    Projecto e construção de uma câmara de cultura celular para uso em microscopia óptica

    O grupo de Engenharia de Tecidos da FCT/UNL desenvolve membranas biodegradáveis que servem de substrato a culturas celulares e que se destinam a substituir temporariamente tecidos humanos danificados. O objectivo desta dissertação é desenvolver uma câmara que permita manter uma cultura celular em incubação por um período de tempo indefinido e que simultaneamente permita a sua observação através de microscopia óptica. Foi projectada e construída uma câmara de cultura compatível com o microscópio disponível no laboratório do Grupo de Engenharia de Tecidos da FCT/UNL, capaz de manter um ambiente interno adequado às necessidades fisiológicas das células: 37 ºC, CO2 a 5% e entre 90 a 100% de humidade sem interferir com a capacidade de formação de imagens do microscópio. Os testes efectuados permitiram verificar que o sistema desenvolvido é capaz de controlar e monitorizar a temperatura e a percentagem de CO2, proporcionando um ambiente adequado para manter as células em incubação por um período de tempo indefinido e permitindo a sua observação num microscópio óptico invertido

    A kinase translocation reporter reveals real-time dynamics of ERK activity in Drosophila

    Extracellular signal-regulated kinase (ERK) lies downstream of a core signalling cascade that controls all aspects of development and adult homeostasis. Recent developments have led to new tools to image and manipulate the pathway. However, visualising ERK activity in vivo with high temporal resolution remains a challenge in Drosophila. We adapted a kinase translocation reporter (KTR) for use in Drosophila, which shuttles out of the nucleus when phosphorylated by ERK. We show that ERK-KTR faithfully reports endogenous ERK signalling activity in developing and adult tissues, and that it responds to genetic perturbations upstream of ERK. Using ERK-KTR in time-lapse imaging, we made two novel observations: firstly, sustained hyperactivation of ERK by expression of dominant-active epidermal growth factor receptor raised the overall level but did not alter the kinetics of ERK activity; secondly, the direction of migration of retinal basal glia correlated with their ERK activity levels, suggesting an explanation for the heterogeneity in ERK activity observed in fixed tissue. Our results show that KTR technology can be applied in Drosophila to monitor ERK activity in real-time and suggest that this modular tool can be further adapted to study other kinases.     Ants as bioindicators of riparian ecological health in catalonian rivers

    In this study, we assess the potential of ants as bioindicators of riparian ecological health in two river types (upland and lowland type) located in the Catalonian region. We proposed to understand to what extent do metrics based on ant responses provide useful information that cannot be presented by traditional biophysical assessments while attempting an approach to creating an ant-based multimetric index (ant-based MMI) of the riparian ecological health. A total of 22 ant species were identified, and 42 metrics related to ant foraging activity, species richness, and functional traits were evaluated as potential core metrics of the index. Riparian features and proximal land use land cover (LULC) were used to distinguish disturbed from less disturbed sites. We found that ant communities strongly responded to human disturbance. When compared with an exclusively physical-based index for the assessment of the riparian health, the ant-based MMI was more sensitive to human disturbance, by also reacting to the effects of the surrounding LULC pressure.     Stitching is used to reduce incomplete infusion of T-joint core (dry-core) and reinforce T-joint structure. However, it may cause new types of flaws, especially submillimeter flaws. Thermographic approaches including microvibrothermography, microlaser line thermography, and microlaser spot thermography on the basis of pulsed and lock-in techniques were proposed. These techniques are used to detect the submillimeter porosities in a stitched T-joint carbon fiber reinforced polymer composite specimen.     Rapid deglaciation during the Bølling-Allerød Interstadial in the Central Pyrenees and associated glacial and periglacial landforms

    The Central Pyrenees hosted a large ice cap during the Late Pleistocene. The cirques under relatively low-altitude peaks (2200-2800 m) include the greatest variety of glacial landforms (moraines, fossil debris-covered glaciers and rock glaciers), but their age and formation process are poorly known. Here, we focus on the headwaters of the Garonne River, namely on the low-altitude Bacivèr Cirque (highest peaks at ~2600 m), with widespread erosive and depositional glacial and periglacial landforms. We reconstruct the pattern of deglaciation from geomorphological observations and a 17-sample dataset of 10Be Cosmic-Ray Exposure (CRE) ages. Ice thickness in the Bacivèr Cirque must have reached ~200 m during the maximum ice extent of the last glacial cycle, when it flowed down towards the Garonne paleoglacier. However, by ~15 ka, during the Bølling-Allerød (B-A) Interstadial, the mouth of the cirque was deglaciated as the tributary glacier shrank and disconnected from the Garonne paleoglacier. Glacial retreat was rapid, and the whole cirque was likely to have been deglaciated in only a few centuries, while paraglacial processes accelerated, leading to the transformation of debris-free glaciers into debriscovered and rock glaciers in their final stages. Climate conditions prevailing at the transition between the B-A and the Younger Dryas (YD) favored glacial growth and the likely development of small moraines within the slopes of the cirque walls by ~12.9 ka, but the dating uncertainties make it impossible to state whether these moraines formed during the B-A or the YD. The melting of these glaciers favored paraglacial dynamics, which promoted the development of rock glaciers as well as debris-covered glaciers. These remained active throughout the Early Holocene until at least ~7 ka. Since then, the landscape of the Bacivèr Cirque has seen a period of relative stability. A similar chronological sequence of deglaciation has been also detected in other cirques of the Pyrenees below 3000 m. As in other mid-latitude mountain regions, the B-A triggered the complete deglaciation of the Garonne paleoglacier and promoted the development of the wide variety of glacial and periglacial landforms existing in the Bacivèr cirque

    The H2020 OCRE Project Opens the Gates of the Commercial Cloud and EO Services Usage to the Research Community

    Cloud and Earth Observation (EO) based services offer the European Research community a wealth of powerful tools. However, for many researchers these tools are currently out of reach. It is difficult to find and select suitable services. Establishing agreements with cloud and EO service providers and ensuring legal and technical compliance requires specialist skills and takes an inordinate amount of time. Equally, service providers find it difficult to reach and meet the needs of the research community in technical, financial and legal areas. The Open Clouds for Research Environments consortium (OCRE) will change this, by putting in place an easy adoption route. In the autumn of 2019, OCRE will run a pan-European tender and establish framework agreements with service providers who meet the requirements of the research community. 10.000 European research and education institutes will be able to directly consume these offerings via the European Open Science Cloud service catalogue, through ready-to-use agreements. They will not have to run a tender of their own. In addition, to stimulate usage, OCRE will make available 9.5 million euro in service credits (vouchers), through adoption funds from the European Commission. OCRE is a pioneer project without precedence, with potentially high impact in the future EO market activities and evolution of service offering, with the objective to burst the usage of EO commercial services by the research environment
