    Propylthiouracil-induced cutaneous vasculitis

    The use of propylthiouracil (PTU) is associated with the development of different auto-antibodies, amongst them are antineutrophil cytoplasmic antibodies (ANCA) that are involved in the pathogenesis of ANCA associated systemic vasculitis. The case of a 46-years old woman who presented cutaneous vasculitis when taking PTU for Graves' disease is reported. Perinuclear ANCA (p-ANCA) was positive with titer > 1/320, but anti-myeloperoxidase antibodies were not detected. Skin biopsy showed leucocytoclastic vasculitis. The patient improved within ten days after withdrawing PTU and the resolution of hyperthyroidism was achieved with radioiodine (131I). The p-ANCA test remained positive > 1/320 eight months and four years after PTU withdrawal.O uso do propiltiouracil (PTU) está associado ao desenvolvimento de diferentes auto-anticorpos, entre eles anticorpos anticitoplasma de neutrófilos (ANCA), que estão envolvidos na patogênese das vasculites sistêmicas ANCA-associadas (VSAA). Será relatado o caso de um paciente do sexo feminino, de 46 anos, que apresentou vasculite cutânea durante o uso de PTU como terapêutica para doença de Graves. O ANCA com padrão perinuclear (p-ANCA) foi positivo em títulos > 1/320, porém anticorpos antimielo-peroxidase (MPO) não foram detectados. A biópsia de pele revelou uma vasculite leucocitoclástica. Houve melhora clínica em dez dias após a retirada do PTU e optou-se pelo iodo radioativo (I131) para o tratamento do hipertireoidismo.     Cross-layer QoS-Aware Resource Allocation for IoT-Enabled Service Choreographies

    International audienceResource allocation for distributed systems is a well-known approach to deal with quality of service requirements. However, existing approaches do not consider the effects of resource allocation at the different levels of a system, especially when considering the end-to-end behavior of distributed compositions such as service choreographies. Based on our previous research on the optimized and QoS-aware resource allocation for service choreographies and publish-subscribe brokers, we outline a novel approach that integrates the treatment of resource allocation at both levels in orderto yield a more precise control over end-to-end QoS. The paper presents an early architecture and discusses some of the main challenges towards realizing the approach

    Design procedures for sustainable structural concretes using wastes and industrial by-products

    The protection of the environment must be a priority in our society, and the construction sector can contribute significantly to this goal. Construction, being one of the industrial sectors that is more demanding in terms of raw materials, must reinforce its effort to implement, in a more profound and systematic way, the paradigm of the circular economy. In this sense, in recent years several studies have been trying to contribute solutions aimed at reintroducing industrial by-products or residues in new products for the construction industry. It should be noted that nowadays it is increasingly important to introduce a higher percentage of recycled materials in concrete. In this context, the present work addresses the appropriateness of a design procedure proposed to maximize the content of electric arc furnace slag (EAFS) and include recycled tire steel fibers (RTSF) in the production of more sustainable structural concretes. For this, the properties of various concrete mixtures at the fresh and hardened state, obtained by the substantial substitution of coarse and fine natural aggregates by EAFS and fly ash (FA), were investigated. The design of EAFS mixtures was based on two conventional reference mixtures (REF1 and REF2), and by using the modified Andreasen and Andersen particle packing model, these were optimized to achieve maximum packing density. Compressive strength, modulus of elasticity behavior, and fresh and physical properties were assessed in order to define the best mix proportions with respect to the predefined requirements of ordinary mixtures. Untreated recycled tire steel fibers (RTSF) were included in the developed sustainable concrete to perform a comparison of the physical properties with unreinforced concretes developed with natural aggregates (REF2) and with EAFS aggregates (EAFS8D1). This incorporation was intended to improve the physical behavior of unreinforced concretes with EAFS aggregates. Mixtures with high percentages of waste aggregates up to 70% (in weight), and 10% (in weight) of FA were obtained, showing competitive mechanical behavior compared to REF1 and REF2. These concrete compositions showed minimum and maximum compressive strengths between 9 MPa and 37 MPa, respectively.     AbstractBackgroundIn BIOACTIVE study, we evaluated vascular responses after the implant of biolimus A9-eluting stent (BES; BioMatrixTM) and the everolimus-eluting stent (EES; XIENCE VTM). In this study, we present the optical coherence tomography analysis (OCT) 6 months post-intervention.MethodsPatients were randomized to treatment with BES (n = 22) or EES (n = 18). The primary outcome was the frequency of non-covered, poorly positioned struts by OCT.ResultsOCT was performed in 26 patients (BES: n = 15; EES: n = 11) and 749 tomographic images and 7,725 stent struts were analyzed. BES and EES showed similar luminal and stent areas. Neointimal hyperplasia area, neointimal thickness and the percentage of in-stent obstruction (8.44 ± 5.10% vs. 9.21 ± 6.36%; p = 0.74) were similar. The rates of not covered struts (BES: 2.10 ± 3.60% vs. ESS: 2.46 ± 2.15%, p = 0.77) and poorly positioned struts (BES: 0.48 ± 1.48% vs. EES 0.44 ± 1.05%, p = 0.94) were similarly low. The frequency of frames with signs consistent with peri-strut inflammatory infiltrate was low and similar between BES (15.53 ± 20.77%) and EES (11.70 ± 27.51%; p = 0.68).ConclusionsThe second-generation drug-eluting stents BES and EES were equally effective at suppressing the neointimal formation after 6 months, with favorable vascular responses. The frequency of frames with peri-strut infiltrate signals per patient was low, and lower than that observed historically with first-generation drug-eluting stents

    Hemoglobinopatias no Distrito Federal, Brasil

    Em uma amostra de 3137 pessoas, residentes no Distrito Federal, foram detectadas as seguintes hemoglobinas: em 3009(95,92%) HbAA; em 91 (2,90%) HbAS; em 20(0,64%) HbAC; em 8 (0,26%) talassemia beta minor; em 5(0,16%) HbAJ alfa; em 3 (0,09%) HbAM e em 1 (0,03%) talassemia major associada com HbAS. A HbAS têm as seguintes freqüências, quanto ao grupo racial: branco - 1,84%; mulato claro 2,55%; mulato médio-3,68%; mulato escuro - 6,80%; negro -10,43% e mestiço de índio -3,85%.The hemoglobins of 3137 persons from the Federal District, Brazil was studied. The identified types of hemoglobins were: HbAA (3009 - 95,92%); HbAS (91 - 2,90%); HbAC (20 - 0,64%) beta thalassemia minor (8 - 0,26%); HbAJ alpha (5 - 0,16%); HbAM (3 - 0,09%) and beta thalassemia major/HbS (1 - 0,03%). The frequencies of sickle cell trait (HbAS) of each racial class were: white (1,84%); light mulatto (2,55%); medium mulatto (3,68%); dark mulatto(6,80%), negro (10,43%)and indian mestizo (3,85%)

    Multichannel arbitrary waveform beamformer transmitter for experimental research activities of ultrasound

    This paper presents the development of a digital beamformer transmitter system for generation of arbitrary ultrasound waveforms, specifically designed for research purposes. The proposed architecture has 8 independent excitation channels and uses an FPGA device (Field Programmable Gated Array) for electronic control channel generation of ultrasonic acoustic beam transmission. The system allows operation in pulse-echo mode, with pulse repetition rate of excitation between 62.5 Hz and 8 kHz, or single pulse, center frequency between 500 kHz and 20 MHz, excitation voltage between -100 and +100 V, and individual control of amplitude apodization, phase angle and time delay trigger. The mathematical method for determining the digital signals of excitation is presented and validated through practical results for waveform generation with Gaussian profile and center frequency of 20 MHz, in a load formed by a 220 pF capacitor in parallel with a resistor of 1 k Ω. The results show that the proposed flexible and fully programmable architecture, can support the development of new algorithms and sophisticated processing techniques of the transmission beamformer.Este trabalho apresenta o desenvolvimento de um sistema digital beamformer de transmissão para geração de formas de onda arbitrárias de ultrassom, projetado especificamente para fins de pesquisa. A arquitetura proposta possui 8 canais de excitação independentes e utiliza um dispositivo FPGA (Field Programmable Gated Array) para controle eletrônico dos canais de geração do feixe acústico ultrassônico de transmissão. O sistema permite operação no modo pulso-eco, com taxa de repetição dos pulsos de excitação entre 62,5 Hz e 8 kHz, ou pulso único, frequência central entre 500 kHz e 20 MHz, tensão de excitação entre -100 V e +100 V, e controle individual da amplitude de apodização, ângulo de fase e atraso temporal de disparo. O método matemático para determinação dos sinais digitais de excitação é apresentado e validado através dos resultados práticos para geração de forma de onda com perfil Gaussiano e frequência central de 20 MHz, em uma carga equivalente formada por um capacitor de 220 pF em paralelo com um resistor de 1 k Ω.     Influence of passage number on the impact of the secretome of adipose tissue stem cells on neural survival, neurodifferentiation and axonal growth

    Mesenchymal stem cells (MSCs), and within them adipose tissue derived stem cells (ASCs), have been shown to have therapeutic effects on central nervous system (CNS) cell populations. Such effects have been mostly attributed to soluble factors, as well as vesicles, present in their secretome. Yet, little is known about the impact that MSC passaging might have in the secretion therapeutic profile. Our aim was to show how human ASCs (hASCs) passage number influences the effect of their secretome in neuronal survival, differentiation and axonal growth. For this purpose, post-natal rat hippocampal primary cultures, human neural progenitor cell (hNPCs) cultures and dorsal root ganglia (DRGs) explants were incubated with secretome, collected as conditioned media (CM), obtained from hASCs in P3, P6, P9 and P12. Results showed no differences when comparing percentages of MAP-2 positive cells (a mature neuronal marker) in neuronal cultures or hNPCs, after incubation with hASCs secretome from different passages. The same was observed regarding DRG neurite outgrowth. In order to characterize the secretomes obtained from different passages, a proteomic analysis was performed, revealing that its composition did not vary significantly with passage number P3 to P12. Results allowed us to identify several key proteins, such as pigment epithelium derived factor (PEDF), DJ-1, interleucin-6 (IL-6) and galectin, all of which have already proven to play neuroprotective and neurodifferentiating roles. Proteins that promote neurite outgrowth were also found present, such as semaphorin 7A and glypican-1. For reasons that remain largely unknown, HPV+OPSCC is significantly more common in men than in women. This study aims to determine the incidence of OPSCC in male and female HPV16-transgenic mice and to explore the role of female sex hormone receptors in the sexual predisposition for HPV+ OPSCC. The tongues of 30-weeks-old HPV16-transgenic male (n = 80) and female (n = 90) and matched wild-type male (n = 10) and female (n = 10) FVB/n mice were screened histologically for intraepithelial and invasive lesions in 2017 at the Centre for the Research and Technology of Agro-Environmental and Biological Sciences (CITAB), Por-tugal. Expression of estrogen receptors alpha (ERα) and beta (ERβ), progesterone receptors (PR) and matrix metalloproteinase 2 (MMP2) was studied immunohistochemically. Collagen remodeling was studied using picrosirius red. Female mice showed robust ERα and ERβ expression in intraepithelial and invasive lesions, which was accompanied by strong MMP2 expression and marked collagen remodeling. Male mice showed minimal ERα, ERβ and MMP2 expression and unaltered collagen patterns. These results confirm the association of HPV16 with tongue base cancer in both sexes. The higher cancer incidence in female versus male mice contrasts with data from OPSCC patients and is associated with enhanced ER expression via MMP2 upregulation.publishersversionpublishe
