    Development of High Technologies as an Indicator of Modern Industry in the Eu

    The article tries to classify the EU states in terms of the advancement of structural changes in their industries on the basis of the increase in the share of advanced technologies in total industrial production, labour, added value, and surplus. In the times of the knowledge-based economy the ability to produce high-tech goods, the demand for which is growing much faster than for traditional goods, indicates the level of modernity of industry. The dynamic growth of the demand for knowledge-based high-tech goods results from the evolution of consumer habits, which are predominantly driven by educated and rich societies who demand that their sophisticated needs met. An important role in stimulating this demand is played by modern media as they instantly deliver information on the latest technical developments and are very efficient in transferring patterns of consumerism. The ability to adapt the structure of production to the needs of the market is crucial in defining a state's position in the international exchange of goods. This is due to the fact that the national technology is tightly interwoven with export abilities

    Is FLT3 internal tandem duplication an unfavorable risk factor for high risk children with acute myeloid leukemia? : Polish experience

    According to the AML-BFM 2004 Interim, a treatment protocol used in Poland since 2005, presence of FLT3 internal tandem duplication (FLT3/ITD) qualifies a patient with acute myeloid leukemia (AML) to a high-risk group (HRG). The present study was aimed to identify the prevalence of FLT3/ITD in children with AML in Poland and to evaluate its prognostic significance in the HRG patients. Out of 291 children with de novo AML treated in 14 Polish centers between January 2006 and December 2012, samples from 174 patients were available for FLT3/ITD analysis. Among study patients 108 children (61.7%) were qualified to HRG. Genomic DNA samples from bone marrow were tested for identification of FLT3/ITD mutation by PCR amplification of exon 14 and 15 of FLT3 gene. Clinical features and treatment outcome in patients with and without FLT3/ITD were analyzed in the study. The FLT3/ITD was found in 14 (12.9%) of 108 HRG children. There were no significant differences between children with and without FLT3/ITD in age and FAB distribution. The white blood cells count in peripheral blood at diagnosis was significantly higher (p <0.01) in the children with FLT3/ITD. Over 5-year overall survival rate for FLT3/ITD positive children was worse (42.4%) comparing to FLT3/ITD negative children (58.9%), but the statistical difference was not significant. However, over 5-year survivals free from treatment failures were similar. The FLT3/ITD rate (12.9%) observed in the study corresponded to the published data. There was no significant impact of FLT3/ITD mutation on survival rates, although further studies are needed on this subject

    The SR Protein B52/SRp55 Is Required for DNA Topoisomerase I Recruitment to Chromatin, mRNA Release and Transcription Shutdown

    DNA- and RNA-processing pathways are integrated and interconnected in the eukaryotic nucleus to allow efficient gene expression and to maintain genomic stability. The recruitment of DNA Topoisomerase I (Topo I), an enzyme controlling DNA supercoiling and acting as a specific kinase for the SR-protein family of splicing factors, to highly transcribed loci represents a mechanism by which transcription and processing can be coordinated and genomic instability avoided. Here we show that Drosophila Topo I associates with and phosphorylates the SR protein B52. Surprisingly, expression of a high-affinity binding site for B52 in transgenic flies restricted localization, not only of B52, but also of Topo I to this single transcription site, whereas B52 RNAi knockdown induced mis-localization of Topo I in the nucleolus. Impaired delivery of Topo I to a heat shock gene caused retention of the mRNA at its site of transcription and delayed gene deactivation after heat shock. Our data show that B52 delivers Topo I to RNA polymerase II-active chromatin loci and provide the first evidence that DNA topology and mRNA release can be coordinated to control gene expression

In the Netherlands, manual therapy as applied according to the Utrecht School of Manual Therapy (MTU) has not been the subject of a randomized controlled trial. MTU differs in diagnoses and treatment from other forms of manual therapy. METHODS/DESIGN: This is a single blind randomized controlled trial in patients with sub-acute and chronic non specific neck pain. Patients with neck complaints existing for two weeks (minimum) till one year (maximum) will participate in the trial. 180 participants will be recruited in thirteen primary health care centres in the Netherlands.The experimental group will be treated with MTU during a six week period. The control group will be treated with physical therapy (standard care, mainly active exercise therapy), also for a period of six weeks.Primary outcomes are Global Perceived Effect (GPE) and functional status (Neck Disability Index (NDI-DV)). Secondary outcomes are neck pain (Numeric Rating Scale (NRS)), Eurocol, costs and quality of life (SF36).     This paper is aimed at analysing geological and engineering conditions in the sur-rounding of Lake Czerniakowskie. Analysed area comprises lands which are the subject of two presently accomplished local plans of spatial management - Lake Czerniakowskie and Southern Czerniaków. The assessment of local conditions was performed after a complex geological and geo-technical studies and analysis of archival materials. The assessment is a base for elaborating local plans of spatial management. Available grounds of the properties favourable for building have been distinguished. The need for protecting water resources of Lake Czerniakowskie restricts a possibility of laying foundations beneath the ground water table

    Klasyczny cykl obiegu wody w zlewni rzecznej polega na przejściu opadu atmosferycznego poprzez infiltrację efektywną do wód gruntowych, za pośrednictwem których definitywnie odpływa ze zlewni. Badania nad obiegiem wody i substancji chemicznych są wykonywane także w Instytucie Melioracji i Użytków Zielonych w Falentach. Prace koncentrują się głównie nad obiegiem substancji biogennych, których udział w różnych fazach obiegu wody w zlewni pozostaje w bezpośrednim związku z produkcją rolniczą, hodowlaną i przeobrażeniami wiejskich jednostek osadniczych. Obiekt doświadczalny w Falentach jest położony w bezpośrednim sąsiedztwie Instytutu w zlewni Raszynki. Prace są wykonywane w mikrozlewni tzw. Cieku Wschodniego spod Laszczek. Opróbowaniu poddano główne fazy cyklu obiegu wody, od opadu atmosferycznego poprzez wody infiltracyjne w strefie saturacji do wód podziemnych. Dzięki opróbowaniu wód infiltracyjnych oraz wód ze strefy saturacji na zróżnicowanych głębokościach uzyskano pełen obraz zmian chemicznego składu wód w profilu pionowym. Wykonane badania potwierdzają zasadność kompleksowego i jednoczesnego rozpoznania jakości wody w poszczególnych fazach krążenia w zlewni rzecznej.A typical water cycle in a river catchment consists in moving atmospheric precipitation through effective infiltration to ground waters, which carry the waters out of the catchment. Studies on the turnover of water and chemical substances are carried out at the Institute for Land Reclamation and Grassland Farming in Falenty. The studies focus mostly on the turnover of nutrients, concentrations of which are closely related to agricultural production, breeding and transformations of rural settlements. Experimental plot of the Institute is situated in its close vicinity in the catchment of the Raszynka River. Studies were carried out in a micro-catchment of the so-called “stream from Laszczki”. Main components of the water cycle, i.e. atmospheric precipitation, infiltration waters in a saturation zone and ground waters were sampled. The effect of entire vertical sampling of the ground-water stratum was achieved thanks to the sampling sites installed at various depths. The studies confirmed a rationale for the complex and simultaneous recognition of water quality in particular phases of cycling in the river catchment

    RhoGAP19D inhibits Cdc42 laterally to control epithelial cell shape and prevent invasion

    Cdc42-GTP is required for apical domain formation in epithelial cells, where it recruits and activates the Par-6-aPKC polarity complex, but how the activity of Cdc42 itself is restricted apically is unclear. We used sequence analysis and 3D structural modeling to determine which Drosophila GTPase-activating proteins (GAPs) are likely to interact with Cdc42 and identified RhoGAP19D as the only high-probability Cdc42GAP required for polarity in the follicular epithelium. RhoGAP19D is recruited by \u3b1-catenin to lateral E-cadherin adhesion complexes, resulting in exclusion of active Cdc42 from the lateral domain. rhogap19d mutants therefore lead to lateral Cdc42 activity, which expands the apical domain through increased Par-6/aPKC activity and stimulates lateral contractility through the myosin light chain kinase, Genghis khan (MRCK). This causes buckling of the epithelium and invasion into the adjacent tissue, a phenotype resembling that of precancerous breast lesions. Thus, RhoGAP19D couples lateral cadherin adhesion to the apical localization of active Cdc42, thereby suppressing epithelial invasion
