    Spatio-temporal modelling of extreme storms

    A flexible spatio-temporal model is implemented to analyse extreme extra-tropical cyclones objectively identified over the Atlantic and Europe in 6-hourly re-analyses from 1979-2009. Spatial variation in the extremal properties of the cyclones is captured using a 150 cell spatial regularisation, latitude as a covariate, and spatial random effects. The North Atlantic Oscillation (NAO) is also used as a covariate and is found to have a significant effect on intensifying extremal storm behaviour, especially over Northern Europe and the Iberian peninsula.     How to enhance crop production and nitrogen fluxes? A result-oriented scheme to evaluate best agri-environmental measures in Veneto Region, Italy

    The cost-effectiveness of adopting agri-environmental measures (AEMs) in Europe, which combine agricultural productions with reduced N losses, is debated due to poorly targeted site-specific funding that is allocated regardless of local variability. An integrated DAYCENT model-GIS platform was developed combining pedo-climatic and agricultural systems information. The aim was to evaluate best strategies to improve N fluxes of agro-ecosystems within a perspective of sustainable intensification. Indicators of agronomic efficiency and environmental quality were considered. The results showed that agronomic benefits were observed with a continuous soil cover (conservation agriculture and cover crops), which enhanced nitrogen use efficiency (+17%) and crop yields (+34%), although in some cases these might be overestimated due to modelling limitations. An overall environmental improvement was found with continuous soil cover and long-term change from mineral to organic inputs (NLeach 45 Mg ha 121), which were effective in the sandy soils of western and eastern Veneto with low SOM, improving the soil-water balance and nutrients availability over time. Results suggest that AEM subsidies should be allocated at a site-specific level that includes pedo-climatic variability, following a result-oriented approach

    Adjuvants : an essential component of neisseria vaccines

    Adjuvants may be classified into delivery systems and immune potentiator or modulator molecules based on their mechanism of action. Neisseria vaccines containing traditional adjuvants such as aluminium salts have existed for long time, but meningitis caused by Neisseria meningitidis serogroups, particularly serogroup B, continues to be a global health problem. Novel strategies have applied in silico and recombinant technologies to develop "universal" antigens (e.g. proteins, peptides and plasmid DNA) for vaccines, but these antigens have been shown to be poorly immunogenic even when alum adjuvanted, implying a need for better vaccine design. In this work we review the use of natural, detoxified, or synthetic molecules in combination with antigens to activate the innate immune system and to modulate the adaptive immune responses. In the main, antigenic and imune potentiator signals are delivered using nano-, micro-particles, alum, or emulsions. The importance of interaction between adjuvants and antigens to activate and target dendritic cells, the bridge between the innate and adaptive immune systems, will be discussed. In addition, nasal vaccine strategies based on the development of mucosal adjuvants and Neisseria derivatives to eliminate the pathogen at the site of infection provide promising adjuvants effective not only against respiratory pathogens, but also against pathogens responsible for enteric and sexually transmitted diseases

    Variable stars in the open cluster NGC 6791 and its surrounding field

    Aims: This work presents a high--precision variability survey in the field of the old, super metal-rich open cluster NGC 6791. Methods: The data sample consists of more than 75,000 high-precision CCD time series measurements in the V band obtained mainly at the Canada-France-Hawaii Telescope, with additional data from S. Pedro Martir and Loiano observatories, over a time span of ten nights. The field covers an area of 42x28 arcmin^2. Results: We have discovered 260 new variables and re-determined periods and amplitudes of 70 known variable stars. By means of a photometric evaluation of the membership in NGC 6791, and a preliminary membership based on the proper motions, we give a full description of the variable content of the cluster and surrounding field in the range 16<V<23.5. Accurate periods can be given for the variables with P<4.0 d, while for ones with longer periods the limited time-baseline hampered precise determinations. We categorized the entire sample as follows: 6 pulsating, 3 irregular, 3 cataclysmic, 89 rotational variables and 61 eclipsing systems; moreover, we detected 168 candidate variables for which we cannot give a variability class since their periods are much longer than our time baseline. Conclusions: On the basis of photometric considerations, and of the positions of the stars with respect to the center of the cluster, we inferred that 11 new variable stars are likely members of the cluster, for 22 stars the membership is doubtful and 137 are likely non-members. We also detected an outburst of about 3 mag in the light curve of a very faint blue star belonging to the cluster and we suggest that this star could be a new U Gem (dwarf nova) cataclysmic variable.Comment: 24 pages, 19 Figures, A&A accepte

    The Marine Fishes of Broward County, Florida: Final Report of 1998-2002 Survey Results

    We inventoried fishes associated with three hardbottom reef tracts that are separated by sand and run parallel to the coast in sequentially deeper water offshore Broward County, Florida. Using SCUBA and the Bohnsack-Bannerot visual point count method, we recorded fish abundance, species richness, sizes (TL), and general habitat characteristics within an imaginary 15m cylinder extending from the substrate to the surface. Sites were sampled along transects at quarter nautical mile intervals along 18 nautical miles of coastline at western and eastern edges, and crest of each of the three reef tracts. A total of 86,463 fishes belonging to 208 species and 52 families was censused from 667 sites over four years (August 1998 to November 2002). Mean species richness, mean total abundance and mean total biomass of fishes increased significantly on each reef tract moving offshore (p Differences were found within reef tracts based on edge or crest sites and position along reefs north or south of Port Everglades and Hillsboro Inlet. Sites within 5 na. mi. south of Port Everglades had lower total abundance and species richness (p Of management interest, was a scarcity or absence of groupers and snappers observed over four years. Although juvenile red grouper were frequently seen (n = 232 at 667 sites), only two were above legal minimum size. No goliath or black grouper were recorded. A total of 10 gag, yellowfin, or scamp grouper was observed; none were legal. Among six snapper species, 219 of 718 were of legal size

    Cochleates derived from Vibrio cholerae O1 proteoliposomes : The impact of structure transformation on mucosal immunisation

    Cochleates are phospholipid-calcium precipitates derived from the interaction of anionic lipid vesicles with divalent cations. Proteoliposomes from bacteria may also be used as a source of negatively charged components, to induce calcium-cochleate formation. In this study, proteoliposomes from V. cholerae O1 (PLc) (sized 160.7±1.6 nm) were transformed into larger (16.3±4.6 µm) cochleate-like structures (named Adjuvant Finlay Cochleate 2, AFCo2) and evaluated by electron microscopy (EM). Measurements from transmission EM (TEM) showed the structures had a similar size to that previously reported using light microscopy, while observations from scanning electron microscopy (SEM) indicated that the structures were multilayered and of cochleate-like formation. The edges of the AFCo2 structures appeared to have spaces that allowed penetration of negative stain or Ovalbumin labeled with Texas Red (OVA-TR) observed by epi-fluorescence microscopy. In addition, freeze fracture electron microscopy confirmed that the AFCo2 structures consisted of multiple overlapping layers, which corresponds to previous descriptions of cochleates. TEM also showed that small vesicles co-existed with the larger cochleate structures, and in vitro treatment with a calcium chelator caused the AFCo2 to unfold and reassemble into small proteoliposome-like structures. Using OVA as a model antigen, we demonstrated the potential loading capacity of a heterologous antigen and in vivo studies showed that with simple admixing and administration via intragastric and intranasal routes AFCo2 provided enhanced adjuvant properties compared with PLc

    Identification of New Genes Related to Virulence of Xanthomonas axonopodis Pv. Citri during Citrus Host Interactions

    A mutant library of the bacterium Xanthomonas citri subsp. citri strain 306 pathotype A (Xac), the causative agent of most aggressive Asiatic type A citrus canker, was screened regarding altered canker symptoms after inoculations into Citrus sinensis and Citrus limonia host leaves. Twenty-six mutants have shown phenotypic virulence changes and have respectively knocked out gene identified by sequencing. In vivo growth curves were obtained for nine mutants to quantify how the mutations could affect pathogen's adaptability to growth inside and attack host plant infected tissue. Among identified genes in mutated strains, we could find those that until now had not been reported as being involved in Xac adaptation and/or virulence, such as predicted to encode for xylose repressor-like protein (XAC Delta xylR), Fe-S oxidoredutase (XAC Delta aslB), helicase IV (XAC Delta helD), ubiquinol cytochrome c oxidoreductase iron-sulfur subunit (XAC Delta petA), chromosome partitioning protein (XAC Delta parB) and cell division protein FtsB (XAC Delta ftsB), in addition to genes predicted to encode for hypothetical proteins.
