    Antioxidant intervention in rheumatoid arthritis: results of an open pilot study

    There is evidence that reactive oxygen species play a causal role in auto-immune diseases, such as rheumatoid arthritis (RA). Despite the supporting evidence for a beneficial effect of antioxidants on clinical characteristics of RA, the right balance for optimal effectiveness of antioxidants is largely unknown. To determine the potential beneficial effects of an antioxidant intervention on clinical parameters for RA, an open pilot study was designed. Eight non-smoking female patients with rheumatoid factor + RA and a Disease Activity Score (DAS 28) higher than 2.5 were enrolled in the study. Patients had to be receiving stable non-steroidal anti-inflammatory drug treatment and/or ‘second line’ medication for at least 3 months. The pilot group consumed 20 g of antioxidant-enriched spread daily during a period of 10 weeks. The intervention was stopped after 10 weeks and was followed by a ‘wash-out’ period of 4 weeks. At t = 0, t = 10 weeks and t = 14 weeks, patients’ condition was assessed by means of DAS. In addition, standard laboratory analyses were performed, and blood-samples for antioxidants were taken. The antioxidant-enriched spread was well tolerated. All laboratory measures of inflammatory activity and oxidative modification were generally unchanged. However, the number of swollen and painful joints were significantly decreased and general health significantly increased, as reflected by a significantly improved (1.6) DAS at t = 10 weeks. The antioxidant effect was considered beneficial as, compared to the scores at t = 0, the DAS significantly reduced at t = 10 weeks. Increase of the DAS (0.7) after the “wash-out period” at t = 14 confirmed a causal relation between changes in clinical condition and antioxidants. This open pilot study aimed to assess the clinical relevance of an antioxidant intervention as a first step in assessing potential beneficial effects of antioxidants on rheumatoid arthritis. These conclusions need to be validated in a larger controlled study population

    Ferromagnetic Semiconductors: Moving Beyond (Ga,Mn)As

    The recent development of MBE techniques for growth of III-V ferromagnetic semiconductors has created materials with exceptional promise in spintronics, i.e. electronics that exploit carrier spin polarization. Among the most carefully studied of these materials is (Ga,Mn)As, in which meticulous optimization of growth techniques has led to reproducible materials properties and ferromagnetic transition temperatures well above 150 K. We review progress in the understanding of this particular material and efforts to address ferromagnetic semiconductors as a class. We then discuss proposals for how these materials might find applications in spintronics. Finally, we propose criteria that can be used to judge the potential utility of newly discovered ferromagnetic semiconductors, and we suggest guidelines that may be helpful in shaping the search for the ideal material.Comment: 37 pages, 4 figure

    Listening In on the Past: What Can Otolith ÎŽ18O Values Really Tell Us about the Environmental History of Fishes?

    Oxygen isotope ratios from fish otoliths are used to discriminate marine stocks and reconstruct past climate, assuming that variations in otolith ÎŽ18O values closely reflect differences in temperature history of fish when accounting for salinity induced variability in water ÎŽ18O. To investigate this, we exploited the environmental and migratory data gathered from a decade using archival tags to study the behaviour of adult plaice (Pleuronectes platessa L.) in the North Sea. Based on the tag-derived monthly distributions of the fish and corresponding temperature and salinity estimates modelled across three consecutive years, we first predicted annual otolith ÎŽ18O values for three geographically discrete offshore sub-stocks, using three alternative plausible scenarios for otolith growth. Comparison of predicted vs. measured annual ÎŽ18O values demonstrated >96% correct prediction of sub-stock membership, irrespective of the otolith growth scenario. Pronounced inter-stock differences in ÎŽ18O values, notably in summer, provide a robust marker for reconstructing broad-scale plaice distribution in the North Sea. However, although largely congruent, measured and predicted annual ÎŽ18O values of did not fully match. Small, but consistent, offsets were also observed between individual high-resolution otolith ÎŽ18O values measured during tag recording time and corresponding ÎŽ18O predictions using concomitant tag-recorded temperatures and location-specific salinity estimates. The nature of the shifts differed among sub-stocks, suggesting specific vital effects linked to variation in physiological response to temperature. Therefore, although otolith ÎŽ18O in free-ranging fish largely reflects environmental temperature and salinity, we counsel prudence when interpreting otolith ÎŽ18O data for stock discrimination or temperature reconstruction until the mechanisms underpinning otolith ÎŽ18O signature acquisition, and associated variation, are clarified

    Discovery of a black smoker vent field and vent fauna at the Arctic Mid-Ocean Ridge

    The Arctic Mid-Ocean Ridge (AMOR) represents one of the most slow-spreading ridge systems on Earth. Previous attempts to locate hydrothermal vent fields and unravel the nature of venting, as well as the provenance of vent fauna at this northern and insular termination of the global ridge system, have been unsuccessful. Here, we report the first discovery of a black smoker vent field at the AMOR. The field is located on the crest of an axial volcanic ridge (AVR) and is associated with an unusually large hydrothermal deposit, which documents that extensive venting and long-lived hydrothermal systems exist at ultraslow-spreading ridges, despite their strongly reduced volcanic activity. The vent field hosts a distinct vent fauna that differs from the fauna to the south along the Mid-Atlantic Ridge.     Mosquito larvicidal activities of Solanum villosum berry extract against the dengue vector Stegomyia aegypti

    <p>Abstract</p> <p>Background</p> <p>Vector control is facing a threat due to the emergence of resistance to synthetic insecticides. Insecticides of botanical origin may serve as suitable alternative biocontrol techniques in the future. Although several plants have been reported for mosquitocidal activity, only a few botanicals have moved from the laboratory to field use, because they are poorly characterized, in most cases active principals are not determined and most of the works are restricted to preliminary screening. <it>Solanum villosum </it>is a common weed distributed in many parts of India with medicinal properties, but the larvicidal activity of this plant has not been reported so far.</p> <p>Methods</p> <p>Aqueous and polar/non-polar solvent extract of fresh, mature, green berries of <it>S. villosum </it>was tested against <it>Stegomyia aegypti</it>, a common vector of dengue fever. A phytochemical analysis of chloroform:methanol extract was performed to search for the active toxic ingredient. The lethal concentration was determined (log probit analysis) and compared with Malathion. The chemical nature of the active substance was also evaluated following ultraviolet-visual (UV-Vis) and infrared (IR) analysis.</p> <p>Results</p> <p>In a 72 hour bioassay experiment with the aqueous extract, the highest mortality was recorded in 0.5% extract. When the mortality of different solvent extracts was compared, the maximum (<it>p </it>< 0.05) mortality was recorded at a concentration of 50 ppm of chloroform:methanol extract (1:1, v/v). The larvicidal activity was lower when compared with the chemical insecticide, Malathion (<it>p </it>< 0.05). Results of regression analysis revealed that the mortality rate (<it>Y</it>) was positively correlated with the period of exposure (<it>X</it>) and the log probit analysis (95% confidence level) recorded lowest value (5.97 ppm) at 72 hours of exposure. Phytochemical analysis of the chlororm:methanol extract reported the presence of many bioactive phytochemicals. Two toxic compounds were detected having <it>R</it><sub>f </sub>= 0.82 (70% and 73.33% mortality in 24 and 48 hours, respectively) and <it>R</it><sub>f </sub>= 0.95 (40% and 50% mortality in 24 and 48 hours, respectively). IR analysis provided preliminary information about the steroidal nature of the active ingredient.</p> <p>Conclusion</p> <p><it>S. villosum </it>offers promise as potential bio control agent against <it>S. aegypti </it>particularly in its markedly larvicidal effect. The extract or isolated bioactive phytochemical could be used in stagnant water bodies for the control of mosquitoes acting as vector for many communicable diseases.</p

    Fermi Large Area Telescope observations of PSR J1836+5925

    The discovery of the gamma-ray pulsar PSR J1836+5925, powering the formerly unidentified EGRET source 3EG J1835+5918, was one of the early accomplishments of the Fermi Large Area Telescope (LAT). Sitting 25 degrees off the Galactic plane, PSR J1836+5925 is a 173 ms pulsar with a characteristic age of 1.8 million years, a spindown luminosity of 1.1×1034\times10^{34} erg s−1^{-1}, and a large off-peak emission component, making it quite unusual among the known gamma-ray pulsar population. We present an analysis of one year of LAT data, including an updated timing solution, detailed spectral results and a long-term light curve showing no indication of variability. No evidence for a surrounding pulsar wind nebula is seen and the spectral characteristics of the off-peak emission indicate it is likely magnetospheric. Analysis of recent XMM observations of the X-ray counterpart yields a detailed characterization of its spectrum, which, like Geminga, is consistent with that of a neutron star showing evidence for both magnetospheric and thermal emission.Comment: Accepted to Astrophysical Journa

    One step closer to understanding the role of bacteria in diabetic foot ulcers: characterising the microbiome of ulcers

    Background: The aim of this study was to characterise the microbiome of new and recurrent diabetic foot ulcers using 16S amplicon sequencing (16S AS), allowing the identification of a wider range of bacterial species that may be important in the development of chronicity in these debilitating wounds. Twenty patients not receiving antibiotics for the past three months were selected, with swabs taken from each individual for culture and 16S AS. DNA was isolated using a combination of bead beating and kit extraction. Samples were sequenced on the Illumina Hiseq 2500 platform. Results: Conventional laboratory culture showed positive growth from only 55 % of the patients, whereas 16S AS was positive for 75 % of the patients (41 unique genera, representing 82 different operational taxonomic units (OTU’s). S. aureus was isolated in 72 % of culture-positive samples, whereas the most commonly detected bacteria in all ulcers were Peptoniphilusspp., Anaerococcus spp. and Corynebacterium spp., with the addition of Staphylococcus spp. in new ulcers. The majority of OTU’s residing in both new and recurrent ulcers (over 67 %) were identified as facultative or strict anaerobic Gram-positive organisms. Principal component analysis (PCA) showed no difference in clustering between the two groups (new and recurrent ulcers). Conclusions: The abundance of anaerobic bacteria has important implications for treatment as it suggests that the microbiome of each ulcer “starts afresh” and that, although diverse, are not distinctly different from one another with respect to new or recurrent ulcers. Therefore, when considering antibiotic therapy the duration of current ulceration may be a more important consideration than a history of healed ulcer

    Rapid metal pollutant deposition from the volcanic plume of Kīlauea, Hawai’i

    AbstractLong-lived basaltic volcanic eruptions are a globally important source of environmentally reactive, volatile metal pollutant elements such as selenium, cadmium and lead. The 2018 eruption of Kīlauea, Hawai’i produced exceptionally high discharge of metal pollutants, and was an unprecedented opportunity to track them from vent to deposition. Here we show, through geochemical sampling of the plume that volatile metal pollutants were depleted in the plume up to 100 times faster than refractory species, such as magnesium and iron. We propose that this rapid wet deposition of complexes containing reactive and potentially toxic volatile metal pollutants may disproportionately impact localised areas close to the vent. We infer that the relationship between volatility and solubility is an important control on the atmospheric behaviour of elements. We suggest that assessment of hazards from volcanic emissions should account for heterogeneous plume depletion of metal pollutants.</jats:p

    Relationships between paranoid thinking, self-esteem and the menstrual cycle.

    This study aimed to investigate whether paranoid experiences and levels of self-esteem fluctuate over the menstrual cycle and whether levels of self-esteem are lower when perceived persecution is felt to be deserved. Measures of anxiety, depression, persecution, deservedness and self-esteem were completed on-line by 278 women over their menstrual cycle. Responses were compared at the paramenstrual (3 days before and after menses onset) and mid-cycle phase. At the paramenstrual phase persecution, negative self-esteem, anxiety and depression were higher and positive self-esteem was lower than at mid-cycle. A greater proportion of women experienced persecution as deserved at the paramenstrual phase. This was associated with higher depression and negative self-esteem scores. Increased levels of deservedness significantly strengthened the relationship between persecution and negative, but not positive, self-esteem. These findings suggest that the paramenstrual phase is a time of vulnerability to increased paranoid experiences, an increased likelihood that feelings of persecution will feel deserved and lowered self-esteem. The findings support the view that interpersonal sensitivities may be key to menstrual cycle symptoms and have an impact on relationships. Further, the study illustrated that ideas developed for psychosis could make a valuable contribution to understanding and managing this aspect of menstruation-related distress
