    Examination of the clinical benefit of adding Reversal Theory concepts to the expressive writing paradigm

    Expressive writing as a form of emotional disclosure produces many physical and psychological benefits (Pennebaker, 2004). Central to the present study are findings that expressive writing decreases symptoms of depression, anxiety, and PTSD (Koopman, Ismailji, Holmes, Classen, Palesh, & Wales, 2005; Opre, Coman, Kallay, Rotaru, & Manier, 2005; Russ, 1992). Reversal Theory (RT) suggests that individuals experience the world from eight different psychological states including serious, playful, conforming, rebellious, mastery, sympathy, self, and other (Apter, 2007). Teaching individuals to be aware of and elicit these states also decreases depression and anxiety (Charat, 2006). The present study sought to determine if the clinical benefit in reducing subclinical symptoms of depression, anxiety, and PTSD in college students could be enhanced by the addition of RT concepts. Theoretically, this addition may increase the benefit derived from expressive writing by augmenting those processes believed to underlie the changes associated with expressive writing. These processes include reduction of inhibition through disclosure, emotional engagement and habituation, and cognitive changes including improved structure for understanding and increased insight (Lepore & Smyth, 2002). The study involved three groups, each participating in a modified version of the traditional expressive writing paradigm. The control group wrote about trivial topics with instructions to be objective; the expressive writing group wrote about a previous traumatic event with instructions to explore deepest thoughts and feelings; the Reversal Theory group wrote about a previous traumatic event with instructions to elicit and write from the perspective of the eight RT states. Participants were measured at the beginning and end of the study for symptoms of depression, anxiety, stress, and event-related stress. Though it was hypothesized that the RT group would show more clinical gain than the expressive writing group and that both experimental groups would fare better than the controls, the results indicated no significant differences among the groups on any of the measures. The author believes the nonsignificant findings most likely resulted from methodological issues including the relatively early collection of post-test data, the wide range of distress scores among participants, and group administration. Other factors may include the limited time for exposure in each RT state and the limited time for participants to develop a narrative of their trauma. Standardization of procedures and future research in this area, especially with regard to the clinical usefulness of Reversal Theory, is recommended

    Flagellin induces β-defensin 2 in human colonic ex vivo infection with enterohemorrhagic Escherichia coli

    Enterohemorrhagic E. coli (EHEC) is an important foodborne pathogen in the developed world and can cause life-threatening disease particularly in children. EHEC persists in the human gut by adhering intimately to colonic epithelium and forming characteristic attaching/effacing lesions. In this study, we investigated the innate immune response to EHEC infection with particular focus on antimicrobial peptide and protein expression by colonic epithelium. Using a novel human colonic biopsy model and polarized T84 colon carcinoma cells, we found that EHEC infection induced expression of human β-defensin 2 (hBD2), whereas hBD1, hBD3, LL-37 and lysozyme remained unchanged. Infection with specific EHEC deletion mutants demonstrated that this was dependent on flagellin, and apical exposure to purified flagellin was sufficient to stimulate hBD2 and also interleukin (IL)-8 expression ex vivo and in vitro. Flagellin-mediated hBD2 induction was significantly reduced by inhibitors of NF-κB, MAP kinase p38 and JNK but not ERK1/2. Interestingly, IL-8 secretion by polarized T84 cells was vectorial depending on the side of stimulation, and apical exposure to EHEC or flagellin resulted in apical IL-8 release. Our results demonstrate that EHEC only induces a modest immune response in human colonic epithelium characterized by flagellin-dependent induction of hBD2 and low levels of IL-8

    The effect of chlorhexidine mouthwash vs propolis mouthwash on the nitrate-reducing activity of oral bacteria and vascular control in healthy individuals

    Nitrate and nitrite molecules are involved in the synthesis of nitric oxide (NO), a gaseous vasodilatory molecule, via the nitrate-nitrite-NO pathway. Oral bacteria are responsible for the reduction of nitrate to nitrite making them important regulators in NO production and vascular control. This paper aimed to investigate whether a laboratory-made propolis mouthwash maintained the oral nitrate reducing capacity (ONRC) of commensal bacteria compared with a chlorhexidine (CHX) mouthwash and the effects of this on vascular function in healthy individuals. Twenty-eight healthy participants visited the laboratory on two occasions where anthropometric, ONRC, blood pressure and vascular function data were collected. Between laboratory visits a CHX mouthwash or a propolis mouthwash were used for seven days, twice-daily. It was found that propolis mouthwash used for seven days maintained the ONRC of commensal bacteria in healthy individuals (pre; 343.4 ±251.8μM vs post; 331.9 ±225.1μM, p=0.71) and that CHX mouthwash significantly abolished this activity, lowering levels by 67.7% in seven days (pre; 399.7 ±356.4μM vs post; 129.1 ±171.3μM, p<0.001). No significant changes in blood pressure or vascular function were seen following mouthwash use despite bacterial changes. It is unclear whether these insignificant findings are down to methodological issues or external physiological pathways. It was concluded that propolis mouthwash and CHX mouthwash affect the ONRC of commensal bacteria differently. Propolis mouthwash seems to preserve the ONRC therefore is less detrimental to the oral microbiome than CHX mouthwash suggesting potential uses in dentistry and as a therapeutic for hypertensive individuals

    Special Education in Juvenile Residential Facilities: Can Animals Help?

    Children with emotional/behavioral disorders (EBD) are arguably one of the highest at-risk groups for dropping out before graduating high school. They are the group of students with disabilities who are most likely to be educated in residential facilities. Residential facilities such as Green Chimneys have incorporated animals into the treatment milieu with success. Animals have been used in various settings to improve the quality of life as well as the emotional and physical needs of people served by these facilities. This article describes the requirements for using animals in residential treatment, the limitations of such programs, and the research findings for the use of animal-assisted therapy or activities (AAT/AAA). Using Gardner’s (1999) iteration of his theory of multiple intelligence, the authors proposed an understanding of how these treatments might be improve the intelligence of a child with EBD. Requirements for effective treatment of special needs children were discussed

    The Talin Head Domain Reinforces Integrin-Mediated Adhesion by Promoting Adhesion Complex Stability and Clustering

    Talin serves an essential function during integrin-mediated adhesion in linking integrins to actin via the intracellular adhesion complex. In addition, the N-terminal head domain of talin regulates the affinity of integrins for their ECM-ligands, a process known as inside-out activation. We previously showed that in Drosophila, mutating the integrin binding site in the talin head domain resulted in weakened adhesion to the ECM. Intriguingly, subsequent studies showed that canonical inside-out activation of integrin might not take place in flies. Consistent with this, a mutation in talin that specifically blocks its ability to activate mammalian integrins does not significantly impinge on talin function during fly development. Here, we describe results suggesting that the talin head domain reinforces and stabilizes the integrin adhesion complex by promoting integrin clustering distinct from its ability to support inside-out activation. Specifically, we show that an allele of talin containing a mutation that disrupts intramolecular interactions within the talin head attenuates the assembly and reinforcement of the integrin adhesion complex. Importantly, we provide evidence that this mutation blocks integrin clustering in vivo. We propose that the talin head domain is essential for regulating integrin avidity in Drosophila and that this is crucial for integrin-mediated adhesion during animal development

    Access to higher education : how might this be achieved for students from economically disadvantaged backgrounds?

    There are significant social inequalities in access to higher education internationally. Students from the most disadvantaged households remain persistently under-represented (Jerrim, Chmielewski, & Parker, 2015), are less likely to enter higher education, and when they do, are more likely to go to further education college rather than university (OECD, 2015; Scottish Funding Council, 2015; Sosu & Ellis, 2014). As a result, governments, supranational organisations such as the EU, and global agencies like UNESCO have expressed ambitions to reduce educational inequality and improve access to higher education (EHEA, 2012; UNESCO, 2015). Several factors such as academic performance, subject choice at secondary school and low motivation have been documented to account for this gap (e.g., Iannelli, Smyth, & Klein, 2015; (Iannelli, Smyth, & Klein, 2015; Chowdry, Crawford, Dearden, Goodman, & Vignoles, 2013; Gorard & Smith, 2006)

    Evaluation of a Head-Worn Display with Ambient Vision Cues for Unusual Attitude Recovery

    A Commercial Aviation Safety Team (CAST) study of 18 loss-of-control events determined that a lack of external visual references was a contributing factor in 17 of these events. CAST recommended that manufacturers should develop and implement virtual day-VMC display systems, such as synthetic vision (SV) or equivalent systems (CAST Safety Enhancement, SE-200). In support of this recommended action, CAST has requested studies to define minimum requirements for virtual day-visual meteorological conditions (VMC) displays to improve flight crew awareness of airplane attitude. NASAs research in Virtual day-VMC displays, known as synthetic vision systems, are intended to support intuitive flight crew attitude awareness similar to a day-VMC-like environment, especially if they could be designed to create visual dominance. A study was conducted to evaluate the utility of ambient vision (AV) cues paired with virtual Head-Up Display (HUD) symbology on a prototype head-worn display (HWD) during recovery from unusual attitudes in a simulated environment. The virtual-HUD component meets the requirement that the HWD may be used as an equivalent display to the HUD. The presence of AV cueing leverages the potential that a HWD has over the HUD for spatial disorientation prevention. The simulation study was conducted as a single-pilot operation, under realistic flight scenarios, with off-nominal events occurring that were capable of inducing unusual attitudes. Independent variables of the experiment included: 1) AV capability (on vs off) 2) AV display opaqueness (transparent vs opaque) and display location (HWD vs traditional head- down displays); AV cues were only present when the HWD was being worn by the subject pilot

    Identification and characterisation of enteroaggregative Escherichia coli subtypes associated with human disease

    Enteroaggregative E. coli (EAEC) are a major cause of diarrhoea worldwide. Due to their heterogeneity and carriage in healthy individuals, identification of diagnostic virulence markers for pathogenic strains has been difficult. In this study, we have determined phenotypic and genotypic differences between EAEC strains of sequence types (STs) epidemiologically associated with asymptomatic carriage (ST31) and diarrhoeal disease (ST40). ST40 strains demonstrated significantly enhanced intestinal adherence, biofilm formation, and pro-inflammatory interleukin-8 secretion compared with ST31 isolates. This was independent of whether strains were derived from diarrhoea patients or healthy controls. Whole genome sequencing revealed differences in putative virulence genes encoding aggregative adherence fimbriae, E. coli common pilus, flagellin and EAEC heat-stable enterotoxin 1. Our results indicate that ST40 strains have a higher intrinsic potential of human pathogenesis due to a specific combination of virulence-related factors which promote host cell colonization and inflammation. These findings may contribute to the development of genotypic and/or phenotypic markers for EAEC strains of high virulence

    Widening Access to Higher Education for Students from Economically Disadvantaged Backgrounds : What Works and Why? [Summary Report]

    There are significant social inequalities in access to higher education internationally. Students from the most disadvantaged backgrounds remain persistently under-represented in higher education (HE). Put simply, university populations fail to reflect their broader societies, with the vast majority of entrants coming from more advantaged backgrounds. Our research contributes to the ongoing effort by systematically examining the evidence for policy and programme interventions that widen access to higher education. We also examined trends in widening access in Scotland, barriers to access, and the impact of the national outreach scheme, the Schools for Higher Education Programme (SHEP), in supporting students to overcome these barriers. We provide recommendations for different stakeholders
