    Effect of Nigella sativa L. on heart rate and some haematological values of alloxan-induced diabetic rabbits

    This study was designed to investigate the effect of an extract of Nigella sativa L. on the heart rate and  some haematological values in alloxan-induced diabetic rabbits. Fifteen New Zealand male rabbits were  divided into three experimental groups: control, diabetic and N. sativa L.-treated diabetic. At the end of the  experimental period (2 months), animals in all three groups were fasted for 12 hours and blood samples  were taken for the determination of glucose levels, RBC and WBC (red and white blood cell) counts,  packed cell volume (PCV), and haemoglobin (Hb) concentration. Heart rates were also measured by a  direct-writing electrocardiograph before the blood withdrawals. It was found that N. sativa L. treatment  increased the lowered RBC and WBC counts, PCV and neutrophil percentage in diabetic rabbits. However,  the WBC count of the N. sativa L. treated diabetic group was still lower than the control. N. sativa L.  treatment also decreased the elevated heart rate and glucose concentration of diabetic rabbits. It is concluded  that oral N. sativa L. treatment might decrease the diabetes-induced disturbances of heart rate and some  haematological parameters of alloxan-induced diabetic rabbits.

    Optimising a nonlinear utility function in multi-objective integer programming

    In this paper we develop an algorithm to optimise a nonlinear utility function of multiple objectives over the integer efficient set. Our approach is based on identifying and updating bounds on the individual objectives as well as the optimal utility value. This is done using already known solutions, linear programming relaxations, utility function inversion, and integer programming.     In this study, monthly streamflow data of eleven stations operated by EIE (ElectricalPower Resources Survey and Development Administration) located at Sakarya river basin wereinvestigated. Three different non-parametric trend tests (Sens T, the Spearmans Rho and the SeasonalKendall tests) were selected to determine linear trends of monthly flow data. The linear slopes of trendswere calculated by using a non-parametric estimator developed by Sen. The beginning of trends wasdetermined by using the Mann-Kendall rank correlation test. In addition, homogeneities in monthlytrends were tested by using a method developed by Van Belle and Hughes. According to the analysescarried out; meaningfull trends are determined (at the 0.05 significance level) in 64% of total 132 monthof 11 stations. Generally, the determined changes are in decreasing trend. The decreasing trend ismaximum in the stations 1221 and 1243 and they are located in northwest of the basin. Maximumnegative slopes are determined in March for all stations except for stations 1216 and 1226. March is theone in which maximum number of trend is determined (10 station) and all of trends are downward. AlsoMarch is the month in which maximum slope is observed. April is the month in which least trend isdetermined (5 station). The beginning of trends was observed in the early 1980s. According to the VanBelle and Hughes Homogeneity test, there is no global trend in the basin.Bu çalõşmada Sakarya havzasõnda bulunan ve EİE tarafõndan işletilen 11 adet akõm gözlemistasyonunun aylõk ortalama akõm verileri kullanõlmõştõr. Aylõk akõm verilerinin lineer trendlerini tespitetmek için, parametrik olmayan üç farklõ metot (Spearmanõn Rho, Senin T ve Mevsimsel Mann-Kendalltestleri) kullanõlmõştõr. Trendlerin lineer eğimleri, Sen tarafõndan geliştirilen ve parametrik olmayan birmetot olan Senin trend eğim metodu kullanõlarak hesaplanmõştõr. Trend tespit edilen aylardaki değişim genellikle azalan yönde olmuştur. 1221ve 1243 numaralõ istasyonlar azalan yönde değişimin en fazla olduğu istasyonlardõr ve bu istasyonlarhavzanõn kuzey batõ kõsmõnda yer almaktadõr. 1216 ve 1226 numaralõ istasyonlar dõşõndaki diğer bütünistasyonlarõn maksimum negatif eğimleri Mart ayõnda tespit edilmiştir. Mart ayõ 10 istasyonla en fazlatrend tespit edilen ay olmuştur ve trendlerin hepsi azalan yöndedir.     Managed honey bee colony losses in Canada, China, Europe, Israel and Turkey, for the winters of 2008-9 and 1009-10

    In 2008 the COLOSS network was formed by honey bee experts from Europe and the USA. The primary objectives set by this scientific network were to explain and to prevent large scale losses of honey bee (Apis mellifera) colonies. In June 2008 COLOSS obtained four years support from the European Union from COST and was designated as COST Action FA0803 – COLOSS (Prevention of honey bee COlony LOSSes). To enable the comparison of loss data between participating countries, a standardized COLOSS questionnaire was developed. Using this questionnaire information on honey bee losses has been collected over two years. Survey data presented in this study were gathered in 2009 from 12 countries and in 2010 from 24 countries. Mean honey bee losses in Europe varied widely, between 7-22% over the 2008-9 winter and between 7-30% over the 2009-10 winter. An important finding is that for all countries which participated in 2008-9, winter losses in 2009-10 were found to be substantially higher. In 2009-10, winter losses in South East Europe were at such a low level that the factors causing the losses in other parts of Europe were absent, or at a level which did not affect colony survival. The five provinces of China, which were included in 2009-10, showed very low mean (4%) A. mellifera winter losses. In six Canadian provinces, mean winter losses in 2010 varied between 16-25%, losses in Nova Scotia (40%) being exceptionally high. In most countries and in both monitoring years, hobbyist beekeepers (1-50 colonies) experienced higher losses than practitioners with intermediate beekeeping operations (51-500 colonies). This relationship between scale of beekeeping and extent of losses effect was also observed in 2009-10, but was less pronounced. In Belgium, Italy, the Netherlands and Poland, 2008-9 mean winter losses for beekeepers who reported ‘disappeared’ colonies were significantly higher compared to mean winter losses of beekeepers who did not report ‘disappeared’ colonies. Mean 2008-9 winter losses for those beekeepers in the Netherlands who reported symptoms similar to “Colony Collapse Disorder” (CCD), namely: 1. no dead bees in or surrounding the hive while; 2. capped brood was present, were significantly higher than mean winter losses for those beekeepers who reported ‘disappeared’ colonies without the presence of capped brood in the empty hives. In the winter of 2009-10 in the majority of participating countries, beekeepers who reported ‘disappeared’ colonies experienced higher winter losses compared with beekeepers, who experienced winter losses but did not report ‘disappeared’ colonies

    A cyclic nucleotide-gated channel (CNGC16) in pollen is critical for stress tolerance in pollen reproductive development

    Cyclic nucleotide-gated channels (CNGCs) have been implicated in diverse aspects of plant growth and development, including responses to biotic and abiotic stress, as well as pollen tube growth and fertility. Here, genetic evidence identifies CNGC16 in Arabidopsis (Arabidopsis thaliana) as critical for pollen fertility under conditions of heat stress and drought. Two independent transfer DNA disruptions of cngc16 resulted in a greater than 10-fold stress-dependent reduction in pollen fitness and seed set. This phenotype was fully rescued through pollen expression of a CNGC16 transgene, indicating that cngc16-1 and 16-2 were both loss-of-function null alleles. The most stress-sensitive period for cngc16 pollen was during germination and the initiation of pollen tube tip growth. Pollen viability assays indicate that mutant pollen are also hypersensitive to external calcium chloride, a phenomenon analogous to calcium chloride hypersensitivities observed in other cngc mutants. A heat stress was found to increase concentrations of 3′,5′-cyclic guanyl monophosphate in both pollen and leaves, as detected using an antibody-binding assay. A quantitative PCR analysis indicates that cngc16 mutant pollen have attenuated expression of several heat-stress response genes, including two heat shock transcription factor genes, HsfA2 and HsfB1. Together, these results provide evidence for a heat stress response pathway in pollen that connects a cyclic nucleotide signal, a Ca(2+)-permeable ion channel, and a signaling network that activates a downstream transcriptional heat shock response

    Potential association of LMNA-associated generalized lipodystrophy with juvenile dermatomyositis

    Abstract Background Juvenile dermatomyositis (JDM) is an auto-immune muscle disease which presents with skin manifestations and muscle weakness. At least 10% of the patients with JDM present with acquired lipodystrophy. Laminopathies are caused by mutations in the lamin genes and cover a wide spectrum of diseases including muscular dystrophies and lipodystrophy. The p.T10I LMNA variant is associated with a phenotype of generalized lipodystrophy that has also been called atypical progeroid syndrome. Case presentation A previously healthy female presented with bilateral proximal lower extremity muscle weakness at age 4. She was diagnosed with JDM based on her clinical presentation, laboratory tests and magnetic resonance imaging (MRI). She had subcutaneous fat loss which started in her extremities and progressed to her whole body. At age 7, she had diabetes, hypertriglyceridemia, low leptin levels and low body fat on dual energy X-ray absorptiometry (DEXA) scan, and was diagnosed with acquired generalized lipodystrophy (AGL). Whole exome sequencing (WES) revealed a heterozygous c.29C > T; p.T10I missense pathogenic variant in LMNA, which encodes lamins A and C. Muscle biopsy confirmed JDM rather than muscular dystrophy, showing perifascicular atrophy and perivascular mononuclear cell infiltration. Immunofluroscence of skin fibroblasts confirmed nuclear atypia and fragmentation. Conclusions This is a unique case with p.T10I LMNA variant displaying concurrent JDM and AGL. This co-occurrence raises the intriguing possibility that LMNA, and possibly p.T10I, may have a pathogenic role in not only the occurrence of generalized lipodystrophy, but also juvenile dermatomyositis.     Post-Transplantation Cyclophosphamide for Graft-versus- Host Disease Prophylaxis in Multiple Myeloma Patients Who Underwent Allogeneic Hematopoietic Cell Transplantation:First Comparison by Donor Type. A Study from the Chronic Malignancies Working Party of the European Society for Blood and Marrow Transplantation

    Graft-versus-host disease (GVHD) remains among the major causes of treatment failure in patients with multiple myeloma (MM) undergoing allogeneic hematopoietic cell transplantation (allo-HCT). The use of post-transplantation cyclophosphamide (PT-Cy) is now a well-established and widely used method for GVHD prophylaxis after HLA haploidentical HCT. However, the rationale for using PT-Cy in the setting of matched donor transplantation is less apparent, given the lesser degree of bidirectional alloreactivity. In this retrospective study, we investigated the role of PT-Cy as GVHD prophylaxis in patients with multiple myeloma underoing allo-HCT, among different donor types, to determine cumulative incidence of acute and chronic GVHD and impact on engraftment, progression-free survival (PFS), GVHD-free/relapse- free survival (GRFS), overall survival (OS), and NRM A total of 295 patients with MM underwent allo-HCT using grafts from a matched related donor (MRD; n = 67), matched unrelated donor (MUD; n = 72), mismatched related or unrelated donor (MMRD/MMUD, 1 antigen; n = 27), or haploidentical donor (haplo; n = 129) using PT-Cy between 2012 and 2018. In addition to PT-Cy, agents used in GVHD prophylaxis included calcineurin inhibitors in 239 patients (81%), with mycophenolate mofetil in 184 of those 239 (77%). For grade II-IV acute GVHD, the cumulative incidence at day +100 was 30% (95% confidence interval [CI], 25% to 36%), 9% (95% CI, 5% to 12%) for grade III-IV acute GVHD, and 27% (95% CI, 21% to 32%) for chronic GVHD (limited, 21%; extensive, 6%), with no differences by donor type. The median time to neutrophil engraftment was 19d (95% CI, 18-19), with no significant difference by donor type. The median time to platelet engraftment was delayed in haploidentical donor graft recipients (27 days versus 21 days; P <.001). Two-year OS, PFS, GRFS, and NRM were 51% (95% CI, 45% to 58%), 26% (95% CI, 20% to 32%), 24% (95% CI, 18% to 30%), and 19% (95% CI, 14% to 24%), respectively, with no significant difference between different donor types. In multivariable analyses, compared with the haplo donors, the use of MRDs was associated with significantly better OS (hazard ratio [HR], 0.6; 95% CI, 0.38 to 0.95; P =.029), and the use of MUDs was associated with a significantly higher GRFS (HR, 0.63; 95% CI, 0.42 to 0.97; P =.034). There was a trend toward improved PFS with use of MUDs (HR, 0.69; 95% CI, 0.46 to 1.04; P =.08). Our data show that PT-Cy in MM patients undergoing allo-HCT resulted in low rates of acute and chronic GVHD and led to favorable survival, especially in the matched related donor setting
