    Inhalation of ZnO nanoparticles: Splice junction expression and alternative splicing in mice

    Despite the wide application of nanomaterials, toxicity studies of nanoparticles (NP) are often limited to in vitro cell models, and the biological impact of NP exposure in mammals has not been thoroughly investigated. Zinc oxide (ZnO) NPs are commonly used in various consumer products. To evaluate the effects of the inhalation of ZnO NP in mice, we studied splice junction expression in the lungs as a proxy to gene expression changes analysis. Female ICR mice were treated with 6.46 x 10(4) and 1.93 x 10(6) NP/cm(3) for 3 days and 3 months, respectively. An analysis of differential expression and alternative splicing events in 298 targets (splice junctions) of 68 genes involved in the processes relevant to the biological effects of ZnO NP was conducted using next-generation sequencing. Three days of exposure resulted in the upregulation of IL-6 and downregulation of BID, GSR, NF-kB2, PTGS2, SLC11A2, and TXNRD1 splice junction expression; 3 months of exposure increased the expression of splice junctions in ALDH3A1, APAF1, BID, CASP3, DHCR7, GCLC, GCLM, GSR, GSS, EHHADH, FAS, HMOX-1, IFN, NF-kB1, NQO-1, PTGS1, PTGS2, RAD51, RIPK2, SRXN1, TRAF6, and TXNRD1. Alternative splicing of TRAF6 and TXNRD1 was induced after 3 days of exposure to 1.93 x 10(6) NP/cm(3). In summary, we observed changes of splice junction expression in genes involved in oxidative stress, apoptosis, immune response, inflammation, and DNA repair, as well as the induction of alternative splicing in genes associated with oxidative stress and inflammation.     Diplomová práce se zabývá mediálním obrazem dětských pěveckých sborů s ohledem na takzvanou "kauzu Kulínský". Cílem práce je zjistit, jak často a jakým způsobem česká tištěná média informují o dětských pěveckých sborech a jakou roli v referování o tématu sehrál případ, v němž byl sbormistr Bohumil Kulínský odsouzen za zneužívání členek dětského sboru Bambini di Praga. V teoretické části jsou vymezeny koncepty z oblasti mediálních teorií a rovněž teoretická východiska související s oborem sborového zpěvu. Ve výzkumné části jsou prezentovány výsledky kvantitativní a kvalitativní obsahové analýzy obsahu tří českých tištěných deníků Mladá fronta DNES, Lidové noviny a Právo se zaměřením na jejich pražská vydání ve dvou časových obdobích. Časová období jsou zvolena s ohledem na vývoj "kauzy Kulínský". Výzkum sleduje nejen četnost textů o dětských pěveckých sborech, ale také užívání žánrů, řazení do rubrik a vyznění textů. Sledovaná období jsou mezi sebou vzájemně porovnána. Část výzkumu se orientuje také na online média a způsob informování o úmrtí sbormistra Bohumila Kulínského. V závěru práce je zhodnoceno, zda informování o dětských pěveckých sborech v českých médiích probíhá v souladu s konceptem zpravodajských hodnot.This thesis focuses on a media image of children's choirs influenced by the so-called "Kulínský Case". The aim of this thesis is to find out how and how often the Czech press informs about children's choirs and if the aforementioned case somehow changed the presentation of this content. The term "Kulínský Case" refers to the controversial case when choirmaster Bohumil Kulínský was found guilty of sexually abusing underage girls. First part of this thesis covers the media theories such as the news values and also describes the concepts linked with choir singing. In the research part, the results of quantitative and qualitative content analysis of three Czech daily newspapers Mladá fronta DNES, Lidové noviny and Právo are presented and interpreted. The analysis is focused on the content published in Prague editions of these three newspapers in two selected time frames. These time frames had been selected in accordance with the development of the "Kulínský Case". The research focuses on the frequency of the published texts, on used genres, newspaper sections and on the meaning and quality of the texts. Afterwards ensues the comparison of the two selected time frames.     Bechyně Chateau during the Reign of Petr Vok from Rožmberk

    This thesis will deal with chateau Bechyně during the period when it was possessed by Petr Vok of Rožmberk. The last Rožmberk bought Bechyně manor from Jindřich of Švamberk in 1569. He let Baldassare Maggi build a new Renaissance chateau which became the centre of Rožmberk court. The whole manor was sold due to enormous debts to Šternberk family in 1596. The author presents personality of Petr Vok in his relation to Bechyně and the life of his court at this chateau. Then she will deal with the building development of the chateau from the late Gothic castle to the Renaissance chateau. Especially, in the next part she will pay attention to painting and stucco decoration of the interiors which reflects Rožmberk spiritual and confessional transformation. In the last chapter, the author will deal also with the decoration of facade of the chateau courtyard which was created in that period

It is striving to find mainly models or inspiration of its painting. After evaluation of the basic literature there is a chapter which introduces castle Blatná as whole. Therefore, it includes the introduction of the history of castle Blatná, its construction development and decoration of the Room of a knight in the Old castle. The next part is already dealing with the green room itself. Gradually, it is searching for connections with graphics, book painting and tapestries. The last chapter contents the stylistic and iconographical-iconological evaluation of the whole decoration. It is striving to identify particular themes of figural paintings and to find a clue to the revealing the background of thought of the whole decoration

Snaží se především najít vzory či inspirace jejích maleb. Po zhodnocení základní literatury následuje kapitola, která představuje zámek Blatnou jako celek. Proto zahrnuje představení historie zámku Blatná, jeho stavební vývoj a výmalbu Rytířského sálu ve Starém paláci. Další část se již věnuje samotné zelené světnici. Postupně hledá souvislosti s grafikou, knižní malbou a tapisériemi. Poslední kapitola obsahuje stylové a ikonograficko - ikonologické zhodnocení celé výzdoby.     School maturity of a child from aspect of mathematics

    Bakalářská práce s názvem Školní zralost z hlediska matematiky, se zabývám v teoretické části školní zralostí, školní připraveností a oblastmi, které k posouzení školní zralostí musíme brát v potaz. Jedná se o fyzickou, psychickou a sociálně emoční zralost. Dále jsem se zaměřila na aspekty školní zralosti z hlediska matematiky. V metodologické části je popsáno 11 úkolů, kdy každý byl zaměřený na jinou oblast matematických schopností a znalostí. V experimentální části je popsána charakteristika mateřské školy, tříd a10 dětí, které byly do výzkumu zapojeny. Dále jsou zde v tabulkách vyhodnoceny činnosti z hlediska dítěte i z hlediska plnění úkolu. Z vyhodnocení vyplývá, že pouze šest dětí z deseti je z hlediska matematiky zralé k nástupu k základnímu vzdělávání.ObhájenoIn the theoretical part of this bachelor's degree work, I am looking into school maturity, school readiness and spheres, which must be consider in the final evaluation. It is a consideration of physical, psychological and socialy emotional maturity. Furthermore, I have focused on aspects of school readiness from mathematical view. The methodological part describes eleven tasks, every single one of them focused on different area of mathematical abilities and knowledge. The experimental part describes characteristics of the kindergarten, classes and ten children which were involved in the study. Also, there are charts evaluating activities from the childs point of view and from the task fulfilment. Following the evaluation, only six out of ten children are prepared for elementary school education from mathematical view
