    Salmonella spp. are the etiologic agents of salmonellosis, a worldwide spread zoonoses causing foodborne outbreaks and clinical diseases. By serological identification, Salmonella enterica subsp. enterica serotype 1,4,[5],12:i:- accounted for 8.8% of human and 1.6% of nonhuman Salmonella strains isolated in São Paulo State, during 1991-2000. A total of 28.6% of them amplified a fragment corresponding to H:1,2 (flagellar phase two) through PCR analysis and were further assigned as S. Typhimurium. Antimicrobial resistance was detected in 36.3% of the 369 PCR-negative strains tested, including the multiresistance to ampicillin, chloramphenicol, sulfonamides, tetracycline, and streptomycin.Salmonella spp. é o agente etiológico da salmonelose, zoonose mundialmente distribuída e responsável por surtos de doenças transmitidas por alimentos e doenças clínicas. Sorologicamente, Salmonella enterica subsp. enterica sorotipo 1,4,[5],12:i:- correspondeu a 8,8% e 1,6% das cepas de Salmonella de origem humana e não-humana, respectivamente, isoladas no Estado de São Paulo, no decênio 1991-2000. Loss of fruit ranges from 10% to 40% and could be reduced if papayas were dried. The process of osmotic dehydration followed by air-drying was studied and modeled for papaya preservation, so it could be optimized. The developed model has been validated with experimental data and simulations have shown how the operating conditions affect the process. An optimization was done using the model in order to search for the best operation condition that would reduce the total processing time

    The Evolution of Helium and Hydrogen Ionization Corrections as HII Regions Age

    Helium and hydrogen recombination lines observed in low-metallicity, extragalactic, HII regions provide the data used to infer the primordial helium mass fraction, Y_P. In deriving abundances from observations, the correction for unseen neutral helium or hydrogen is usually assumed to be absent; i.e., the ionization correction factor is taken to be unity (icf = 1). In a previous paper (VGS), we revisited the question of the icf, confirming a "reverse" ionization correction: icf < 1. In VGS the icf was calculated using more nearly realistic models of inhomogeneous HII regions, suggesting that the published values of Y_P needed to be reduced by an amount of order 0.003. As star clusters age, their stellar spectra evolve and so, too, will their icfs. Here the evolution of the icf is studied, along with that of two, alternate, measures of the "hardness" of the radiation spectrum. The differences between the icf for radiation-bounded and matter-bounded models are also explored, along with the effect on the icf of the He/H ratio (since He and H compete for some of the same ionizing photons). Particular attention is paid to the amount of doubly-ionized helium predicted, leading us to suggest that observations of, or bounds to, He++ may help to discriminate among models of HII regions ionized by starbursts of different ages and spectra. We apply our analysis to the Izotov & Thuan (IT) data set utilizing the radiation softness parameter, the [OIII]/[OI] ratio, and the presence or absence of He++ to find 0.95 < icf < 0.99. This suggests that the IT estimate of the primordial helium abundance should be reduced by Delta-Y = 0.006 +- 0.002, from 0.244 +- 0.002 to 0.238 +- 0.003.Comment: 27 double-spaced pages, 11 figures, 5 equations; revised to match the version accepted for publication in the Ap

    Dual-stage sugar substitution in strawberries with a \u3ci\u3eStevia\u3c/i\u3e-based sweetener

    The present study introduces and analyzes a new process denominated dual-stage sugar substitution (D3S). This process aims to induce sugar substitution in strawberries. In a first stage, high-calorie sugars (sucrose, fructose and glucose) are partially removed from the fruit samples and in a second stage, low-calorie sugar (stevioside and rebaudioside) is incorporated to the fruit to maintain its sweetness. The process was evaluated by studying the use of ultrasound application in one or both stages of the D3S process. Best performance of the process was obtained by subjecting the fruit samples to ultrasound in the sugar removal stage followed by immersion of the samples in Stevia-based solution without application of ultrasound in the sweetener incorporation stage. These operating conditions result in the highest sugar removal during the first stage, highest water loss during the process and highest sweetener incorporation during the second stage of the D3S process. The work described in this research is relevant to the production of dried fruits. A process to produce low-calorie dried fruit is presented. The process removes high-calorie sugars from the fruit and replaces it with a natural low-calorie sugar restoring the sweetness of the fruit

    Esterase production by Aureobasidium pullulans URM 7059 in stirred tank and airlift bioreactors using residual biodiesel glycerol as substrate

    Supplementary material related to this article can be found, in the online version, at doi:https://doi.org/10.1016/j.bej.2021.107954.Aureobasidium pullulans URM 7059 produced esterase using residual glycerol from biodiesel as the sole carbon source. The culture medium containing residual glycerol (0.1% v/v), (NH4)2SO4 (4g/L), and yeast extract (8g/L) resulted in the highest esterase production using shake-flasks. The enzyme exhibited a molar mass of 50kDa and was stable at neutral pH and temperatures below 30°C. The cations Cu2+ and Al3+ did not affect the esterase activity, while Ca2+ promoted the highest activity loss. The enzyme kinetic parameters were determined using different substrates (p-nitrophenylcaprylate and p-nitrophenylbutyrate). Km and Vmax were 1.4mM and 218µmolmin-1 for p-NPC, and 1.55mM and 76.7µmolmin-1 for p-NPB. The esterase production was further evaluated using stirred tank and 2-L airlift bioreactors. The airlift reactor operating at the highest air flow rate (8L/min) increased the enzyme productivity 3-fold compared to the shake-flasks experiments. However, the crude enzymatic extract showed 3 active protein bands by zymography with molecular masses of 172kDa, 66kDa, and 40kDa approximately, suggesting that the pattern of enzyme production changed due to aeration. The crude enzyme degraded the MACO-Sty biopolymer in 14 days, being stable in a wide range of pH (7.0 9.0) and temperatures (40°C 80°C).     Ionization Corrections For Low-Metallicity H II Regions and the Primordial Helium Abundance

    Helium and hydrogen recombination lines observed in low-metallicity, extragalactic H II regions provide the data used to infer the primordial helium mass fraction, Y_P. The ionization corrections for unseen neutral helium (or hydrogen) are usually assumed to be absent; i.e., the ionization correction factor is taken to be unity (icf = 1). In this paper we revisit the question of the icf for H II regions ionized by clusters of young, hot, metal-poor stars. Our key result is that for the H II regions used in the determination of Y_P, there is a ``reverse'' ionization correction: icf < 1. We explore the effect on the icf of more realistic inhomogeneous H II region models and find that for those regions ionized by young stars, with ``hard'' radiation spectra, the icf is reduced further below unity. In Monte Carlos using H II region data from the literature (Izotov and Thuan 1998) we estimate a reduction in the published value of Y_P of order 0.003, which is roughly twice as large as the quoted statistical error in the Y_P determination.Comment: 23 pages, 2 postscript figures; ApJ accepted; minor change

    A salmonelose permanece uma importante causa de doença diarréica em humanos no Estado de São Paulo, Brasil. Nesse estudo foram identificados 3554 isolados de Salmonella de infecções humanas, durante o período 1996-2003. Entre os 68 diferentes sorovares determinados, S. Enteritidis foi predominante em infecções gastrointestinais e extra-intestinais, responsável por 67.4% de todos os isolados. S. Typhimurium e S. enterica subsp. enterica (4,5,12:i:-) foram mais freqüentemente isolados de crianças com idade de < 1 a 4 anos, em contraste, infecções por S. Enteritidis foram predominantes em pessoas entre 20 e 50 anos. A contínua vigilância laboratorial das salmoneloses, incluindo a sorotipagem, tem evidenciado os sorovares de Salmonella causando infecções humanas em nossa área geográfica ao longo do tempo.Salmonellosis remains an important cause of diarrheal illness in humans in São Paulo State, Brazil. In this study were identified 3554 Salmonella isolates from human infections, during the period 1996-2003. Among 68 different serovars determined, S. Enteritidis was the most frequent one in gastrointestinal and extra-intestinal infections accounting for 67.4% of all isolates. S. Typhimurium and S. enterica subsp. enterica (4,5,12:i:-) were most frequently isolated from children aged < 1-4 year-old, in contrast, people with S. Enteritidis infections were most likely to be 20-50 year-old. In our geographic area the continued laboratorial surveillance of salmonellosis, including serotyping, has showed the trends in Salmonella serovars causing infections in humans throughout the time

    <p>Abstract</p> <p>Background</p> <p><it>Paracoccidioides brasiliensis </it>is a dimorphic fungus that causes the most prevalent systemic mycosis in Latin America. The response to heat shock is involved in pathogenesis, as this pathogen switches from mycelium to yeast forms in a temperature dependent fashion that is essential to establish infection. HSP90 is a molecular chaperone that helps in the folding and stabilization of selected polypeptides. HSP90 family members have been shown to present important roles in fungi, especially in the pathogenic species, as an immunodominant antigen and also as a potential antifungal therapeutic target.</p> <p>Results</p> <p>In this work, we decided to further study the <it>Pbhsp90 </it>gene, its expression and role in cell viability because it plays important roles in fungal physiology and pathogenesis. Thus, we have sequenced a <it>Pbhsp90 </it>cDNA and shown that this gene is present on the genome as a single copy. We have also confirmed its preferential expression in the yeast phase and its overexpression during dimorphic transition and oxidative stress. Treatment of the yeast with the specific HSP90 inhibitors geldanamycin and radicicol inhibited growth at 2 and 10 μM, respectively.</p> <p>Conclusion</p> <p>The data confirm that the <it>Pbhsp90 </it>gene encodes a morphologically regulated and stress-responsive protein whose function is essential to cell viability of this pathogen. This work also enforces the potential of HSP90 as a target for antifungal therapies, since the use of HSP90 inhibitors is lethal to the <it>P. brasiliensis </it>yeast cells in a dose-responsive manner.</p

    Fagotipos de Salmonella Enteritidis isoladas de amostras clínicas, alimentos e carcassas de frangos no sul do Brasil

    Neste trabalho utilizou-se 272 isolados de Salmonella Enteritidis, dos quais 111 foram isolados de carcaças de frango congeladas, 126 de alimentos e material biológico de humanos envolvidos em episódios de toxinfecções alimentares e 35 de diferentes materiais de origem avícola Destas amostras, 111 foram fagotipadas, sendo 57,65% classificadas como fagotipo 4, 32,43% como fagotipo 4a, 3,60% como fagotipo 6a e 0,90% como fagotipo 7, enquanto 5,40% das amostras não foram fagotipáveis A predominância do fagotipo 4 está em concordância com os resultados publicados ao redor do mundo, e reforça a necessidade de estudos relacionados ao significado epidemiológico destes achados.272 isolates of Salmonella Enteritidis (111 isolated from frozen broiler chicken carcasses, 126 from human food and other biological materials involved in food poisoning outbreaks and 35 from different poultry materials) were selected for phage typing. From these, 111 were phage typed, 57.65% being classified as phage type 4, 32.43% as phage type 4a, 3.60% as phage type 6a and 0.90% as phage type 7, whereas 5.40% samples were not phage typeable. The predominance of phage type 4 is in agreement with the results published worldwide, and reinforces the need for studies related to the epidemiological meaning of these findings
