    Long-term reproductive costs of snare injuries in a keystone terrestrial by-catch species

    Extensive bushmeat hunting is a major threat to wildlife conservation worldwide, particularly when unselective methods such as wire snares kill target and non-target species (by-catch). Animals that escape from snares have injuries of varying severity, with effects on performance that are largely unknown, as most studies typically focus on immediate mortality caused by snaring. Here, we assessed the life-history costs of debilitating snare injuries in individually known female spotted hyenas Crocuta crocuta in three clans in the Serengeti National Park, Tanzania. This keystone predator is a regular by-catch of illegal bushmeat hunting of herbivores in the Serengeti ecosystem. We monitored individuals which escaped from snares between May 1987 and March 2020 and survived long enough to return to their clan territories from commuting trips in the park and surrounding protected areas. Snares that inflicted debilitating injuries on females did not reduce longevity but did delay age at first reproduction and reduced both litter size and offspring survival to the age of 1 year. This long-term decrease in reproductive performance likely resulted from increased inflammatory and immune responses to the snare injury and/or a decreased ability to travel the long distances necessary to feed on migratory herbivores. While our results are based on a relatively small sample of females with debilitating injuries, they suggest that the total population-level costs of wire snares in terrestrial by-catch species may be underestimated and that future studies may need to account for the potential reproductive costs of sublethal snare injuries

    A randomised trial evaluating Bevacizumab as adjuvant therapy following resection of AJCC stage IIB, IIC and III cutaneous melanoma : an update

    At present, there are no standard therapies for the adjuvant treatment of malignant melanoma. Patients with primary tumours with a high-Breslow thickness (stages IIB and IIC) or with resected loco-regional nodal disease (stage III) are at high risk of developing metastasis and subsequent disease-related death. Given this, it is important that novel therapies are investigated in the adjuvant melanoma setting. Since angiogenesis is essential for primary tumour growth and the development of metastasis, anti-angiogenic agents are attractive potential therapeutic candidates for clinical trials in the adjuvant setting. Therefore, we initiated a phase II trial in resected high-risk cutaneous melanoma, assessing the efficacy of bevacizumab versus observation. In the interim safety data analysis, we demonstrate that bevacizumab is a safe therapy in the adjuvant melanoma setting with no apparent increase in the surgical complication rate after either primary tumour resection and/or loco-regional lymphadenectomy

    A role for TSPO in mitochondrial Ca2+ homeostasis and redox stress signaling

    The 18 kDa translocator protein TSPO localizes on the outer mitochondrial membrane (OMM). Systematically overexpressed at sites of neuroinflammation it is adopted as a biomarker of brain conditions. TSPO inhibits the autophagic removal of mitochondria by limiting PARK2-mediated mitochondrial ubiquitination via a peri-organelle accumulation of reactive oxygen species (ROS). Here we describe that TSPO deregulates mitochondrial Ca2+ signaling leading to a parallel increase in the cytosolic Ca2+ pools that activate the Ca2+-dependent NADPH oxidase (NOX) thereby increasing ROS. The inhibition of mitochondrial Ca2+ uptake by TSPO is a consequence of the phosphorylation of the voltage-dependent anion channel (VDAC1) by the protein kinase A (PKA), which is recruited to the mitochondria, in complex with the Acyl-CoA binding domain containing 3 (ACBD3). Notably, the neurotransmitter glutamate, which contributes neuronal toxicity in age-dependent conditions, triggers this TSPO-dependent mechanism of cell signaling leading to cellular demise. TSPO is therefore proposed as a novel OMM-based pathway to control intracellular Ca2+ dynamics and redox transients in neuronal cytotoxicity

    Women's Experiences of Preeclampsia: Australian Action on Preeclampsia Survey of Women and Their Confidants

    Introduction. The experience of normal pregnancy is often disrupted for women with preeclampsia (PE). Materials and Methods. Postal survey of the 112 members of the consumer group, Australian Action on Pre-Eclampsia (AAPEC). Results. Surveys were returned by 68 women (61% response rate) and from 64 (57%) partners, close relatives or friends. Respondents reported experiencing pre-eclampsia (n = 53), eclampsia (n = 5), and/or Hemolysis, Elevated Liver enzymes, and Low Platelets (HELLP syndrome) (n = 26). Many women had no knowledge of PE prior to diagnosis (77%) and, once diagnosed, did not appreciate how serious or life threatening it was (50%). Women wanted access to information about PE. Their experience contributed substantial anxiety towards future pregnancies. Partners/friends/relatives expressed fear for the woman and/or her baby and had no prior understanding of PE. Conclusions. The PE experience had a substantial effect on women, their confidants, and their babies and affected their approach to future pregnancies. Access to information about PE was viewed as very important

The well-documented EEG "signature" of preparation for a task switch-a protracted positive-polarity modulation over the posterior scalp-has thus far not been reported in language switching, and the interpretation of previously reported effects of preparation on language switching performance is complicated by confounding factors. In an experiment using event-related potentials (ERPs) and an optimized picture-naming paradigm that addressed these confounds the language was specified by an auditory cue on every trial and changed unpredictably. There were two key manipulations. First, the cue-stimulus interval allowed either generous (1,500 ms) or little (100 ms) opportunity for preparation. Second, to explore the interplay between bottom-up and top-down language selection, we compared a highly transparent and familiar "supercue"-the name of the language spoken in that language to a relatively opaque cue (short speeded-up fragment of national anthem). Preparation for a switch elicited a brain potential strongly reminiscent of the posterior switch positivity documented in task switching. As previously shown in task switching, its amplitude inversely predicted the performance "switch cost," demonstrated by our ERP analyses contingent on reaction time (RT). This overlap in the electrophysiological correlates of preparing to switch tasks and languages suggests domain-general processes for top-down selection of task-set and language for production. But, the surprisingly small language switch cost following the supercue in the short CSI suggests that rapid and (possibly automatic) bottom-up selection-not typically observed in task switching-may also occur.     Using machine learning to parametrize postmerger signals from binary neutron stars

    There is growing interest in the detection and characterization of gravitational waves from postmerger oscillations of binary neutron stars. These signals contain information about the nature of the remnant and the high-density and out-of-equilibrium physics of the postmerger processes, which would complement any electromagnetic signal. However, the construction of binary neutron star postmerger waveforms is much more complicated than for binary black holes: (i) there are theoretical uncertainties in the neutron-star equation of state and other aspects of the high-density physics, (ii) numerical simulations are expensive and available ones only cover a small fraction of the parameter space with limited numerical accuracy, and (iii) it is unclear how to parametrize the theoretical uncertainties and interpolate across parameter space. In this work, we describe the use of a machine-learning method called a conditional variational autoencoder (CVAE) to construct postmerger models for hyper/massive neutron star remnant signals based on numerical-relativity simulations. The CVAE provides a probabilistic model, which encodes uncertainties in the training data within a set of latent parameters. We estimate that training such a model will ultimately require 104\sim 10^4 waveforms. However, using synthetic training waveforms as a proof-of-principle, we show that the CVAE can be used as an accurate generative model and that it encodes the equation of state in a useful latent representation

    Nonlinear effects in the black hole ringdown: absorption-induced mode excitation

    Gravitational-wave observations of black hole ringdowns are commonly used to characterize binary merger remnants and to test general relativity. These analyses assume linear black hole perturbation theory, in particular that the ringdown can be described in terms of quasinormal modes even for times approaching the merger. Here we investigate a nonlinear effect during the ringdown, namely how a mode excited at early times can excite additional modes as it is absorbed by the black hole. This is a third-order secular effect: the change in the black-hole mass causes a shift in the mode spectrum, so that the original mode is projected onto the new ones. Using nonlinear simulations, we study the ringdown of a spherically-symmetric scalar field around an asymptotically anti-de Sitter black hole, and we find that this "absorption-induced mode excitation" (AIME) is the dominant nonlinear effect. We show that this effect takes place well within the nonadiabatic regime, so we can analytically estimate it using a sudden mass-change approximation. Adapting our estimation technique to asymptotically-flat Schwarzschild black holes, we expect AIME to play a role in the analysis and interpretation of current and future gravitational wave observations
