Biomolecules of Muscle Fatigue in Metabolic Myopathies
Metabolic myopathies are a group of genetic disorders that affect the normal functioning of muscles due to abnormalities in metabolic pathways. These conditions result in impaired energy production and utilization within muscle cells, leading to limitations in muscle function with concomitant occurrence of related signs and symptoms, among which fatigue is one of the most frequently reported. Understanding the underlying molecular mechanisms of muscle fatigue in these conditions is challenging for the development of an effective diagnostic and prognostic approach to test targeted therapeutic interventions. This paper outlines the key biomolecules involved in muscle fatigue in metabolic myopathies, including energy substrates, enzymes, ion channels, and signaling molecules. Potential future research directions in this field are also discussed
Metodología del aula invertida y competencia indaga mediante métodos científicos en estudiantes de una institución educativa pública de Ica, 2023
El docente debe estar preparado para aplicar metodologías activas que
generen y motiven el pensamiento crítico y autonomía en los estudiantes este
estudio tuvo la finalidad de determinar la relación entre la metodología del aula
invertida y la competencia indaga mediante métodos científicos en estudiantes del
tercer grado de secundaria de una Institución Educativa Pública de Ica, 2023.
Metodológicamente este estudio sigue la cuantitativa de tipo básico y nivel
descriptivo. El diseño fue no experimental. La población quedó constituida por 160
estudiantes entre 12 y 15 años de tercer grado de secundaria. La muestra del
estudio quedó constituida por 66 estudiantes de tercer grado. Se usó el muestreo
probabilístico al azar simple. La técnica la encuesta y la observación con sus
respectivos instrumentos Cuestionario sobre la aplicación de la metodología del
aula invertida, Cuestionario de autoevaluación de la competencia la competencia
indaga mediante métodos científicos con validez y confiabilidad. Los resultados
manifiestan que existe relación significativa entre la metodología del aula invertida
y la competencia indaga mediante métodos científicos en estudiantes (α=0.05, Sig.
Bilateral=0,000, r=0.570). En conclusión, existe una relación moderada entre la
variable metodología del aula invertida y la competencia indaga mediante métodos
científicos
Decision Making by a Neuromorphic Network of Volatile Resistive Switching Memories
The necessity of having an electronic device working in relevant biological
time scales with a small footprint boosted the research of a new class of
emerging memories. Ag-based volatile resistive switching memories (RRAMs)
feature a spontaneous change of device conductance with a similarity to
biological mechanisms. They rely on the formation and self-disruption of a
metallic conductive filament through an oxide layer, with a retention time
ranging from a few milliseconds to several seconds, greatly tunable according
to the maximum current which is flowing through the device. Here we prove a
neuromorphic system based on volatile-RRAMs able to mimic the principles of
biological decision-making behavior and tackle the Two-Alternative Forced
Choice problem, where a subject is asked to make a choice between two possible
alternatives not relying on a precise knowledge of the problem, rather on noisy
perceptions

The European project SUREBridge – A case study in Tuscany
The European project SUREBridge (Sustainable Refurbishment of Existing Bridges) is develop-ing a new concept for the refurbishment of road bridges. The proposed technique takes ad-vantage of the peculiarities of fibre-reinforced materials to perform upgrading, repair, and strengthening in an effective and efficient way in terms of resource consumption, waste produc-tion, construction time, and traffic disruption.
The technique applies to bridges with reinforced concrete slab and longitudinal girders made of either reinforced concrete or steel. Longitudinal girders are strengthened by bonding carbon fibre-reinforced polymer (CFRP) laminates to their bottom surfaces. Higher structural perfor-mances are achieved by pre-stressing the CFRP laminates. The existing concrete slab is not demolished, with savings in both construction time and waste production. Instead, tailor-made glass fibre-reinforced polymer (GFRP) panels are connected to the deck to increase its overall bending strength. Furthermore, GFRP panels enable the widening of the road section, if neces-sary to upgrade the bridge to increased traffic demand.
This paper presents the application of the SUREBridge technique to a real bridge located in San Miniato, Tuscany, Italy. The designed intervention includes both the widening of the road section and the structural strengthening of the deck to comply with current traffic loads
A Novel Nonsense Pathogenic TTN Variant Identified in a Patient with Severe Dilated Cardiomyopathy
Both genetic and environmental factors contribute to the development of dilated
cardiomyopathy. Among the genes involved, TTN mutations, including truncated variants, explain
25% of DCM cases. We performed genetic counseling and analysis on a 57-year-old woman
diagnosed with severe DCM and presenting relevant acquired risk factors for DCM (hypertension,
diabetes, smoking habit, and/or previous alcohol and cocaine abuse) and with a family history of
both DCM and sudden cardiac death. The left ventricular systolic function, as assessed by
standard echocardiography, was 20%. The genetic analysis performed using TruSight Cardio
panel, including 174 genes related to cardiac genetic diseases, revealed a novel nonsense TTN
variant (TTN:c.103591A > T, p.Lys34531*), falling within the M-band region of the titin protein. This
region is known for its important role in maintaining the structure of the sarcomere and in
promoting sarcomerogenesis. The identified variant was classified as likely pathogenic based on
ACMG criteria. The current results support the need of genetic analysis in the presence of a family
history, even when relevant acquired risk factors for DCM may have contributed to the severity of
the disease
Cerebral Palsy:Early Markers of Clinical Phenotype and Functional Outcome
The Prechtl General Movement Assessment (GMA) has become a cornerstone assessment in early identification of cerebral palsy (CP), particularly during the fidgety movement period at 3-5 months of age. Additionally, assessment of motor repertoire, such as antigravity movements and postural patterns, which form the Motor Optimality Score (MOS), may provide insight into an infant's later motor function. This study aimed to identify early specific markers for ambulation, gross motor function (using the Gross Motor Function Classification System, GMFCS), topography (unilateral, bilateral), and type (spastic, dyskinetic, ataxic, and hypotonic) of CP in a large worldwide cohort of 468 infants. We found that 95% of children with CP did not have fidgety movements, with 100% having non-optimal MOS. GMFCS level was strongly correlated to MOS. An MOS > 14 was most likely associated with GMFCS outcomes I or II, whereas GMFCS outcomes IV or V were hardly ever associated with an MOS > 8. A number of different movement patterns were associated with more severe functional impairment (GMFCS III-V), including atypical arching and persistent cramped-synchronized movements. Asymmetrical segmental movements were strongly associated with unilateral CP. Circular arm movements were associated with dyskinetic CP. This study demonstrated that use of the MOS contributes to understanding later CP prognosis, including early markers for type and severity

Consensus guidelines for the detection of immunogenic cell death
none82siApoptotic cells have long been considered as intrinsically tolerogenic or unable to elicit immune responses specific for dead cell-associated antigens. However, multiple stimuli can trigger a functionally peculiar type of apoptotic demise that does not go unnoticed by the adaptive arm of the immune system, which we named "immunogenic cell death" (ICD). ICD is preceded or accompanied by the emission of a series of immunostimulatory damage-associated molecular patterns (DAMPs) in a precise spatiotemporal configuration. Several anticancer agents that have been successfully employed in the clinic for decades, including various chemotherapeutics and radiotherapy, can elicit ICD. Moreover, defects in the components that underlie the capacity of the immune system to perceive cell death as immunogenic negatively influence disease outcome among cancer patients treated with ICD inducers. Thus, ICD has profound clinical and therapeutic implications. Unfortunately, the gold-standard approach to detect ICD relies on vaccination experiments involving immunocompetent murine models and syngeneic cancer cells, an approach that is incompatible with large screening campaigns. Here, we outline strategies conceived to detect surrogate markers of ICD in vitro and to screen large chemical libraries for putative ICD inducers, based on a high-content, high-throughput platform that we recently developed. Such a platform allows for the detection of multiple DAMPs, like cell surface-exposed calreticulin, extracellular ATP and high mobility group box 1 (HMGB1), and/or the processes that underlie their emission, such as endoplasmic reticulum stress, autophagy and necrotic plasma membrane permeabilization.
