    Enhancing methane production from food waste fermentate using biochar. The added value of electrochemical testing in pre-selecting the most effective type of biochar

    Background: Recent studies have suggested that addition of electrically conductive biochar particles is an effective strategy to improve the methanogenic conversion of waste organic substrates, by promoting syntrophic associations between acetogenic and methanogenic organisms based on interspecies electron transfer processes. However, the underlying fundamentals of the process are still largely speculative and, therefore, a priori identification, screening, and even design of suitable biochar materials for a given biotechnological process are not yet possible. Results: Here, three charcoal-like products (i.e., biochars) obtained from the pyrolysis of different lignocellulosic materials, (i.e., wheat bran pellets, coppiced woodlands, and orchard pruning) were tested for their capacity to enhance methane production from a food waste fermentate. In all biochar-supplemented (25 g/L) batch experiments, the complete methanogenic conversion of fermentate volatile fatty acids proceeded at a rate that was up to 5 times higher than that observed in the unamended (or sand-supplemented) controls. Fluorescent in situ hybridization analysis coupled with confocal laser scanning microscopy revealed an intimate association between archaea and bacteria around the biochar particles and provided a clear indication that biochar also shaped the composition of the microbial consortium. Based on the application of a suite of physico-chemical and electrochemical characterization techniques, we demonstrated that the positive effect of biochar is directly related to the electron-donating capacity (EDC) of the material, but is independent of its bulk electrical conductivity and specific surface area. The latter properties were all previously hypothesized to play a major role in the biochar-mediated interspecies electron transfer process in methanogenic consortia. Conclusions: Collectively, these results of this study suggest that for biochar addition in anaerobic digester operation, the screening and identification of the most suitable biochar material should be based on EDC determination, via simple electrochemical tests. © 2017 The Author(s)

    LICIACube on DART Mission: An Asteroid Impact Captured by Italian Small Satellite Technology

    In the frame of the Planetary Defense program, NASA developed the Double Asteroid Redirection Test (DART) mission and the Italian Space Agency joined the effort. DART’s spacecraft will act as a kinetic impactor by deliberately crashing into the moonlet of Didymos binary system (i.e. Didymos-B) while the effects of the impact will be observed by a small satellite, the Light Italian CubeSat for Imaging of Asteroid (LICIACube) and ground-based telescopes. LICIACube, an Italian Space Agency (ASI) mission, will fly with a relative velocity of approximately 6.5 km/s and it will document the effects of the impact, the crater and the evolution of the plume generated by the collision. LICIACube will have to maintain the asteroid\u27s pointing at an angular speed of approximately 10 deg/s to fly-by the asteroid close to the Didymos-B surface. The images acquired by LICIACube will be processed onboard through the autonomous navigation algorithm to identify the asteroid system and control the satellite attitude. They will also help the scientific community and provide feedback to the Planetary Defense program, pioneered by the Space Agencies.     Plant diversity changes in a nature reserve: a probabilistic sampling method for quantitative assessments

    Species pool conservation is critical for the stability of ecosystem processes. However, climate and land use changes will likely affect biodiversity, and managers of protected areas are under increasing pressure to monitor native species diversity changes by approaches that are scientifically sound and comparable over time. Here we describe a plant diversity monitoring system in use since 2002 in the “Montagna di Torricchio” Nature Reserve (LTER_EU_IT_033), a Central Apennines representative area of 317 ha, most of which is under strict protection. The aim of this paper was to assess changes in plant species richness over time and to deduce the patterns of species assemblage. The monitoring system was based on a probabilistic sampling design representative of the different physiognomic vegetation types occurring in the Reserve. A total of 34 plots (10×10m) were sampled in 2002, 2003 and 2015, and their species presence/absence and relative coverage were estimated. Repeated measure ANOVA was used to test for plot-level and ecosystem-based changes in species richness along the study period. Temporal nestedness and temporal turnover metrics were used to assess patterns of species’ compositional changes. The results showed significantly different levels of species richness depending on the year, with the lowest value in 2003, probably linked to extreme drought events. Forest systems were comparatively stable, demonstrating the capacity to buffer interannual climate variability. Regarding compositional changes along the entire period (2002–2015), we found random patterns of both temporal nestedness and turnover, indicating stability in species composition. However, we also showed the contemporary occurrence of species loss and species replacement processes, considering the dry year 2003, a finding which should be further explored through fine-scale studies to unravel mechanisms of community assembly under drought. The use of a probabilistic sampling design representative of the different physiognomic vegetation types proved to be advantageous in monitoring the Nature Reserve vegetation and collecting reliable quantitative information. This data, in turn, provides the basis for improvements in management practices and proposed adaptation measures

    Role of Daptomycin in Cutaneous Wound Healing: A Narrative Review

    Daptomycin is active against Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus (MRSA) and the on-label indications for its use include complicated skin and skin structure infections (cSSSI). We performed a narrative review of the literature with the aim to evaluate the role of daptomycin in the skin wound healing process, proposing our point of view on the possible association with other molecules that could improve the skin healing process. Daptomycin may improve wound healing in MRSA-infected burns, surgical wounds, and diabetic feet, but further studies in humans with histological examination are needed. In the future, the combination of daptomycin with other molecules with synergistic action, such as vitamin E and derivates, IB-367, RNA III-inhibiting peptide (RIP), and palladium nanoflowers, may help to improve wound healing and overcome forms of antibiotic resistanc

    Weight of risk factors for mortality and short-term mortality displacement during the COVID-19 pandemic.

    Background: We conducted a population-based cohort study to estimate mortality before, during and after the COVID-19 peak and to compare mortality in 2020 with rates reported in previous years, with a view to helping decision makers to apply containment measures for high-risk groups. Methods: All deaths were collected between 2015 and 2020 from municipal registry database. In 2020, weeks 1-26 were stratified in three periods: before, during and after the COVID mortality peak. The Poisson Generalized Linear regression Model showed the “harvesting effect”. Three logistic regressions for 8 dependent variables (age and comorbidities) and a t-test of  differences described all-cause mortality risk factors in 2019 and 2020 and differences between COVID and non-COVID patients. Results: A total of 47,876 deaths were collected. All-cause deaths increased by 38.5% during the COVID peak and decreased by 18% during the post-peak period in comparison with the average registered during the control period (2015-19), with significant mortality displacement in 2020. Except for chronic renal injuries in subjects aged 45-64 years, diabetes and chronic cardiovascular diseases in those aged 65-84 years, and neuropathies in those aged >84 years, the weight of comorbidities in deaths was similar or lower in COVID subjects than in non-COVID subjects. Discussions: Surprisingly, the weight of comorbidities in death, compared to weight in non-COVID subjects allows you to highlight some surprising results such as COPD, IBD and Cancer. The excess mortality that we observed in the entire period were modest in comparison with initial estimates during the peak, owing to the mild influenza season and the harvesting effect starting from the second half of May.   &nbsp

    Transcutaneous vagus nerve stimulation combined with robotic rehabilitation improves upper limb function after stroke

    The efficacy of standard rehabilitative therapy for improving upper limb functions after stroke is limited; thus, alternative strategies are needed. Vagus nerve stimulation (VNS) paired with rehabilitation is a promising approach, but the invasiveness of this technique limits its clinical application. Recently, a noninvasive method to stimulate vagus nerve has been developed. The aim of the present study was to explore whether noninvasive VNS combined with robotic rehabilitation can enhance upper limb functionality in chronic stroke. Safety and efficacy of this combination have been assessed within a proof-of-principle, double-blind, semirandomized, sham-controlled trial. Fourteen patients with either ischemic or haemorrhagic chronic stroke were randomized to robot-assisted therapy associated with real or sham VNS, delivered for 10 working days. Efficacy was evaluated by change in upper extremity Fugl-Meyer score. After intervention, there were no adverse events and Fugl-Meyer scores were significantly better in the real group compared to the sham group. Our pilot study confirms that VNS is feasible in stroke patients and can produce a slight clinical improvement in association to robotic rehabilitation. Compared to traditional stimulation, noninvasive VNS seems to be safer and more tolerable. Further studies are needed to confirm the efficacy of this innovative approach

    Microsatellite Constellation for Mars Communication and Navigation

    Exploration of Mars and establishment of human settlement have been of sharp interest for several decades. Since the turn of the century, efforts have been ramped up to make these a reality. With the execution of multiple robotic exploration missions and several more planned missions in the next two decades, as well as serious plans for human landing missions, a key need is the establishment of accurate, reliable, expansive, and cost-effective positioning and communication service for several users in the Mars environment. The Mars Communication and Navigation (MCN) mission is a multi-satellite constellation at Mars that shall provide data relay and positioning services for the identified possible users, that are orbiters, landers, ascenders, autonomous rovers, and human landing missions. The aim of MCN is to investigate and prototype key technologies for a Mars positioning and communication system using small satellites, in order to enable the development and operations of a wide range of Mars missions, providing a backbone Earth–Mars communication and navigation infrastructure. This work focuses on the critical architectural aspects of the MCN. The end-to-end (E2E) system architecture is presented, in order to provide an overview of the space and ground segments along with the operations concepts. Concerning the orbital configuration, the constellation and its deployment strategy are discussed. The MCN constellation baseline comprises 24 microsatellites operating in a Walker-like orbital configuration at Mars to provide service for more than 70 users potentially. Moreover, a Relay/Gateway link is utilized to serve as a communication bridge between Earth ground segment and the MCN constellation. Concerning the communication and navigation aspects, their architectures and possible solutions are highlighted, together with an overview of the related critical technologies required to achieve the mission objectives

    Carfilzomib, lenalidomide, and dexamethasone in relapsed refractory multiple myeloma: a prospective real-life experience of the Regional Tuscan Myeloma Network

    IntroductionCarfilzomib, a potent, irreversible, selective proteasome inhibitor has demonstrated consistent results in relapsed/refractory multiple myeloma (RRMM) combined with lenalidomide and dexamethasone (KRd). No prospective studies are yet available that analyzed the efficacy of the KRd combination.MethodsHerein, we report a multicenter prospective observational study on 85 patients who were treated with KRd combination as the second or third line of treatment, according to standard practice.ResultsThe median age was 61 years; high-risk cytogenetic was found in 26% and renal impairment (estimated glomerular filtration rate (eGFR) <60 ml/min) in 17%. After a median follow-up of 40 months, patients received a median number of 16 cycles of KRd, with a median duration of treatment (DoT) of 18 months (range, 16.1–19.2 months). The overall response rate was 95%, with a high-quality response (≥very good partial remission [VGPR]) in 57% of the patients. The median progression-free survival (PFS) was 36 months (range, 29.1–43.2 months). Achievement of at least VGPR and a previous autologous stem cell transplantation (ASCT) were associated with longer PFS. The median overall survival (OS) was not reached (NR); the 5-year OS rate was 73%. Nineteen patients underwent KRd treatment as a bridge to autologous transplantation, obtaining a post-transplant minimal residual disease (MRD) negativity in 65% of cases. The most common adverse events were hematological, followed by infection and cardiovascular events, rarely G3 or higher, with a discontinuation rate for toxicities of 6%.
