Hematodinium infection seasonality in the Firth of Clyde (Scotland) nephrops norvegicus population: a re-evaluation
Hematodinium infections in Norway lobster Nephrops norvegicus from the Clyde Sea area (CSA) population, Scotland, UK, have previously been undetected in summer. This study aimed to establish if the CSA is actually devoid of infected N. norvegicus in this season. Two PCR assays, an ELISA and 2 tests that detect only patent infection (pleopod and body colour methods) were applied in a 21 mo study. Patent infection was seasonal, appearing predominantly in spring, while subpatent infection diagnosed by ELISA and PCR was highly prevalent in all seasons. Generalised linear modelling supported this assertion, as sampling in September and February significantly increased the probability of finding infected N. norvegicus (p < 0.01); infections were predominantly subpatent and patent respectively, at these times. Therefore, Hematodinium seasonality in N. norvegicus populations is likely to have been an artefact of insensitive diagnostic tests. Light Hematodinium infections were found using PCR assays when patent infections were at their most prevalent and intense, suggesting that infection develops at different rates in different N. norvegicus individuals and that only a portion of the total number of infected N. norvegicus die within a single year. These new data were added to a long-term data series for the CSA (1990 to 2008), which showed that after an initial 5 yr epidemic period, prevalence stabilised at 20 to 25%. Comparisons with ‘susceptible-infected-recovered/removed’ (SIR) models suggest that this high prevalence is maintained through high birth rates of susceptible host N. norvegicus
A cross sectional study of the prevalence, risk factors and population attributable fractions for limb and body lesions in lactating sows on commercial farms in England
Background: Lesions on sows' limbs and bodies are an abnormality that might impact on their welfare. The prevalence of and risks for limb and body lesions on lactating sows on commercial English pig farms were investigated using direct observation of the sows and their housing.
Results: The prevalence of lesions on the limbs and body were 93% (260/279) and 20% (57/288) respectively. The prevalence of limb and body lesions was significantly lower in outdoor-housed sows compared with indoor-housed sows. Indoor-housed sows had an increased risk of wounds (OR 6.8), calluses (OR 8.8) and capped hock (OR 3.8) on their limbs when housed on fully slatted floors compared with solid concrete floors. In addition, there was an increased risk of bursitis (OR 2.7), capped hock (OR 2.3) and shoulder lesions (OR 4.8) in sows that were unwilling to rise to their feet. There was a decreased risk of shoulder lesions (OR 0.3) and lesions elsewhere on the body (OR 0.2) in sows with more than 20 cm between their tail and the back of the crate compared with sows with less than 10 cm.
Conclusion: The sample of outdoor housed sows in this study had the lowest prevalence of limb and body lesions. In lactating sows housed indoors there was a general trend for an increased risk of limb and body lesions in sows housed on slatted floors compared with those housed on solid concrete floors with bedding. Sows that were less responsive to human presence and sows that had the least space to move within their crates had an additional increased risk of lesions

Developing theory and practice: Creation of a Community of Practice through Action Research produced excellence in stroke care
Much emphasis is placed on expert knowledge such as evidence based stroke guidelines, but not enough attention is paid to the processes that are required to translate this into delivery of everyday good care. This paper highlights the worth of creating a Community of Practice (CoP) as a means to achieve this. Drawing on findings from a study conducted 2000 - 2002 of the processes involved in establishment of a nationally lauded high quality Stroke Unit, it demonstrates how successful development of a new service was linked to creation of a CoP.
Recent literature suggests CoPs have a key role in the implementation of evidence-based practice; this study supports this claim whilst revealing for the first time the practical knowledge and skills required to develop this style of working. Findings also indicate that the participatory and democratic characteristics of Action Research are congruent with the collaborative approach required for the development of a CoP. The study is an exemplar of how practitioner researchers can capture learning from changing practice, thus contributing to evidence-based healthcare with theoretical as well as practical knowledge. Findings are relevant to those developing stroke services globally but also to those interested in evidence-based practice. (198
High-level inhibition of mitochondrial complexes III and IV is required to increase glutamate release from the nerve terminal
<p>Abstract</p> <p>Background</p> <p>The activities of mitochondrial complex III (ubiquinol-cytochrome <it>c </it>reductase, EC 1.10.2.2) and complex IV (cytochrome <it>c </it>oxidase EC 1.9.3.1) are reduced by 30-70% in Huntington's disease and Alzheimer's disease, respectively, and are associated with excitotoxic cell death in these disorders. In this study, we investigated the control that complexes III and complex IV exert on glutamate release from the isolated nerve terminal.</p> <p>Results</p> <p>Inhibition of complex III activity by 60-90% was necessary for a major increase in the rate of Ca<sup>2+</sup>-independent glutamate release to occur from isolated nerve terminals (synaptosomes) depolarized with 4-aminopyridine or KCl. Similarly, an 85-90% inhibition of complex IV activity was required before a major increase in the rate of Ca<sup>2+</sup>-independent glutamate release from depolarized synaptosomes was observed. Inhibition of complex III and IV activities by ~ 60% and above was required before rates of glutamate efflux from polarized synaptosomes were increased.</p> <p>Conclusions</p> <p>These results suggest that nerve terminal mitochondria possess high reserves of complex III and IV activity and that high inhibition thresholds must be reached before excess glutamate is released from the nerve terminal. The implications of the results in the context of the relationship between electron transport chain enzyme deficiencies and excitotoxicity in neurodegenerative disorders are discussed.</p
A qualitative exploration of living with chronic neuropathic pain after spinal cord injury: an Italian perspective
Purpose: To understand how people with spinal cord injury (SCI) in Italy experienced and managed chronic neuropathic pain (CNP), and their perspectives of Italian healthcare services.
Method: Nine people with SCI participated. Two focus groups (3 and 4 individuals respectively) and one semi-structured interview were audio-recorded and transcribed. One “virtual interview” was conducted via email. A qualitative thematic analysis was undertaken.
Results:
Three main themes were identified. Firstly, participants experienced pain as a powerful, intrusive and, at times, inescapable force, with the potential to overwhelm the sense of self, and place limits on enjoyable experiences. Secondly, participants recounted a strong desire to understand CNP, and, in the absence of expert guidance, used trial-and-error methods to find ways of relieving pain. Thirdly, healthcare practice was perceived as pharmacologically-focused and lacking specialist knowledge. Practitioners were described as reluctant to explore alternative therapies or participate in collaborative, patient-centered care.
Conclusions:
This study reveals SCI-related CNP as a deeply troubling and psychologically distressing condition impacting widely on everyday life. Specialist, collaborative, individually-tailored rehabilitation approaches that attend to patients’ priorities and experiences, include education about CNP, and offer opportunities to explore complementary treatments, may be welcomed by people living with this condition in Italy

Sport workforce strategy in the United Kingdom (UK) has identified the occupational therapy profession as being ideally positioned to contribute to public health agendas relating to tackling physical inactivity amongst marginalised populations, such as disabled people and people with experience of mental distress. However, a robust understanding of the enablers, restrictions, and exclusions such groups encounter when seeking to participate in sport and physical activity is currently lacking.
Methods:
This study aimed to gain an in-depth understanding of the different ways people with experience of mental distress talked about their participation in a community-based football league in England, in the UK. Nine people took part in this strand of a larger participatory action research (PAR) study, which used go-along interviews as the method of data collection. In alignment with PAR seeking to address power imbalances, the data from the go-along interviews were analysed through a Foucauldian lens using a collaboratively produced analytic framework.
Findings:
Participants constructed the community-based football league as fostering feelings of purpose and belonging, against a backdrop of them describing experiencing stigma and exclusion when seeking to be active in their wider communities. They used the concept of occupational marginalisation to further interpret their situation.
Conclusion:
Understanding why and how people participate in football extends beyond seeing it as an individual exercise to shared social lives and occupations. With this perspective, occupational therapists could address occupational marginalisation in partnership with community sports organisations, collaborating for wider social change beyond specialist services.Funding information: Royal College of Occupational Therapists' Annual Award; Constance Owens Trust; Elizabeth Casson Trus
Gated rotation mechanism of site-specific recombination by ϕC31 integrase
Integrases, such as that of the Streptomyces temperate bacteriophage ϕC31, promote site-specific recombination between DNA sequences in the bacteriophage and bacterial genomes to integrate or excise the phage DNA. ϕC31 integrase belongs to the serine recombinase family, a large group of structurally related enzymes with diverse biological functions. It has been proposed that serine integrases use a “subunit rotation” mechanism to exchange DNA strands after double-strand DNA cleavage at the two recombining att sites, and that many rounds of subunit rotation can occur before the strands are religated. We have analyzed the mechanism of ϕC31 integrase-mediated recombination in a topologically constrained experimental system using hybrid “phes” recombination sites, each of which comprises a ϕC31 att site positioned adjacent to a regulatory sequence recognized by Tn3 resolvase. The topologies of reaction products from circular substrates containing two phes sites support a right-handed subunit rotation mechanism for catalysis of both integrative and excisive recombination. Strand exchange usually terminates after a single round of 180° rotation. However, multiple processive “360° rotation” rounds of strand exchange can be observed, if the recombining sites have nonidentical base pairs at their centers. We propose that a regulatory “gating” mechanism normally blocks multiple rounds of strand exchange and triggers product release after a single round

Effect of Palmitic Acid on the Electrical Conductivity of Carbon Nanotubes−Epoxy Resin Composites
We found that the palmitic acid allows an efficient dispersion of carbon nanotubes in the epoxy matrix. We have set up an experimental protocol in order to enhance the CNTs dispersion in epoxy resin. Electrical conductivity is optimal using a 1:1 CNTs to palmitic acid weight ratio. The associated percolation threshold is found between 0.05 and 0.1 wt % CNTs, i.e., between 0.03 and 0.06 vol %. The SEM image shows essentially individual CNTs which is inagreement with conductivity measurements. In comparison with composites without palmitic acid, the use of palmitic acid improves the electrical properties of CNTs-epoxy resin composites
Radius and chirality dependent conformation of polymer molecule at nanotube interface
Temperature dependent conformations of linear polymer molecules adsorbed at
carbon nanotube (CNT) interfaces are investigated through molecule dynamics
simulations. Model polyethylene (PE) molecules are shown to have selective
conformations on CNT surface, controlled by atomic structures of CNT lattice
and geometric coiling energy. PE molecules form entropy driven assembly
domains, and their preferred wrapping angles around large radius CNT (40, 40)
reflect the molecule configurations with energy minimums on a graphite plane.
While PE molecules prefer wrapping on small radius armchair CNT (5, 5)
predominantly at low temperatures, their configurations are shifted to larger
wrapping angle ones on a similar radius zigzag CNT (10, 0). A nematic
transformation around 280 K is identified through Landau-deGennes theory, with
