    Action of HMGB1 on miR221/222 cluster in neuroblastoma cell lines

    microRNA (miR/miRNA) are small non-coding RNAs that control gene expression at the post-transcriptional level by targeting mRNAs. Aberrant expression of miRNAs is often observed in different types of cancer. Specific miRNAs function as tumor suppressors or oncogenes and interfere with various aspects of carcinogenesis, including differentiation, proliferation and invasion. Upregulation of miRNAs 221 and 222 has been shown to induce a malignant phenotype in numerous human cancers via inhibition of phosphatase and tensin homolog (PTEN) expression. Neuroblastoma is the most common extracranial solid malignancy in children, which is characterized by cellular heterogeneity that corresponds to different clinical outcomes. The different cellular phenotypes are associated with different gene mutations and miRs that control genetic and epigenetic factors. For this reason miRs are considered a potential therapeutic target in neuroblastoma. The aim of the present study was to investigate the mechanisms by which extracellular high mobility group box 1 (HMGB1) promotes cell growth in neuroblastoma. SK-N-BE(2) and SH-SY5Y neuroblastoma derived cell lines were transfected with the antisense oligonucleotides, anti-miR-221 and -222, followed by treatment with HMGB1 to investigate the expression of the oncosuppressor PTEN. In this study, it was demonstrated that HMGB1, which is released by damaged cells and tumor cells, upregulates miR-221/222 oncogenic clusters in the two human neuroblastoma derived cell lines. The results revealed that the oncogenic cluster miRs 221/222 were more highly expressed by the most undifferentiated cell line [SK-N-BE(2)] compared with the the less tumorigenic cell line (SH-SY5Y) and that exogenous HMGB1 increases this expression. In addition, HMGB1 modulates PTEN expression via miR-221/222, as demonstrated by transiently blocking miR-221/222 with anti-sense oligonucleotides. These results may lead to the development of novel therapeutic strategies for neuroblastoma

    Case Study on Soft Soil Improvement Using Innovative and Cost-Effective Reinforcing Techniques

    This paper describes, discusses and compares three new innovations of reinforcement systems for soft soils: (a) a combination of gridded matrass and piles of bamboo, (b) a combination of matrass and piles of bamboo, and (c) a group of mini piles connected by small H-beams with a compacted top layer to hold the top mini piles. First, illustrations and applications of the three types of reinforcement are described from three full-scale field works. Technical bases for the three types are presented, while technical comparisons are discussed next. Finally, conclusions are developed. The case studies, analysis results, and full-scale fieldwork verifications show that the three reinforcement systems have worked properly. Each system has its own advantages and disadvantages in terms of construction duration and cost, capability/effectiveness and material availability, especially in rural areas

    Behaviors of Pipe-Soil Interaction on Unstable Slopes by Finite Element Simulation

    Pipe-soil interaction plays an important role to the pipe stress analysis subjected to soil movement due to slope instability and/or slope failure. As the soils / slopes begin to fail, a single pipe or a group of pipes buried inside failure zone will bear additional loads which frequently lead to overstress or buckling. To study this phenomena, a case study was carried out by modelling a group of pipes subjected to ground movement in lateral direction. ABAQUS, a finite element software was employed to establish a 3 D numerical model of pipe-soil interaction during landslide event. Discussions of the results of this analysis are presented by focusing on the behaviors of displacement and stress of the pipes due to slope failure. Based on the analysis results, the length of failure zone becomes the major cause of the location of maximum stress during slope failure, it should be assessed carefully

    Sistem pengelolaan arsip memegang peranan penting bagi jalannya suatu organisasi yang dapat bermanfaat sebagai sumber informasi yang dapat dijadikan bahan penilaian dan pengambilan keputusan untuk penyusunan program pengembangan organisasi. Pengarsipan berkas pertanggungjawaban anggaran belanja derah yang dilakukan di Dinas Pemuda dan Olahraga Provinsi Kepulauan Bangka Belitung belum begitu baik dimana sering terjadi kehilangan berkas dan tidak pernah dilakukan backup data sehingga berkas harus dibuat ulang. Untuk itu dirasakan perlu adanya sebuah sistem informasi yang dapat membantu mengatasi permasalahan yang ada. Analisa dan perancangan sistem dalam penelitian ini menggunakan metode berorientasi objek dengan pemodelan UML dan menggunakan tools Rational Rose. Penelitian ini menghasilkan desain sistem informasi pengarsipan yang mampu menghasilkan informasi yang cepat dan akurat bagi pengambilan keputusan

    HMGB1-Induced Cross Talk between PTEN and miRs 221/222 in Thyroid Cancer

    High mobility group box 1 (HMGB1) is an ubiquitous protein that plays different roles in the nucleus, cytoplasm and extra-cellular space. It is an important DAMP molecule that allows communication between damaged or tumor cells and the immune system. Tumor cells exploit HMGB1’s ability to activate intracellular pathways that lead to cell growth and migration. Papillary thyroid cancer is a well differentiated tumor and is often used to study relationships between cells and the inflammatory microenvironment as the latter is characterized by high levels of inflammatory cells and cytokines. Anaplastic thyroid cancer is one of the most lethal human cancers in which many microRNAs and tumor suppressor genes are de-regulated. Up-regulation of microRNAs 221 and 222 has been shown to induce the malignant phenotype in many human cancers via inhibition of PTEN expression. In this study we suggest that extracellular HMGB1 interaction with RAGE enhances expression of oncogenic cluster miR221/222 that in turn inhibits tumor suppressor gene PTEN in two cell lines derived from human thyroid anaplastic and papillary cancers. The newly identified pathway HMGB1/RAGE/miR 221/222 may represent an effective way of tumor escape from immune surveillance that could be used to develop new therapeutic strategies against anaplastic tumors

    Fused Filament Fabrication and Computer Numerical Control Milling in Cultural Heritage Conservation

    This paper reports a comparison between the advantages and disadvantages of fused filament fabrication (FFF) and computer numerical control (CNC) milling, when applied to a specific case of conservation of cultural heritage: the reproduction of four missing columns of a 17th-century tabernacle. To make the replica prototypes, European pine wood (the original material) was used for CNC milling, while polyethylene terephthalate glycol (PETG) was used for FFF printing. Neat materials were chemically and structurally characterized (FTIR, XRD, DSC, contact angle measurement, colorimetry, and bending tests) before and after artificial aging, in order to study their durability. The comparison showed that although both materials are subject to a decrease in crystallinity (an increase in amorphous bands in XRD diffractograms) and mechanical performance with aging, these characteristics are less evident in PETG (E = 1.13 +/- 0.01 GPa and sigma = 60.20 +/- 2.11 MPa after aging), which retains water repellent (ca = 95.96 +/- 5.56 degrees) and colorimetric ( increment E = 2.6) properties. Furthermore, the increase in flexural strain (%) in pine wood, from 3.71 +/- 0.03% to 4.11 +/- 0.02%, makes it not suitable for purpose. Both techniques were then used to produce the same column, showing that for this specific application CNC milling is quicker than FFF, but, at the same time, it is also much more expensive and produces a huge amount of waste material compared to FFF printing. Based on these results, it was assessed that FFF is more suitable for the replication of the specific column. For this reason, only the 3D-printed PETG column was used for the subsequent conservative restoration

    Numerical and Experimental Studies of Wave Propagation Induced by Pile Driving

    . This paper presents results of numerical and experimental studies to predict the peak particle velocity (ppv) induced by a pile driving. By utilizing a professional finite element software, Plaxis 2D Dynamic, this study analyzed ppv due to pile driving in clays for various soil stiffness and various embedded pile lengths. For verification, a full scale field test of pile driving was performed in East Kalimantan with installed instrumentations of accelerations. Results of both instrumentation and numerical analysis show that ppv depends on distance and soil rigidity. The closer the object to pile tip, the larger the ppv that will be produced. The more rigid the soils at the pile tip, the larger the ppv, too. The results also show that both field test and numerical analysis results are comparable. Finally, this paper proposes a chart to predict the ppv of soils due to pile driving in clays. The output of the proposed method is the predicted ppv for various distances from pile driving location

    ARCHAEOLOGICAL MATERIALS FROM GABII (CENTRAL ITALY): KNOWLEDGE OF OFFERINGS AND RITUALS AT THE INFANT BURIALS THROUGH AN INTEGRATED APPROACH

    The ancient Latin city of Gabii is situated 18 km (11.2 miles) to the east of Rome (Central Italy) along the modern Via Prenestina. Gabii was a renowned city in Roman times, particularly during the Republican period and there are various influences in the site that can be identified in Roman culture itself. Gabii is also one of the most significant and important archaeological sites in the territory of the Municipality of Rome and due to its characteristics, it represents today an extraordinary research context. From the excavations carried out in the past it is possible to see how, under the soil, the main structures and buildings of the ancient city are still largely preserved. Among the various testimonies of the past, the tombs, and the micro and macro remains that these contain, represent an opportunity to investigate such practices in the context of Early Iron Age and Orientalizing Latium. In particular, the finds from the Area D baby burials of Gabii enriched the existing dataset so far significantly, allowing us to explore funerary ritual behavior in a more systematic way. This work reports the results of the detailed examination of four tombs (Tombs 30, 50, 51 and 52) of archaeological site. The field strategy for the excavation of the tombs was geared from the start towards both the systematic retrieval of archaeobotanical and zooarchaeological remains and the sampling for organic residue analysis. Aiming for total recovery, the sediments from the tomb fills were sifted in their entirety as their stratigraphic excavation progressed, and samples were taken for flotation. This careful screening allowed for the detection of concentrations of organic material that represent plant and/or animal depositions. The excavation and removal of the grave goods was carried out following strict protocols for residue sampling, minimizing the risk of organic contamination. Samples were analysed by High Temperature Gas Chromatography/Mass Spectrometry (HTGC/MS) and Gas chromatography/Combustion/Isotope ratio mass spectrometry (GC-C-IRMS). For each burial, a subset of vessels including both closed and open shapes was selected, such as cups, open bowl without foot, amphoretta, amphora with dots, Kantharos, plate on a foot, olla, and olpe in bucchero. The results demonstrate the still largely unexploited potential of this sort of integrated studies, encouraging us to expand the application of chemical methods to contexts from other well–controlled excavations

    Scutellaria caucasica A. Ham.: Morphological features and headspace characterization

    In the context of a wide research project, a micromorphological and phytochemical characterization was performed on the vegetative and reproductive organs of Scutellaria caucasica A. Ham. (Lamiaceae), cultivated at the Ghirardi Botanic Garden (Toscolano Maderno, BS, Lombardy, Italy). The morphological survey revealed the presence of both non-glandular and glandular trichomes. The latter belonged to three different morphotypes: peltate, short-stalked and long-stalked capitate. Histochemical assays demonstrated that the terpenes biosynthesis mainly took place in the peltates, while short-stalked capitates secreted only polysaccharides; the long-stalked ones mainly produced polysaccharides, coupled with terpene and polyphenolic fractions. An element of novelty was represented by the characterization of the VOC emission profile. Leaves and flowers showed differences in their emissions: the floral profile had a higher number of compounds than that of the leaves (37 vs 29), with a higher heterogeneity. The almost totality of the leaf profile was characterized by sesquiterpene hydrocarbons (98.8 %), while the flowers presented a more varied composition, with sesquiterpene hydrocarbons (87.2 %), monoterpenes (10.4 % oxygenated, 1.8 % hydrocarbons) and non-terpenes derivatives (0.6 %). The most abundant compounds were \u3b3-muurolene (42.6 %) and \u3b2-caryophyllene (35.0 %) in the leaves and in the flowers, respectively. In the flower headspace, 16 exclusive compounds were identified, among which germacrene D (31.7 %) dominated; leaves had 8 exclusive compounds, with valencene (1.8 %) as the most represented one. 21 common compounds were revealed: \u3b2-caryophyllene (34.1 % leaves; 35.0 % flowers), \u3b1-humulene (3.0 % leaves; 3.1 % flowers), alloaromadendrene (2.4 % leaves; 1.0 % flowers), \u3b1-copaene (2.1 % leaves; 2.7 % flowers) and \u3b2-copaene (2.2 % leaves; 1.5 % flowers) were the most abundant ones. \u3b3-Muurolene relative abundances (42.6 % leaves; 0.7 % flowers) were very different between the two profiles. Overall, this work represented the first multidisciplinary study on S. caucasica, combining a scientific research approach with the policies of the Open Science
