    Multiple Centroid Methodology to analyze genotype adaptability

    This study was carried out to propose a modification of the centroid method. The method is modified in thechoice of the ideotype, to be defined according to the researcher's interest, using the bi-segmented regression model, ratherthan based on experimental data, which limit the comparison of genotypes. To illustrate the method, one trait was simulatedin 25 genotypes in 8 environments. For the simulations and statistical analyses the software package GENES was used. Themultiple centroid is more flexible than the original centroid method, since it can be used according to the researcher's objectiveand the desired recommendation strategy. It is also readily interpretable for recommendation and unambiguous. Besides, thenumber of genotypes of interest can be compared, making a detailed study possible, by separating the genotypes intorecommendation classes

    DNA de alta qualidade isolado a partir do córtex de Euterpe edulis Mart. (Arecaceae)

    Euterpe edulis is an arboreal species of high size that occurs naturally in the Atlantic Forest and in ‘Cerrado’ forests. For these characteristics, it is difficult to collect young, healthy and fresh leaves to obtain the genomic DNA of sufficient quality for the species, required for molecular biology studies. To facilitate the collection of biological material field, this study proposed the isolation from plant tissue in the cortex. Isolation and analyzes were carried out compared with the DNA obtained from young leaves coming from plants and tissue of the cortex of adult plants. The integrity of the DNA obtained was confirmed by electrophoresis, revealing the genomic DNA degradation and the low quality of the purification was checked by absorbance ratios criteria 260/280 nm and 260/230 nm, with a similar pattern in both types of tissues evaluated. DNA concentrations ranged from 81.9 to 310.1 ng/µl and cortex, from 105.2 to 404.8 ng/µl. The confirmation of the quality of the DNA obtained was determined by microsatellite amplification, where the products showed bands defined for both plant materials. Thus, the genomic DNA isolated from the cortex of Euterpe edulis, constitutes an additional plant tissue to obtain a genomic DNA of equal quality, quantity and integrity of the obtained sheets with the advantage of the collecting being facilitated in the field.Euterpe edulis (palmiteiro juçara) é uma arbórea de porte alto que ocorre naturalmente em florestas da Mata Atlântica e Cerrado. Por estas características é difícil coletar folhas jovens, sadias e frescas para obtenção de DNA genômico de boa qualidade para a espécie, necessário para estudos de biologia molecular. Visando facilitar coletas a campo de material biológico, neste estudo foi proposta uma nova metodologia de extração a partir de tecidos vegetais do córtex. Foram comparadas extrações e análises realizadas com DNA obtido de folhas jovens oriundas de mudas e de tecido do córtex de plantas adultas. A integridade do DNA obtido foi confirmada por eletroforese, revelando DNA genômico com pouca degradação e a qualidade da purificação foi verificada pelos critérios de razões de absorbâncias de 260/280 nm e 260/230 nm, apresentando padrão similar nos dois tipos de tecidos avaliados. As concentrações de DNA em folha variaram de 81,9 a 310,1 ng/μL, e no córtex, de 105,2 a 404,8 ng/μL. A confirmação da qualidade do DNA obtido foi determinada por amplificação de microssatélites onde os produtos apresentaram bandas definidas para os dois materiais vegetais. Assim o DNA genômico extraído a partir do córtex de Euterpe edulis, constitui um tecido vegetal adicional para obtenção de DNA genômico de igual qualidade, quantidade e integridade que o obtido de folhas, com a vantagem da coleta ser facilitada a campo

    Validation of microsatellite markers for assisted selection of soybean resistance to cyst nematode races 3 and 14

    O objetivo deste trabalho foi validar marcadores microssatélites associados à resistência às raças 3 e 14 do nematóide-de-cisto (Heterodera glycines Ichinohe) da soja (Glycine max L.), para serem utilizados em programas de seleção assistida por marcadores moleculares (SAM). Microssatélites dos grupos de ligação A2, D2 e G da soja foram testados em duas populações, e suas eficiências de seleção foram determinadas. As populações foram 65 famílias F2:3, do cruzamento Msoy8001 (resistente) x Conquista (suscetível), e 66 famílias F2:3, do cruzamento S5995 (resistente) x Renascença (suscetível), avaliadas para a resistência às raças 3 e 14, respectivamente. Famílias com índice de fêmeas de até 30% foram consideradas moderadamente resistentes. Marcadores dos grupos de ligação A2 e G apresentaram associação com a resistência à raça 3. Os marcadores Satt309 e GMENOD2B explicaram a maior proporção da variância fenotípica nos diferentes grupos. As combinações Satt309+GMENOD2B e Satt309+Satt187 apresentaram eficiência de seleção de 100%. A resistência à raça 14 foi associada com marcadores do grupo de ligação G, e a eficiência de seleção da combinação Satt309+Satt356 foi de 100%. Os diferenciais de seleção fenotípica e de seleção assistida mostraram que os dois tipos de seleção podem proporcionar ganhos similares.The objective of this work was to validate microsatellite markers associated with resistance to soybean cyst nematode (Heterodera glycines Ichinohe) races 3 and 14, in soybean (Glycine max L.) genotypes, for use in marker-assisted selection (MAS) programs. Microsatellites of soybean linkage groups A2, D2 and G were tested in two populations, and their selection efficiencies were determined. The populations were 65 F2:3 families from Msoy8001 (resistant) x Conquista (susceptible) cross, and 66 F2:3 families of S5995 (resistant) x Renascença (susceptible) cross, evaluated for resistance to races 3 and 14, respectively. Families with female index up to 30% were considered moderately resistant. Markers of A2 and G linkage groups were associated with resistance to race 3. Markers Satt309 and GMENOD2B explained the greatest proportion of phenotypic variance in the different groups. The combinations Satt309+GMENOD2B and Satt309+Satt187 presented 100% selection efficiency. Resistance to race 14 was associated with markers of G linkage group, and selection efficiency in the Satt309+Satt356 combination was 100%. The selection differential obtained by phenotypic and marker assisted selection showed that both can result in similar gains

    Alternative methodology for Scott-Knott test

    The test proposed by Scott Knott (1974), a procedure of means grouping, is an effective alternative to performprocedures of multiple comparisons without ambiguity. This study aimed to propose a modification related to the partitioningand means grouping in the said procedure, to obtain results without ambiguity among treatments, organized in morehomogeneous groups. In the proposed methodology, treatments that did not participate in the initial group are joined for a newanalysis, which allows for a better group distribution. In a comparative study, four experiments were simulated in a randomizedcomplete block design. The first consisted of 10 and the other 3 of 100 treatments. All experiments were performed in threereplications at a significance level of 0.05 for the means grouping test. Only in the third experiment of those of 100 treatmentsthe groups formed by Scott-Knott did not differ from the methodology proposed here. The proposed methodology is consideredeffective, aiming at the identification of elite cultivar groups for recommendation

    Physiological quality of ‘BRS’ passion fruit seeds grown at different altitudes and storage environments

    The aim of this work was to study the physiological quality of ‘BRS’ passion fruit seeds grown at different altitudes stored in cold chamber and in natural environment for six months.  Hybrid passion fruit seeds (BRS Gigante Amarelo, BRS Sol do Cerrado and BRS Rubi do Cerrado) grown at three altitudes (104; 711; 1016 meters) in the southern region of the state of Espírito Santo were used. Germination was carried out in germitest paper rolls, placed in BOD-type germination chambers at temperature of 20-30 ºC. The following parameters were analyzed: germination, germination speed index, shoot and root length and dry mass of seedlings. The experimental design used was completely randomized in a factorial scheme with split plot in time 4x(3x3) (four bimonthly analyses x three hybrids x three altitudes), with two independent storage environments and three replicates of 100 seeds. Hybrid Passiflora edulis f. flavicarpa seeds can be stored in natural environment or cold chamber. Hybrid yellow passion fruit seeds grown at altitudes of 104 and 711 meters show higher germination and vigor percentages. Hybrid ‘Gigante Amarelo’ seeds had lower averages for variables under study, regardless of altitude. Hybrid ‘Rubi do Cerrado’ seeds grown at altitude of 1016 meters and hybrid ‘Sol do Cerrado’ seeds grown at altitudes of 104 and 711 meters have higher germination and vigor percentages and can be stored for 180 days in natural environment or cold chamber without viability loss.     Vegetative growth of 28 genotypes of Coffea canephora at 850 meters of altitude

    Coffea canephora plants grown at low or elevated temperatures undergo changes in photosynthetic processes, which have a direct impact on vegetative growth. This species shows plants with distinct stress tolerance mechanisms and a great genetic diversity; however, there are no studies concerning the study of altitude on plant growth for these genotypes. The objective of this study was to evaluate the growth rates of orthotropic and plagiotropic branches from 28 Coffea canephora genotypes, ca. 3 years of age, cultivated at 850 m of altitude. The experimental design was a randomized complete block design with four repetitions. Each repetition was compounded by five plants. The period of the experimental evaluation was from 01/23/2013 to 07/07/2013. We selected and identified the orthotropic and plagiotropic branches, which were evaluated bimonthly according to their length (mm), assessing the growth rates. The results were associated with climatic factors that occurred during the experimental period. The genetic material of Coffea canephora evaluated in the study shows genetic diversity to the characteristics growth rate of orthotropic and plagiotropic branches, thus, it is possible to select genotypes with potential for cultivation at 850 m of altitude. In general, genotypes of Coffea canephora exhibited variations in growth rates, mainly in response to changes in temperature, with a sharp and constant drop in growth rates between the period from 05/05/2013 to 05/ 23/2013, when the recorded temperatures were always below 13 °C. Genotypes A1, NV2 and NV1 can be pointed as the ones which showed the best vegetative growth results at 850 meters of altitude.publishersversionpublishe

    Variability of Root System Size and Distribution among Coffea canephora Genotypes

    This work aimed to evaluate the variability in the distribution of the root system among genotypes of C. canephora cv. Conilon and indicate management strategies for a more efficient mineral fertilization. Root distribution was evaluated in six genotypes. The experimental design was in randomized blocks with three replications. Soil monoliths measuring about 27 cm3 were collected at six different soil depths, at three row distances and nine distances of inter-row planting. The collections were carried out in one plant of each repetition. In total, 1296 samples were evaluated. The roots were washed, digitized and processed to quantify length density, volume, surface area and diameter. The distribution of the root system was characterized using semivariograms. It was observed that the highest concentration of roots occurred in the distances close to the irrigation drippers. There was variation in the distribution of the root system among the genotypes. However, in general, the root system is concentrated at a depth of 0 to 20 cm in the soil, at distances up to 50 cm in the planting row and up to 60 cm in inter-rows.     Genetic divergence toward the selection of promising bean progenitors via mixed multivariate models

    Abstract The genetic variability present in the bean (Phaseolus vulgaris L.) germplasm that is currently used as an agricultural crop has been shown to be stable in production and is acceptable for human sustenance. Accordingly, to maintain as much of the available variability as possible, this study aimed to examine the genetic divergence in the bean using multivariate analysis to identify the sources of genetic variability and enable breeders to recognize genetic combinations that have a greater chances of success before crossings are performed. This study was conducted in a randomized block design with three replications in the agricultural year 2015. The agronomic traits evaluated were the stem diameter (DIAM) in millimeters, plant height (PH) in centimeters, number of seedsper plant (NS), protein percentage (PROT), height of the first pod (HFP) in centimeters, pod number (PN), grain mass per plant (GM) in g plant−1, grain yield (GY) in kg ha−1, and straw yield (SY) in kg ha−1. To enable selection of the most divergent genotypes, twenty different genotypes were analyzed via clustering according to the average linkage criterion (UPGMA) using a matrix of the mean standardized Euclidean distances and principal component analysis based on the values predicted via a multivariate mixed model. The results obtained in this study revealed a high degree of genetic divergence and allowed the progenies to be allocated into different groups, as well as recommended crossings for future bean breeding programs.Resumen La variabilidad genética presente en el germoplasma de frijol (Phaseolus vulgaris L.) actualmente utilizada en la agricultura es la garantía más pronunciada de estabilidad de la producción y sustento humano en relación con este cultivo. En consecuencia, para mantener la mayor variabilidad disponible posible, este estudio tuvo como objetivo examinar la divergencia genética mediante análisis multivariante para identificar fuentes de variabilidad genética y permitir a los mejoradores reconocer las combinaciones genéticas con mayores posibilidades de éxito antes de que se realicen los cruces. El experimento se realizó en diseño de bloques al azar con tres repeticiones en el año agrícola 2015. Los caracteres agronómicos evaluados fueron: diámetro del tallo (DIAM) en mm; altura (ALT) en cm; cantidad de semillas (CS); porcentaje de proteína (PROT); altura de la primera vaina (APV) en cm; número de pod (NP); masa de grano por planta (MG) en g planta−1; rendimiento de grano (RG) en kg ha−1; y rendimiento de paja (RP) en kg ha−1. Para seleccionar los genotipos más divergentes, se analizaron 20 diferentes agrupando según el criterio de ligamiento promedio (UPGMA) usando la matriz de distancias euclidianas estandarizadas medias, y el análisis de componentes principales en base a los valores predichos mediante el modelo mixto multivariante. Los resultados obtenidos en este estudio revelaron un alto grado de divergencia genética y permitieron la asignación de las progenies en diferentes grupos, así como recomendaciones para cruces en futuros programas de mejoramiento de frijol

    Strategies for sampling and establishment of core collections

    O objetivo deste trabalho foi avaliar a influência da intensidade de amostragem, do tamanho da coleção de germoplasma inicial e da variância da amostragem sobre a qualidade das respectivas coleções nucleares, quanto à representatividade das coleções iniciais. Foram simulados sete tamanhos de coleções iniciais e utilizadas seis intensidades de amostragem para estabelecimento de coleções nucleares, utilizando caracteres morfoagronômicos. Determinaram-se o número de grupos formados, o coeficiente de coincidência entre a coleção nuclear e a coleção inicial e o coeficiente de determinação dos acessos amostrados para comporem a coleção nuclear. Também foi proposto o uso de uma estratégia alternativa para estabelecer coleções nucleares, de forma a maximizar a diversidade entre os acessos. O tamanho da coleção inicial influencia a intensidade de amostragem empregada na obtenção da coleção nuclear. A amostragem de acessos pelo método de Tocher, com critério de aglomeração inverso, mostrou-se eficiente na obtenção de coleções nucleares. As diferentes magnitudes de variância das coleções iniciais não influenciaram os coeficientes de determinação (R2) nem os coeficientes de coincidência entre a coleção inicial e as respectivas coleções nucleares.The objective of this work was to evaluate the effect of sampling intensity, initial collection's size and its variance over the quality of the respective core collections, in terms of representation of the initial collections. Therefore, simulations were performed for seven sizes of the initial collections and six sampling intensities for the establishment of core collections were used, comprising morpho-agronomic traits. Groups formed were determined, as well as the coincidence coefficient between the core and the initial collection, and the determination coefficient of the accessions sampling to compose the core collection. The use of an alternative strategy was proposed to establish core collections, this strategy maximizes the diversity among the accessions. The size of initial collections affects the sampling intensity to be used to obtain a core collection. The selection of accessions with the method of Tocher, comprising criteria of inverse grouping, was efficient for the formation of the core collections. The different magnitudes of variance of the initial collections did not affect either the coefficient of determination (R2) or the coincidence rate between initial collections and respective core collection
