    Valuing Mortality Risk in China: Comparing Stated-Preference Estimates from 2005 and 2016

    We estimate the marginal rate of substitution of income for reduction in current annual mortality risk (the “value per statistical life” or VSL) using stated-preference surveys administered to independent samples of the general population of Chengdu China in 2005 and 2016. We evaluate the quality of estimates by the theoretical criterion that WTP for risk reduction should be strictly positive and nearly proportional to the magnitude of the risk reduction (evaluated by comparing answers between respondents). We test the effect of excluding respondents whose answers violate these validity criteria. For subsamples of respondents that satisfy the criteria, point estimates of the sensitivity of WTP to risk reduction are consistent with theory and yield estimates of VSL that are two to three times larger than estimated using the full samples. Between 2005 and 2016, estimated VSL increased sharply, from about 22,000 USD in 2005 to 550,000 USD in 2016. Income also increased substantially over this period. Attributing the change in VSL solely to the change in income implies an income elasticity of about 2.5

    Arctic Ocean Simulations in the CMIP6 Ocean Model Intercomparison Project (OMIP)

    oai:publications.copernicus.org:gmdd107357Arctic Ocean simulations in 19 global ocean-sea ice models participating in the Ocean Model Intercomparison Project (OMIP) of the CMIP6 are evaluated in this paper. Our results indicate that no significant improvements were achieved in the Arctic Ocean simulations from the previous Coordinated Ocean-ice Reference Experiments phase II (CORE-II) to the current OMIP. Large model biases and inter-model spread exist in the simulated mean state of the halocline and Atlantic Water layer in the OMIP models. Most of the OMIP models suffer from too thick and deep Atlantic Water layer, too deep halocline base, and large fresh biases in the halocline. The OMIP models largely agree on the inter-annual and decadal variability of the Arctic Ocean freshwater content and volume/heat/freshwater transports through the Arctic Ocean gateways. The models can reproduce observed changes in volume, heat and freshwater transports through the gateways except for the Bering Strait. Overall, the performance of the Arctic Ocean simulations is similar between the CORE2-forced OMIP-1 and JRA55-do-forced OMIP-2.</p

    High-fiber-diet-related metabolites improve neurodegenerative symptoms in patients with obesity with diabetes mellitus by modulating the hippocampal–hypothalamic endocrine axis

    ObjectiveThrough transcriptomic and metabolomic analyses, this study examined the role of high-fiber diet in obesity complicated by diabetes and neurodegenerative symptoms.MethodThe expression matrix of high-fiber-diet-related metabolites, blood methylation profile associated with pre-symptomatic dementia in elderly patients with type 2 diabetes mellitus (T2DM), and high-throughput single-cell sequencing data of hippocampal samples from patients with Alzheimer's disease (AD) were retrieved from the Gene Expression Omnibus (GEO) database and through a literature search. Data were analyzed using principal component analysis (PCA) after quality control and data filtering to identify different cell clusters and candidate markers. A protein–protein interaction network was mapped using the STRING database. To further investigate the interaction among high-fiber-diet-related metabolites, methylation-related DEGs related to T2DM, and single-cell marker genes related to AD, AutoDock was used for semi-flexible molecular docking.ResultBased on GEO database data and previous studies, 24 marker genes associated with high-fiber diet, T2DM, and AD were identified. Top 10 core genes include SYNE1, ANK2, SPEG, PDZD2, KALRN, PTPRM, PTPRK, BIN1, DOCK9, and NPNT, and their functions are primarily related to autophagy. According to molecular docking analysis, acetamidobenzoic acid, the most substantially altered metabolic marker associated with a high-fiber diet, had the strongest binding affinity for SPEG.ConclusionBy targeting the SPEG protein in the hippocampus, acetamidobenzoic acid, a metabolite associated with high-fiber diet, may improve diabetic and neurodegenerative diseases in obese people

    Tourism experiences reduce the risk of cognitive impairment in the Chinese older adult: A prospective cohort study

    Background: Given the etiological complexity of cognitive impairment, no effective cure currently exists for precise treatment of dementia. Although scholars have noted tourism’s potential role in managing cognitive impairment and mild dementia, more robust empirical investigation is needed in this area. This study aimed to examine the associations between tourism and cognitive impairment and dementia in older Chinese adults. Method: From a nationwide community-based cohort, 6,717 individuals aged ≥ 60 were recruited from 2011 to 2014, of whom 669 (9.96%) had had at least one tourism experience in the 2 years prior to enrollment. All the participants were then prospectively followed up until 2018. The association between tourism and cognitive impairment was examined by the Cox proportional hazards regression model. The adjusted hazard ratio (aHR) and its 95% confidence interval (CI) were calculated to evaluate the effect of tourism experience on cognitive impairment and dementia. Results: A total of 1,416 individuals were newly diagnosed with cognitive impairment and 139 individuals with dementia onset during follow-up. The incidence of cognitive impairment was significantly lower among participants with tourism experiences (316.94 per 10,000 person-years) than those without such experiences (552.38 per 10,000 person-years). Cox regression showed that tourism decreased the risk of cognitive impairment (aHR = 0.69, 95% CI: 0.41–0.62) when adjusted for behavioral covariates and characteristics. Compared with participants without tourism experiences, those with 1, 2, and ≥ 3 tourism experiences had a lower risk of cognitive impairment with the aHRs of 0.72 (95% CI: 0.52–0.99), 0.65 (0.42–1.01), and 0.68 (0.44–0.98), respectively. Tourism experiences also reduced participants’ risk of dementia (aHR = 0.41, 95% CI: 0.19–0.89). Conclusion: Our findings demonstrated associations between tourism and reduced risks of cognitive impairment and dementia in older Chinese adults. Thus, tourism could serve as a novel approach to dementia prevention

    Classic and targeted anti-leukaemic agents interfere with the cholesterol biogenesis metagene in acute myeloid leukaemia: Therapeutic implications

    Despite significant advances in deciphering the molecular landscape of acute myeloid leukaemia (AML), therapeutic outcomes of this haematological malignancy have only modestly improved over the past decades. Drug resistance and disease recurrence almost invariably occur, highlighting the need for a deeper understanding of these processes. While low O2 compartments, such as bone marrow (BM) niches, are well‐recognized hosts of drug‐resistant leukaemic cells, standard in vitro studies are routinely performed under supra‐physiologic (21% O2, ambient air) conditions, which limits clinical translatability. We hereby identify molecular pathways enriched in AML cells that survive acute challenges with classic or targeted therapeutic agents. Experiments took into account variations in O2 tension encountered by leukaemic cells in clinical settings. Integrated RNA and protein profiles revealed that lipid biosynthesis, and particularly the cholesterol biogenesis branch, is a particularly therapy‐induced vulnerability in AML cells under low O2 states. We also demonstrate that the impact of the cytotoxic agent cytarabine is selectively enhanced by a high‐potency statin. The cholesterol biosynthesis programme is amenable to additional translational opportunities within the expanding AML therapeutic landscape. Our findings support the further investigation of higher‐potency statin (eg rosuvastatin)–based combination therapies to enhance targeting residual AML cells that reside in low O2 environments

    Overview and prospect of the detection capability of China's first precipitation measurement satellite FY-3G

    Based on introducing the technical characteristics of FY-3G, which is China's first precipitation measurement satellite and successfully launched at 09∶36 BT on April 16 in 2023, this paper focuses on the precipitation detection capabilities and application prospect in rainstorm monitoring of FY-3G. The results show that, with an orbit at 407 km and an inclination angle of 50°, and equipped with a dual-frequency Ka/Ku band precipitation measurement radar, microwave, and optical imaging instruments, the FY-3G satellite can detect the three-dimensional structure of disastrous weather systems such as typhoon, heavy rainfall, and other strong convection events in most of China. At the design level, FY-3G has precipitation detection capabilities comparable to the current US Second Generation Global Precipitation Measurement Program (GPM) Core Satellite (GPMCO), but better payload types, quantities, and channel settings compared with the GPMCO satellite. After the service operation, the FY-3G satellite, together with other polar-orbiting meteorological satellites such as FY-3 AM, PM, and EM, as well as high-orbit geostationary satellites, will form the Fengyun precipitation detection constellation system, which will improve the overall precipitation detection capability of the Fengyun Satellite constellation and provide stronger basic support for meteorological disaster prevention and mitigation

    Multiple Kinases Involved in the Nicotinic Modulation of Gamma Oscillations in the Rat Hippocampal CA3 Area

    Neuronal synchronization at gamma band frequency (20–80 Hz, γ oscillations) is closely associated with higher brain function, such as learning, memory and attention. Nicotinic acetylcholine receptors (nAChRs) are highly expressed in the hippocampus, and modulate hippocampal γ oscillations, but the intracellular mechanism underlying such modulation remains elusive. We explored multiple kinases by which nicotine can modulate γ oscillations induced by kainate in rat hippocampal area CA3 in vitro. We found that inhibitors of cyclic AMP dependent kinase (protein kinase A, PKA), protein kinase C (PKC), N-methyl-D-aspartate receptor (NMDA) receptors, Phosphoinositide 3-kinase (PI3K) and extracellular signal-related kinases (ERK), each individually could prevent the γ oscillation-enhancing effect of 1 μM nicotine, whereas none of them affected baseline γ oscillation strength. Inhibition of the serine/threonine kinase Akt increased baseline γ oscillations and partially blocked its nicotinic enhancement. We propose that the PKA-NMDAR-PI3K-ERK pathway modifies cellular properties required for the nicotinic enhancement of γ oscillations, dependent on a PKC-ERK mediated pathway. These signaling pathways provide clues for restoring γ oscillations in pathological conditions affecting cognition. The suppression of γ oscillations at 100 μM nicotine was only dependent on PKA-NMDAR activation and may be due to very high intracellular calcium levels
