    Towards a holistic understanding of pastoralism

    Pastoralism is a globally-important livelihood, with large social, environmental and economic importance across much of our planet. Yet, it is also a vulnerable practice with widespread crises, urgently calling for better systemic understanding. The current disciplinary compartmentalization of research not only hampers this but allows perpetuation of unfortunate misconceptions. Furthermore, a long-standing marginalization of the livelihood prevails, with pastoralism being largely overlooked in international environmental and economic forums or sustainability agendas. Here we call for transformative approaches to pastoralism research that can advance an integrated understanding of these social-ecological systems through a comparative lens. We develop a framework that uses: i) timescales from the distant past to the present, ii)social, economic and environmental dimensions, and iii) diverse geographic contexts and scales, to capture emerging properties allowing for cross-cultural comparisons. We provide specific guidelines for formally developing a coherent set of sustainability indicators that are transferable across time and space, and can track sustainability. In an exploratory exercise, we also show that very distinct pastoralist systems have undergone similar transitions across time, approaching critical thresholds and then either collapsing or recovering. An integrated view of the interactions between the environmental, social and economic dimensions of these transitions allows for an improved understanding of potential tipping points, hence supporting more proactive and informed decision-making. We conclude that the need for a paradigm shift in pastoralism science and policy is pressing. Determining when, where and how is pastoralism not only sustainable, but also the most adaptive livelihood, has become a priority. This paper is based on Manzano et al. (2021).Peer reviewe

    Glioblastoma on a microfluidic chip: Generating pseudopalisades and enhancing aggressiveness through blood vessel obstruction events

    Background: Glioblastoma (GBM) is one of the most lethal tumor types. Hypercellular regions, named pseudo- palisades, are characteristic in these tumors and have been hypothesized to be waves of migrating glioblastoma cells.These “waves” of cells are thought to be induced by oxygen and nutrient depletion caused by tumor-induced blood vessel occlusion. Although the universal presence of these structures in GBM tumors suggests that they may play an instrumental role in GBM’s spread and invasion, the recreation of these structures in vitro has remained challenging. Methods: Here we present a new microfluidic model of GBM that mimics the dynamics of pseudopalisade forma- tion.To do this, we embedded U-251 MG cells within a collagen hydrogel in a custom-designed microfluidic device. By controlling the medium flow through lateral microchannels, we can mimic and control blood-vessel obstruction events associated with this disease. Results: Through the use of this new system, we show that nutrient and oxygen starvation triggers a strong migratory process leading to pseudopalisade generation in vitro.These results validate the hypothesis of pseudo- palisade formation and show an excellent agreement with a systems-biology model based on a hypoxia-driven phenomenon. Conclusions: This paper shows the potential of microfluidic devices as advanced artificial systems capable of mod- eling in vivo nutrient and oxygen gradients during tumor evolution

    Unique case of esophageal rupture after a fall from height

    <p>Abstract</p> <p>Background</p> <p>Traumatic ruptures of the esophagus are relatively rare. This condition is associated with high morbidity and mortality. Most traumatic ruptures occur after motor vehicle accidents.</p> <p>Case Presentation</p> <p>We describe a unique case of a 23 year old woman that presented at our trauma resuscitation room after a fall from 8 meters. During physical examination there were no clinical signs of life-threatening injuries. She did however have a massive amount of subcutaneous emphysema of the chest and neck and pneumomediastinum. Flexible laryngoscopy revealed a lesion in the upper esophagus just below the level of the upper esophageal sphincter. Despite preventive administration of intravenous antibiotics and nutrition via a nasogastric tube, the patient developed a cervical abscess, which drained spontaneously. Normal diet was gradually resumed after 2.5 weeks and the patient was discharged in a reasonable condition 3 weeks after the accident.</p> <p>Conclusions</p> <p>This case report presents a high cervical esophageal rupture without associated local injuries after a fall from height.</p

    Global importance of Indigenous Peoples, their lands, and knowledge systems for saving the world's primates from extinction

    Publisher Copyright: Copyright © 2022 The Authors, some rights reserved.Primates, represented by 521 species, are distributed across 91 countries primarily in the Neotropic, Afrotropic, and Indo-Malayan realms. Primates inhabit a wide range of habitats and play critical roles in sustaining healthy ecosystems that benefit human and nonhuman communities. Approximately 68% of primate species are threatened with extinction because of global pressures to convert their habitats for agricultural production and the extraction of natural resources. Here, we review the scientific literature and conduct a spatial analysis to assess the significance of Indigenous Peoples' lands in safeguarding primate biodiversity. We found that Indigenous Peoples' lands account for 30% of the primate range, and 71% of primate species inhabit these lands. As their range on these lands increases, primate species are less likely to be classified as threatened or have declining populations. Safeguarding Indigenous Peoples' lands, languages, and cultures represents our greatest chance to prevent the extinction of the world's primates.Peer reviewe

    PLK1 facilitates chromosome biorientation by suppressing centromere disintegration driven by BLM-mediated unwinding and spindle pulling

    Centromeres provide a pivotal function for faithful chromosome segregation. They serve as a foundation for the assembly of the kinetochore complex and spindle connection, which is essential for chromosome biorientation. Cells lacking Polo-like kinase 1 (PLK1) activity suffer severe chromosome alignment defects, which is believed primarily due to unstable kinetochore-microtubule attachment. Here, we reveal a previously undescribed mechanism named ‘centromere disintegration’ that drives chromosome misalignment in PLK1-inactivated cells. We find that PLK1 inhibition does not necessarily compromise metaphase establishment, but instead its maintenance. We demonstrate that this is caused by unlawful unwinding of DNA by BLM helicase at a specific centromere domain underneath kinetochores. Under bipolar spindle pulling, the distorted centromeres are promptly decompacted into DNA threadlike molecules, leading to centromere rupture and whole-chromosome arm splitting. Consequently, chromosome alignment collapses. Our study unveils an unexpected role of PLK1 as a chromosome guardian to maintain centromere integrity for chromosome biorientation

    The life history of human foraging: Cross-cultural and individual variation

    Human adaptation depends on the integration of slow life history, complex production skills, and extensive sociality. Refining and testing models of the evolution of human life history and cultural learning benefit from increasingly accurate measurement of knowledge, skills, and rates of production with age. We pursue this goal by inferring hunters' increases and declines of skill from approximately 23,000 hunting records generated by more than 1800 individuals at 40 locations. The data reveal an average age of peak productivity between 30 and 35 years of age, although high skill is maintained throughout much of adulthood. In addition, there is substantial variation both among individuals and sites. Within study sites, variation among individuals depends more on heterogeneity in rates of decline than in rates of increase. This analysis sharpens questions about the coevolution of human life history and cultural adaptation

    Evolution of the use of corticosteroids for the treatment of hospitalised COVID-19 patients in Spain between March and November 2020: SEMI-COVID national registry

    Objectives: Since the results of the RECOVERY trial, WHO recommendations about the use of corticosteroids (CTs) in COVID-19 have changed. The aim of the study is to analyse the evolutive use of CTs in Spain during the pandemic to assess the potential influence of new recommendations. Material and methods: A retrospective, descriptive, and observational study was conducted on adults hospitalised due to COVID-19 in Spain who were included in the SEMI-COVID- 19 Registry from March to November 2020. Results: CTs were used in 6053 (36.21%) of the included patients. The patients were older (mean (SD)) (69.6 (14.6) vs. 66.0 (16.8) years; p < 0.001), with hypertension (57.0% vs. 47.7%; p < 0.001), obesity (26.4% vs. 19.3%; p < 0.0001), and multimorbidity prevalence (20.6% vs. 16.1%; p < 0.001). These patients had higher values (mean (95% CI)) of C-reactive protein (CRP) (86 (32.7-160) vs. 49.3 (16-109) mg/dL; p < 0.001), ferritin (791 (393-1534) vs. 470 (236- 996) ”g/dL; p < 0.001), D dimer (750 (430-1400) vs. 617 (345-1180) ”g/dL; p < 0.001), and lower Sp02/Fi02 (266 (91.1) vs. 301 (101); p < 0.001). Since June 2020, there was an increment in the use of CTs (March vs. September; p < 0.001). Overall, 20% did not receive steroids, and 40% received less than 200 mg accumulated prednisone equivalent dose (APED). Severe patients are treated with higher doses. The mortality benefit was observed in patients with oxygen saturation </=90%. Conclusions: Patients with greater comorbidity, severity, and inflammatory markers were those treated with CTs.     In the present study, scanning electron microscopy with energy-dispersive X-ray spectroscopy (SEM–EDS) was used in conjunction with the sequential elution technique (SET) to evaluate the efficacy of extraction of Zn from the moss Pseudoscleropodium purum (Hedw.) M. Fleisch. Moss samples were collected from two areas, a contaminated area (with high concentrations of Zn) in the surroundings of a steel works, and an uncontaminated area. Some samples from the uncontaminated area were used as controls, and others were incubated in a solution of ZnCl2 for subsequent analysis of uptake and location of Zn. The concentrations of Zn in the different cell compartments of the moss were determined by use of the SET, and different points on the surface of the moss and in deposited particles were analysed by SEM–EDS. The results showed that the extracellular fraction obtained by SET includes the Zn bound to cation exchange sites in the moss and Zn bound to particles on the moss surface, which leads to overestimation of extracellular Zn. After extracellular extraction, Zn was not detected on the surface of the moss or in the particles. To avoid problems associated with the presence of particles on the moss surface, a washing procedure should be used to remove such particles before application of the SET. Another possibility is to use the intracellular concentration, as this is not affected by the metal load in the particles and better reflects those contaminants that affect the moss metabolism
