulmón. Mediante secuenciación se confirmó la presencia de Mycoplasma arginini y Mycoplasma bovis en la glándula mamaria y pulmón, respectivamente. Los hallazgos patológicos fueron similares a los descriptos en casos de agalactia contagiosa, por lo que fue necesario determinar que especies de Mycoplasma estaban presentes para poder descartar la presencia de esta enfermedad exótica.EEA BalcarceFil: Cantón, Germán José.     Diferentes estrategias para el diagnóstico de la brucelosis ovina por brucella ovis en una majada endémica de Argentina

    Brucella ovis es el agente etiológico de la epididimitis contagiosa del carnero. El diagnóstico se basa en el examen clínico, la serología y en el aislamiento de B. ovis a partir de semen. El análisis del semen mediante PCR complementa el diagnóstico directo para la detección de B. ovis y de otros patógenos reproductivos de los ovinos. En este trabajo se comparan diferentes estrategias para el diagnóstico de la epididimitis contagiosa del carnero. Veintidós carneros fueron examinados clínicamente. Las muestras de suero fueron analizadas con diferentes pruebas serológicas. Las muestras de semen se cultivaron y evaluaron mediante PCR multiplex. Los carneros con cultivo positivo fueron sacrificados y se tomaron muestras de testículos, epididídimos, vesículas seminales y ampollas del conducto deferente para el estudio por histopatológico. La presencia de B. ovis en semen se asoció significativamente con la seropositividad y con lesiones histopatológicas en los órganos estudiados. La multiplex PCR mostró una sensibilidad similar al cultivo de semen y podría ser usada para el diagnóstico directo de B. ovis. Aunque este estudio fue realizado en pocos animales, este provee información útil para el diagnóstico de la brucelosis ovina en una majada endémica y refuerza la necesidad de dos controles por año.Brucella ovis is the etiological agent of contagious epididymitis in rams. Prevalence increases when biannual control was not performed in B. ovis endemic flocks. Diagnosis is based on clinical examination, serological tests and bacteriological isolation of B. ovis from semen. Semen analysis by PCR technique complements the direct diagnosis for the detection of B. ovis or other pathogens of reproduction in sheep. In this work, different strategies for the diagnosis of contagious epididymitis in a flock, which this disease have been apparently controlled, were compared. Twenty-two rams were clinically examined and serum samples were analyzed different serological tests. Semen samples were cultured in bacteriological media and were evaluated by multiplex PCR. Rams with a positive culture were necropsied and testes, epididymides, vesicular glands and ampullaes were taken and studied by histopathology. B. ovis shedding in semen was significantly associated with the seropositivity and genital tract histopathological lesions. In our work, multiplex PCR showed a similar sensitivity to semen culture and could be used as a complementary test for the direct diagnosis of B. ovis. Although this study involved a low number of animals, it provides useful information for the diagnosis of ovine brucellosis in an endemic flock and reinforces the need for biannual control.Fil: Fiorentino, Maria Andrea.     Immune response and reproductive consequences in experimentally infected ewes with Brucella ovis during late pregnancy

    La brucelosis ovina por Brucella ovis es una enfermedad de prevalencia alta en Argentina. Para evaluar la patogenicidad de B. ovis y la respuesta serológica durante el último mes de gestación, 6 ovejas se distribuyeron en dos grupos: G1, ovejas preñadas, n = 4 y G2, ovejas no preñadas, n = 2. Tres ovejas del G1 (15 días preparto) y una del G2 fueron inoculadas con B. ovis. Se analizaron muestras de suero mediante diferentes pruebas serológicas. Se realizó aislamiento y PCR a partir de mucus cérvico-vaginal (mcv), placenta y leche. En las muestras de placenta se realizó histopatología. Las hembras del G1 parieron corderos vivos; se detectaron anticuerpos en las ovejas desafiadas del G1 a partir de los 5 días posinoculación. El mcv de las ovejas desafiadas resultó negativo al aislamiento en ambos grupos. Las muestras de leche del G1 fueron positivas por cultivo y PCR a B. ovis. La técnica de PCR resultó positiva en las placentas de las ovejas desafiadas del G1. La histopatología reveló una placentitis necrótica supurativaen una de las ovejas desafiadas. El desafío con B. ovis preparto resultó en la invasión de la placenta y de la glándula mamaria, con la consecuente excreción de la bacteria por leche. La infección con B. ovis indujo una respuesta humoral temprana en las ovejas. La colonización de la placenta por B. ovis y la excreción de la bacteria por la leche sugieren un potencial riesgo de infección activa para los corderos y la posibilidad de que estos se comporten como portadores latentes de la infección.Ovine brucellosis by Brucella ovis is a highly prevalent disease in Argentina. This study aimed to evaluate the pathogenicity of B. ovis and the serological response in ewes during late pregnancy and in their offspring. Six adult ewes were distributed in two groupsGI (pregnant females, n = 4) and G2 (nonpregnant females, n = 2). Three pregnant ewes at 15 days prepartum and one nonpregnant eve were inoculated with B. ovis. Sera of sheep and their offspring were analyzed by different serological tests. Samples of cervicovaginal mucus, placenta and milk were studied by bacteriology. A Brucella genus-specific PCR assay was carried out in placenta and milk samples. Placenta samples were hystopathologically processed. G1 females gave birth to live lambs, but one died hours postpartum. Serological techniques employed detected antibodies in serum of inoculated pregnant animal 5 days postchallenge. Sera of female controls G1 and G2 remained negative throughout the study. Cervicovaginal mucus of infected ewes in G1 and G2 yielded negative results to bacteriology, but B. ovis was isolated from milk. The PCR assay was positive for the placenta and milk from inoculated pregnant ewes. Histopathology revealed necrotic suppurative placentitis in one placenta. However, although results demonstrated that B. ovis can invade the placenta and mammary gland, this bacterium did not cause abortion when it was inoculated intravenously at 15 days prepartum. B. ovis infection induced an early humoral response in pregnant ewes, but their lambs remained seronegative, indicating that there was no transfer of antibodies in infancy.     Evaluation of direct immunofluerescence test for Campylobacter fetus in bull experimentally infected and commensal bacteria from the reproductive tract of bulls

    Campylobacter fetus is a Gram negative bacterium, with 2 relevant subspecies for cattle health: C. fetus subsp. fetus (Cff) and C. fetus subsp. venerealis (Cfv) which cause important reproductive losses. In the present work, four groups of bulls were used: one group to characterize the genital microbiota (G1; n=3) and the other three to evaluate the direct immunofluorescence (DIF) test for the diagnosis of bovine campylobacteriosis in preputial samples (PS) from experimentally infected bulls with Cff (G2; n=3) and Cfv (G3; n=3), and controls (G4; n=4). Seven bacterial strains of different genera were identified in the PS from G1. Other 11 laboratory bacterial strains were included: Cff, Cfv and Cfv biovar Intermedius, 3 Campylobacter non-fetus, 3 Gram negative and 2 Gram positive. Only the C. fetus strains presented positive immunostaining. The sensitivity and specificity of the DIF test were 79 % and 100 %, respectively. Concordance between DIF test and bacteriological culture was 83 % (Kappa index: 0.65). The present study shows the high specificity of the DIF test for detection of C. fetus using a fluorescent conjugate elaborated in Argentina, and highlights the relevance of no cross reaction against 15 bacterial strains normally present in bull preputial microbiota.Campylobacter fetus es una bacteria Gram negativa que incluye 2 subespecies de relevancia sanitaria para el bovino: C. fetus subsp. fetus (Cff) y C. fetus subsp. venerealis (Cfv), causantes de importantes pérdidas reproductivas. En el presente estudio, se utilizaron cuatro grupos de toros: un grupo (G1) para caracterizar la microbiota genital, y otros tres (G2, G3, G4) (n=10) para evaluar el test de inmunofluorescencia directa (IFD) para el diagnóstico de la campilobacteriosis bovina en esmegma prepucial (EP) de toros experimentalmente infectados con Cff y Cfv. Se identificaron siete cepas bacterianas de distinto género del G1. Otras 11 cepas bacterianas de laboratorio fueron incluidas: Cff, Cfv, Cfv biovar Intermedius, 3 Campylobacter no-fetus, 3 Gram negativo y 2 Gram positivo. Solo las cepas de C. fetus presentaron inmunofluorescencia positiva. La sensibilidad y especificidad de la IFD fue de 79 % y 100 %, respectivamente. La concordancia entre la IFD y el cultivo bacteriológico fue de 83 % (Kappa: 0,65). Se demuestra la alta especificidad de la IFD para la detección de C. fetus con un conjugado fluorescente elaborado en Argentina, y se destaca la no reacción cruzada contra 15 cepas bacterianas normalmente presentes en la microbiota prepucial del toro.Fil: García, Juan Agustín.     Neospora caninum and Toxoplasma gondii as causes of reproductive losses in commercial sheep flocks from Argentina

    The aim of this study is to provide preliminary data about the occurrence of N. caninum- and T. gondii-related abortions and perinatal deaths in sheep from Argentina. Thirty ovine aborted foetuses and 33 perinatal deaths were submitted to the Veterinary Diagnostic Service at INTA EEA Balcarce (Argentina) during 2017–2019. A complete necropsy was performed on all specimens submitted, and foetal and placental tissues were examined. Foetal cavity fluids were collected for assessment of antibodies to N. caninum and T. gondii by indirect fluorescent antibody technique (IFAT). Placental and foetal tissue samples were collected for DNA extraction and histopathological analysis. The differential diagnosis with other causes of abortion was carried out. Of the sampled specimens, 20.63% (13/63) displayed evidence for N. caninum infection by IFAT and PCR, and in 61.5% (8/13) of the positive specimens the parasite was confirmed as the cause of abortion/perinatal death based on the presence of compatible histological lesions and/or positive immunohistochemistry test, positive PCR and/or positive IFAT, and no other infectious agents diagnosed. In contrast, T. gondii infection was confirmed in 9.52% (6/63) of the analysed specimens, but only in 2 lambs T. gondii was determined as the death cause. Neospora caninum and T. gondii co-infections were confirmed in 4 analysed specimens (2 aborted foetuses and 2 perinatal deaths). These results demonstrated that N. caninum is efficiently transmitted and a frequent cause of ovine reproductive failure in the commercial analysed flocks compared with T. gondii. Despite T. gondii congenital infection was detected in some specimens (6/63), it was confirmed as the cause of death in only two of them. Thus, and considering the limited availability of confirmed samples, we could not determine whether toxoplasmosis is a major problem in Argentinian sheep flocks or not. More studies on a greater number of specimens from different ovine production systems under different management conditions are necessary to assess the real impact of neosporosis and toxoplasmosis in Argentina.EEA MercedesFil: Della Rosa, Paola. Of the sampled specimens, 20.63% (13/63) displayed evidence for N. caninum infection by IFAT and PCR, and in 61.5% (8/13) of the positive specimens the parasite was confirmed as the cause of abortion/perinatal death based on the presence of compatible histological lesions and/or positive immunohistochemistry test, positive PCR and/or positive IFAT, and no other infectious agents diagnosed. In contrast, T. gondii infection was confirmed in 9.52% (6/63) of the analysed specimens, but only in 2 lambs T. gondii was determined as the death cause. Neospora caninum and T. gondii co-infections were confirmed in 4 analysed specimens (2 aborted foetuses and 2 perinatal deaths). These results demonstrated that N. caninum is efficiently transmitted and a frequent cause of ovine reproductive failure in the commercial analysed flocks compared with T. gondii. Despite T. gondii congenital infection was detected in some specimens (6/63), it was confirmed as the cause of death in only two of them. Thus, and considering the limited availability of confirmed samples, we could not determine whether toxoplasmosis is a major problem in Argentinian sheep flocks or not. More studies on a greater number of specimens from different ovine production systems under different management conditions are necessary to assess the real impact of neosporosis and toxoplasmosis in Argentina.Fil: Della Rosa, Paola.     Brote de abortos ovinos por Campylobacter fetus fetus en Argentina

    Campylobacter infection is one of the major causes of ovine abortionsworldwide. However there are no previous reports of Campylobacter fetussubspecies fetus associated to outbreak ovine abortion in Argentina. Thisstudy constitutes the first report of C. fetus fetus outbreak ovine abortion innaturally infected ewes in Argentina. The problem was presented in a dairyflock of 205 pampinta ewes. In one week 7 abortions were recorded. Some ofthe aborted sheep also retained placentas and showed vaginal discharges.Twins and three placentas were sampled to determine the cause of theabortions. C. fetus fetus was isolated in lung, liver and abomasal fluids fromboth fetuses and in 2 out of 3 placentas. C. fetus fetus DNA was detected in2/3 placentas tested and in lungs of both fetuses. Additionally, N. caninumDNA was amplified in a ewe placenta. No N. caninum or T. gondii serologicaltests were carried out in the ewes, therefore these protozoa infection couldnot be ruled out in the flock. Toxoplasma and Neospora infection are relativelycommon in Argentinean flocks. However, protozoan abortion in smallruminants has not been extensively reported, therefore it is important to ruleout the presence of these pathogens when ovine abortions are registered.Further work is needed in order to quantify the impact of C. fetus fetusinfection in sheep production systems of the region.Fil: Fiorentino, Maria Andrea.     Discovering Europe's seabed geology: the EMODnet concept of uniform collection and harmonization of marine data

    Maritime spatial planning, management of marine resources, environmental assessments and forecasting all require good seabed maps. Similarly there is a need to support the objectives to achieve Good Environmental Status in Europe's seas by 2020, set up by the European Commission's Marine Strategy Framework Directive. Hence the European Commission established the European Marine Observation and Data Network (EMODnet) programme in 2009, which is now in its fourth phase (2019–21). The programme is designed to assemble existing, but fragmented and partly inaccessible, marine data and to create contiguous and publicly available information layers which are interoperable and free of restrictions on use, and which encompass whole marine basins. The EMODnet Geology project is delivering integrated geological map products that include seabed substrates, sedimentation rates, seafloor geology, Quaternary geology, geomorphology, coastal behaviour, geological events such as submarine landslides and earthquakes, and marine mineral occurrences. Additionally, as a new product during the ongoing and preceding phase of the project, map products on submerged landscapes of the European continental shelf have been compiled at various time frames. All new map products have a resolution of 1:100 000, although finer resolution is presented where the underlying data permit. A multi-scale approach is adopted whenever possible. Numerous national seabed mapping programmes worldwide have demonstrated the necessity for proper knowledge of the seafloor. Acting on this, the European Commission established the European Marine Observation and Data Network (EMODnet) programme in 2009. The national geological survey organizations of Europe have a strong network of marine geological teams through the Marine Geology Expert Group of the association of European geological surveys (Eurogeosurveys). This network was the foundation of the EMODnet Geology consortium which today consists of the national geological surveys of Finland, the UK, Sweden, Norway, Denmark, Estonia, Latvia, Lithuania, Poland, The Netherlands, Belgium, France, Ireland, Spain, Italy, Slovenia, Croatia, Albania, Greece, Cyprus, Malta, Russia, Germany, Montenegro and Iceland, as well as marine teams of research organizations in Portugal (IPMA), Bulgaria (IO-BAS), Romania (GeoEcoMar), the UK (CEFAS), Greece (HCMR) and Ukraine (PSRGE, replaced in the fourth phase by Institute of Geological Sciences, NAS of Ukraine). The consortium is further strengthened with experts from six universities: Edge Hill University (UK), Sapienza University of Rome (Italy), University of Tartu (Estonia), University of Crete through FORTH-ICS, Institute of Marine Science and Technology of Dokuz Eylul University (Turkey), and EMCOL Research Centre of Istanbul Technical University – altogether, 30 partners and nine subcontractors. The EMODnet Geology programme is now in its fourth phase, which started in September 2019. In addition to geological information, the wider EMODnet programme aims to also bring together information from European seas on seabed habitats, physical properties, chemistry, biology, human activities and hydrography. This paper describes the EMODnet Geology project and the different end products which were delivered in the end of the third phase and will be further developed during the recent fourth phase of the project

    Obesity elicits immune cell infiltration of adipose tissue provoking chronic low-grade inflammation. Regulatory T cells (Tregs) are specifically reduced in adipose tissue of obese animals. Since interleukin (IL)-21 plays an important role in inducing and maintaining immune-mediated chronic inflammatory processes and negatively regulates Treg differentiation/activity, we hypothesized that it could play a role in obesity-induced insulin resistance. We found IL-21 and IL-21R mRNA expression upregulated in adipose tissue of high-fat diet (HFD) wild-type (WT) mice and in stromal vascular fraction from human obese subjects in parallel to macrophage and inflammatory markers. Interestingly, a larger infiltration of Treg cells was seen in the adipose tissue of IL-21 knockout (KO) mice compared with WT animals fed both normal diet and HFD. In a context of diet-induced obesity, IL-21 KO mice, compared with WT animals, exhibited lower body weight, improved insulin sensitivity, and decreased adipose and hepatic inflammation. This metabolic phenotype is accompanied by a higher induction of interferon regulatory factor 4 (IRF4), a transcriptional regulator of fasting lipolysis in adipose tissue. Our data suggest that IL-21 exerts negative regulation on IRF4 and Treg activity, developing and maintaining adipose tissue inflammation in the obesity state

    Clinical and molecular epidemiology of erythropoietic protoporphyria in Italy

    Background: Erythropoietic protoporphyria (EPP) is a rare inherited disease associated with heme metabolism, characterized by severe life-long photosensitivity and liver involvement. Objectives: To provide epidemiological data of EPP in Italy. Materials and Methods: Prospective/retrospective data of EPP patients were collected by an Italian network of porphyria specialist centres (Gruppo Italiano Porfiria, GrIP) over a 20-year period (1996-2017). Results: In total, 179 patients (79 females) with a clinical and biochemical diagnosis of EPP were assessed, revealing a prevalence of 3.15 cases per million persons and an incidence of 0.13 cases per million persons/year. Incidence significantly increased after 2009 (due to the availability of alfa-melanotide, which effectively limits skin photosensitivity). Mean age at diagnosis was 28 years, with only 22 patients (12.2%) diagnosed 6410 years old. Gene mutations were assessed in 173 (96.6%) patients; most (164; 91.3%) were FECH mutations on one allele in association with the hypomorphic variant, c.315-48C, on the other (classic EPP), and nine (5.2%) were ALAS2 mutations (X-linked EPP). Only one case of autosomal recessive EPP was observed. Of the 42 different FECH mutations, 15 are novel, three mutations collectively accounted for 45.9% (75/164) of the mutations (c.215dupT [27.2%], c.901_902delTG [11.5%] and c.67\u2009+\u20095G\u2009>\u2009A [7.2%]), and frameshift mutations were prevalent (33.3%). A form of light protection was used by 109/179 (60.8%) patients, and 100 (56%) had at least one \u3b1-melanotide implant. Three cases of severe acute liver involvement, requiring OLT, were observed. Conclusions: These data define, for the first time, the clinical and molecular epidemiology of EPP in Italy
