    Spread of epileptiform activity in the immature rat neocortex studied with voltage-sensitive dyes and laser scanning microscopy

    1. Adult rats and rats with a postnatal age of 3-29 days (PN 3-29) were used for the preparation of in vitro slices of the frontal neocortex. Epileptiform activity was induced by bath application of the gamma-aminobutyric acid-A (GABAA) receptor antagonists bicuculline or picrotoxin. 2. The voltage-sensitive dye RH 414 and a laser scanning microscope were used for multiple-site optical recordings of membrane potential changes associated with epileptiform activity. Optical signals were compared with simultaneously measured extra-cellular field potentials. 3. Optical signals could be reliably recorded for the duration of the experiments (2-4 h). Extracellular recordings of convulsant-induced paroxysmal depolarizing shifts (PDSs) in slices stained with RH 414 were comparable with those obtained in unstained slices. Changes in dye signals in response to reductions in extracellular calcium, addition of tetrodotoxin (TTX), or application of excitatory amino acid receptor antagonists indicate that the fluorescence changes correlate well with established electrophysiological measures of epileptiform activity. 4. In slices from adult animals, dye signals were observed at all recording sites. The response with the shortest latency occurred invariably at the site of stimulation, and activity spread rapidly in both vertical and horizontal directions. Spread was significantly faster in the vertical than in the horizontal direction. 5. Epileptiform activity was absent or only weakly expressed in slices from PN 3-9 animals. Activity was detectable predominantly in upper cortical layers. 6. Dye signals were observed at all measurement points in slices from PN 10-19 animals. In this age group, peak amplitude increased with spread of activity from lower to upper cortical layers. There was no significant difference between the speed of propagation in the vertical and in the horizontal directions. Spontaneous epileptiform activity occurred at a high rate in the PN 10-19 age group, and signals associated with spontaneous epileptiform events were largest in upper layers. 7. In the PN 10-19 age group, optical signals were characterized by the repetitive occurrence of PDS discharges superimposed on a sustained response. The amplitude of the sustained response decreased with increasing distance from the site of stimulation. Analysis of the latencies revealed that the superimposed PDS-like events were generated at multiple sites within the scanning area.     Abstract BACKGROUND: The number of homeless pregnant women is continuing to increase around the world. Homeless women are experiencing difficulties with their pregnancy that others may not face, including birth abnormalities and not seeking appropriate care due to cost and stigmatization. Nurses need to educate homeless pregnant women on what is needed for a healthy pregnancy and childbirth. OBJECTIVE: This exploratory research describes the needs for advocacy in women who are experiencing homelessness while pregnant and how homelessness affects pregnancy, childbirth, and postpartum care. LITERATURE REVIEW: The search methods used was CINAHL and ETSU Library. Keywords: homelessness, childbirth, homeless women, nursing. There were ten studies established all together. FINDINGS: Homeless pregnant women are not educated on contraceptives and what they need to do to have a healthy pregnancy. For example, they don’t know that being homeless and absence of proper care results in abnormalities such as low birth weights, longer NICU stays, and premature childbirths. CONCLUSION: We conclude that homeless pregnant women should be advocated for in hopes they will feel comfortable and seek proper care for a safe pregnancy.     High performance, large cross section S bender for neutron polarization

The results of the experiment show for the wavelength of 4.4 amp; 730;A a homogenous polarization of more than 98 across the bender cross section and a high transmission of above 65 of the spin up component of the neutron beam.     Smoothness-constrained time-lapse inversion of data from 3D resistivity surveys

    Three-dimensional resistivity surveys and their associated inversion models are required to accurately resolve structures exhibiting very complex geology. In the same light, 3D resistivity surveys collected at multiple times are required to resolve temporally varying conditions. In this work we present 3D data sets, both synthetic and real, collected at different times. The large spatio-temporal data sets are then inverted simultaneously using a least-squares methodology that incorporates roughness filters in both the space and time domains. The spatial roughness filter constrains the model resistivity to vary smoothly in the x-, y- and z-directions. A temporal roughness filter is also applied that minimizes changes in the resistivity between successive temporal inversion models and the L-curve method is used to determine the optimum weights for both spatial and temporal roughness filters. We show that the use of the temporal roughness filter can accurately resolve changes in the resistivity even in the presence of noise. The L1- and L2-norm constraints for the temporal roughness filter are first examined using a synthetic model. The synthetic data test shows that the L1-norm temporal constraint produces significantly more accurate results when the resistivity changes abruptly with time. The model obtained with the L1-norm temporal constraint is also less sensitive to random noise compared with independent inversions (i.e., without any temporal constraint) and the L2-norm temporal constraint. Anomalies that are common in models using independent inversions and the L2-norm and L1-norm temporal constraints are likely to be real. In contrast, anomalies present in a model using independent inversions but that are significantly reduced with the L2-norm and L1-norm constraints are likely artefacts. For field data sets, the method successfully recovered temporal changes in the subsurface resistivity from a landfill monitoring survey due to rainwater infiltration, as well as from an experiment to map the migration of sodium cyanide solution from an injection well using surface and borehole electrodes in an area with significant topography

    Arrested Cracks in Nonlinear Lattice Models of Brittle Fracture

    We generalize lattice models of brittle fracture to arbitrary nonlinear force laws and study the existence of arrested semi-infinite cracks. Unlike what is seen in the discontinuous case studied to date, the range in driving displacement for which these arrested cracks exist is very small. Also, our results indicate that small changes in the vicinity of the crack tip can have an extremely large effect on arrested cracks.     Addressing moderated mediation hypotheses: Theory, methods, and prescriptions

    This article provides researchers with a guide to properly construe and conduct analyses of conditional indirect effects, commonly known as moderated mediation effects. We disentangle conflicting definitions of moderated mediation and describe approaches for estimating and testing a variety of hypotheses involving conditional indirect effects. We introduce standard errors for hypothesis testing and construction of confidence intervals in large samples but advocate that researchers use bootstrapping whenever possible. We also describe methods for probing significant conditional indirect effects by employing direct extensions of the simple slopes method and Johnson-Neyman technique for probing significant interactions. Finally, we provide an SPSS macro to facilitate the implementation of the recommended asymptotic and bootstrapping methods. We illustrate the application of these methods with an example drawn from the Michigan Study of Adolescent Life Transitions, showing that the indirect effect of intrinsic student interest on mathematics performance through teacher perceptions of talent is moderated by student math self-concept

    Human Mars Mission Design - The Ultimate Systems Challenge

    A human mission to Mars will occur at some time in the coming decades. When it does, it will be the end result of a complex network of interconnected design choices, systems analyses, technical optimizations, and non-technical compromises. This mission will extend the technologies, engineering design, and systems analyses to new limits, and may very well be the most complex undertaking in human history. It can be illustrated as a large menu, or as a large decision tree. Whatever the visualization tool, there are numerous design decisions required to assemble a human Mars mission, and many of these interconnect with one another. This paper examines these many decisions and further details a number of choices that are highly interwoven throughout the mission design. The large quantity of variables and their interconnectedness results in a highly complex systems challenge, and the paper illustrates how a change in one variable results in ripples (sometimes unintended) throughout many other facets of the design. The paper concludes with a discussion of some mission design variables that can be addressed first, and those that have already been addressed as a result of ongoing National Aeronautics and Space Administration (NASA) developments, or as a result of decisions outside the technical arena. It advocates the need for a 'reference design' that can be used as a point of comparison, and to illustrate the system-wide impacts as design variables change

    An observational study of safety and clinical outcome measures across patient groups in the United Kingdom Medical Cannabis Registry.

    BACKGROUND: There is a paucity of high-quality data on patient outcomes and safety after initiating treatment with cannabis-based medicinal products (CBMPs). The aim of this study was to assess the clinical outcomes and safety of CBMPs by analyzing patient-reported outcome measures and adverse events across a broad spectrum of chronic conditions. RESEARCH DESIGN AND METHODS: This study analyzed patients enrolled in the UK Medical Cannabis Registry. Participants completed the EQ-5D-5L to assess health-related quality of life, Generalized Anxiety Disorder-7 (GAD-7) questionnaire to measure anxiety severity, and the Single-item Sleep Quality Scale (SQS) to rate sleep quality at baseline and follow-up after 1, 3, 6, and 12 months. RESULTS: A total of 2833 participants met inclusion criteria. The EQ-5D-5L index value, GAD-7, and SQS all improved at each follow-up (p  0.050). Adverse events were reported by 474 (16.73%) participants. CONCLUSIONS: This study suggests that CBMPs are associated with an improvement in health-related quality of life in UK patients with chronic diseases. Treatment was tolerated well by most participants, but adverse events were more common in female and cannabis-naïve patients
