    Characterization of \u3cem\u3eGlomerella\u3c/em\u3e Strains Recovered from Anthracnose Lesions on Common Bean Plants in Brazil

    Anthracnose caused by Colletotrichum lindemuthianum is an important disease of common bean, resulting in major economic losses worldwide. Genetic diversity of the C. lindemuthianum population contributes to its ability to adapt rapidly to new sources of host resistance. The origin of this diversity is unknown, but sexual recombination, via the Glomerella teleomorph, is one possibility. This study tested the hypothesis that Glomerella strains that are frequently recovered from bean anthracnose lesions represent the teleomorph of C. lindemuthianum. A large collection of Glomerella isolates could be separated into two groups based on phylogenetic analysis, morphology, and pathogenicity to beans. Both groups were unrelated to C. lindemuthianum. One group clustered with the C. gloeosporioides species complex and produced mild symptoms on bean tissues. The other group, which belonged to a clade that included the cucurbit anthracnose pathogen C. magna, caused no symptoms. Individual ascospores recovered from Glomerella perithecia gave rise to either fertile (perithecial) or infertile (conidial) colonies. Some pairings of perithecial and conidial strains resulted in induced homothallism in the conidial partner, while others led to apparent heterothallic matings. Pairings involving two perithecial, or two conidial, colonies produced neither outcome. Conidia efficiently formed conidial anastomosis tubes (CATs), but ascospores never formed CATs. The Glomerella strains formed appressoria and hyphae on the plant surface, but did not penetrate or form infection structures within the tissues. Their behavior was similar whether the beans were susceptible or resistant to anthracnose. These same Glomerella strains produced thick intracellular hyphae, and eventually acervuli, if host cell death was induced. When Glomerella was co-inoculated with C. lindemuthianum, it readily invaded anthracnose lesions. Thus, the hypothesis was not supported: Glomerella strains from anthracnose lesions do not represent the teleomorphic phase of C. lindemuthianum, and instead appear to be bean epiphytes that opportunistically invade and sporulate in the lesions

    The acute phase response is a systemic reaction to inflammatory processes characterized by multiple physiological adaptations, including the hepatic synthesis of acute-phase proteins. In humans, serum amyloid A (SAA) is one of the most prominent of these proteins. Despite the huge increase of serum levels of SAA in inflammation, its biological role remains to be elucidated, even though SAA is undoubtedly active in neutrophils. In a previous study, we reported that SAA induces the release of tumor necrosis factor-alpha, interleukin (IL)-1beta and IL-8 from human blood neutrophils. Here, we extend our earlier study, focusing on the effect of SAA on neutrophil IL-8 transcription and on the signaling pathways involved. We demonstrate herein that SAA, in relatively low concentrations (0.4-100 microg/ml) compared with those found in plasma in inflammatory conditions, induces a dose-dependent release of IL-8 from neutrophils. The p38 mitogen-activated protein kinase inhibitor SB 203580 inhibits the IL-8 mRNA expression and the release of protein from neutrophils. The release of IL-8 from SAA-stimulated neutrophils is strongly suppressed by the addition of N-acetyl-l-cysteine, alpha-mercaptoethanol, glutathione, and dexamethasone. SAA also induces IL-8 expression and release from monocytes. In conclusion, SAA appears to be an important mediator of the inflammatory process, possibly contributing to the pool of IL-8 produced in chronic diseases, which may play a role in degenerative diseases

    Virtual Environment, Digital Hypertext, Reading and Writing in Foreign Language

    This work intends to analyze - in an activity carried out with students from the third period of the Language (Spanish) undergraduate course of a college located in Itaperuna, a town in the State of Rio de Janeiro, Brazil - how the digital hyper textual reading can facilitate the selection of information in order to facilitate the writing process of texts in Spanish that can be broadcasted in the virtual environment, verifying how this may happen, so that these productions are shared, also allowing the interaction of the subjects with the language and with their peers. For this objective, we used the qualitative methodology (Erickson, 1986) with action research, seeking foundation in what theorists such as Lévy (1996, 1999), Coscarelli (2006, 2009), Gomes et al. (2015), Bannell et al. (2016), among others, investigate. As results, we emphasize that the subjects of the research actively participated in the construction of their own learning regarding the aspects covered in the foreign language class, and with this they were able to practice reading, writing, and the USAge of the vocabulary and grammar studied. Finally, we conclude that the hypertext worked here as an inclusive device, facilitator of reading and propitiator of writing, making the participant students authors of digital texts that provided them with learning throughout all the process they have been through

    Aplicação dos Princípios Analítico-Comportamentais para Alterar o Comportamento de uma Esquizofrênica

    The present study investigated the behavior of a chronic schizophrenic using the principles of behavior analysis. The participant was female, age 57, single and has had multiple hospitalizations since the age of 18. The types of problematic behavior that received intervention were: eye contact, self isolation in the yard, participation in the events of the institution, and others. To control the effect of the intervention procedures the ABAB reversal design followed by a follow-up was used. The experiment started with the baseline of phase I. In intervention phases I and II positive reinforcement, modeling and extinction procedures were used. The results indicated that the procedures of behavior analysis were effective to control the problematic-behavior displayed by the participant.O objetivo do presente estudo foi investigar o comportamento de uma pessoa diagnosticada como esquizofrênica crônica e retardo mental no contexto psiquiátrico, em uma unidade particular conveniada com o SUS. Para essa finalidade foram utilizados os princípios da análise do comportamento aplicada. A participante foi uma pessoa do sexo feminino, com 57 anos, solteira, semi-analfabeta, descendente de família de parcos recursos econômicos e que houvera sido internada em várias instituições especializadas para tratamentos psiquiátricos, desde os 18 anos de idade. Foram selecionadas sete classes de comportamentos-problema para sofrerem intervenção como, por exemplo, manter contato olho a olho, permanecer isolada sentada num banco do pátio, participar dos eventos da instituição etc. Para o controle dos procedimentos foi utilizado o delineamento de reversão do tipo ABAB, seguido por follow-up. O delineamento foi iniciado com a coleta de dados de linha de base I. A seguir foram realizadas as sessões nas fases de intervenções I e II. Nas intervenções foram utilizados procedimentos de reforçamento positivo, modelagem e extinção.  Os resultados demonstraram que os procedimentos da análise do comportamento aplicada foram efetivos para controlar os comportamentos-problema apresentados pela participante

    Chamomile gel versus urea cream to prevent acute radiation dermatitis in head and neck cancer patients : results from a preliminary clinical trial

    We assessed safety and potential efficacy of a chamomile gel compared with urea cream to prevent acute radiation dermatitis in head and neck cancer patients. We assessed safety and potential efficacy of the chamomile gel in escalating concentrations of 2.50%, 5.00% and 8.35% of chamomile. Concentration of 8.35% was chosen for a randomized trial comparing chamomile gel (8.35%) with urea cream (n=24per group), for potential efficacy to delay or prevent radiation dermatitis in these patients. Preliminary results demonstrate a delayed onset of dermatitis, with onset of Grade 2 dermatitis at 5.1 (1.3) weeks in the chamomile group and 4.5 (1.3) weeks in the urea group (effect size of 0.46). Itching, burning and hyperpigmentation were more frequently reported in the urea group. Results indicates a potential efficacy of the chamomile gel. Further studies are needed to confirm the effect of the chamomile gel in reducing or delaying the occurrence of radiation dermatitis

    Dairy Foods in a Moderate Energy Restricted Diet Do Not Enhance Central Fat, Weight, and Intra-Abdominal Adipose Tissue Losses nor Reduce Adipocyte Size or Inflammatory Markers in Overweight and Obese Adults: A Controlled Feeding Study

    Background. Research on dairy foods to enhance weight and fat loss when incorporated into a modest weight loss diet has had mixed results. Objective. A 15-week controlled feeding study to determine if dairy foods enhance central fat and weight loss when incorporated in a modest energy restricted diet of overweight and obese adults. Design. A 3-week run-in to establish energy needs; a 12-week 500 kcal/d energy reduction with 71 low-dairy-consuming overweight and obese adults randomly assigned to diets: ≤1 serving dairy/d (low dairy, LD) or ≤4 servings dairy/d (adequate dairy, AD). All foods were weighed and provided by the metabolic kitchen. Weight, fat, intra-abdominal adipose tissue (IAAT), subcutaneous adipose tissue (SAT) macrophage number, SAT inflammatory gene expression, and circulating cytokines were measured. Results. No diet differences were observed in weight, fat, or IAAT loss; nor SAT mRNA expression of inflammation, circulating cytokines, fasting lipids, glucose, or insulin. There was a significant increase (P = 0.02) in serum 25-hydroxyvitamin D in the AD group. Conclusion. Whether increased dairy intake during weight loss results in greater weight and fat loss for individuals with metabolic syndrome deserves investigation. Assessment of appetite, hunger, and satiety with followup on weight regain should be considered

    A Revolução Industrial iniciou um longo processo de alteração na produção de mercadorias e consequentemente nos padrões de consumo mundial. Devido os grandes investimentos em tecnologia, a  oferta  e  demanda  de  bens  atinge  grandes  proporções.  Esse  processo  contínuo  passou  por  várias modificações e aperfeiçoamentos, mas sempre com um crescimento da quantidade de mercadorias. As grandes indústrias hoje alcançam lugares antes inimagináveis para ofertar o seu produto e influenciar as decisões  do  consumidor.  Com  o  apoio  da  mídia,  propagam-se  os  padrões  de  consumo  dos  países desenvolvidos nos lugares mais remotos, como no interior das florestas. O exacerbado consumo de bens nessas localidades leva a um problema  antes característico do meio urbano: o destino dos resíduos oriundos  do  consumo.  O  objetivo  deste  trabalho  é  avaliar  a  geração  e  destino  do  lixo  nas  florestas acreanas. Para tanto, utilizaram-se materiais e métodos desenvolvidos pelo projeto ASPF, capitaneado pelo Departamento de Economia da UFAC. Assim, observou-se uma preocupante elevação da produção de lixo na floresta acreana, que pode contribuir para aumentar a degradação ambiental, colocando em xeque a sustentabilidade do sistema em questão

    In vitro and in vivo assessment of the anti-malarial activity of Caesalpinia pluviosa

    <p>Abstract</p> <p>Background</p> <p>To overcome the problem of increasing drug resistance, traditional medicines are an important source for potential new anti-malarials. <it>Caesalpinia pluviosa</it>, commonly named "sibipiruna", originates from Brazil and possess multiple therapeutic properties, including anti-malarial activity.</p> <p>Methods</p> <p>Crude extract (CE) was obtained from stem bark by purification using different solvents, resulting in seven fractions. An MTT assay was performed to evaluate cytotoxicity in MCF-7 cells. The CE and its fractions were tested <it>in vitro </it>against chloroquine-sensitive (3D7) and -resistant (S20) strains of <it>Plasmodium falciparum</it> and <it>in vivo </it>in <it>Plasmodium chabaudi</it>-infected mice. <it>In vitro </it>interaction with artesunate and the active <it>C. pluviosa </it>fractions was assessed, and mass spectrometry analyses were conducted.</p> <p>Results</p> <p>At non-toxic concentrations, the 100% ethanolic (F4) and 50% methanolic (F5) fractions possessed significant anti-malarial activity against both 3D7 and S20 strains. Drug interaction assays with artesunate showed a synergistic interaction with the F4. Four days of treatment with this fraction significantly inhibited parasitaemia in mice in a dose-dependent manner. Mass spectrometry analyses revealed the presence of an ion corresponding to <it>m/z </it>303.0450, suggesting the presence of quercetin. However, a second set of analyses, with a quercetin standard, showed distinct ions of <it>m/z </it>137 and 153.</p> <p>Conclusions</p> <p>The findings show that the F4 fraction of <it>C. pluviosa </it>exhibits anti-malarial activity <it>in vitro </it>at non-toxic concentrations, which was potentiated in the presence of artesunate. Moreover, this anti-malarial activity was also sustained <it>in vivo </it>after treatment of infected mice. Finally, mass spectrometry analyses suggest that a new compound, most likely an isomer of quercetin, is responsible for the anti-malarial activity of the F4.</p
