    Rapid asssessment of drinking-water quality: a handbook for implementation

    The JMP Rapid assessment of drinking-water quality (RADWQ) was conceived by WHO and UNICEF to explore the quality of drinking-water from "improved" sources. Results in five pilot countries showed a wide range of conditions, from full compliance with the WHO Drinking-water Quality Guidelines to specific sources in a given country only meeting standards in 34% of the samples. Further efforts along the lines of RADWQ will take global water quality testing out of the strict sphere of monitoring into the broader field of capacity development. The RADWQ handbook describes the methods and procedures applied by WHO and UNICEF in the five countries, for adoption by any authority or institution that wants to prepare a snapshot of the quality of "improved" sources of drinking-water, as a first step towards strengthening drinking-water quality regulations

    Root cortex development is fine-tuned by the interplay of MIGs, SCL3 and DELLAs during arbuscular mycorrhizal symbiosis

    Root development is a crucial process that determines the ability of plants to acquire nutrients, adapt to the substrate and withstand changing environmental conditions. Root plasticity is controlled by a plethora of transcriptional regulators that allow, in contrast to tissue development in animals, post-embryonic changes that give rise to new tissue and specialized cells. One of these changes is the accommodation in the cortex of hyperbranched hyphae of symbiotic arbuscular mycorrhizal (AM) fungi, called arbuscules. Arbuscule-containing cells undergo massive reprogramming to coordinate developmental changes with transport processes. Here we describe a novel negative regulator of arbuscule development, MIG3. MIG3 induces and interacts with SCL3, both of which modulate the activity of the central regulator DELLA, restraining cortical cell growth. As in a tug-of-war, MIG3-SCL3 antagonizes the function of the complex MIG1-DELLA, which promotes the cell expansion required for arbuscule development, adjusting cell size during the dynamic processes of the arbuscule life cycle. Our results in the legume plant Medicago truncatula advance the knowledge of root development in dicot plants, showing the existence of additional regulatory elements not present in Arabidopsis that fine-tune the activity of conserved central modules

    Stable longitudinal associations of family income with children's hippocampal volume and memory persist after controlling for polygenic scores of educational attainment

    Despite common notion that the correlation of socioeconomic status with child cognitive performance may be driven by both environmentally- and genetically-mediated transactional pathways, there is a lack of longitudinal and genetically informed research that examines these postulated associations. The present study addresses whether family income predicts associative memory growth and hippocampal development in middle childhood and tests whether these associations persist when controlling for DNA-based polygenic scores of educational attainment. Participants were 142 6-to-7-year-old children, of which 127 returned when they were 8-to-9 years old. Longitudinal analyses indicated that the association of family income with children's memory performance and hippocampal volume remained stable over this age range and did not predict change. On average, children from economically disadvantaged background showed lower memory performance and had a smaller hippocampal volume. There was no evidence to suggest that differences in memory performance were mediated by differences in hippocampal volume. Further exploratory results suggested that the relationship of income with hippocampal volume and memory in middle childhood is not primarily driven by genetic variance captured by polygenic scores of educational attainment, despite the fact that polygenic scores significantly predicted family income

    A versatile toolkit for high throughput functional genomics with Trichoderma reesei

    <p>Abstract</p> <p>Background</p> <p>The ascomycete fungus, <it>Trichoderma reesei </it>(anamorph of <it>Hypocrea jecorina</it>), represents a biotechnological workhorse and is currently one of the most proficient cellulase producers. While strain improvement was traditionally accomplished by random mutagenesis, a detailed understanding of cellulase regulation can only be gained using recombinant technologies.</p> <p>Results</p> <p>Aiming at high efficiency and high throughput methods, we present here a construction kit for gene knock out in <it>T. reesei</it>. We provide a primer database for gene deletion using the <it>pyr4, amdS </it>and <it>hph </it>selection markers. For high throughput generation of gene knock outs, we constructed vectors using yeast mediated recombination and then transformed a <it>T. reesei </it>strain deficient in non-homologous end joining (NHEJ) by spore electroporation. This NHEJ-defect was subsequently removed by crossing of mutants with a sexually competent strain derived from the parental strain, QM9414.</p> <p>Conclusions</p> <p>Using this strategy and the materials provided, high throughput gene deletion in <it>T. reesei </it>becomes feasible. Moreover, with the application of sexual development, the NHEJ-defect can be removed efficiently and without the need for additional selection markers. The same advantages apply for the construction of multiple mutants by crossing of strains with different gene deletions, which is now possible with considerably less hands-on time and minimal screening effort compared to a transformation approach. Consequently this toolkit can considerably boost research towards efficient exploitation of the resources of <it>T. reesei </it>for cellulase expression and hence second generation biofuel production.</p

    Effects of Growth Hormone Treatment in Adults with Prader-Willi Syndrome

    OBJECTIVE: Since limited data exist on adults with Prader-Willi syndrome (PWS) and growth hormone (GH) treatment, we report our experience on the effects of treatment for one year on body composition, physical activity, strength and energy expenditure, diet, general chemistry and endocrine data with quality of life measures. DESIGN: We studied 11 adults with PWS (6F:5M; average age = 32 yrs) over a 2 year period with GH treatment during the first year only. Electrolytes, IGF-I, glucose, thyroid, insulin, lipids, body composition, physical activity and strength, diet, energy expenditure and quality of life data were collected and analyzed statistically using linear modeling at baseline, at 12 months following GH therapy and at 24 months after treatment cessation for 12 months. RESULTS: Total lean muscle mass was significantly increased (p < 0.05) during GH treatment along with moderate-vigorous physical activity and plasma IGF-I and HDL levels, but returned to near baseline after treatment. Percent body fat decreased during the 12 months of GH treatment but increased after treatment. CONCLUSIONS: Previously reported beneficial effects of GH treatment in children with PWS were found in our adults regarding body composition, physical activity and plasma HDL and IGF-I levels. Several beneficial effects diminished to near baseline after cessation of GH treatment for 12 months supporting the continuation of treatment in PWS into adulthood and possibly adults not previously treated during childhood

    Colonization of Vitis vinifera L. by the Endophyte Trichoderma sp. Strain T154: Biocontrol Activity Against Phaeoacremonium minimum

    [EN] Trichoderma strains used in biological control products usually exhibit high efficiency in the control of plant diseases. However, their behavior under field conditions is difficult to predict. In addition, the potential of indigenous strains has been poorly assayed as well as their possible behavior as endophytes. Hence, niche colonization is a key feature for an effective protection. In this study, we aimed to: (i) explore the possibility of using a new Trichoderma strain isolated from vine to control pathogens, (ii) study the in planta interaction with the pathogen Phaeoacremonium minimum W. Gams, Crous, M.J. Wingf. & L. Mugnai (formerly Phaeoacremonium aleophilum), a pioneer fungus involved in Grapevine Trunk Diseases (GTDs) such as esca. For this purpose, fluorescently tagged Trichoderma sp. T154 and a P. minimum strain were used for scanning electron microscopy and confocal scanning laser microscopy analyses. Data showed that the Trichoderma strain is able to colonize plants up to 12 weeks post inoculation and is located in xylem, fibers, as well as in parenchymatic tissues inside the wood. The beneficial fungus reduced colonization of the esca-related pathogen colonizing the same niches. The main observed mechanism involved in biocontrol of Trichoderma against the esca pathogen was spore adhesion, niche exclusion and only few typical hypha coiling was found between Trichoderma and the pathogen. These results suggest that the Trichoderma strain has potential for reducing the colonization of Phaeoacremonium minimum and thus, an inoculation of this biological control agent can protect the plant by limiting the development of GTD, and the strain can behave as an endophyte.SIThe grant awarded to GC-H (FPU15/04681) comes from the Ministry of Education, Culture, and Sport (Spain).     Diffractive orbits in isospectral billiards

    Isospectral domains are non-isometric regions of space for which the spectra of the Laplace-Beltrami operator coincide. In the two-dimensional Euclidean space, instances of such domains have been given. It has been proved for these examples that the length spectrum, that is the set of the lengths of all periodic trajectories, coincides as well. However there is no one-to-one correspondence between the diffractive trajectories. It will be shown here how the diffractive contributions to the Green functions match nevertheless in a ''one-to-three'' correspondence.Comment: 20 pages, 6 figure

    Constraining the Spin of the Black Hole in Fairall 9 with Suzaku

    We report on the results of spectral fits made to data obtained from a 168 ksec Suzaku observation of the Seyfert-1 galaxy Fairall 9. The source is clearly detected out to 30 keV. The observed spectrum is fairly simple; it is well-described by a power-law with a soft excess and disk reflection. A broad iron line is detected, and easily separated from distinct narrow components owing to the resolution of the CCDs in the X-ray Imaging Spectrometer (XIS). The broad line is revealed to be asymmetric, consistent with a disk origin. We fit the XIS and Hard X-ray Detector (HXD) spectra with relativistically-blurred disk reflection models. With the assumption that the inner disk extends to the innermost stable circular orbit, the best-fit model implies a black hole spin parameter of a = 0.60(7) and excludes extremal values at a high level of confidence. We discuss this result in the context of Seyfert observations and models of the cosmic distribution of black hole spin.Comment: Accepted for publication in Ap
