Tutkielmassa laadullisen tutkimusaineiston muodostaa 12 Herttoniemen asuntolan miehen puolistrukturoitua haastattelua. Sekundaariaineisto sisĂ€ltÀÀ asiakastietojĂ€rjestelmÀÀn (ATJ) kirjattuja rekisteritietoja ja asuntolassa tehtyjĂ€ havaintoja. Tutkimusaineisto on osa laajempaa pÀÀkaupunkiseudun Sosiaalialan osaamiskeskus SOCCAn Asunnottomien palvelujen kehittĂ€misyksikkö -hankkeessa vuosina 2007-2008 kerĂ€ttyĂ€ selvitysaineistoa. Aineisto on kerĂ€tty yhdessĂ€ lÀÀketieteen lisensiaatti Agnes Stenius-Ayoaden kanssa. Aineistosta julkaistiin lokakuussa 2009 selvitys 'Asunnottomuus vastaanottoyksiköissĂ€. Asunnottomien vastaanottoyksiköiden asiakkaiden sosiaalinen tilanne ja terveydentila pÀÀkaupunkiseudulla'. Tutkielmassa pyritÀÀn syventĂ€mÀÀn ja tĂ€ydentĂ€mÀÀn pÀÀkaupunkiseudun vastaanottoyksiköiden asunnottomien sosiaalista tilannetta ja terveydentilaa koskevan selvityksen tuloksia. Herttoniemen miesten asuntolassa majailevien miesten sosiaalisen tilanteen rakentumista tarkastellaan tutkielmassa sekĂ€ yksilöiden psykososiaaliseen tilanteeseen ja tarpeisiin kiinnittyvĂ€stĂ€ ettĂ€ rakenteellisesta nĂ€kökulmasta. Aineistoa lĂ€hestytÀÀn sen luennassa faktanĂ€kökulmasta. TĂ€llöin aineistoa luetaan Pertti Alasuutarin mÀÀritelmin aineiston ulkopuolista todellisuutta heijastavana tai siitĂ€ jotakin vĂ€ittĂ€vĂ€nĂ€. Tutkielmassa Herttoniemen asuntolan miesten sosiaalinen tilanne rakentuu kuvaukseksi miesten asuntolassa kokemasta turvattomuudesta ja turvattomien asuntola olosuhteiden pĂ€ihteiden kĂ€yttöÀ yllĂ€pitĂ€vĂ€stĂ€ kulttuurista. Asuntolan olosuhteet ja pĂ€ihteiden kĂ€yttö olivat myös suurimmat miesten arkeen vaikuttavat tekijĂ€t lĂ€hes kaikkien elĂ€mĂ€n alueiden, kuten hygienian hoidon, ruokailun, sosiaalisten suhteiden ja asioiden hoidon suhteen. Asuntolassa asumiseen ja pĂ€ihteiden kĂ€yttöön liittyi monella vaikeudet huolehtia omasta taloudesta, varkauksien ja vĂ€kivallan kohteeksi joutuminen, yksinĂ€isyys ja luottamuksellisten ihmissuhteiden puute. Turvattoman asuntolan olosuhteissa miesten ainoaksi vaihtoehdoksi nĂ€ytti muodostuvan pullo ja puhdas selviytymisen pakko. Toisaalta asuntola nĂ€yttĂ€ytyi miehille tilana, jossa edes jonkinlainen hyvĂ€ksytyksi tuleminen oli mahdollista suhteessa asuntolan ulkopuoliseen maailmaan. Siten irtautuminen asuntolan kiinnipitĂ€vĂ€stĂ€ sosiaalisesta ympĂ€ristöstĂ€ ja suhteista oli heille vaikeaa.     Children of the Lord’s Resistant Army; An exploration of social relations from the perspective of Ugandan former child combatants

    Ever since 1987 the Lord’s Resistant Army (LRA), a theocratic rebel force, has been abducting children in Uganda to partake in the conflict against the Ugandan government and its forces. The children’s lives are drastically changed in a matter of minutes; they are suddenly forced to whiteness and take part in ruthless brutalities. Some of them live for months, others for years, under dictatorial rule and order. This research is set to explore the multiple challenges regarding social relations faced by child combatants both during and after life with the LRA. Fundamental notions such as, self-perception, perception of other, role taking and group formation are questioned. The research is based in Husserlian phenomenological theory, providing understanding of the children’s/adolescence’s conscious reflections on their daily experiences. Five different documents containing citations of former child combatants in Uganda are used for analysis in both Sphinx Lexica and Minerva, which is based in phenomenological theory of Meaning Constitution Analysis (MCA). The results show that the children/adolescents feel confused and uncertain about their roles and relations due to the drastic changes in their social networks. The argument follows that the impacts need to be considered as a significant part of their reintegration process

    This paper presents Espoo as a sustainable learning city, recognised by UNESCO as one of twelve pioneering Learning Cities in 2015. The vision of the city states that Espoo is a good place to live, learn, work and do business, and where residents can have their say in matters. Dynamic co-creation processes with residents, partners, universities, and enterprises have helped create and implement the Espoo Story and to find sustainable solutions for the city and its residents. The city of Espoo is preparing its citizens for the future by empowering all kinds of learners to be at the centre of their own learning processes. Espoo provides continuous learning paths leading from early childhood education, to capacity building in the workplace, and to learning in later life. Flexible learning spaces and digitalisation are expanding opportunities to participate and have fun when learning.Članek predstavlja mesto Espoo kot trajnostno učeče se mesto, ki ga je leta 2015 Unesco prepoznal kot enega od 12 pionirskih učečih se mest. Vizija mesta je Espoo kot odlično mesto za ĆŸivljenje, učenje, delo in poslovanje ter kot mesto, kjer imajo prebivalci svoj glas pri odločanju. Dinamični postopki soustvarjanja, v katere se vključujejo prebivalci, partnerji, univerze in podjetja, so pomagali ustvariti in uresničiti zgodbo mesta Espoo ter poiskati trajnostne reĆĄitve za kraj in njegove prebivalce. Espoo pripravlja svoje prebivalce na prihodnost s tem, da opolnomoča vse učeče se, da so sami v srediơču svojih lastnih učnih procesov. Espoo jim ponuja učno pot, ki se razteza od izobraĆŸevanja v zgodnjem otroĆĄtvu do krepitve zmogljivosti na delovnem mestu in končno tudi do učenja v poznejĆĄih ĆŸivljenjskih obdobjih. Poleg tega fleksibilni učni prostor in digitalizacija dajeta vedno večje moĆŸnosti za sodelovanje in zabavno učenje

    Perceptuell inlÀrning av bedömning av bakbenshÀlta hos hÀst

    Syftet med studien var att undersöka om det gĂ„r att förbĂ€ttra studenters förmĂ„ga att bedöma bakbenshĂ€ltor hos hĂ€star pĂ„ video, med hjĂ€lp av tvĂ„ olika datorbaserade inlĂ€rningsverktyg som ocksĂ„ jĂ€mfördes med avseende pĂ„ deras effekt pĂ„ perceptuell inlĂ€rning. En specialgjord trĂ€ningsmodul för perceptuell inlĂ€rning av bedömning av bakbenshĂ€ltor skapades pĂ„ student-webbportalen Canvas. Modulen byggdes upp av videomaterial pĂ„ hĂ€star som travar pĂ„ rakt spĂ„r. Instruerande auditiv feedback och slowmotion-repriser pĂ„ videorna visades efter varje trĂ€ningsomgĂ„ng. För att skapa videorna till inlĂ€rningsverktyget, mĂ€ttes 50 hĂ€star med objektiv rörelseanalys och filmades efterĂ„t. Av dessa valdes 27 hĂ€star ut, baserat pĂ„ förekomst av unilateral bakbenshĂ€lta eller om de var ohalta. Det andra inlĂ€rningsverktyget (LamenessTrainerÂź), var tillgĂ€ngligt online och baseras pĂ„ animerade hĂ€star som springer pĂ„ rakt spĂ„r, utan nĂ„gra föremĂ„l i bakgrunden och utan variation i rörelsemönster förutom grader av asymmetri (hĂ€lta). Verktyget gav direktfeedback under trĂ€ningen om vilket ben som var halt samt grad av asymmetri. De tvĂ„ inlĂ€rningsverktygen testades pĂ„ studenter frĂ„n program pĂ„ Sveriges Lantbruksuniversitet med nĂ„gon form av koppling till djurvetenskap. Studenterna kunde frivilligt anmĂ€la sig till studien genom att svara pĂ„ en massutskickad e-postinbjudan. Studenterna delades slumpmĂ€ssigt in till att anvĂ€nda ett av de tvĂ„ inlĂ€rningsverktygen. Tre digitala möten hölls under en tidsperiod pĂ„ Ă„tta dagar. Innan trĂ€ningen började vid det första mötet, fick studenterna fylla i en enkĂ€t med frĂ„gor om vilket program de studerar, deras hĂ€sterfarenhet utanför skolan och hur de sjĂ€lvskattade sin förmĂ„ga att bedöma bakbenshĂ€ltor hos hĂ€st. Alla studenter fick sedan utföra samma diagnostiska prov, baserat pĂ„ filmade hĂ€star, för att mĂ€ta en basnivĂ„ pĂ„ deras kunskap. Efter detta fick de trĂ€na i nĂ„gon av de tvĂ„ trĂ€ningsmoduler. Det andra trĂ€ningstillfĂ€llet hölls fyra dagar senare. Vid det sista mötet den Ă„ttonde dagen, gjorde studenterna om samma diagnostiska test för att se om hade förbĂ€ttrat sina resultat. Ingen signifikant skillnad kunde ses mellan provresultaten över de bĂ„da provomgĂ„ngarna, vare sig för gruppen som anvĂ€nde sig av Canvas eller den som anvĂ€nde LamenessTrainerÂź. Signifikant skillnad kunde ses hos studenter som hade sjĂ€lvskattat sin förmĂ„ga att bedöma bakbenshĂ€ltor som liten (p=0, 0263) och hos studenter som hade viss erfarenhet av hĂ€star (p=0,025). Inom dessa mindre grupper var det möjligt att se tendens till signifikant skillnad (p=0,0514 och p=0,052) för de studenter som hade anvĂ€nt sig av trĂ€ningsmodulen i Canvas. Slutsatsen var att studenter finner det svĂ„rt att bedöma bakbenshĂ€ltor och att förbĂ€ttringen efter tvĂ„ trĂ€ningstillfĂ€llen med tvĂ„ olika digitala trĂ€ningsverktyg inte var signifikanta. Endast ca 50 % av hĂ€starna bedömdes korrekt efter trĂ€ningstillfĂ€llena. Detta understryker behovet av, speciellt för veterinĂ€rstudenter, mer trĂ€ning i att bedöma bakbenshĂ€ltor. Den videobaserade trĂ€ningsmodulen i Canvas hade en tendens att vara av tydligare positiv anvĂ€ndning för studenter med generellt mindre hĂ€sterfarenhet och för studenter som tycker sig ha mindre förmĂ„ga att bedöma bakbenhĂ€ltor.The aim of this study was to investigate if it is possible to improve students’ ability to evaluate hindlimb lameness in horses from video using two different computer aided learning tools and to compare the learning results from the two methods. One of the two evaluated tools was a custom-made training module for perceptual learning of hindlimb lameness, created in the student web portal Canvas. This tool was based on video material of real horses trotting in a straight line, with instructive auditory feedback and slow-motion replay of the videos presented after each evaluation. To create the videos, 50 horses were measured with objective gait analysis and filmed. From these, 27 were selected based the presence of either unilateral hindlimb lameness or no lameness. The other learning tool (LamenessTrainerÂź), was available online and was based on computer-animated horses running straight, with no objects in the background and no variation in the motion pattern except for the degree of asymmetry (lameness). Direct feedback of the degree of asymmetry and affected limb was provided during the training as the answer was submitted, either as correct or incorrect. Eligible participants were students from programs at the Swedish University of Agricultural Sciences with any sort of connection animal science, and that responded to a mailing-list invitation. The students were randomly assigned to use either one of the two learning tools. Three digital meetings were held with four days in between. All students perform the same diagnostic test in the first and third meetings (answers were not revealed initially), and they did training-sessions using the tool they were assigned on the first and second meeting occasion. Before the training began in the first meeting, the students filled out a survey including basic background data, and self-assessment regarding experience with horses, and hindlimb lameness assessment skills. No significant difference was found between the test results before and after training, either for the group using Canvas or the one using LamenessTrainerÂź. Significant difference was, however, found among the subset of students who had self-evaluated their ability of hindlimb lameness assessment as small (p=0,0263), and among students who had some experience of horses (p=0,025), ignoring tool. Splitting the latter two groups by training tool, a tendency towards significant difference of improvement (p=0,0514 and p=0,052) was seen among students who had been using Canvas. The conclusion was that students find it hard to evaluate hindlimb lameness and in general show no significant improvement after two trainings sessions with the two computer aided learning tools. Only approximately 50% of the horses shown to the students was diagnosed correctly, even after training. This supports the need for especially veterinary students, to receive more training in evaluating hindlimb lameness. Training with computer aided learning tools seemed useful for students with less general horse experience and for students that themselves believe they have small abilities to evaluate hindlimb lameness, and the tool with real horses (in Canvas) might have an advantage, but this needs further study

    The generation of a T reporter cell line based screening method for T-cell receptor functional testing

    The immune system recognizes foreign entities in the body and induces protective responses. Sometimes, the immune system does not work optimally, which can lead to different clinical manifestations, one of which is cancer. T cells are a part of the immune system and has the ability to recognize and eliminate the body’s own cells that have transformed into cancer cells. In the field of immuno-oncology, a vast body of research is focusing on understanding the relationship between cancer and the immune system, as well as developing strategies to eliminate cancer by utilizing the immune system. The rise of computational approaches, that can analyze large amounts of data from single-cell sequencing techniques, have enabled the field to conduct research on a new level of complexity. However, this powerful tool has not yet reached its full potential, one major challenge is the lack of functional data of T-cell receptor recognition. In this thesis, we are focusing on the T-cell receptor mediated therapies against cancer, and work toward setting up a screen that assesses T-cell receptor specificity and functionality, which is a powerful tool to acquire knowledge of T-cell biology and improve immunotherapies. One can postulate that the functional data produced by functional screens could be used to train prediction algorithms to a degree, where we could predict the specificity of a TCR based on raw sequencing data.Immunsystemet kĂ€nner igen kroppsfrĂ€mmande komponenter och aktiverar skyddsmekanismer för att eliminera inkrĂ€ktaren. Ibland fungerar inte immunsystemet optimalt, vilket kan leda till olika kliniska manifestationer, var av en Ă€r cancer. T-lymfocyter Ă€r en del av immunsystemet och har en förmĂ„ga att kĂ€nna igen och eliminera kroppsegna celler som har förĂ€ndrats till cancerceller. Inom den heta forskningsomrĂ„det immunonkologi, arbetar forskare mot att förstĂ„ relationen mellan cancerceller och immunsystemet, samt utvecklar immunoterapier som strĂ€var till att eliminera cancer genom att anvĂ€nda sig av immunsystemets komponenter. Framsteg i bioinformatik har möjliggjort hantering och analys av stora mĂ€ngder data frĂ„n sekvensering av enskilda celler, och dĂ€rmed tagit forskningen till en ny nivĂ„ av komplexitet. Metoden har dock inte Ă€n nĂ„tt sin fulla potential, en utmaning Ă€r bristen av funktionell data av T-cellreceptorn. I denna avhandling, fokuserar vi pĂ„ T-cell-medierade immunoterapier och arbetar mot att bygga upp en screeningmetod för att undersöka T-cellreceptorns igenkĂ€nningsförmĂ„ga och funktion. Denna kraftiga metod kan anvĂ€ndas för att fĂ„ baskunskap om T-cellens biologi och utveckla immunoterapier. Man kan Ă€ven postulera, att med hjĂ€lp av den funktionella data som screeningmetoden ger, Ă€r det möjligt att trĂ€na algoritmer till den grad, att vi kan förutsĂ€ga en viss T-cellreceptors specificitet bara pĂ„ basis av den sekvenserade receptorns rĂ„data
