    Evidence for the maintenance of slowly varying equatorial currents by intraseasonal variability

    Recent evidence from mooring data in the equatorial Atlantic reveals that semi-annual and longer time scale ocean current variability is close to being resonant with equatorial basin modes. Here we show that intraseasonal variability, with time scales of 10's of days, provides the energy to maintain these resonant basin modes against dissipation. The mechanism is analogous to that by which storm systems in the atmosphere act to maintain the atmospheric jet stream. We demonstrate the mechanism using an idealised model set-up that exhibits equatorial deep jets. The results are supported by direct analysis of available mooring data from the equatorial Atlantic Ocean covering a depth range of several thousand meters. The analysis of the mooring data suggests that the same mechanism also helps maintain the seasonal variability

    A longitudinal study of gene expression in healthy individuals

    <p>Abstract</p> <p>Background</p> <p>The use of gene expression in venous blood either as a pharmacodynamic marker in clinical trials of drugs or as a diagnostic test requires knowledge of the variability in expression over time in healthy volunteers. Here we defined a normal range of gene expression over 6 months in the blood of four cohorts of healthy men and women who were stratified by age (22–55 years and > 55 years) and gender.</p> <p>Methods</p> <p>Eleven immunomodulatory genes likely to play important roles in inflammatory conditions such as rheumatoid arthritis and infection in addition to four genes typically used as reference genes were examined by quantitative reverse transcription-polymerase chain reaction (qRT-PCR), as well as the full genome as represented by Affymetrix HG U133 Plus 2.0 microarrays.</p> <p>Results</p> <p>Gene expression levels as assessed by qRT-PCR and microarray were relatively stable over time with ~2% of genes as measured by microarray showing intra-subject differences over time periods longer than one month. Fifteen genes varied by gender. The eleven genes examined by qRT-PCR remained within a limited dynamic range for all individuals. Specifically, for the seven most stably expressed genes (CXCL1, HMOX1, IL1RN, IL1B, IL6R, PTGS2, and TNF), 95% of all samples profiled fell within 1.5–2.5 Ct, the equivalent of a 4- to 6-fold dynamic range. Two subjects who experienced severe adverse events of cancer and anemia, had microarray gene expression profiles that were distinct from normal while subjects who experienced an infection had only slightly elevated levels of inflammatory markers.</p> <p>Conclusion</p> <p>This study defines the range and variability of gene expression in healthy men and women over a six-month period. These parameters can be used to estimate the number of subjects needed to observe significant differences from normal gene expression in clinical studies. A set of genes that varied by gender was also identified as were a set of genes with elevated expression in a subject with iron deficiency anemia and another subject being treated for lung cancer.</p

    Identification of novel loci associated with gastrointestinal parasite resistance in a Red Maasai x Dorper backcross population

    Gastrointestinal (GI) parasitic infection is the main health constraint for small ruminant production, causing loss of weight and/or death. Red Maasai sheep have adapted to a tropical environment where extreme parasite exposure is a constant, especially with highly pathogenic Haemonchus contortus. This breed has been reported to be resistant to gastrointestinal parasite infection, hence it is considered an invaluable resource to study associations between host genetics and resistance. The aim of this study was to identify polymorphisms strongly associated with host resistance in a double backcross population derived from Red Maasai and Dorper sheep using a SNP-based GWAS analysis. The animals that were genotyped represented the most resistant and susceptible individuals based on the tails of phenotypic distribution (10% each) for average faecal egg counts (AVFEC). AVFEC, packed cell volume (AVPCV), and live weight (AVLWT) were adjusted for fixed effects and co-variables, and an association analysis was run using EMMAX. Revised significance levels were calculated using 100,000 permutation tests. The top five significant SNP markers with - log10 p-values >3.794 were observed on five different chromosomes for AVFEC, and BLUPPf90/PostGSf90 results confirmed EMMAX significant regions for this trait. One of these regions included a cluster of significant SNP on chromosome (Chr) 6 not in linkage disequilibrium to each other. This genomic location contains annotated genes involved in cytokine signalling, haemostasis and mucus biosynthesis. Only one association detected on Chr 7 was significant for both AVPCV and AVLWT. The results generated here reveal candidate immune variants for genes involved in differential response to infection and provide additional SNP marker information that has potential to aid selection of resistance to gastrointestinal parasites in sheep of a similar genetic background to the double backcross population

    Canadian university students\u27 gendered attitudes toward plagiarism

    Prior research on plagiarism has indicated that men may have a greater predisposition toward academic dishonesty than women. However, little research has been conducted using psychometrically tested instruments to validate such claims. To address this gap, a survey was conducted with 377 undergraduate students at a Canadian university on their attitudes toward plagiarism using a psychometrically validated instrument (the Attitudes Toward Plagiarism Questionnaire - Revised). Using differential item functioning/Rasch analysis, no overall differences in attitudes toward plagiarism based on gender were found. A descriptive analysis of both men and women revealed that while only a concerning minority of students reported engaging in plagiarist behaviours; there was a tendency for students to take a permissive stance on plagiarism. These results are discussed within the wider context of plagiarism research in higher education

    Teacher Disposition Scale (TDS): construction and psychometric validation

    Despite the growing prominence and importance of the field of teacher dispositions for addressing selection criteria and assessment in educational research, there is an absence of psychometrically evaluated instruments to facilitate investigations. Furthermore, research thus far has not explored the relationship between teacher dispositions and better student outcomes. To address this paucity, key dispositional factors conducive to successful teaching and learning were identified through interviewing high performing teachers. Based on the interview, data a 24-item Teacher Disposition Scale (TDS) was constructed consisting of five dimensions: Motivation to teach; Teacher efficacy; Willingness to learn; Conscientiousness; and Interpersonal and communication skill. The scale was psychometrically tested on 179 pre-service undergraduate students at an Australian university. Both factor analysis and Rasch measurement modelling indicated evidence of a reliable and valid multi-dimensional scale. The article concludes with a discussion on the implications of the scale for future research

    Abstract Background The health benefits of polyunsaturated fatty acids (PUFAs), particularly those of the n-3 series are well documented. The mechanisms by which these effects are mediated are not fully clarified. Methods We used microarrays to assess the effects on gene expression in HT29 colon adenocarcinoma cells of exposure to the n-3 fatty acid eicosapentaenoic acid (EPA). HT29 cells were cultured with EPA (150 μM) for up to 24 hr prior to harvesting and isolation of RNA. Microarray results were analyzed within the statistical package 'R', and GeneGo MetaCore was used to identify key pathways of altered gene expression. Results EphB4, Vav2 and EphA1 gene expression were identified as significantly altered by EPA treatment. Statistically significant changes in gene expression after HT29 exposure to EPA were confirmed in a second experiment by real-time RT-PCR (TaqMan), This experiment also compared the effects of exposure to EPA to arachadonic acid (AA, n-6). Corresponding changes in protein expression were also assessed by Western blotting. Conclusions Eph receptor mediated signaling is an entirely novel signaling pathway through which EPA may promote a wide range of health benefits, in particular in relation to reduction of colorectal cancer progression.</p
