    Modeling Aggregation of Proliferating Microglia in Response to Amyloid-beta in Dementia

    Amyloid-beta plaques are prominent biological markers in dementia brains. In response to plaque formation, the brain\u27s immune cells, microglia, become reactive. Microglia are measurable cells that surround amyloid-beta plaques, indicating their location. A system of partial differential equations describes the concentration of microglia in dementia brains by incorporating chemotactic signaling. However, this system fails to incorporate increasing numbers of reactive microglia cells in response to amyloid-beta aggregation. A system of ordinary differential equations describing the number of significant cells and proteins in the brain suggests the amount of reactive microglia increases significantly during the progression of dementia. We couple these two systems to examine the influence of increasing amounts of reactive microglia on microglia concentration, which is indicative of amyloid-beta plaques. We compare the results of these systems to data and confirm the ability to predict the width of the aggregated microglia clusters when increasing amounts of microglia are included

    Time-of-day effects in arousal: disrupted diurnal cortisol profiles in children with ADHD

    Background: Fluctuations in attention-deficit hyperactivity disorder (ADHD) symptoms related to regulatory deficits in arousal states are themselves characterized by circadian rhythms. Although cortisol is an important circadian arousal-related marker, studies focusing on across-the-day cortisol variations in ADHD are scarce. There is no study with multiple measurements to take into account interday and intraday variability. Methods: Salivary cortisol was sampled five times a day (awakening, 30 min after awakening, noon, 4 p.m., 8 p.m.) across five consecutive days in 33 children with ADHD (22 with and 11 without oppositional defiant disorder; ODD) and 33 class- and sex-matched controls (aged 612). The cortisol awakening response (increase from awakening to 30 min after awakening) and the diurnal cortisol profile (across-the-day variations) were compared for ADHD with ODD (ADHD + ODD) and without ODD (ADHD) subgroups and the control group. Results: The cortisol awakening response was not significantly different between groups. However, longitudinal analyses to evaluate cortisol profiles across the day revealed a significant Group x Time effect (p < .001). More specifically, compared to each other, the ADHD subgroup showed a flatter slope with relative morning hypo-arousal and evening hyperarousal, whereas the ADHD + ODD subgroup showed a steeper slope with relative morning hyperarousal and evening hypo-arousal (p < .001). Conclusions: Findings support time-related arousal disruptions in children with ADHD associated with the presence or absence of ODD comorbidity. We recommend research on cortisol in larger samples for a better understanding of arousal mechanisms involved in ADHD not only with and without ODD but also with other comorbidities which may have implications for timing of arousal-based treatments

    Particle physics in the sky and astrophysics underground : connecting the universe's largest and smallest scales

The certified thesis is available in the Institute Archives and Special Collections.Includes bibliographical references (p. 151-169).Particles have tremendous potential as astronomical messengers, and conversely, studying the universe as a whole also teaches us about particle physics. This thesis encompasses both of these research directions. Many models predict a diffuse flux of high energy neutrinos from active galactic nuclei and other astrophysical sources. The "Astrophysics Underground" portion of this thesis describes a search for this neutrino flux performed by looking for extremely high energy upward-going muons using the Super-Kamiokande detector, and comparing the observed flux to the expected background. We use our results to to set an upper limit on the diffuse neutrino flux from astrophysical sources. In addition to using particles to do astronomy, we can also use the universe itself as a particle physics lab. Cosmology provides new insights that could never be observed in terrestrial laboratories. The "Particle Physics in the Sky" portion of this thesis focuses on extracting cosmological information from galaxy surveys. To overcome technical challenges faced by the latest galaxy surveys, we produced a comprehensive upgrade to mangle, a software package that processes the angular masks defining the survey area on the sky. We added dramatically faster algorithms and new useful features to this software that are necessary for managing complex masks of the Sloan Digital Sky Survey (SDSS) and will be invaluable for future surveys as well. With this software in hand, we utilized galaxy clustering data from SDSS to investigate the relation between galaxies and dark matter by studying relative bias, i.e., the relation between different types of galaxies. If all galaxies were perfect tracers of dark matter, different subpopulations would trace each other perfectly as well. However, separating galaxies by their luminosities and colors reveals a complicated picture: red galaxies are clustered more strongly than blue galaxies, with both the brightest and the faintest red galaxies showing the strongest clustering. Furthermore, red and blue galaxies tend to occupy different regions of space, effectively introducing an element of stochasticity (randomness) when modeling their relative distributions.     Ultrahigh-temperature granulite-facies metamorphism and exhumation of deep crust in a migmatite dome during late- to post-orogenic collapse and extension in the central Adirondack Highlands (New York, USA)

    This study combines field observations, mineral and whole-rock geochemistry, phase equilibrium modeling, and U-Pb sensitive high-resolution ion microprobe (SHRIMP) zircon geochronology to investigate sillimanite-bearing felsic migmatites exposed on Ledge Mountain in the central Adirondack Highlands (New York, USA), part of an extensive belt of mid-crustal rocks comprising the hinterland of the Mesoproterozoic Grenville orogen. Phase equilibrium modeling suggests minimum peak metamorphic conditions of 960-1025 °C and 11-12.5 kbar during the Ottawan orogeny—sig-nificantly higher pressure-temperature conditions than previously determined—followed by a period of near-isothermal decompression, then isobaric cooling. Petrography reveals abundant melt-related microstructures, and pseudosection models show the presence of at least ~15%-30% melt during buoyancy-driven exhumation and decompression. New zircon data document late Ottawan (re)crys-tallization at ca. 1047 ± 5 to 1035 ± 2 Ma following ultrahigh-temperature (UHT) metamorphism and anatexis on the retrograde cooling path. Inherited zircon cores give a mean date of 1136 ± 5 Ma, which suggests derivation of these felsic granulites by partial melting of older igneous rocks. The ferroan, anhydrous character of the granulites is similar to that of the ca. 1050 Ma Lyon Mountain Granite and consistent with origin in a late- to post-Ottawan extensional environment. We present a model for development of a late Ottawan migmatitic gneiss dome in the central Adirondacks that exhumed deep crustal rocks including the Snowy Mountain and Oregon anorthosite massifs with UHT Ledge Mountain migmatites. Recognition of deep crustal meta-plutonic rocks recording UHT metamorphism in a migmatite gneiss dome has significant implications for crustal behavior in this formerly thickened orogen

    Traveling Waves for Conservation Laws with Cubic Nonlinearity and BBM Type Dispersion

    Scalar conservation laws with non-convex fluxes have shock wave solutions that violate the Lax entropy condition. In this paper, such solutions are selected by showing that some of them have corresponding traveling waves for the equation supplemented with dissipative and dispersive higher-order terms. For a cubic flux, traveling waves can be calculated explicitly for linear dissipative and dispersive terms. Information about their existence can be used to solve the Riemann problem, in which we find solutions for some data that are different from the classical Lax-Oleinik construction. We consider dispersive terms of a BBM type and show that the calculation of traveling waves is somewhat more intricate than for a KdV-type dispersion. The explicit calculation is based upon the calculation of parabolic invariant manifolds for the associated ODE describing traveling waves. The results extend to the p-system of one-dimensional elasticity with a cubic stress-strain law.Comment: 17 pages, 5 figure

    The International Scientific Association for Probiotics and Prebiotics (ISAPP) consensus statement on the definition and scope of synbiotics

    In May 2019, the International Scientific Association for Probiotics and Prebiotics (ISAPP) convened a panel of nutritionists, physiologists and microbiologists to review the definition and scope of synbiotics. The panel updated the definition of a synbiotic to “a mixture comprising live microorganisms and substrate(s) selectively utilized by host microorganisms that confers a health benefit on the host”. The panel concluded that defining synbiotics as simply a mixture of probiotics and prebiotics could suppress the innovation of synbiotics that are designed to function cooperatively. Requiring that each component must meet the evidence and dose requirements for probiotics and prebiotics individually could also present an obstacle. Rather, the panel clarified that a complementary synbiotic, which has not been designed so that its component parts function cooperatively, must be composed of a probiotic plus a prebiotic, whereas a synergistic synbiotic does not need to be so. A synergistic synbiotic is a synbiotic for which the substrate is designed to be selectively utilized by the co-administered microorganisms. This Consensus Statement further explores the levels of evidence (existing and required), safety, effects upon targets and implications for stakeholders of the synbiotic concept
