    Play’s the Thing: Community Adventure Play Experiences

    The authors conducted a participatory action community-based research project that included the development, implementation and evaluation of 36 Community Adventure Play Experiences (CAPEs) in a variety of communities in the United States and abroad. In CAPEs, the emphasis is on free play created by children using “loose parts” that are readily available in their communities such as cardboard boxes, tires, fabric, recyclables, sand, wood, and water. During these play experiences children use their imaginations, and take the initiative to collaborate and problem-solve with found materials. Play facilitators ensure a safe physical and emotional space and assist children in their play as directed by the children. The authors studied CAPEs in rural, urban and peri-urban settings in the United States, Tanzania, Malawi, and Zimbabwe. As part of the investigation, they interviewed 225 caregivers and teachers in the United States, Tanzania, Malawi and Zimbabwe for information about their perceptions of CAPEs

    The Center for the Urban River at Beczak is a 3900-square-foot environmental education and research facility located on 2 acres of Hudson riverfront park in downtown Yonkers. It is operated by Sarah Lawrence College in cooperation with the Beczak Environmental Education Center. The objective of this study was to measure the effects of a field trip to CURB on students’ environmental empathy, environmental engagement, cultural awareness, and interest in CURB. This was achieved with qualitative and quantitative measures, including a multi-case study (Bogdan & Biklen, 1998) and a quantitative survey. The qualitative multi-case study, in the field of participatory action research (Denzin & Lincoln, 2000), included note-taking and observation of students attending CURB programs

    Globular clusters in the outer halo of M31: the survey

    We report the discovery of 40 new globular clusters (GCs) that have been found in surveys of the halo of M31 based on INT/WFC and CHFT/Megacam imagery. A subset of these these new GCs are of an extended, diffuse nature, and include those already found in Huxor et al. (2005). The search strategy is described and basic positional and V and I photometric data are presented for each cluster. For a subset of these clusters, K-band photometry is also given. The new clusters continue to be found to the limit of the survey area (~100 kpc), revealing that the GC system of M31 is much more extended than previously realised. The new clusters increase the total number of confirmed GCs in M31 by approximately 10% and the number of confirmed GCs beyond 1 degree (~14 kpc) by more than 75%. We have also used the survey imagery as well recent HST archival data to update the Revised Bologna Catalogue (RBC) of M31 globular clusters.Comment: Accepted to MNRA

    Biogenesis of the inner membrane complex is dependent on vesicular transport by the alveolate specific GTPase Rab11B

    Apicomplexan parasites belong to a recently recognised group of protozoa referred to as Alveolata. These protists contain membranous sacs (alveoli) beneath the plasma membrane, termed the Inner Membrane Complex (IMC) in the case of Apicomplexa. During parasite replication the IMC is formed de novo within the mother cell in a process described as internal budding. We hypothesized that an alveolate specific factor is involved in the specific transport of vesicles from the Golgi to the IMC and identified the small GTPase Rab11B as an alveolate specific Rab-GTPase that localises to the growing end of the IMC during replication of Toxoplasma gondii. Conditional interference with Rab11B function leads to a profound defect in IMC biogenesis, indicating that Rab11B is required for the transport of Golgi derived vesicles to the nascent IMC of the daughter cell. Curiously, a block in IMC biogenesis did not affect formation of sub-pellicular microtubules, indicating that IMC biogenesis and formation of sub-pellicular microtubules is not mechanistically linked. We propose a model where Rab11B specifically transports vesicles derived from the Golgi to the immature IMC of the growing daughter parasites

    Three dimensional tracking for volumetric spectral-domain optical coherence tomography

    We present a three-dimensional (3D) tracker for a clinical ophthalmic spectral domain optical coherence tomography (SD-OCT) system that combines depth-tracking with lateral tracking, providing a stabilized reference frame for 3D data recording and post acquisition analysis. The depth-tracking system is implemented through a real-time dynamic feedback mechanism to compensate for motion artifact in the axial direction. Active monitoring of the retina and adapting the reference arm of the interferometer allowed the whole thickness of the retina to be stabilized to within +/- 100 mu m. We achieve a relatively constant SNR from image to image by stabilizing the image of the retina with respect to the depth dependent sensitivity of SD-OCT. The depth tracking range of our system is 5.2 mm in air and the depth is adjusted every frame. Enhancement in the stability of the images with the depth-tracking algorithm is demonstrated on a healthy volunteer.X1119sciescopu

    Outlying HII Regions in HI-Selected Galaxies

    We present results from the first systematic search for outlying HII regions, as part of a sample of 96 emission-line point sources (referred to as ELdots - emission-line dots) derived from the NOAO Survey for Ionization in Neutral Gas Galaxies (SINGG). Our automated ELdot-finder searches SINGG narrow-band and continuum images for high equivalent width point sources outside the optical radius of the target galaxy (> 2 X r25 in the R-band). Follow-up longslit spectroscopy and deep GALEX images (exposure time > 1000 s) distinguish outlying HII regions from background galaxies whose strong emission lines ([OIII], Hbeta or [OII]) have been redshifted into the SINGG bandpass. We find that these deep GALEX images can serve as a substitute for spectroscopic follow-up because outlying HII regions separate cleanly from background galaxies in color-color space. We identify seven SINGG systems with outlying massive star formation that span a large range in Halpha luminosities corresponding to a few O stars in the most nearby cases, and unresolved dwarf satellite companion galaxies in the most distant cases. Six of these seven systems feature galaxies with nearby companions or interacting galaxies. Furthermore, our results indicate that some outlying HII regions are linked to the extended-UV disks discovered by GALEX, representing emission from the most massive O stars among a more abundant population of lower mass (or older) star clusters. The overall frequency of outlying HII regions in this sample of gas-rich galaxies is 8 - 11% when we correct for background emission-line galaxy contamination (~75% of ELdots).Comment: 20 pages, 14 Figures, Accepted by A

    A mechanism for the inhibition of DNA-PK-mediated DNA sensing by a virus

    The innate immune system is critical in the response to infection by pathogens and it is activated by pattern recognition receptors (PRRs) binding to pathogen associated molecular patterns (PAMPs). During viral infection, the direct recognition of the viral nucleic acids, such as the genomes of DNA viruses, is very important for activation of innate immunity. Recently, DNA-dependent protein kinase (DNA-PK), a heterotrimeric complex consisting of the Ku70/Ku80 heterodimer and the catalytic subunit DNA-PKcs was identified as a cytoplasmic PRR for DNA that is important for the innate immune response to intracellular DNA and DNA virus infection. Here we show that vaccinia virus (VACV) has evolved to inhibit this function of DNA-PK by expression of a highly conserved protein called C16, which was known to contribute to virulence but by an unknown mechanism. Data presented show that C16 binds directly to the Ku heterodimer and thereby inhibits the innate immune response to DNA in fibroblasts, characterised by the decreased production of cytokines and chemokines. Mechanistically, C16 acts by blocking DNA-PK binding to DNA, which correlates with reduced DNA-PK-dependent DNA sensing. The C-terminal region of C16 is sufficient for binding Ku and this activity is conserved in the variola virus (VARV) orthologue of C16. In contrast, deletion of 5 amino acids in this domain is enough to knockout this function from the attenuated vaccine strain modified vaccinia virus Ankara (MVA). In vivo a VACV mutant lacking C16 induced higher levels of cytokines and chemokines early after infection compared to control viruses, confirming the role of this virulence factor in attenuating the innate immune response. Overall this study describes the inhibition of DNA-PK-dependent DNA sensing by a poxvirus protein, adding to the evidence that DNA-PK is a critical component of innate immunity to DNA viruses

    Second trimester inflammatory and metabolic markers in women delivering preterm with and without preeclampsia.

    ObjectiveInflammatory and metabolic pathways are implicated in preterm birth and preeclampsia. However, studies rarely compare second trimester inflammatory and metabolic markers between women who deliver preterm with and without preeclampsia.Study designA sample of 129 women (43 with preeclampsia) with preterm delivery was obtained from an existing population-based birth cohort. Banked second trimester serum samples were assayed for 267 inflammatory and metabolic markers. Backwards-stepwise logistic regression models were used to calculate odds ratios.ResultsHigher 5-α-pregnan-3β,20α-diol disulfate, and lower 1-linoleoylglycerophosphoethanolamine and octadecanedioate, predicted increased odds of preeclampsia.ConclusionsAmong women with preterm births, those who developed preeclampsia differed with respect metabolic markers. These findings point to potential etiologic underpinnings for preeclampsia as a precursor to preterm birth

    Ferredoxin containing bacteriocins suggest a novel mechanism of iron uptake in <i>Pectobacterium spp</i>

    In order to kill competing strains of the same or closely related bacterial species, many bacteria produce potent narrow-spectrum protein antibiotics known as bacteriocins. Two sequenced strains of the phytopathogenic bacterium &lt;i&gt;Pectobacterium carotovorum&lt;/i&gt; carry genes encoding putative bacteriocins which have seemingly evolved through a recombination event to encode proteins containing an N-terminal domain with extensive similarity to a [2Fe-2S] plant ferredoxin and a C-terminal colicin M-like catalytic domain. In this work, we show that these genes encode active bacteriocins, pectocin M1 and M2, which target strains of &lt;i&gt;Pectobacterium carotovorum&lt;/i&gt; and &lt;i&gt;Pectobacterium atrosepticum&lt;/i&gt; with increased potency under iron limiting conditions. The activity of pectocin M1 and M2 can be inhibited by the addition of spinach ferredoxin, indicating that the ferredoxin domain of these proteins acts as a receptor binding domain. This effect is not observed with the mammalian ferredoxin protein adrenodoxin, indicating that &lt;i&gt;Pectobacterium spp.&lt;/i&gt; carries a specific receptor for plant ferredoxins and that these plant pathogens may acquire iron from the host through the uptake of ferredoxin. In further support of this hypothesis we show that the growth of strains of &lt;i&gt;Pectobacterium carotovorum&lt;/i&gt; and &lt;i&gt;atrosepticum&lt;/i&gt; that are not sensitive to the cytotoxic effects of pectocin M1 is enhanced in the presence of pectocin M1 and M2 under iron limiting conditions. A similar growth enhancement under iron limiting conditions is observed with spinach ferrodoxin, but not with adrenodoxin. Our data indicate that pectocin M1 and M2 have evolved to parasitise an existing iron uptake pathway by using a ferredoxin-containing receptor binding domain as a Trojan horse to gain entry into susceptible cells
