    Probing the strength of the Iberian lithosphere

    Como indicador de la resistencia de la litosfera a largo plazo, el espesor elástico efectivo (Te) se puede utilizar para entender la relación entre la reología de la litosfera y la evolución geodinámica de ambientes tectónicos complejos. En este trabajo presentamos, por primera vez, un mapa de alta resolución de la variación espacial de Te en la península ibérica, obtenido a partir del análisis espectral de la coherencia entre la topografía y anomalía de Bouguer, con el objetivo de mejorar nuestro conocimiento de la variación regional en la reología y el comportamiento mecánico a largo plazo de su litosfera. La mayor parte de la región se caracteriza por valores de espesor elástico efectivo bajos (Te < 20 km), destacando un domino de valores de Te moderados y altos en la zona centro y sudoeste peninsular, que se prolonga hacia el norte de África, limitado por fuertes gradientes de Te, especialmente en la zona sur.As a proxy for long-term lithospheric strength, the effective elastic thickness (Te) can be used to understand the relationship between lithospheric rheology and geodynamic evolution of complex tectonic settings. Here we investigate the lithospheric strength of the Iberian Peninsula in order to improve our knowledge of the regional variations of the rheology and long-term mechanical behavior of the lithosphere in the study area. For this we present, for the first time, a high-resolution map of spatial variations of Te in the Iberian Peninsula from the spectral analysis of the Bouguer coherence.     CALIFA, the Calar Alto Legacy Integral Field Area survey: IV. Third public data release

    This paper describes the third public data release (DR3) of the Calar Alto Legacy Integral Field Area (CALIFA) survey. Science-grade quality data for 667 galaxies are made public, including the 200 galaxies of the second public data release (DR2). Data were obtained with the integral-field spectrograph PMAS/PPak mounted on the 3.5 m telescope at the Calar Alto Observatory. Three different spectral setups are available: i) a low-resolution V500 setup covering the wavelength range 3745-7500 Å (4240-7140 Å unvignetted) with a spectral resolution of 6.0 Å (FWHM) for 646 galaxies, ii) a medium-resolution V1200 setup covering the wavelength range 3650-4840 Å (3650-4620 Å unvignetted) with a spectral resolution of 2.3 Å (FWHM) for 484 galaxies, and iii) the combination of the cubes from both setups (called COMBO) with a spectral resolution of 6.0 Å and a wavelength range between 3700-7500 Å (3700-7140 Å unvignetted) for 446 galaxies. The Main Sample, selected and observed according to the CALIFA survey strategy covers a redshift range between 0.005 and 0.03, spans the color-magnitude diagram and probes a wide range of stellar masses, ionization conditions, and morphological types. The Extension Sample covers several types of galaxies that are rare in the overall galaxy population and are therefore not numerous or absent in the CALIFA Main Sample. All the cubes in the data release were processed using the latest pipeline, which includes improved versions of the calibration frames and an even further improved image reconstruction quality.     Oncogenic Role of Secreted Engrailed Homeobox 2 (EN2) in Prostate Cancer

    Engrailed variant-2 (EN2) has been suggested as a potential diagnostic biomarker; however, its presence and functional role in prostate cancer (PCa) cells is still controversial or unknown. Here, we analyzed 1) the expression/secretion profile of EN2 in five independent samples cohorts from PCa patients and controls (prostate tissues and/or urine) to determine its utility as a PCa biomarker; and 2) the functional role of EN2 in normal (RWPE1) and tumor (LNCaP/22Rv1/PC3) prostate cells to explore its potential value as therapeutic target. EN2 was overexpressed in our two cohorts of PCa tissues compared to control and in tumor cell lines compared with normal-like prostate cells. This profile was corroborated in silico in three independent data sets [The Cancer Genome Atlas(TCGA)/Memorial Sloan Kettering Cancer Center (MSKCC)/Grasso]. Consistently, urine EN2 levels were elevated and enabled discrimination between PCa and control patients. EN2 treatment increased cell proliferation in LNCaP/22Rv1/PC3 cells, migration in RWPE1/PC3 cells, and PSA secretion in LNCaP cells. These effects were associated, at least in the androgen-sensitive LNCaP cells, with increased AKT and androgen-receptor phosphorylation levels and with modulation of key cancer-associated genes. Consistently, EN2 treatment also regulated androgen-receptor activity (full-length and splicing variants) in androgen-sensitive 22Rv1 cells. Altogether, this study demonstrates the potential utility of EN2 as a non-invasive diagnostic biomarker for PCa and provides novel and valuable information to further investigate its putative utility to develop new therapeutic tools in PCa

    Usual dietary intake, nutritional adequacy and food sources of calcium, phosphorus, magnesium and vitamin D of spanish children aged one to dagger

    Bone problems in the population begin to be establish in childhood. The present study aims to assess the usual calcium, phosphorus, magnesium, and vitamin D intakes, along with the food sources of these nutrients, in Spanish children participating in the EsNuPI (Estudio Nutricional en Población Infantil Española) study. Two 24 h dietary recalls were applied to 1448 children (1 to <10 years) divided into two sub-samples: one reference sample (RS) of the general population [n = 707] and another sample which exclusively included children consuming enriched or fortified milks, here called “adapted milks” (AMS) [n = 741]. Estimation of the usual intake shows that nutrient intake increased with age for all nutrients except vitamin D. Using as reference the Dietary Reference Values from the European Food Safety Authority (EFSA), calcium and magnesium intakes were found to be below the average requirement (AR) and adequate intake (AI), respectively, in a considerable percentage of children. Furthermore, phosphorus exceeded the AI in 100% of individuals and vitamin D was lower than the AI in almost all children studied. The results were very similar when considering only plausible reporters. When analyzing the food sources of the nutrients studied, milk and dairy products contributed the most to calcium, phosphorus, magnesium, and vitamin D. Other sources of calcium were cereals and vegetables; for phosphorus: meat, meat products, and cereals; for magnesium: cereals and fruits; and, for vitamin D: fish and eggs. These results highlight the desirability of improving the intake concerning these nutrients, which are involved in bone and metabolic health in children. The AMS group appeared to contribute better to the adequacy of those nutrients than the RS group, but both still need further improvement.     NGS-Based Molecular Karyotyping of Multiple Myeloma: Results from the GEM12 Clinical Trial

    Simple Summary Multiple Myeloma (MM) is considered an incurable chronic disease, which prognosis depends on the presence of different genomic alterations. To accomplish a complete molecular diagnosis in a single essay, we have designed and validated a capture-based NGS approach to reliably identify pathogenic mutations (SNVs and indels), genomic alterations (CNVs and chromosomic translocations), and IGH rearrangements. We have observed a good correlation of the results obtained using our capture panel with data obtained by both FISH and WES techniques. In this study, the molecular classification performed using our approach was significantly associated with the stratification and outcome of MM patients. Additionally, this panel has been proven to detect specific IGH rearrangements that could be used as biomarkers in patient follow-ups through minimal residual disease (MRD) assays. In conclusion, we think that MM patients could benefit from the use of this capture-based NGS approach with a more accurate, single-essay molecular diagnosis. Next-generation sequencing (NGS) has greatly improved our ability to detect the genomic aberrations occurring in multiple myeloma (MM); however, its transfer to routine clinical labs and its validation in clinical trials remains to be established. We designed a capture-based NGS targeted panel to identify, in a single assay, known genetic alterations for the prognostic stratification of MM. The NGS panel was designed for the simultaneous study of single nucleotide and copy number variations, insertions and deletions, chromosomal translocations and V(D)J rearrangements. The panel was validated using a cohort of 149 MM patients enrolled in the GEM2012MENOS65 clinical trial. The results showed great global accuracy, with positive and negative predictive values close to 90% when compared with available data from fluorescence in situ hybridization and whole-exome sequencing. While the treatments used in the clinical trial showed high efficacy, patients defined as high-risk by the panel had shorter progression-free survival (p = 0.0015). As expected, the mutational status of TP53 was significant in predicting patient outcomes (p = 0.021). The NGS panel also efficiently detected clonal IGH rearrangements in 81% of patients. In conclusion, molecular karyotyping using a targeted NGS panel can identify relevant prognostic chromosomal abnormalities and translocations for the clinical management of MM patients

    The dependence of oxygen and nitrogen abundances on stellar mass from the CALIFA survey

    Context. The study of the integrated properties of star-forming galaxies is central to understand their formation and evolution. Some of these properties are extensive and therefore their analysis require totally covering and spatially resolved observations. Among these properties, metallicity can be defined in spiral discs by means of integral field spectroscopy (IFS) of individual H ii regions. The simultaneous analysis of the abundances of primary elements, as oxygen, and secondary, as nitrogen, also provides clues about the star formation history and the processes that shape the build-up of spiral discs. Aims. Our main aim is to analyse simultaneously O/H and N/O abundance ratios in H ii regions in different radial positions of the discs in a large sample of spiral galaxies to obtain the slopes and the characteristic abundance ratios that can be related to their integrated properties. Methods. We analysed the optical spectra of individual selected H ii regions extracted from a sample of 350 spiral galaxies of the CALIFA survey. We calculated total O/H abundances and, for the first time, N/O ratios using the semi-empirical routine Hii-Chi-mistry, which, according to Pérez-Montero (2014, MNRAS, 441, 2663), is consistent with the direct method and reduces the uncertainty in the O/H derivation using [N ii] lines owing to the dispersion in the O/H-N/O relation. Then we performed linear fittings to the abundances as a function of the de-projected galactocentric distances. Results. The analysis of the radial distribution both for O/H and N/O in the non-interacting galaxies reveals that both average slopes are negative, but a non-negligible fraction of objects have a flat or even a positive gradient (at least 10% for O/H and 4% for N/O). The slopes normalised to the effective radius appear to have a slight dependence on the total stellar mass and the morphological type, as late low-mass objects tend to have flatter slopes. No clear relation is found, however, to explain the presence of inverted gradients in this sample, and there is no dependence between the average slopes and the presence of a bar. The relation between the resulting O/H and N/O linear fittings at the effective radius is much tighter (correlation coefficient ρ = 0.80) than between O/H and N/O slopes (ρ = 0.39) or for O/H and N/O in the individual H ii regions (ρ = 0.37). These O/H and N/O values at the effective radius also correlate very tightly (less than 0.03 dex of dispersion) with total luminosity and stellar mass.     Genetic landscape of 6089 inherited retinal dystrophies affected cases in Spain and their therapeutic and extended epidemiological implications

    ESRETNET Study Group, The ERDC Study Group, The Associated Clinical Study Group.Inherited retinal diseases (IRDs), defined by dysfunction or progressive loss of photoreceptors, are disorders characterized by elevated heterogeneity, both at the clinical and genetic levels. Our main goal was to address the genetic landscape of IRD in the largest cohort of Spanish patients reported to date. A retrospective hospital-based cross-sectional study was carried out on 6089 IRD affected individuals (from 4403 unrelated families), referred for genetic testing from all the Spanish autonomous communities. Clinical, demographic and familiar data were collected from each patient, including family pedigree, age of appearance of visual symptoms, presence of any systemic findings and geographical origin. Genetic studies were performed to the 3951 families with available DNA using different molecular techniques. Overall, 53.2% (2100/3951) of the studied families were genetically characterized, and 1549 different likely causative variants in 142 genes were identified. The most common phenotype encountered is retinitis pigmentosa (RP) (55.6% of families, 2447/4403). The most recurrently mutated genes were PRPH2, ABCA4 and RS1 in autosomal dominant (AD), autosomal recessive (AR) and X-linked (XL) NON-RP cases, respectively; RHO, USH2A and RPGR in AD, AR and XL for non-syndromic RP; and USH2A and MYO7A in syndromic IRD. Pathogenic variants c.3386G > T (p.Arg1129Leu) in ABCA4 and c.2276G > T (p.Cys759Phe) in USH2A were the most frequent variants identified. Our study provides the general landscape for IRD in Spain, reporting the largest cohort ever presented.
