    A physics-based theory to navigate across risks and opportunities in the performance space: Application to crisis management

    This article presents an original theory for system management, based on physics principles. That theory considers that risks and opportunity can be seen as forces pushing or pulling a system with regards to its objective and its KPIs. Based on that proposal, this article presents the theory, based on (i) identification of susceptibility of systems to internal and external characteristics (danger, favorable conditions), thus creating forces (risks and opportunities), and (ii) evaluation of the sensibility of systems to these forces, thus creating consequences (damages or benefits). This article also presents the practical vision of that theory by detailing the way to observe the force-inducted trajectory of the considered system with regards to its KPIs. An illustrative example and discussions about the perspectives conclude the article

    Precision-medicine findings from the FACE-SZ cohort to develop motivation-enhancing programs in real-world schizophrenia

    Background: In people with schizophrenia, major areas of everyday life are impaired, including independent living, productive activities, social relationships and overall quality of life. Enhanced understanding of factors that hinder real-life functioning is vital for treatments to translate into more positive outcomes. Aim: The goal of the present study was to identify factors associated with motivation deficits in real-life schizophrenia, and to assess its contribution to impaired functioning and quality of life. Methods: Based on previous literature and clinical experience, several factors were selected and grouped into factors potentially explaining motivation deficits. Some of these variables were never investigated before in relationship with motivation deficits. Results: In 561 patients with schizophrenia of the national FACE-SZ cohort living in the community, 235 (41.9%) reported severe motivation deficits. These deficits were found to be significantly associated with impaired socially useful activities, psychological and physical quality of life (in almost all domains), alcohol use disorder (aOR = 2.141, p = 0.021), severe nicotine dependence (aOR = 2.906, p < 0.001) independently of age and sex. No significant association was found for body mass index, metabolic syndrome or physical activity level. In the second model, we identified the following modifiable factors associated with motivation deficits: history of suicide attempt (aOR = 2.297, p = 0.001), positive symptoms (aOR = 1.052, p = 0.006), current major depressive episode (aOR = 2.627, p < 0.001), sleep disorders (aOR = 1.474, p = 0.024) and lower medication adherence (aOR = 0.836, p = 0.001) independently of gender, current alcohol use disorder, second-generation antipsychotics and akathisia. No significant association was found for negative symptoms, childhood trauma and inflammation. These results were maintained after removing patients with schizoaffective disorders or those with major depressive disorder. Interpretation: Motivation deficits are frequent and remain persistent unmet need in real-world schizophrenia that should be addressed in future guidelines. Based on our results, literature and clinical experience, we recommend to address in priority major depression, sleep, suicide, positive symptoms (when present and as early as possible) and medication adherence to improve motivation deficits of schizophrenia. © 2022 Informa UK Limited, trading as Taylor & Francis Group.Sorbonne Universités à Paris pour l'Enseignement et la RechercheFondaMental-Cohorte

    Hyperconnected City Logistics: Capillary Network Design and Management

    In hyperconnected city logistics, this dissertation focuses on capillary networks which enable first-and-last mile logistics and fulfillment activities and customer interfaces with logistics systems. Capillary logistics networks are central to several innovations aiming to enable fast and convenient service to consumers while reducing cost and negative externalities associated with logistics activities in urban environments under the pressure of e-commerce growth. In this dissertation, we examine two types of urban capillary logistics networks: smart locker bank networks and access hub networks. Smart locker banks enable the aggregation of customer locations into a network of unattended pickup-and-delivery points. Access hubs serve as consolidation and transshipment points for logistics service providers at the neighborhood level. Our objective is to provide a set of methods to design and manage capillary networks and identify key managerial insights to shape urban logistics. In doing so, we leverage concepts of the Physical Internet to examine modularity, hyperconnectivity and mobility solutions for smart locker bank and access hub networks.     The hypoxia-inducible factor α pathway couples angiogenesis to osteogenesis during skeletal development

    Skeletal development and turnover occur in close spatial and temporal association with angiogenesis. Osteoblasts are ideally situated in bone to sense oxygen tension and respond to hypoxia by activating the hypoxia-inducible factor α (HIFα) pathway. Here we provide evidence that HIFα promotes angiogenesis and osteogenesis by elevating VEGF levels in osteoblasts. Mice overexpressing HIFα in osteoblasts through selective deletion of the von Hippel–Lindau gene (Vhl) expressed high levels of Vegf and developed extremely dense, heavily vascularized long bones. By contrast, mice lacking Hif1a in osteoblasts had the reverse skeletal phenotype of that of the Vhl mutants: long bones were significantly thinner and less vascularized than those of controls. Loss of Vhl in osteoblasts increased endothelial sprouting from the embryonic metatarsals in vitro but had little effect on osteoblast function in the absence of blood vessels. Mice lacking both Vhl and Hif1a had a bone phenotype intermediate between those of the single mutants, suggesting overlapping functions of HIFs in bone. These studies suggest that activation of the HIFα pathway in developing bone increases bone modeling events through cell-nonautonomous mechanisms to coordinate the timing, direction, and degree of new blood vessel formation in bone
