An Experiment in Scaling Impact: Assessing the Growth Capital Aggregation Pilot
This report presents an assessment of the Growth Capital Aggregation Pilot. It was commissioned by the Edna McConnell Clark Foundation, founder and lead investor of the grantmaking initiative.Starting in 2000, The Edna McConnell Clark Foundation (Clark) adopted an investment approach to grantmaking that focused on providing growth capital to youth-serving organizations with demonstrated commitments to evaluation and measurable outcomes. For grantees, the strategy meant larger, longer-term, unrestricted investments, complemented by extensive access to consulting and technical assistance to strengthen their organizations.This approach helped Clark grantees across the portfolio increase the numbers of youth they served (for example, by 18 percent between 2005 and 2006) and achieve annual revenue gains (averaging 19 percent over the four years prior to the founding of GCAP). At the same time, the Foundation concluded that more capital would be required if its grantees and other promising youth-serving organizations were to realize their ultimate scale and sustainability potential

spin foam amplitude via a lattice discretisation. Furthermore, by analysing the
supersymmetric quantum amplitudes, we show that they can be re-interpreted as

Training Restricted Boltzmann Machines on Word Observations
The restricted Boltzmann machine (RBM) is a flexible tool for modeling
complex data, however there have been significant computational difficulties in
using RBMs to model high-dimensional multinomial observations. In natural
language processing applications, words are naturally modeled by K-ary discrete
distributions, where K is determined by the vocabulary size and can easily be
in the hundreds of thousands. The conventional approach to training RBMs on
word observations is limited because it requires sampling the states of K-way
softmax visible units during block Gibbs updates, an operation that takes time
linear in K. In this work, we address this issue by employing a more general
class of Markov chain Monte Carlo operators on the visible units, yielding
updates with computational complexity independent of K. We demonstrate the
success of our approach by training RBMs on hundreds of millions of word
n-grams using larger vocabularies than previously feasible and using the
learned features to improve performance on chunking and sentiment
classification tasks, achieving state-of-the-art results on the latter
Recent occurrence of Cylindrospermopsis raciborskii, in Waikato lakes of New Zealand.
Cylindrospermopsis raciborskii is a toxin-producing species of cyanobacteria that in autumn 2003 was recorded for the first time in three shallow (max. depth â€5 m) Waikato lakes and a hydro-electric dam on the Waikato River, New Zealand. It formed water blooms at densities >100 000 cells/ml in Lakes Waahi and Whangape. Net rates of population growth >0.2 day-1 were recorded for C. raciborskii in Lakes Ngaroto, Waahi, and Karapiro, based on comparisons of low numbers (detection of cells/ml) from initial samples and its presence at bloom densities (>15 000 cells/ml) in the subsequent sample "x"-"y" days later. C. raciborskii may be well adapted to rapid proliferation in the Waikato lakes, which are eutrophic to hypertrophic, with high light attenuation, and where nitrogen (N) fixation may provide it with a competitive advantage over non-nitrogen fixing algae under N-limited conditions

Gaining assurance in a voter-verifiable voting system
The literature on e-voting systems has many examples of discussion of the correctness of the computer and communication algorithms of such systems, as well as discussions of their vulnerabilities. However, a gap in the literature concerns the practical need (before adoption of a specific e-voting system) for a complete case demonstrating that the system as a whole has sufficiently high probability of exhibiting the desired properties when in use in an actual election. This paper discusses the problem of producing such a case, with reference to a specific system: a version of the PrĂȘt Ă Voter scheme for voter-verifiable e-voting. We show a possible organisation of a case in terms of four main requirements â accuracy, privacy, termination and âtrustednessââ and show some of the detailed organisation that such a case should have, the diverse kinds of evidence that needs to be gathered and some of the interesting difficulties that arise
Intravital Multiphoton Microscopy with Fluorescent Bile Salts in Rats as an In Vivo Biomarker for Hepatobiliary Transport Inhibition
The bile salt export pump (BSEP) is expressed at the canalicular domain of hepatocytes, where it mediates the elimination of monovalent bile salts into the bile. Inhibition of BSEP is considered a susceptibility factor for drug-induced liver injury that often goes undetected during nonclinical testing. Although in vitro assays exist for screening BSEP inhibition, a reliable and specific method for confirming Bsep inhibition in vivo would be a valuable follow up to a BSEP screening strategy, helping to put a translatable context around in vitro inhibition data, incorporating processes such as metabolism, protein binding, and other exposure properties that are lacking in most in vitro BSEP models. Here, we describe studies in which methods of quantitative intravital microscopy were used to identify dose-dependent effects of two known BSEP/Bsep inhibitors, 2-[4-[4-(butylcarbamoyl)-2-[(2,4-dichlorophenyl)sulfonylamino]phenoxy]-3-methoxyphenyl]acetic acid (AMG-009) and bosentan, on hepatocellular transport of the fluorescent bile salts cholylglycyl amidofluorescein and cholyl-lysyl-fluorescein in rats. Results of these studies demonstrate that the intravital microscopy approach is capable of detecting Bsep inhibition at drug doses well below those found to increase serum bile acid levels, and also indicate that basolateral efflux transporters play a significant role in preventing cytosolic accumulation of bile acids under conditions of Bsep inhibition in rats. Studies of this kind can both improve our understanding of exposures needed to inhibit Bsep in vivo and provide unique insights into drug effects in ways that can improve our ability interpret animal studies for the prediction of human drug hepatotoxicity
