    Fibrinogen Sevilla, a congenital dysfibrinogenemia characterized by an abnormal monomer aggregation and a defective plasmin lysis

    A dysfibrinogenemia (fibrinogen Sevilla) was detected in a 64-yr-old woman with no previous history of hemorrhagic diathesis or thrombosis. Thrombin and reptilase times were prolonged. The aggregation of fibrin monomers showed a prolonged latency time with a defective slope although fibrinopeptide release and clot stabilization were found to be normal. Plasmin proteolysis was abnormal with a much slower plasmic degradation in patient's purified fibrinogen. By chromatofocussing the patient's fibrinogen showed an abnormality in pattern elution with a second peak eluting at a pH slightly more basic than the normal one (pH 5.5). Likewise, the isoelectrofocussing of purified non-reduced patient's fibrinogen in agarose gel showed an abnormal distribution in its focussed bands, especially in a group which focussed in a pI-interval between 5.20-5.85. By two-dimensional electrophoresis we did not find any abnormality in the fibrinogen-reduced chains. These results could indicate that the abnormal monomer aggregation, as well as the defective plasmin lysis, could be due to conformational aspects of fibrinogen rather than to structural defects

    Identificación de genotipos de papa con tolerancia al déficit hídrico

    Con el objetivo de identificar genotipos de papa (Solanum spp.) con tolerancia al déficit hídrico, se implementó un ensayo en invernadero en la EESC del INIAP. Se evaluaron 39 genotipos con y sin déficit hídrico. Las variables evaluadas fueron: potencial de recuperación (PR), contenido relativo de agua (CRA), potencial hídrico (Ψ h), número de tubérculos por planta (NTP), rendimiento por planta (RP) y promedio geométrico del rendimiento (PGR). Se encontró un efecto significativo de genotipos (G), déficit hídrico (E) y su interacción G x E. La variedad INIAP-Josefina mostró tolerancia al déficit hídrico en la mayoría de variables. Esta investigación mostró genotipos con un mejor PR que otros, a los 16 días de déficit hídric o y a las 24 horas de recuperación. Para CRA se encontraron genotipos a los 13 y 16 días de déficit hídrico con valores superiores a los 66,98 y 62,98% respectivamente. Los clones 11-9-108, 12-4-145 e INIAP-Josefina con más de 9 tubérculos por planta se ubicaron en los primeros rangos, mientras INIAP-Josefina, 11-9-85, 11-9-45 y 12-4-50 presentaron rendimientos superiores a 132 g/planta. El PGR estableció a INIAP-Josefina, 11-9-45 y 11-9-85 en los primeros rangos con valores superiores a 122,54. Al evaluar el Ψh con déficit hídrico se encontró a INIAP-Catalina, INIAP-Estela y los clones 11-9-66, 11-9-85, 11-9-28 y 11-9-92 mostraron el menor efecto del estrés hídrico con valores menores. Existió variación en la respuesta de los genotipos al déficit hídrico, se han identificado genotipos con tolerancia que continuarán el proceso de evaluación dentro del esquema de mejoramiento

    Endothelial Progenitor Cells as a Potential Biomarker in Interstitial Lung Disease Associated with Rheumatoid Arthritis

    Interstitial lung disease (ILD) increases morbidity and mortality in patients with rheumatoid arthritis (RA). Although the pathogenesis of ILD associated with RA (RA-ILD(+)) remains poorly defined, vascular tissue is crucial in lung physiology. In this context, endothelial progenitor cells (EPC) are involved in endothelial tissue repair. However, little is known about their implication in RA-ILD(+). Accordingly, we aimed to investigate the potential role of EPC related to endothelial damage in RA-ILD(+). EPC quantification in peripheral blood from 80 individuals (20 RA-ILD(+) patients, 25 RA-ILD(-) patients, 21 idiopathic pulmonary fibrosis (IPF) patients, and 14 healthy controls) was performed by flow cytometry. EPC were considered as CD34(+), CD45(low), CD309(+) and CD133(+). A significant increase in EPC frequency in RA-ILD(+) patients, as well as in RA-ILD(-) and IPF patients, was found when compared with controls (p < 0.001, p = 0.02 and p < 0.001, respectively). RA-ILD(+) patients exhibited a higher EPC frequency than the RA-ILD(-) ones (p = 0.003), but lower than IPF patients (p < 0.001). Our results suggest that EPC increase may represent a reparative compensatory mechanism in patients with RA-ILD(+). The degree of EPC frequency may help to identify the presence of ILD in RA patients and to discriminate RA-ILD(+) from IPF

    Spatial surface-pattern analyses and boundary detection techniques applied in forest ecology

    [EN] We review methods for uni- and multivariate surface pattern analysis and boundary detection used in forest ecology. A continuous surface pattern is defined as the locations of points (trees) in the space and the associated variable or variables. We illustrate useful methods to describe spatial patterns and infer the generating processes. We show the statistical basis and applied examples of univariate methods for binary (join counts) and quantitative variables (Moran and Geary correlograms, semivariograms, fractal dimension). We explain the calculus and interpretation of multivariate methods to describe surface patterns (Mantel test and correlogram) and their relationships with ordination methods. Finally, we show examples of techniques for boundary detection. Most analysed patterns corresponded to Pinus uncinata forests from the upper altitudinal limit in the Pyrenees or from a relict population. We discuss the advantages and disadvantages of each methodology and their applications in forest ecology.[ES] En este trabajo se revisan los métodos de análisis univariable y multivariable de los patrones de superficies y de detección de fronteras más utilizados en ecología forestal. El patrón de superficies es un patrón espacial continuo definido por las posiciones de los puntos (árboles) en el espacio y una o varias variables asociadas a cada punto. Se ilustran métodos útiles para describir patrones espaciales e inferir los procesos que los generaron. Se muestra el fundamento estadístico y ejemplos aplicados de métodos de análisis univariables para variables binarias (conteo contiguo) y cuantitativas (correlogramas de Moran y Geary, semivariogramas, dimensión fractal). Se detalla el cálculo e interpretación de métodos multivariables para la descripción de patrones de superficies (correlograma y test de Mantel) y su relación con los métodos de ordenación. Finalmente, se muestran ejemplos de métodos para la detección de fronteras. La mayor parte de los patrones reales analizados provienen de bosques de Pinus uncinata del límite altitudinal superior en los Pirineos o bien de una población relíctica.     A topolographical approach to infrastructure:Political topography, topology and the port of Dar es Salaam

    Economic infrastructure hubs, such as ports, are crucial sites for exploring new political geographies. In such environments, mobilities are enabled and rigidly channelled premised on the stasis of the port-as-checkpoint. Such nodes are part of an ever-growing political geography of zones that requires more attention. This article proposes a ‘topolographical’ approach – a combined heuristic drawing from political topography and topology – to comprehend more fully the transformations in the political geographies of large-scale infrastructures. The cardinal nature of the port of Dar es Salaam makes it a crucial site through which to illustrate the purchase of this framework. The topographical analysis puts forward the port of Dar as ‘archipelago of global territories’, within which heterogeneous actors claim graduated authority. Drawing on topology, the article shows what is folded into the port, constantly shaping not only who governs but, more importantly, how power and authority are exercised. It will be shown how imaginaries of the port - as gateway, seamless space, and modernity ‘from scratch’ - as much as new technological devices work to produce historically and geographically distinct political geographies, and indeed bring new ones into being

    Single-molecule experiments in biological physics: methods and applications

    I review single-molecule experiments (SME) in biological physics. Recent technological developments have provided the tools to design and build scientific instruments of high enough sensitivity and precision to manipulate and visualize individual molecules and measure microscopic forces. Using SME it is possible to: manipulate molecules one at a time and measure distributions describing molecular properties; characterize the kinetics of biomolecular reactions and; detect molecular intermediates. SME provide the additional information about thermodynamics and kinetics of biomolecular processes. This complements information obtained in traditional bulk assays. In SME it is also possible to measure small energies and detect large Brownian deviations in biomolecular reactions, thereby offering new methods and systems to scrutinize the basic foundations of statistical mechanics. This review is written at a very introductory level emphasizing the importance of SME to scientists interested in knowing the common playground of ideas and the interdisciplinary topics accessible by these techniques. The review discusses SME from an experimental perspective, first exposing the most common experimental methodologies and later presenting various molecular systems where such techniques have been applied. I briefly discuss experimental techniques such as atomic-force microscopy (AFM), laser optical tweezers (LOT), magnetic tweezers (MT), biomembrane force probe (BFP) and single-molecule fluorescence (SMF). I then present several applications of SME to the study of nucleic acids (DNA, RNA and DNA condensation), proteins (protein-protein interactions, protein folding and molecular motors). Finally, I discuss applications of SME to the study of the nonequilibrium thermodynamics of small systems and the experimental verification of fluctuation theorems.     The Mexican consensus on the diagnosis, treatment, and prevention of NSAID-induced gastropathy and enteropathy

    Más de 30 millones de personas consumen diariamente antiinflamatorios noesteroideos (AINE) en el mundo, y este consumo se ve incrementado anualmente. Aunque losAINE poseen propiedades analgésicas y antiinflamatorias, sus eventos adversos gastrointesti-nales son bien reconocidos. En nuestro país no existía un consenso respecto al diagnóstico,tratamiento y prevención de la gastropatía y la enteropatía por AINE, por lo que la AsociaciónMexicana de Gastroenterología reunió a un grupo de expertos para establecer recomendacionesde utilidad para la comunidad médica. En este consenso se emitieron 33 recomendaciones. Elconsenso destaca que el riesgo de toxicidad gastrointestinal de los AINE varía según el fármacoempleado y su farmacocinética, lo cual debe ser considerado al momento de su prescripción. Losfactores de riesgo de complicación gastroduodenal por AINE son: antecedente de úlcera pép-tica, edad mayor a 65 a˜nos, dosis altas del AINE, infección por Helicobacter pylori (H.pylori), ypresencia de comorbilidades graves. Los síntomas y el da˜no gastroduodenal inducido por AINEson variables ya que puede cursar asintomático o manifestarse como anemia por deficiencia dehierro, hemorragia, estenosis y perforación. La cápsula endoscópica y la enteroscopia son méto-dos diagnósticos directos en la enteropatía por AINE. Respecto a la prevención, se recomiendaprescribir la dosis mínima necesaria de un AINE para obtener el efecto deseado y durante elmenor tiempo. Finalmente, los inhibidores de la bomba de protones (IBP) representan el están-dar de oro para la profilaxis y tratamiento de los efectos gastroduodenales, mas no son útilesen la enteropatía

    Efficient Excitation of Channel Plasmons in Tailored, UV-Lithography-Defined V-Grooves

    [Image: see text] We demonstrate the highly efficient (>50%) conversion of freely propagating light to channel plasmon-polaritons (CPPs) in gold V-groove waveguides using compact 1.6 μm long waveguide-termination coupling mirrors. Our straightforward fabrication process, involving UV-lithography and crystallographic silicon etching, forms the coupling mirrors innately and ensures exceptional-quality, wafer-scale device production. We tailor the V-shaped profiles by thermal silicon oxidation in order to shift initially wedge-located modes downward into the V-grooves, resulting in well-confined CPPs suitable for nanophotonic applications

    Normal reference values of strength in pelvic floor muscle of women: a descriptive and inferential study

    Background: To describe the clinical, functional and quality of life characteristics in women with Stress Urinary Incontinence (SUI). In addition, to analyse the relationship between the variables reported by the patients and those informed by the clinicians, and the relationship between instrumented variables and the manual pelvic floor strength assessment.Methods: Two hundred and eighteen women participated in this observational, analytical study. An interview about Urinary Incontinence and the quality of life questionnaires (EuroQoL-5D and SF-12) were developed as outcomes reported by the patients. Manual muscle testing and perineometry as outcomes informed by the clinician were assessed. Descriptive and correlation analysis were carried out.Results: The average age of the subjects was (39.93 ± 12.27 years), (24.49 ± 3.54 BMI). The strength evaluated by manual testing of the right levator ani muscles was 7.79 ± 2.88, the strength of left levator ani muscles was 7.51 ± 2.91 and the strength assessed with the perineometer was 7.64 ± 2.55. A positive correlation was found between manual muscle testing and perineometry of the pelvic floor muscles (p < .001). No correlation was found between outcomes of quality of life reported by the patients and outcomes of functional capacity informed by the physiotherapist.Conclusion: A stratification of the strength of pelvic floor muscles in a normal distribution of a large sample of women with SUI was done, which provided the clinic with a baseline. There is a relationship between the strength of the pelvic muscles assessed manually and that obtained by a perineometer in women with SUI. There was no relationship between these values of strength and quality of life perceived

    HLA association with the susceptibility to anti-synthetase syndrome

    Objective: To investigate the human leukocyte antigen (HLA) association with anti-synthetase syndrome (ASSD). Methods: We conducted the largest immunogenetic HLA-DRB1 and HLA-B study to date in a homogeneous cohort of 168 Caucasian patients with ASSD and 486 ethnically matched healthy controls by sequencing-based-typing. Results: A statistically significant increase of HLA-DRB1*03:01 and HLA-B*08:01 alleles in patients with ASSD compared to healthy controls was disclosed (26.2% versus 12.2%, P = 1.56E–09, odds ratio–OR [95% confidence interval–CI] = 2.54 [1.84–3.50] and 21.4% versus 5.5%, P = 18.95E–18, OR [95% CI] = 4.73 [3.18–7.05]; respectively). Additionally, HLA-DRB1*07:01 allele was significantly decreased in patients with ASSD compared to controls (9.2% versus 17.5%, P = 0.0003, OR [95% CI] = 0.48 [0.31–0.72]). Moreover, a statistically significant increase of HLA-DRB1*03:01 allele in anti-Jo-1 positive compared to anti-Jo-1 negative patients with ASSD was observed (31.8% versus 15.5%, P = 0.001, OR [95% CI] = 2.54 [1.39–4.81]). Similar findings were observed when HLA carrier frequencies were assessed. The HLA-DRB1*03:01 association with anti-Jo-1 was unrelated to smoking history. No HLA differences in patients with ASSD stratified according to the presence/absence of the most representative non-anti-Jo-1 anti-synthetase autoantibodies (anti-PL-12 and anti-PL-7), arthritis, myositis or interstitial lung disease were observed. Conclusions: Our results support the association of the HLA complex with the susceptibility to ASSD
