    Systemic lupus erythematosus and implications for the oral cavity

    Systemic Lupus Erythematosus affects several people around the world and because it is characterized as a chronic inflammatory disease of multifactorial origin, and with systemic impairment, great attention must be paid from diagnosis to treatment in order to optimize the entire follow-up of the patient. The dental doctor plays an important role in the diagnosis of the condition and must be attentive to the early signs that can appear in the oral cavity with a frequency of up to 21%. In this way, through this bibliographic review, which has as main goal to correlate Systemic Lupus Erythematosus with its direct consequences in the oral cavity, it will be possible to help dentists in the diagnosis, to understand in detail the development of the disease and what attitude should be taken in its presence.info:eu-repo/semantics/publishedVersio

    Evaluation of Approaches to Identify the Targets of Cellular Immunity on a Proteome-Wide Scale

    Background: Vaccine development against malaria and other complex diseases remains a challenge for the scientific community. The recent elucidation of the genome, proteome and transcriptome of many of these complex pathogens provides the basis for rational vaccine design by identifying, on a proteome-wide scale, novel target antigens that are recognized by T cells and antibodies from exposed individuals. However, there is currently no algorithm to effectively identify important target antigens from genome sequence data; this is especially challenging for T cell targets. Furthermore, for some of these pathogens, such as Plasmodium, protein expression using conventional platforms has been problematic but cell-free in vitro transcription translation (IVTT) strategies have recently proved successful. Herein, we report a novel approach for proteome-wide scale identification of the antigenic targets of T cell responses using IVTT products. Principal Findings: We conducted a series of in vitro and in vivo experiments using IVTT proteins either unpurified, absorbed to carboxylated polybeads, or affinity purified through nickel resin or magnetic beads. In vitro studies in humans using CMV, EBV, and Influenza A virus proteins showed antigen-specific cytokine production in ELIspot and Cytometric Bead Array assays with cells stimulated with purified or unpurified IVTT antigens. In vitro and in vivo studies in mice immunized with the Plasmodium yoelii circumsporozoite DNA vaccine with or without IVTT protein boost showed antigen-specific cytokine production using purified IVTT antigens only. Overall, the nickel resin method of IVTT antigen purification proved optimal in both human and murine systems. Conclusions: This work provides proof of concept for the potential of high-throughput approaches to identify T cell targets of complex parasitic, viral or bacterial pathogens from genomic sequence data, for rational vaccine development against emerging and re-emerging diseases that pose a threat to public health

    Força dos músculos cérvico-escapulares e a severidade da disfunção temporomandibular em mulheres com cervicalgia mecânica

    Introdução: modificações da função muscular cervical têm sido verificadas em pacientes com cervicalgia e DTM. Entretanto, ainda não é conhecida a relação entre a severidade da DTM e a força muscular cervical na presença/ausência de cervicalgia. Objetivo: verificar a prevalência de DTM em mulheres com e sem cervicalgia mecânica, avaliar a força dos músculos cérvico-escapulares e sua associação com a severidade da DTM. Métodos: participaram 15 voluntárias sem dor cervical (GC) e 14 mulheres com cervicalgia mecânica (GCM), selecionadas por meio do Índice de Disfunção Relacionada ao Pescoço. O diagnóstico e a gravidade da DTM foram determinados pelos Critérios diagnósticos para pesquisa em desordens temporomandibulares e Índice Temporomandibular (IT), respectivamente. A força dos músculos trapézio superior, flexores e extensores cervicais foi aferida por dinamometria digital manual. Resultados: 64,5% das mulheres com cervicalgia e 33,3% das sem dor cervical apresentaram diagnóstico de DTM (p = 0,095). O GCM apresentou menor força dos músculos flexores (p = 0,044) e extensores cervicais (p = 0,006) e maior IT (p = 0,038) que o GC. Também foi verificada correlação negativa moderada entre o IT e força dos músculos trapézio superior dominante (p = 0,046, r = -0,547), não dominante (p = 0,007, r = -0,695) e flexores cervicais (p = 0,023, r = -0,606) no GCM. Conclusão: não houve diferença na prevalência de DTM entre mulheres com e sem cervicalgia. Entretanto, mulheres com cervicalgia apresentaram menor força muscular cervical (comparadas às sem cervicalgia) que esteve associada a maior severidade da DTM. Assim, em mulheres com cervicalgia associada à DTM, é recomendável avaliar e abordar a severidade desta disfunção e o comprometimento dos músculos cérvico-escapulares.Introduction: changes in cervical muscle function have been observed in patients with neck pain (NP) and TMD. However, the relationship between TMD severity and neck muscle strength in the presence/absence of NP is unknown. Objective: to determine the prevalence of TMD in women with and without mechanical NP and assess the cervical-scapular muscle strength and its association with TMD severity. Methods: fifteen volunteers without neck pain (CG) and 14 women with mechanical neck pain (NPG) took part and were selected by the Neck Disability Index. The diagnosis and severity of TMD were determined by the Research Diagnostic Criteria for TMD and Temporomandibular Index (TI), respectively. The strength of the upper trapezius muscle, and cervical flexor and extensor muscles was measured by digital hand dynamometer. Results: 64.5% of women with NP and 33.3% without NP were diagnosed with TMD (p = 0.095). The NPG showed lower strength of the cervical flexor (p = 0.044) and extensor (p=0.006) muscles, and higher TI (p = 0.038) than in the CG. It was also verified moderate negative correlation between TI and the strength of dominant (p = 0.046, r = -0.547) and non-dominant (p = 0.007, r = -0.695) upper trapezius, and cervical flexors (p = 0.023, r = -0.606) in the NPG. Conclusion: there was no difference in the prevalence of TMD in women with and without NP. However, women with NP have lower cervical muscle strength - compared to those without NP - which was associated with greater severity of TMD. Thus, in women with NP associated with TMD, it is advisable to assess and address the severity of this dysfunction and identify the cervical-scapular muscles compromise

    Ethyl-cellulose rumen-protected methionine enhances performance during the periparturient period and early lactation in Holstein dairy cows

    The onset of lactation in dairy cows is characterized by severe negative energy and protein balance. Increasing Met availability during this time may improve milk production, hepatic lipid metabolism, and immune function. The aim of this study was to evaluate the effect of feeding ethyl-cellulose rumen-protected methionine (RPM; Mepron, Evonik Nutrition and Care GmbH, Hanau-Wolfgang, Germany) on the performance of dairy cows during prepartum and early-lactation periods. Sixty multiparous Holstein cows were used in a block design and assigned to either a control or an ethyl-cellulose RPM diet. Ethyl-cellulose RPM was supplied from -28 to 60 d relative to parturition at a rate of 0.09% and 0.10% of dry matter during the prepartum and postpartum periods, respectively. That rate ensured that the ratio of Lys to Met in metabolizable protein was close to 2.8:1. Cows fed ethyl-cellulose RPM had dry matter intakes (DMI) that were 1.2 kg/d greater during the prepartum period and consequently had overall greater cumulative DMI than cows in the control group. Compared with controls, during the fresh period (1-30 d in milk; DIM) feeding ethyl-cellulose RPM increased DMI by 1.7 kg/d, milk yield by 4.1 kg/d, fat yield by 0.17 kg/d, milk protein yield by 0.20 kg/d, 3.5% fat-corrected milk by 4.3 kg/d, and energy-corrected milk by 4.4 kg/d. Although ethyl-cellulose RPM supplementation increased milk protein content by 0.16 percentage units compared with the control during the fresh period, no differences were observed for milk fat, lactose, and milk urea nitrogen concentration. During the high-producing period (31-60 DIM), cows fed ethyl-cellulose RPM increased DMI and milk yield by 1.45 and 4.4 kg/d, respectively. Ethyl-cellulose RPM also increased fat yield by 0.19 kg/d, milk protein yield by 0.17 kg/d, 3.5% fat-corrected milk by 4.7 kg/d, and energy-corrected milk by 4.8 kg/d compared with controls. Ethyl-cellulose RPM supplementation reduced plasma fatty acids in the fresh period and decreased Îł-glutamyl transferase, indicating better liver function. In conclusion, when lysine was adequate, feeding ethyl-cellulose RPM to achieve a ratio close to 2.8:1 in metabolizable protein improved dairy cow performance from parturition through 60 DIM. The greater milk production was, at least in part, driven by the greater voluntary DMI and better liver function

    The periparturient period is the most critical phase in the productive cycle of dairy cows and is characterized by impairment of the immune system. Our objective was to evaluate the effect of feeding ethyl-cellulose rumen-protected methionine (RPM) starting at d -28 from expected parturition through 60 d in milk on biomarkers of inflammation, oxidative stress, and liver function as well as leukocyte function. Sixty multiparous Holstein cows were used in a block design and assigned to either a control or the control plus ethyl-cellulose RPM (Mepron, Evonik Nutrition & Care GmbH). Mepron was supplied from -28 to 60 d in milk at a rate of 0.09% and 0.10% dry matter during the prepartum and postpartum period. That rate ensured that the ratio of Lys to Met in the metabolizable protein was close to 2.8:1. Blood samples from 15 clinically healthy cows per treatment were collected at d -30, -14, 1, 7, 21, 30, and 60 and analyzed for biomarkers of liver function, inflammation, and oxidative stress. Neutrophil and monocyte function in whole blood was measured in vitro at -14, 1, 7, 21, and 30 d in milk. The statistical model included the random effect of block and fixed effect of treatment, time, and its interaction. Compared with control, ethyl-cellulose RPM increased plasma cholesterol and paraoxonase after parturition. Among the inflammation biomarkers measured, ethyl-cellulose RPM led to greater albumin (negative acute-phase protein) and lower haptoglobin than control cows. Although concentration of IL-1β was not affected by treatments, greater IL-6 concentration was detected in response to ethyl-cellulose RPM. Cows supplemented with ethyl-cellulose RPM had greater plasma concentration of ferric-reducing antioxidant power, β-carotene, tocopherol, and total and reduced glutathione, whereas reactive oxygen metabolites were lower compared with control cows. Compared with control, ethyl-cellulose RPM enhanced neutrophil phagocytosis and oxidative burst. Overall, the results indicate that ethyl-cellulose RPM supply to obtain a Lys-to-Met ratio of 2.8:1 in the metabolizable protein during the periparturient period and early lactation is an effective approach to help mitigate oxidative stress and inflammation as well as enhance liver and neutrophil function in dairy cows

    Voltage-Gated Sodium Channel Modulation by a New Spider Toxin Ssp1a Isolated From an Australian Theraphosid

    Given the important role of voltage-gated sodium (NaV) channel-modulating spider toxins in elucidating the function, pharmacology, and mechanism of action of therapeutically relevant NaV channels, we screened the venom from Australian theraphosid species against the human pain target hNaV1.7. Using assay-guided fractionation, we isolated a 33-residue inhibitor cystine knot (ICK) peptide (Ssp1a) belonging to the NaSpTx1 family. Recombinant Ssp1a (rSsp1a) inhibited neuronal hNaV subtypes with a rank order of potency hNaV1.7 > 1.6 > 1.2 > 1.3 > 1.1. rSsp1a inhibited hNaV1.7, hNaV1.2 and hNaV1.3 without significantly altering the voltage-dependence of activation, inactivation, or delay in recovery from inactivation. However, rSsp1a demonstrated voltage-dependent inhibition at hNaV1.7 and rSsp1a-bound hNaV1.7 opened at extreme depolarizations, suggesting rSsp1a likely interacted with voltage-sensing domain II (VSD II) of hNaV1.7 to trap the channel in its resting state. Nuclear magnetic resonance spectroscopy revealed key structural features of Ssp1a, including an amphipathic surface with hydrophobic and charged patches shown by docking studies to comprise the interacting surface. This study provides the basis for future structure-function studies to guide the development of subtype selective inhibitors

    A multivalent chimeric vaccine composed of Schistosoma mansoni SmTSP-2 and Sm29 was able to induce protection against infection in mice

    Schistosoma mansoni is a blood fluke parasite responsible for schistosomiasis. The best long-term strategy to control schistosomiasis is through immunization combined with drug treatment. In this study, we cloned, expressed and purified SmTSP-2 fused to the N- and C-terminal halves of Sm29 and tested these chimeras as vaccine candidates using an adjuvant approved to be used in humans. The results demonstrated that vaccination with SmTSP-2 fused to N- or C-terminus of Sm29-induced reduction in worm burden and liver pathology when compared to control animals. Additionally, we detected high levels of mouse-specific IgG, IgG1 and IgG2a against both chimeras and significant amounts of IFN-γ and TNF-α and no IL-4. Finally, studies with sera from patients resistant to infection and living in schistosomiasis endemic areas revealed high levels of specific IgG to both chimeras when compared to healthy individuals. In conclusion, SmTSP-2/Sm29 chimeras tested here induced partial protection against infection and might be a potential vaccine candidate

    Schistosoma mansoni Tegument Protein Sm29 Is Able to Induce a Th1-Type of Immune Response and Protection against Parasite Infection

    Schistosomiasis is the most important human helminth infection in terms of morbidity and mortality. Although the efforts to develop a vaccine against this disease have experienced failures, a new generation of surface antigens revealed by proteomic studies changed this scenario. Our group has characterized the protein Sm29 described previously as one of the most exposed and expressed antigens in the outer tegument of Schistosoma mansoni. Studies in patients living in endemic areas for schistosomiasis revealed high levels of IgG1 and IgG3 anti-Sm29 in resistant individuals. In this study, confocal microscope analysis showed Sm29 present in the surface of lung-stage schistosoluma and adult worms. Recombinant Sm29, when used as vaccine candidate, induced high levels of protection in mice. This protection was associated with a typical Th1 immune response and reduction of worm burden, liver granulomas and in intestinal eggs. Further, microarray analysis of worms recovered from vaccinated mice showed significant down-regulation of several genes encoding previously characterized vaccine candidates and/or molecules exposed on the surface, suggesting an immune evasion strategy of schistosomes under immune attack. These results demonstrated that Sm29 as one of the important antigens with potential to compose a vaccine against schistosomiasis

    Study design: Integrative literature review. Aims: To summarize the main results of research and publications related to the use of alternative communication technologies for people with restrictions on their communication skills, particularly those laryngectomized with head and neck cancer. Method: This study was based on bibliographic research in electronic databases: PubMed, LILACS, SCIELO and CINAHL through the use of the following descriptors: “Head and Neck Neoplasms”, “Head and Neck Cancer”, “Quality of Life” and “Communication” , their corresponding in Portuguese and their crosses. Results: Selected 14 articles published in the period from 2004 to 2014, categorized into two units of analysis: I: Illness and treatment impact on quality of life and physical aspects, functional and symptoms (N = 5), II: Psychosocial aspects related to the treatment of head and neck cancer (N = 9). The articles indicated that the disease and the treatment of cancer cause body changes, at times traumatic, with important impacts on the functional and esthetic aspects, with speech, nutrition and physical appearance alterations, besides affecting the psychological factors. The laryngectomy is highly stressing and compromises the communication and social interactions, particularly in relation to their families and the health team. The consequences are manifested in everyday life of these subjects, including emotional responses ranging in a singular way to each individual. The use of alternative communication resources can encourage active social participation by people with communication difficulties and favor therefore their quality of life (QoL). Conclusion: The use of alternative resources for communication, by partial or total laryngectomized favors the exercise of the autonomy, maintaining communication skills, active social participation and improves QoL in person’s with cancerModelo do estudo: Revisão Integrativa da Literatura. Método: estudo baseado em levantamento bibliográfico nas bases de dados eletrônicas: PubMed, LILACS, SCIELO e CINAHL, através do uso dos descritores: “Head and Neck Neoplasms”, “Head and Neck Cancer”, “Quality of Life” e “Communication”, seus correspondentes em português e seus cruzamentos. Resultados: Foram selecionados 14 artigos, publicados no período de 2004 à 2014, categorizados em duas unidades de análise: I: Impacto do adoecimento e do tratamento na qualidade de vida e aspectos físicos, funcionais e sintomas (N=5), II: Aspectos psicossociais relacionados ao tratamento de câncer de cabeça e pescoço (N=9). Os artigos indicaram que o adoecimento e o tratamento do câncer causam alterações corporais, por vezes traumáticas, com impactos importantes sobre os aspectos funcionais e estéticos, com alterações da fala, da alimentação e aparência física, além de afetar fatores psicológicos. A laringectomia é altamente estressante e compromete a comunicação e as interações sociais, particularmente em relação a seus familiares e equipe de saúde. Essas consequências manifestam-se na vida cotidiana desses sujeitos, incluindo respostas emocionais que variam de forma singular a cada individuo. O uso de recursos de comunicação alternativa pode favorecer a participação social ativa por parte de pessoas com dificuldades de comunicação e favorecer, assim, sua qualidade de vida (QV).
