    SKY analysis revealed recurrent numerical and structural chromosome changes in BDII rat endometrial carcinomas

    <p>Abstract</p> <p>Background</p> <p>Genomic alterations are common features of cancer cells, and some of these changes are proven to be neoplastic-specific. Such alterations may serve as valuable tools for diagnosis and classification of tumors, prediction of clinical outcome, disease monitoring, and choice of therapy as well as for providing clues to the location of crucial cancer-related genes.</p> <p>Endometrial carcinoma (EC) is the most frequently diagnosed malignancy of the female genital tract, ranking fourth among all invasive tumors affecting women. Cytogenetic studies of human ECs have not produced very conclusive data, since many of these studies are based on karyotyping of limited number of cases and no really specific karyotypic changes have yet been identified. As the majority of the genes are conserved among mammals, the use of inbred animal model systems may serve as a tool for identification of underlying genes and pathways involved in tumorigenesis in humans. In the present work we used spectral karyotyping (SKY) to identify cancer-related aberrations in a well-characterized experimental model for spontaneous endometrial carcinoma in the BDII rat tumor model.</p> <p>Results</p> <p>Analysis of 21 experimental ECs revealed specific nonrandom numerical and structural chromosomal changes. The most recurrent numerical alterations were gains in rat chromosome 4 (RNO4) and losses in RNO15. The most commonly structural changes were mainly in form of chromosomal translocations and were detected in RNO3, RNO6, RNO10, RNO11, RNO12, and RNO20. Unbalanced chromosomal translocations involving RNO3p was the most commonly observed structural changes in this material followed by RNO11p and RNO10 translocations.</p> <p>Conclusion</p> <p>The non-random nature of these events, as documented by their high frequencies of incidence, is suggesting for dynamic selection of these changes during experimental EC tumorigenesis and therefore for their potential contribution into development of this malignancy. Comparative molecular analysis of the identified genetic changes in this tumor model with those reported in the human ECs may provide new insights into underlying genetic changes involved in EC development and tumorigenesis.</p

    Pharmacological strategies to reduce pruritus during postoperative epidural analgesia after lumbar fusion surgery - a prospective randomized trial in 150 patients

    BACKGROUND: Epidural analgesia with bupivacain, epinephrine and fentanyl provides excellent pain control after lumbar fusion surgery, but pruritus and motor block are frequent side effects. Theoretically epidural ropivacain combined with oral oxycodone could decrease the incidence of these side effects. The two regimens were compared in a prospective randomized trial. PATIENTS AND METHODS: 150 patients (87 women) treated with posterior instrumented lumbar fusion were included. The mean age was 51 +/- 11 years. 76 were randomized to bupivacain, epinephrine and fentanyl (group B) and 74 to ropivacain and oxycodone (group R). Pruritus, motor block and pain were measured 6 hours after surgery, thereafter 6 times per day for 5 days. Any pain breakthrough episode was registered whenever it occurred. RESULTS: The epidural treatment could be performed in 143 patients (72 in group B and 71 in group R). Disturbing pruritus occurred in 53 patients in group B compared to 12 in group R (p &lt; 0.0001). Motor blockade was most frequent on day 1, occurring in 45% of the patients with no difference between the groups. Both regimes gave good pain control with average VAS under 40, but the pain relief was statistically better in group B. The number of pain breakthrough episodes did not differ between the groups. CONCLUSIONS: Pruritus could be reduced with a combination of epidural ropivacain and oral oxycodone, at the price of a slightly higher pain level. Ropivacaine was not found to be superior to bupivacaine with regard to motor blocks

    Clustering of energy balance-related behaviours, sleep, and overweight among Finnish adolescents

    To examine how clusters of energy balance-related behaviours (EBRBs), including sleep related factors, were associated with overweight among adolescents. In Finland, 4262 adolescents, aged 13-15, participated in the cross-national Health Behaviour in School-aged Children study. The adolescents completed questionnaires assessing EBRBs [sleep duration, discrepancy and quality, physical activity (PA), screen time, junk food, fruit, and vegetable intake] and height and weight. Clusters were identified with kappa-means cluster analysis and their associations with overweight with logistic regression analyses. Common clusters for boys and girls were labelled "Healthy lifestyle" and "High screen time, unhealthy lifestyle". In addition, the cluster "Low/moderate screen time, unhealthy lifestyle" was identified among boys, and the cluster "Poor sleep, unhealthy lifestyle" among girls. Only girls in the cluster "High screen time, unhealthy lifestyle" were at increased risk for overweight. Girls, whose EBRB was characterized by high screen time and low PA, but not with poor sleep, were at increased risk for overweight. Future studies should examine ways to promote PA among adolescent girls with high interest in screen-based activities.Peer reviewe

    Voidaanko kouluinterventiolla lisätä lasten kasvisten ja hedelmien kulutusta – intervention toteutuksen merkitys

    Lasten vähäistä kasvisten ja hedelmien kulutusta on pyritty lisäämään erilaisilla kouluinterventioilla, mutta Suomessa interventioita on tehty vain muutama. Tarkoituksena oli tutkia, onnistutaanko kouluinterventiolla lisäämään 11-vuotiaiden lasten kasvisten ja hedelmien kulutusta. Tutkimus on osa eurooppalaista Pro Greens -projektia ja Suomessa tutkimus tehtiin ruotsinkielisissä kouluissa Folkhälsanin järjestämänä. Lukuvuoden kestäneen kouluintervention jälkeen interventiokoulujen oppilaat olivat lisänneet hedelmien, mutta ei juurikaan kasvisten kulutustaan kontrollikouluihin verrattuna. Intervention vaikutukset vaihtelivat intervention toteutusasteen mukaan. Suuren toteutusasteen luokkien lapset lisäsivät hedelmien kulutustaan, kun taas vähäisen toteutusasteen luokkien lapset eivät eronneet kontrolliryhmästä. Intervention toteutusaste vaihteli paljon koululuokittain ja sillä oli suuri merkitys intervention onnistumisessa

    Parental Education and Pre-School Children's Objectively Measured Sedentary Time : The Role of Co-Participation in Physical Activity

    Parental co-participation in physical activity (PA) may be a beneficial parenting practice for diminishing children's sedentary time (ST). Less information is available, however, on the explanatory role of co-participation in PA regarding parental educational differences in children's ST. Preschool-aged children (N = 864, mean age 4.8, 52% boys) with their parents participated in a cross-sectional DAGIS (Increased Health and Wellbeing in Pre-schools) study between years 2015 and 2016. Children (N = 821) wore an accelerometer for one week. Parents were informed of their educational background, and the frequency of visits with their child in nature, to parks or playgrounds, their own yard, and indoor sport facilities (N = 808). Testing the associations required multiple regression analyses. Parents with a low educational background reported more frequent visits with their child to their own yard, and these visits were associated with children's lower ST. More highly educated parents co-visited indoor sport facilities more frequently, although this did not have a significant association with children's ST. More frequent visits in nature were associated with a lower ST at weekdays, regardless of educational background. Future health promotion strategies should inform parents that frequent co-participation in PA, for example, in one's own yard, is beneficial for lowering children's ST.Peer reviewe

    Are associations between home environment and preschool children’s sedentary time influenced by parental educational level in a cross-sectional survey?

    Background: Childhood obesity is a major public health concern, especially in low socioeconomic groups. Sedentary time (SED) is an important predictor of obesity. To be able to diminish SED it is important to find modifiable predictors of sedentary behavior. The home environment associated with children's SED may vary by parental socioeconomic status. This study aims to clarify the association between parental educational level (PEL) and the home environment of 3-6-year-old children, and to examine how home environment associates with children's SED, and whether PEL modifies these associations. Methods: A cross-sectional Increased health and wellbeing in preschools (DAGIS) study was conducted in 2015-2016 in Finland. The parents (n = 809) filled in questionnaires assessing PEL, and the home physical and social environment related to children's SED. Children's SED was measured with accelerometers, which the children (n = 745) wore for 1 week. Results: High PEL was associated with a home environment restraining sedentary behaviour compared with low PEL. Stricter descriptive norms about screen time, considering it important to limit the child's screen time, and satisfaction about the child's screen time associated with children's lower SED. The association with parental psychological control and SED was influenced by PEL. In the PEL stratified analyses, however, the associations between psychological control and SED did not reach statistical significance. Conclusions: Future interventions aiming to decrease SED should pay attention to relevant factors in children's sedentary behaviour home environment. It is important to acknowledge the possible PEL differences in these factors.Peer reviewe
